X-linked spinal and bulbar muscular atrophy (SBMA) occurs due to an expansion of the trinucleotide repeat (CAG)n in the androgen receptor gene. Anticipation is relatively rare in SBMA in contrast to spinocerebellar ataxia type 1 (SCAl), and dentatorubral and pallidoluysian atrophy (DRPLA) which show obvious paternal anticipation. The differences in the CAG repeat number were compared among sperm, leukocytes and skeletal muscles of SBMA patients. In SBMA, the sperm of most patients and the skeletal muscle of all patients showed the same repeat number as their leukocytes, whereas the increase in

Amyotrophic lateral sclerosis (ALS), although a disease that has been well recognized for nearly 150 years, still causes problems of diagnosis and management, as there is no definitive diagnostic test, and the disease is pleomorphic. Research criteria were developed for the categorization of definite, probable, and possible ALS at the El Escorial Workshop (published in 1994). The principal features are upper and lower motor neuron signs at several levels of the neuraxis, without involvement of other neurological systems. Separation of ALS or motor neuron disease (MND) from the spinal muscular

To investigate genetic factors involved in the pathogenesis of Hashimoto's thyroiditis (HT; goitrous autoimmune thyroiditis), HLA class I and class II antigens were analyzed in both seropositive HT (99 patients) and seronegative HT (43 patients). The frequency of HLA-DRw53 antigen was increased significantly in both seropositive HT (antigen frequency, 0.83; relative risk, 33.3; P less than 0.0002; corrected P less than 0.001) and seronegative HT (antigen frequency, 0.81; relative risk, 3.02; P less than 0.01; corrected P less than 0.05). The etiological fraction values for HLA-DRw53 in seropos

There is growing evidence for genetic causes of Alzheimer's disease; therefore, consideration of its age at onset among genetically predisposed individuals is timely. The authors studied the onset of Alzheimer-like dementia in families of 79 probands ascertained clinically for the Bronx Veterans Administration/Mt. Sinai School of Medicine longitudinal studies of Alzheimer's disease. Analyses among their 379 first-degree relatives, living and decreased, suggested that 1) early onsets form a distribution that is distinct from that of later onsets, and thus, different causes or mechanisms may ope

The ability of anti-idiotypic (anti-Id) antibodies to immunize mice against African trypanosomiasis independent of antigen has been confirmed. Of three allogeneic anti-Id antibodies raised against three protective monoclonal antibodies, each with specificity for the variant surface antigen of a clone of Trypanosoma rhodesiense, only one (anti-7H11 Id) was effective in immunizing BALB/c mice against homologous challenge. The immunity was associated with the more rapid and enhanced expression of the corresponding Id after infection. The immunity was restricted to mice bearing genes linked to Igh

To determine the hereditary pattern of acute anterior uveitis, we examined seven families-one family with four cases, one family with three cases, and four families with two cases of acute anterior uveitis. A total of 16 patients with uveitis and 28 relatives underwent ocular examinations and serologic determination of HLA antigens, protease inhibitor alpha 1-antitrypsin typing, and X-ray examination of sacroiliac joints. Sacroiliitis was found in 11 of 16 patients (68.8%) and incomplete Reiter's disease in seven of 16 (43.8%). HLA-B27 antigen was found in all patients with uveitis and in six

BACKGROUND: Dominant optic atrophy (DOA) is an inherited optic neuropathy caused by mutations of the OPA1 gene. Patients with DOA have a gradual loss of vision that is often detected in early life. While most cases stabilize at around a decimal best-corrected visual acuity (BCVA) of 0.1, some show mild impairments without visual field abnormalities. This then makes the diagnosis of DOA difficult. We report our longitudinal findings of a 56-year-old man with autosomal dominant DOA whose visual functions remained relatively good and genetic testing was needed for the diagnosis of DOA.

Blood α-tocopherol (α-Toc) concentrations decline gradually throughout the prepartum period, reaching the nadir after calving in dairy cows. The 6 α-Toc-related molecules [α-Toc transfer protein (TTPA); afamin; scavenger receptor class B, Type I; ATP-binding cassette transporter A1; tocopherol-associated protein (SEC14L2); and cytochrome P450 family 4, subfamily F, polypeptide 2 (CYP4F2)] are expressed in liver and other peripheral tissues. These molecules could regulate α-Toc transport, blood concentrations, and metabolism of α-Toc. Therefore, the aim of this study was to evaluate the changes

High temperature growth/survival was revealed in a phylogenetic relative (SMMA_5) of the mesophilic Paracoccus isolated from the 78 to 85°C water of a Trans-Himalayan sulfur-borax spring. After 12 h at 50°C, or 45 min at 70°C, in mineral salts thiosulfate (MST) medium, SMMA_5 retained ~2% colony forming units (CFUs), whereas comparator Paracoccus had 1.5% and 0% CFU left at 50°C and 70°C, respectively. After 12 h at 50°C, the thermally conditioned sibling SMMA_5_TC exhibited an ~1.5 time increase in CFU count; after 45 min at 70°C, SMMA_5_TC had 7% of the initial CFU count. 1,000-times diluted

Inorganic phosphate consumption in adults is two-to-three-fold higher than the recommended daily allowance due to its widespread use as a preservative and flavor enhancer in processed foods. We previously reported that 12 weeks of high-phosphate diet (HPD) reduced VO2max and altered metabolic gene expression and mitochondrial dysfunction in mouse skeletal muscle. Here, we investigated whether exercise training reverses the negative consequences of HPD. Male mice were fed normal (NPD) or HPD for 5 weeks, then continued on NPD or HPD with 9 weeks of voluntary exercise training. Serum, urine, and

Viruses, a group of non-cellular parasites, pose serious threats to human health and agriculture. While viral genetic variations modulating virus adaptation to biotic stresses have been reported, whether and how they affect virus interactions with abiotic stresses such as high temperature remain unknown. Here, we report the impact of AV2 mutations on the life cycle of squash leaf curl China virus (SLCCNV), a bipartite begomoviruses. We first conducted sequence analysis and identified a naturally-occurring mutation at position 35 in AV2 that changes the codon TGG to TAG. Followed analysis revea

Maternally inherited symbionts are central to arthropod biology, functioning both as mutualistic partners and as reproductive parasites. Genomic analyses provide critical insight into these interactions. Here, we sequenced, assembled and examined the genomes of Spiroplasma and Rickettsia co-infecting the lacewing Mallada desjardinsi, with the aim of elucidating the male-killing phenotype of Spiroplasma and predicting potential phenotypes for Rickettsia. In Spiroplasma, we identified a set of candidate effector genes. However, Spaid-like genes, where present, lacked the functional domains previ

Nitrates and antibiotics like tetracycline (TC) coexist in wastewater and inhibit nitrite (NO₂--N) accumulation during partial denitrification (PD), restricting anammox coupling. A moving bed biofilm reactor (PD-MBBR) and a sequencing batch reactor (PD-SBR) were compared under TC stress (0-8 mg/L). The PD-MBBR proved more robust, sustaining a high nitrate transformation ratio (NTR) of 95.11% and ∼53% TC removal. Metagenomic sequencing, quantitative polymerase chain reaction (qPCR), and molecular docking revealed this tolerance stemmed from physical shielding and metabolic compensation. Carrier

Acinetobacter baumannii is a Gram-negative nosocomial bacterium that is a member of the ESKAPE group of pathogens, notable for its virulence and intrinsic antibiotic resistance. It causes diverse infections, including respiratory and soft tissue disease, that are increasingly difficult to treat. Glutathione (GSH), the major intracellular redox buffer, is known in other bacteria to protect against stress and influence physiological processes such as biofilm formation. To investigate the roles of GSH in A. baumannii, we analyzed transposon mutant strains lacking gshA and gshB genes, which encode

Horizontal gene transfer (HGT) can accelerate plant adaptation, but the evolutionary fate of newly acquired nuclear genes is often unclear. Here, we analyzed a papain-like cysteine peptidase horizontally transferred from Panicoideae to wild Hordeum (Poaceae). Using chromosome-level assemblies of 21 diploid Hordeum species, we assessed presence/absence, copy-number variation, coding diversity, selection, predicted protein functionality, and expression. To infer donor relationships, we screened 77 Panicoideae accessions (48 species, 20 genera) by polymerase chain reaction and sequenced positive

5-oxoproline (5-OP) or pyroglutamic acid is an intermediate in the degradation arc of the glutathione cycle. It is metabolized into glutamate through the action of the 5-oxoprolinase enzyme, the only enzyme known to act on this metabolite. 5-OP has long been known to be relatively inert with a proposed role as an osomoprotectant. Recent studies on the 5-oxoprolinase enzyme in mammalian cells have, however, shown that knockdown or deletion of 5-oxoprolinase makes mice (and humans) prone to heart failure, an effect ascribed to oxidative stress caused by a twofold increase in 5-OP. To examine the

As the hybrid offspring of cattle and yak, cattle-yaks suffer from male sterility, manifesting as cascading spermatogenic failure. Despite the Y chromosome's pivotal role in spermatogenesis, the absence of a high-quality yak Y assembly has long impeded mechanistic understandings from this perspective. Here, a near-complete 42.4 Mb yak Y chromosome is constructed through a multi-stage assembly strategy that integrates de novo assembly with pangenome graph construction and Hi-C guided refinement. By developing a rigorously standardized gene annotation pipeline for precise cross-species compariso

Carboxylic acid reductases (CARs) function as versatile biocatalysts with a broad substrate range. However, CARs still struggled with relatively low activity in many enzyme reactions. While extensive efforts have focused on engineering CAR itself, the influence of phosphopantetheinyl transferase (Sfp), which is essential for CAR activation, has received significantly less attention. Consequently, information regarding the optimal ratio of CAR to Sfp for efficient biotransformation remains limited. To touch this issue, we investigated the effect of tandem sfp genes to CAR by introducing CAR-med
