The human cytochrome P450 19A1 (CYP19A1, aromatase) is a heme-containing protein catalyzing the final steps of the biosynthesis of the steroid hormone 17β-estradiol. It is a key target for the treatment of sex-hormone-related disorders due to its role in mediating the conversion of androgens to estrogens. Here, we report the development of a virtual screening workflow incorporating machine learning and structure-based modeling that has led to the discovery of new CYP19A1 inhibitors. The machine learning models were built on comprehensive CYP19A1 data sets extracted from the ChEMBL and PubChem

Severe Fever with Thrombocytopenia Syndrome Virus (SFTSV) is a highly pathogenic tick-borne virus that causes hemorrhagic fever in humans, with a mortality rate of up to 30 %. SFTSV has been primarily detected in arachnid vectors, particularly Haemaphysalis longicornis. While the majority of human SFTSV infections are linked to tick exposure, a significant proportion of cases have no reported history of tick bites, suggesting the existence of alternative vectors. Here, we report the first detection of SFTSV RNA in deer ked (Lipoptena fortisetosa, family Hippoboscidae) using reverse transcripti

Pituitary tumor transforming gene 1 (Pttg1) is upregulated in cirrhosis and hepatocarcinoma (HCC). We assessed the therapeutic effect of liver-targeted Pttg1 siRNA Retinol (Ret) pBAE nanoparticles (NPs) to treat these disturbances. Fibrosis was induced in Wistar rats by carbon tetrachloride inhalation and HCC by diethylnitrosamine injection. Ret pBAE NPs accumulated in hepatic tissue, close to zones positive for αSMA staining. Pttg1 interference increased mean arterial pressure, reduced portal hypertension and decreased collagen accumulation and inflammatory infiltrate in fibrotic rats. In HCC

Respiratory coronaviruses pose significant health threats to both humans and animals. Porcine respiratory coronavirus (PRCV) typically causes mild respiratory infections in pigs and serves as a valuable model for human respiratory coronavirus. However, the investigation of PRCV pathogenesis remains limited. This study investigates the prevalence, genetic diversity, and pathogenesis of PRCV in China. From 2022 to 2024, 1186 pig tracheal samples were collected across 10 provinces, revealing a widespread presence of PRCV with an 11.8 % overall prevalence. For the first time, we isolated a PRCV st

Parasitic worms (helminths) release extracellular vesicles (EVs) -membrane-surrounded, nanosized structures loaded with a repertoire of active biomolecules- as part of their excretory/secretory products. These particles play key roles in intercellular communication, not only within multicellular organisms but also between organisms that establish stable biological associations, as it occurs during parasitism. Since their "rediscovery" in 2012, the field of helminth EVs has expanded significantly, with a wealth of research conducted both in model organisms and parasites of human and veterinary

Neurocysticercosis is caused by the establishment of Taenia solium cysticerci in the central nervous system. The extraparenchymal form (ExP-NCC) is the most severe clinical presentation that may remain asymptomatic for years. Current treatment involves cysticidal drugs (albendazole and/or praziquantel) combined with glucocorticoids to manage the associated neuroinflammation; however, only ∼30% of patients respond effectively. This highlights the need to improve therapeutic strategies. Herein, the experimental murine model of human ExP-NCC was further characterized to improve its usefulness in

DC inhibitory receptor (DCIR) is a C-type lectin receptor selectively expressed on myeloid cells, including monocytes, macrophages, DCs, and neutrophils. Its role in immune regulation has been implicated in murine models and human genome-wide association studies, suggesting defective DCIR function associates with increased susceptibility to autoimmune diseases such as rheumatoid arthritis, lupus, and Sjögren's syndrome. However, little is known about the mechanisms underlying DCIR activation to dampen inflammation. Here, we developed anti-DCIR agonistic antibodies that promote phosphorylation

Congenital toxoplasmosis in humans and in other mammalian species, such as small ruminants, is a well-known cause of abortion and fetal malformations. The calcium-dependent protein kinase 1 (CDPK1) inhibitor BKI-1748 has shown a promising safety profile for its use in humans and a good efficacy against Toxoplasma gondii infection in vitro and in mouse models. Ten doses of BKI-1748 given every other day orally in sheep at 15 mg/kg did not show systemic or pregnancy-related toxicity. In sheep experimentally infected at 90 days of pregnancy with 1000 TgShSp1 oocysts, the BKI-1748 treatment admini

In contrast to the extensive evidence from animal studies, only few human data are available on the relation of vascular growth factors and collateral function as well as on the conditions which may modify their release or function. In 31 patients with total coronary occlusion (TCOs) blood was collected from distal to the occlusion site (collateral circulation) and from the aortic root (systemic circulation). Serum was used to assess its mitogenic potential in [3H]-thymidine incorporation assay on human umbilical vein endothelial cells. Serum from patients with the duration of occlusion betwee

We aimed to develop and validate a Loop-mediated Isothermal Amplification (LAMP) assay to Sporothrix brasiliensis. LAMP reaction was developed using six primers designed based on calmodulin gene. In the LAMP reaction, we tested twenty isolates of S. brasiliensis from animals and humans, along with ten tissue samples extracted from the left footpad of mice that had been experimentally infected with S. brasiliensis. In addition, it included DNA samples from various other fungal species for specificity evaluation. All S. brasiliensis isolates yielded positive results in the LAMP, and the limit of

Antibody-based blocking of vascular endothelial growth factor (VEGF) reduces choroidal neovascularization (CNV) and retinal edema, rescuing vision in patients with neovascular age-related macular degeneration (nAMD). However, poor response and resistance to anti-VEGF treatment occurs. We report that targeting the Notch ligand Jagged1 by a monoclonal antibody reduces neovascular lesion size, number of activated phagocytes and inflammatory markers and vascular leakage in an experimental CNV mouse model. Additionally, we demonstrate that Jagged1 is expressed in mouse and human eyes, and that Jagg

Kudoa septempunctata is a myxosporean parasite that infects the trunk muscles of olive flounder (Paralichthys olivaceus) and has been reported to cause foodborne illnesses in humans. However, the molecular mechanisms underlying K. septempunctata spore toxicity remain largely unknown. In this study, the gastroenteropathy of K. septempunctata was examined in human colon adenocarcinoma cells as well as experimental mice inoculated with spores. We found that K. septempunctata decreased transepithelial resistance and disrupted epithelial tight junctions by deleting ZO-1 in Caco-2 monolayers. Additi

Several clinical trials and experimental studies have recently shown that vitamin K (VK) supplementation benefits the human body. Specifically, VK participates in coagulation and is associated with cellular senescence and cancer. VK has a potential anticancer effect in various cancers, such as pancreatic and prostate cancers. Through anti-inflammatory and antioxidant effects, VK can prevent senescence and inhibit cancer metastasis. Therefore, cancer prognosis can be improved by preventing cellular senescence. In addition, VK can inhibit the proliferation, growth, and differentiation of cancer

Aortic valve stenosis secondary to aortic valve calcification is the most common valve disease in the Western world. Calcification is a result of pathological proliferation and osteogenic differentiation of resident valve interstitial cells. To develop non-surgical treatments, the molecular and cellular mechanisms of pathological calcification must be revealed. In the current overview, we present methods for evaluation of calcification in different ex vivo, in vitro and in vivo situations including imaging in patients. The latter include echocardiography, scanning with computed tomography and

The growth and maintenance of nearly every tissue in the body is influenced by systemic hormones during embryonic development through puberty and into adulthood. Of the ~130 different hormones expressed in the human body, steroid hormones and peptide hormones are highly abundant in circulation and are known to regulate anabolic processes and wound healing in a tissue-dependent manner. Of interest, differential levels of sex hormones have been associated with ocular pathologies, including dry eye disease and keratoconus. In this review, we discuss key studies that have revealed a role for andro

Glaucoma is an asymptotic condition that damages the optic nerves of a human eye. Glaucoma is frequently caused due to abnormally high pressure in an eye that leads to permanent blindness. Detecting glaucoma at an initial phase has the possibility of curing this disease, but diagnosing accurately is considered as a challenging task. Therefore, this paper proposes a novel method known as a glaucoma detection system that performs the diagnosis of glaucoma by exploiting the prescribed characteristics. The significant intention of this paper involves diagnosing the glaucoma disease present at the

The involvement of the central nervous system (CNS) during human acute and chronic Chagas disease (CD) has been largely reported. Meningoencephalitis is a frequent finding during the acute infection, while during chronic phase the CNS involvement is often accompanied by behavioral and cognitive impairments. In the same vein, several studies have shown that rodents infected with Trypanosoma cruzi (T. cruzi) display behavior abnormalities, accompanied by brain inflammation, in situ production of pro-inflammatory cytokines and parasitism in diverse cerebral areas, with involvement of microglia, m

Epstein-Barr virus (EBV) is a cancer-associated pathogen responsible for 165,000 deaths annually. EBV is also the etiological agent of infectious mononucleosis and is linked to multiple sclerosis and rheumatoid arthritis. Thus, an EBV vaccine would have a significant global health impact. EBV is orally transmitted and has tropism for epithelial and B cells. Therefore, a vaccine would need to prevent infection of both in the oral cavity. Passive transfer of monoclonal antibodies against the gH/gL glycoprotein complex prevent experimental EBV infection in humanized mice and rhesus macaques, sugg

Currently, Chlamydia trachomatis still possesses a significant impact on public health, with more than 130 million new cases each year, alongside a high prevalence of asymptomatic infections (approximately 80% in women and 50% in men). C. trachomatis infection involves a wide range of different cell types, from cervical epithelial cells, testicular Sertoli cells to Synovial cells, leading to a broad spectrum of pathologies of varying severity both in women and in men. Several two-dimensional in vitro cellular models have been employed for investigating C. trachomatis host-cell interaction, alt
