HLA class I typing in Egyptian childhood minimal change nephrotic syndrome.

BACKGROUND: Reports on the association between human leukocyte antigens (HLAs) and childhood minimal change nephrotic syndrome (MCNS) are variable. The association between HLA class I and MCNS has not been tested among Egyptian children. METHODS: We studied such associations in a group of steroid-responsive MCNS Egyptian children (n=37) in comparison to a group of healthy Egyptians (n=100). Typing was performed using standard lymphocytotoxicity assay. RESULTS: The frequency of HLA-B27 was remarkably higher among MCNS children (18.9%) compared with the control group (1%) (p<0.001, risk ratio

Dietary intake of heterocyclic amines and cancers of the esophagus and gastric cardia.

The results of two epidemiological studies suggest that high intake of heterocyclic amines, which are formed on the surface of meats cooked at high temperatures, might be associated with increased risk of esophageal or cardia cancers. Our aim was to further investigate heterocyclic amine intake and risk of these cancers. We examined data from a nationwide, population-based, case-control study of risk factors for adenocarcinoma of the esophagus and gastric cardia and squamous cell carcinoma of the esophagus in Sweden, with 185, 258, and 165 cases, respectively, and 815 controls. Heterocyclic am

Antimicrobial susceptibilities of Shiga toxin-producing Escherichia coli isolates from pigs with edema disease in Japan.

Fifty-seven Shiga toxin-producing Escherichia coli (STEC) strains isolated from pigs with edema disease (ED) from 1997 to 2001 in Japan were examined for antimicrobial susceptibilities. The susceptibilities were compared with those of E. coli ATCC 23546 isolated from pig with ED in the 1950's. Consequently, the isolated STECs showed high susceptibility to peptides and bicozamycin in a way similar to the reference strain. On the other hand, the STECs showed low susceptibility to beta-lactams, tetracyclines, novobiocin, fosfomycin, trimethoprim, and old quinolones. It became clear that the susce

Usefulness of lanoconazole (Astat) cream in the treatment of hyperkeratotic type tinea pedis. Comparative study of monotherapy and combination therapy with 10% urea ointment (Pastaron).

Hyperkeratotic type tinea pedis is a refractory type of superficial dermatomycosis. Treatment for hyperkeratotic type tinea pedis is mainly with oral antimycotics, such as griseofulvin, and healing is generally considered to be difficult with only topical antimycotics. In this randomized comparative study, the usefulness of a topical application of 1% lanoconazole cream (Astat) monotherapy (group I) was compared with that of combination therapy with 1% lanoconazole cream and 10% urea ointment (Pastaron) (group II) in a series of patients with hyperkeratotic type tinea pedis. The clinical impro

Strategies for enhancement of radioimmunotherapy.

Two new strategies for increasing tumor uptake have been investigated. First the effect of interleukin-2 (IL-2) on tumor uptake of 125I-Lym-1 antibody in nude mice was investigated. Secondly, the use of 67Cu-labeled Lym-1 was evaluated in patients. In nude mice implanted with Raji human lymphoma, a greater than 2-fold enhancement of tumor uptake of 125I-Lym-1 was observed after administration of PEG-interleukin-2 (PEG-IL-2). The macrocycle 1,4,8,11-tetraazacylcotetradecane-N,N',N",N"'-tetraacetic acid (TETA), synthesized specifically for copper chelation, has been conjugated to Lym-1 for 67Cu

A Di-Rhamnolipid (Rha2-C10-C10) Inhibits Colorectal Cancer Motility and Immune Evasion Through AKT-Associated Signaling and PD-L1 Regulation.

Colorectal cancer (CRC) progression is driven by metastatic potential, metabolic reprogramming, and immune evasion. In this study, we investigated the effects of a di-rhamnolipid, Rha2-C10-C10 (1), on CRC cell motility, energy metabolism, and immune-related signaling. Compound 1 exhibited minimal cytotoxicity in AGS, A549, and MDA-MB-231 cells, while modestly reducing viability in Caco2 cells at higher concentrations. Notably, it significantly suppressed invasion and migration, with the most pronounced effects observed in Caco2 cells. These effects were associated with downregulation of mesenc

Task-Specific Effects of mGlu2/3 Receptor Agonist LY379268 on MK-801-Induced Behavioral and Neural Dysfunctions in Rats.

NMDA receptor hypofunction can lead to behavioral and cognitive disturbances, including hyperlocomotion, and is considered a core pathophysiological mechanism underlying cognitive and negative symptoms in schizophrenia. This study examined whether treatment with the mGlu2/3 receptor agonist LY379268 (1 and 2 mg/kg) could counteract such disruptions induced by the NMDA antagonist MK-801 (0.1 mg/kg). Rats were tested under two conditions: an aversive learning task (active place avoidance on a rotating arena) and a non-aversive open field test. Additionally, local field potentials were recorded f

Long-term benefits of upadacitinib for Atopic Dermatitis: deep responses in patient-reported outcomes over 140 weeks from the Measure Up 1 and Measure Up 2 clinical trials.

OBJECTIVES: Atopic dermatitis (AD), a chronic, immune-mediated inflammatory disease, is characterized by intense itch, eczematous rash, and skin pain, which can have negative impacts to quality-of-life (QoL), sleep, and mental health (especially anxiety and depression). Evaluation of the impacts of AD on the patient's lived experience are most accurately assessed by the patient, making measures of patient-reported outcomes (PROs) indispensable. The objective of the current study was to assess the long-term impact of upadacitinib, a once-daily oral selective Janus kinase inhibitor approved for

A Preclinical Study of [64Cu]Cu-NOTA-KN035 for Molecular Imaging of PD-L1 in Tumors.

The programmed cell death protein-1/programmed cell death ligand-1 (PD-1/PD-L1) axis plays a central role in tumor immune regulation, with PD-L1 expression serving as a critical biomarker for patient stratification and response prediction. Accurate, noninvasive assessment of PD-L1 expression is, therefore, essential for guiding clinical decision-making. KN035 is an ∼79.6 kDa fusion protein comprising a humanized single-domain antibody linked to an Fc fragment, offering a smaller molecular size than conventional monoclonal antibodies. In this study, KN035 was conjugated with p-SCN-Bn-NOTA and r

Efficacy and safety of Venetoclax combined with Azacitidine in the treatment of relapsed acute myeloid leukemia after allogeneic hematopoietic stem cell transplantation - a single-center retrospective study.

BACKGROUND: Venetoclax in combination with azacitidine (VEN-AZA) has become the standard of care for acute myeloid leukemia (AML) patients who are elderly or unfit to receive intense chemotherapy. However, the efficacy and safety of this regimen in AML patients who relapse after allogeneic hematopoietic stem cell transplantation (allo-HSCT) still needs more exploration. METHODS: We conducted a retrospective analysis of the data of 9 patients with relapsed AML after allo-HSCT treated with VEN-AZA from April 2021 to May 2023 at our center. Of these, 6 had molecular relapses (including persisten

Galactose-decorated lipid nanoparticle-mediated delivery of a selective NLRP3 inhibitor attenuates hepatic inflammation in metabolic dysfunction-associated steatotic liver disease.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a metabolic stress-induced hepatic injury closely associated with insulin resistance and genetic predisposition, affecting about 25% of the global population and becoming the leading cause of chronic liver disease. Therapeutic interventions targeting the regulation of inflammation-related pathways have shown potential for significant improvements in MASLD treatment. Herein, we developed galactose-decorated lipid nanoparticles for selective NLRP3 inhibitor targeted liver delivery (MCC950@Gal-LNPs). The galactose modification en

Wnt-dependent modulation of alveolar epithelial phenotypes and barrier function in human progenitor-like cells.

Alveolar regeneration requires coordinated alveolar type 1 (AT1)/alveolar type 2 (AT2) responses and an intact epithelial barrier. In vitro systems, however, rarely couple lineage control with high resistance. Here we establish a concise workflow in which human AT2-like progenitors are expanded in a medium containing a small-molecule cocktail (Y-27632/A-83-01/CHIR99021; YAC), followed by air-liquid interface (ALI) culture in medium without the YAC cocktail (hereafter referred to as the YAC(-) condition). Bimodal Fluorescent Conjugate Based on Prostate-Specific Membrane Antigen Ligands with the Chelating Agent DOTA and SulfoCy5 Dye: Synthesis, Radiolabeling, and Biological Activity.

Prostate-specific membrane antigen (PSMA) is an essential zinc-dependent metalloprotease classified within the type II transmembrane protein family, often referred to as glutamate carboxypeptidase II (GCPII). PSMA is recognized as a particularly promising target for both the diagnosis and therapeutic intervention of prostate cancer. In this study, we designed and synthesized PSMA-targeted DOTA-loaded bimodal conjugate 11 with SulfoCy5 fluorescent dye, performed in vitro characterization, and analyzed biodistribution in vivo. At 40-100 nM concentrations, the resulting conjugate demonstrated rel
