Koumine inhibits osteoclastogenesis and prevents ovariectomy-induced bone loss via suppression of the MAPK signaling pathways.

Excessive osteoclast formation drives osteolytic bone diseases such as osteoporosis. Koumine (KM), an alkaloid derived from Gelsemium elegans, exhibits various bioactivities; however, its role in bone homeostasis remains unknown. This study investigated the effects of KM on RANKL-induced osteoclastogenesis in bone marrow-derived macrophages (BMMs). Cell viability, differentiation (TRAcP staining), and function (F-actin ring formation, bone resorption pit assay) were assessed. The underlying mechanisms were explored using Western blot and qPCR. The in vivo efficacy of KM was evaluated in an ova

Phase separation-based HTS identifies cobimetinib as a YAP-TEAD inhibitor that suppresses hyperactivated YAP-induced cancer progression.

The Hippo signaling pathway prevents unchecked cell growth, coordinates apoptosis, and preserves proper organ function. Dysregulation of this pathway has been implicated in a myriad of diseases, particularly in cancer. The YAP (Yes-associated protein)-TEAD (TEA domain transcription factor) complex, the key transcriptional downstream effector of the Hippo pathway, hence stands out as an appealing target for therapeutic intervention. In this study, we developed a high-throughput screening (HTS) assay leveraging phase separation principles and found that the US Food and Drug Administration-approv

Phellopterin Inhibits Inflammation and Relieves Cartilage Degradation by Suppressing MAPK Signaling in Osteoarthritis.

Osteoarthritis (OA) is thought to be the most common joint disorder and a major cause of disability and socioeconomic burden worldwide. Phellopterin has been demonstrated to exert anti-inflammatory effects in various diseases. However, the function of phellopterin in OA remains unclear. In this study, we aimed to investigate the role and mechanism of action of phellopterin in OA. We found that phellopterin promoted the proliferative ability and repressed the apoptotic ability of interleukin-1β (IL-1β)-treated ATDC5 cells. Our data also confirmed that phellopterin relieved inflammatory mediator

Betulin inhibits titanium particle-induced osteolysis and attenuates RANKL-associated osteoclastogenesis by suppressing the MAPK pathway.

Wear particle-induced aseptic loosening is an important factor compromising the long-term success of total joint replacement. It is often associated with osteolysis and inflammatory cellular responses within the joint. Betulin, a pentacyclic triterpenoid, promotes osteoblast differentiation and suppresses osteoclast formation. This study aimed to explore the therapeutic effect of betulin on aseptic loosening induced by wear particles and its underlying mechanism. A mouse calvarial model was used to study the effects of betulin on titanium particle-induced osteolysis in vivo. RAW264.7 murine ma

Comparative Analysis of Primary and Liver Fibroblasts Reveals MET as a Potent Target in Pancreatic Cancer Metastasis.

BACKGROUND & AIMS: The tumor microenvironment drives many malignant features of pancreatic ductal adenocarcinoma. The fibroblasts within pancreatic tumors promote tissue remodeling, immune suppression, and resistance to therapy. However, the interactions between stromal populations and pancreatic cancer cells are less understood in the liver, the most frequent site of pancreatic ductal adenocarcinoma metastasis. METHODS: To address this, we employed single-cell transcriptomics to compare primary pancreatic vs liver pancreatic ductal adenocarcinoma lesions. Using a ligand-receptor interaction

Exploring the immune-modulatory role of melatonin in head kidney and spleen cells of freshwater snakehead teleost, Channa punctatus.

Melatonin, the ubiquitous hormone marks its presence from plants to animals with remarkable antioxidant and anti-inflammatory properties. In the teleost, melatonin is secreted from the pineal gland, which functions as a modulator of circadian rhythm, being homologous to the suprachiasmatic nuclei of mammals. The current study revolves around evaluating the role of in vitro melatonin in the immune mechanism of the snakehead fish, Channa punctatus. Reactive oxygen and nitrogen species production was assayed by respiratory burst and nitrite assay. Mitogen-stimulated lymphocyte proliferation was d

Dual inhibition of PP2A and WEE1 induces apoptosis and mitotic catastrophe in cancer cells.

The serine/threonine phosphatase-2 (PP2A) controls mitogen-associated signaling and DNA replication. The WEE1 protein tyrosine kinase controls S phase progression and G2/M phase transition. Databases disclose that the levels of PP2A and WEE1 are associated with poor prognosis of leukemic patients (p = 0,0017/0,025; n = 54-163). We applied advanced, clinically used drugs that block PP2A and WEE1 to human cultured leukemia cells, primary pre-leukemic and chronic myeloid leukemia cells, and pancreatic ductal adenocarcinoma cells. Combined application of the drugs depleted cells in all cell cycle

Erdheim-Chester Disease: A Diagnostic Challenge.

Erdheim-Chester disease (ECD) is a rare, non-Langerhans cell histiocytosis characterized by excessive production and accumulation of histiocytes within various tissues. The discovery of activating mutations within the Mitogen-Activated Protein Kinase (MAPK) pathway has established its neoplastic nature and enabled the use of targeted therapeutic approaches. Despite these advances, ECD remains a diagnostic challenge, often due to its nonspecific symptoms, multisystemic involvement, and consequent delay in diagnosis. We report three cases of ECD illustrating its diverse clinical presentations an

Atherosclerosis: Molecular Pathogenesis and Therapeutic Interventions.

Atherosclerosis is a multifactor condition and driving force behind many cardiovascular diseases. Moreover, it remains a leading cause of mortality worldwide. The current therapies, including statins and fibrates, focus mainly on lipid lowering. However, this condition is characterized by various biological pathways, particularly the oxidized LDL uptake by macrophages, chronic inflammation, or vascular smooth muscle cells proliferation, migration, and phenotypic switching, which forces the researchers to explore the numerous molecular pathways standing behind those processes. Despite a lot res

Pharmacological inhibition of MKP-1 exacerbates central post-stroke pain and neuroinflammation in mice.

Central post-stroke pain (CPSP) remains difficult to treat due to limited understanding of its underlying mechanisms. Emerging evidence implicates microglia-driven neuroinflammation in CPSP development. Mitogen-activated protein kinase phosphatase-1 (MKP-1) negatively regulates MAPK signaling and modulates inflammation in neurological disorders, yet its role in CPSP is unclear. Using a mouse model of thalamic hemorrhage, we examined MKP-1 expression and function in CPSP. Behavioral testing assessed pain hypersensitivity and affective disturbances. Immunofluorescence and western blot evaluated

Development of receptor and oncogene-responsive luciferase reporter vectors that are activated via kinase signaling to AP-1.

This study aimed to generate lentiviral vectors carrying an array of AP-1 motifs driving luciferase gene expression as reporters of mitogen-activated protein kinase (MAPK) activity. We created a series of vectors based on LeGO-iG that were used to generate stably transduced leukemia cell lines. A vector termed LEGO-AP1×6-GM55 containing an array of 6 AP-1 sites linked to the minimal CSF2 promoter was sufficient to support high levels of MAPK-inducible luciferase activity in leukemic cell lines that was suppressed by MAPK inhibitors. The inclusion of a putative chromatin priming element encompa

Targeting MAPK Pathways in Skin, Thyroid, and Pancreatic Cancer: A Perspective on Synthetic Inhibitors and Natural Modulators.

Cancer frequently arises from the impaired functioning of the Mitogen-activated protein kinase (MAPK) signaling system, driven by mutation or overexpression of key signaling components. RAF, MEK, ERK, and KRAS inhibitors have significantly improved clinical outcomes, but efficacy is still restricted due to pathway reactivation, adaptive resistance, and signaling cross-talk. These limitations have led to the search for novel therapeutic approaches and molecular targets in the MAPK network. Melanoma, thyroid carcinoma, and pancreatic adenocarcinoma were selected because MAPK pathway alterations

An integrative multiparametric approach stratifies putative distinct phenotypes of blast phase chronic myelomonocytic leukemia.

Approximately 30% of patients with chronic myelomonocytic leukemia (CMML) undergo transformation to a chemo-refractory blastic phase (BP-CMML). Seeking novel therapeutic approaches, we profiled blast transcriptomes from 42 BP-CMMLs, observing extensive transcriptional heterogeneity and poor alignment to current acute myeloid leukemia (AML) classifications. BP-CMMLs display distinctive transcriptomic profiles, including enrichment for quiescence and variability in drug response signatures. Integrating clinical, immunophenotype, and transcriptome parameters, Random Forest unsupervised clustering

NEK7-induced phosphorylation of EGFR on serine 1070 drives the acquired lenvatinib resistance in hepatocellular carcinoma.

BACKGROUND AND AIMS: Lenvatinib is recognized as a first-line therapy for inoperable hepatocellular carcinoma (HCC) patients. Growing evidence indicates that lenvatinib resistance can be acquired in HCC cells via kinase rewiring. APPROACH AND RESULTS: We established acquired lenvatinib-resistant organoids and HCC cell lines. NIMA-related coiled-coil kinase 7 (NEK7) was identified as an HCC lenvatinib acquired resistance gene by kinase CRISPR-Cas9 genetic screen. Functional analyses demonstrate that NEK7 enhanced lenvatinib resistance in HCC, and NEK7 knockdown or knockout displays the antitum

ITGAV and SMAD4 influence the progression and clinical outcome of pancreatic ductal adenocarcinoma.

Pancreatic ductal adenocarcinoma (PDAC) is a very aggressive and lethal malignancy with limited treatment options, a fact that underscores the urgent need for more effective therapies to improve patient outcomes. Preclinical studies have shown promise for αV integrin-targeted therapies; however, clinical trials have been disappointing, highlighting the need for further research. In this study, we demonstrate that integrin subunit alpha V (ITGAV) signals through both mothers against decapentaplegic homolog 4 (SMAD4)-dependent or SMAD4-independent pathways, depending on the genetic context. Sodium new houttuyfonate affects tumor angiogenesis by suppressing FGF1 expression and the p38/MAPK signaling pathway in pancreatic cancer.

Pancreatic cancer is one of the deadliest cancers in the world. Phytochemicals are considered as potential anticancer drugs because of their safety and low cost. Sodium new houttuyfonate (SNH) is derived from Houttuynia cordata Thunb. (HCT), which is a famous edible and medicinal plant in China. Although research findings suggest that SNH possesses anticancer properties, its role and mechanisms in pancreatic cancer remain unclear. Using network pharmacology, we identified 49 potential targets of SNH action in pancreatic cancer. In vitro, we confirmed that SNH inhibited the proliferation and in

Opposing roles of p38α-mediated phosphorylation and PRMT1-mediated arginine methylation in driving TDP-43 proteinopathy.

Amyotrophic lateral sclerosis (ALS) is a devastating neurodegenerative disorder typically characterized by insoluble inclusions of hyperphosphorylated TDP-43. The mechanisms underlying toxic TDP-43 accumulation are not understood. Persistent activation of p38 mitogen-activated protein kinase (MAPK) is implicated in ALS. However, it is unclear how p38 MAPK affects TDP-43 proteinopathy. Here, we show that p38α MAPK inhibition reduces pathological TDP-43 phosphorylation, aggregation, cytoplasmic mislocalization, and neurotoxicity. Remarkably, p38α MAPK inhibition mitigates aberrant TDP-43 phenoty

MEK1/2 inhibitors suppress pathological α-synuclein and neurotoxicity in cell models and a humanized mouse model of Parkinson's disease.

The abnormal accumulation of misfolded proteins is a common hallmark of many neurodegenerative disorders. Among these proteins, α-synuclein (αsyn) is a well-characterized pathogenic protein in Parkinson's disease (PD) and other synucleinopathies. αsyn can be hyperphosphorylated and form pathological aggregates, leading to neurodegeneration. Thus, chemical modulators of pathological αsyn may suppress its downstream toxicity and provide entry points to therapeutic intervention. Here, we identified mitogen-activated protein kinase kinase 1/2 (MEK1/2) inhibitors as negative modulators of basal αsy

SRC at the crossroads of KRAS inhibitor resistance: Mechanisms and therapeutic opportunities.

Kirsten rat sarcoma (KRAS) mutations are among the most common oncogenic drivers in human cancers, particularly in non-small cell lung cancer (NSCLC), colorectal cancer (CRC), and pancreatic ductal adenocarcinoma (PDAC). The development of allele-specific KRAS inhibitors, especially those targeting the KRASG12C variant, represents a landmark achievement in precision oncology. Yet their therapeutic benefit is often transient, as tumors rapidly develop seemingly heterogeneous resistance mechanisms. Increasing evidence implicates SRC, a non-receptor tyrosine kinase frequently hyperactivated in KR
