Matrix stiffness drives squamous cell carcinoma progression via a Piezo1-mediated mechanotransduction feedback loop.

INTRODUCTION: Squamous cell carcinoma (SCC) is one of the most frequent solid tumor accounting for more than one million cancer deaths annually. Emerging evidence highlights the critical role of mechanical signaling in tumor progression, yet the mechanobiological interplay between extracellular matrix (ECM) stiffness and SCC pathogenesis remains poorly understood. OBJECTIVES: This study aimed to investigate how matrix stiffness drives SCC progression via Piezo1-mediated mechanotransduction and its clinical implications. METHODS: Atomic force microscopy quantified tissue stiffness in human cu

Human Papillomavirus Infection Is a Favorable Prognostic Factor for Patients With Stages I to IVA Esophageal Squamous Cell Carcinoma but not Adenocarcinoma.

With its 2 distinct histological subtypes (squamous cell carcinoma [SCC] and adenocarcinoma [ADC]), its increasing incidence in both high- and low-income countries and its elevated 5-year mortality rate, esophageal cancer still represents a significant global health challenge. Although the implication of high-risk human papillomaviruses (HPV) in both anogenital and head and neck carcinogenesis is well-established, the association between these mucosotropic viruses and esophageal cancers has been a subject of debate for nearly 2 decades. In an effort to resolve this unclear situation and advanc

Comparison between community-acquired pneumonia and post-obstructive pneumonia associated with endobronchial tumors.

BACKGROUND: Endobronchial tumors can infiltrate the bronchial wall or protrude into the bronchial lumen, causing post-obstructive pneumonia (POP). Differentiating between POP and community-acquired pneumonia (CAP) is challenging due to similar clinical, laboratory, and imaging findings, which can delay the diagnosis and treatment of endobronchial tumors. METHODS: We compared general demographic information, laboratory test results, lung CT images, bronchoscopic observations, pathological findings between the POP group and the CAP group. RESULTS: (1) The POP group consisted mainly of older in

Non-canonical Platelet activation by oral squamous cell carcinoma extracellular vesicles through cooperative thrombin formation.

Coagulation contributes to cancer mortality, and tumor cells activate platelets to release growth factors. However, this activation is non-canonical, employs unknown agonists and, uniquely, is not immediate. Platelets are activated by human cell lines derived from Oral Squamous Cell Carcinomas (OSCC) with this atypical delay. The stimulatory activity was not a soluble factor, but rather was completely recovered in spontaneously shed extracellular vesicles (EV). EV did not contain established platelet agonists. Instead, chemical and pharmacologic inhibitors show OSCC EV interacted with quiescen

Surgical removal of unexpected oral squamous cell carcinoma arising from a maxillary odontogenic keratocyst in a young female patient: a case report and literature review.

BACKGROUND: Although odontogenic cysts in the maxilla are often seen, malignant changes inside these cysts are rather uncommon. According to reports, the occurrence of carcinomas developing in odontogenic cysts ranges from 1 to 2 per 1,000. Squamous cell carcinoma (SCC) arising from an odontogenic keratocyst (OKC) is a rare malignant transformation that typically presents in the jaws. This form of carcinoma is locally aggressive and often associated with a poor prognosis, especially when the diagnosis is delayed. CASE PRESENTATION: This report discusses a healthy 13-year-old girl with no medic

Acidic plasma-activated povidone-iodine induces copper-dependent oxidative death in oral squamous cell carcinoma cells.

Oral squamous cell carcinoma (OSCC) requires therapeutic strategies that preserve oral function while providing effective tumor control. Here we demonstrate that non-thermal plasma (NTP) irradiation of a clinically established povidone-iodine solution generates a potent cytotoxic formulation termed plasma-activated povidone-iodine (PAI). PAI selectively reduced viability of OSCC cell lines (SAS, HSC-2, HSC-4) compared with normal fibroblasts. Mechanistically, PAI activity strictly depended on its acidic pH (3.0 or below) and hydrogen peroxide (H2O2) generated during NTP irradiation. Neutraliza

Inflammation as a master regulator of immunotherapy response in head and neck squamous cell carcinoma: from malignant transformation to ecology-aware precision combinations.

In the era of immunotherapy, head and neck squamous cell carcinoma (HNSCC) has demonstrated clear benefits from immune-based treatments and is widely regarded as a tumor type with high immunotherapeutic potential. These tumors are characterized by robust and persistent inflammatory responses that actively drive tumor initiation and progression while concurrently shaping their sensitivity or resistance to therapy. Inflammation simultaneously creates therapeutic vulnerabilities and barriers by altering tumor behavior and reprogramming the immune microenvironment. This review examined HNSCC throu

Metaplastic-Like and Squamous Cell Carcinoma-Like Cells in Liquid-Based Cytology Associated With Small Cell Neuroendocrine Carcinoma of the Uterine Cervix.

Metaplastic-like cells exhibit an eccentric nucleus. SCC-like cells are planar aggregates with prominent atypical nuclei, similar to SCCs. These squamous epithelium changes may serve as cytologic clues to the early detection of small cell neuroendocrine carcinoma of the uterine cervix with predominant subepithelial growth. The authors identified metaplastic-like and SCC-like cells in cervical SCNEC cytology. These findings offer vital clues for early detection, even in cases with predominant subepithelial growth.

Gelsemine induces apoptosis through a reactive oxygen species- and MAPK/JNK1/2-coregulated pathway in tongue squamous cell carcinoma cells.

Tongue cancer is recognized globally as a prevalent oral cancer, with squamous cell carcinoma accounting for over 90% of oral cancer diagnoses. Tongue squamous cell carcinoma (SCC) has a poor survival rate and is highly invasive. The present study aimed to investigate the effect of the therapeutic agent gelsemine on SAS cells line derived from tongue squamous. Gelsemine was found to induce significant cell shrinkage and morphological changes. In addition, gelsemine significantly increased apoptosis, cytotoxicity and expression of signaling proteins related to apoptosis, namely cleaved PARP and

Clinical and experimental aspects of tirbanibulin treatments.

Tirbanibulin is a topical medication approved in 2020 as a 5 day topical treatment for actinic keratosis, a precancerous skin lesion characterized by the hyperproliferation of atypical keratinocytes. Tirbanibulin exerts its effects through dual mechanisms: microtubule inhibition, which disrupts cell division, and Src kinase inhibition, which interferes with cancer-associated signaling pathways. Although its primary use is in AK, these mechanisms suggested potential therapeutic applications in a broader range of epithelial cancers. In this literature review, we explored tirbanibulin's applicati

Autologous extracellular vesicles derived from conjunctival squamous cell carcinoma deliver therapeutic microRNAs to induce apoptosis in originating cancer.

There are significant challenges in treating advanced and recurrent conjunctival squamous cell carcinoma (cSCC). Therapeutic miRNAs directed at cSCC may have anticancer potential, but questions remain regarding efficiency of their targeted delivery. In this study, we address limitations of miRNA delivery by engineering autologous extracellular vesicles (EVs) of ∼130 nm sizes derived from cSCC UMSCC9 cells and tumors (cSCC-EVs) using a microfluidic based reconstruction system. We ICG-labelled these cSCC-EVs to enable subsequent tracking of miRNA delivery to tumors in vivo, and loaded them with

Single cell transcriptomics reveals dysregulated immnue homeostasis in different stages in HPV-induced cutaneous squamous cell carcinoma.

In order to explore the huge impact of impaired immnue homeostasis on the occurrence and development of cutaneous squamous cell carcinoma (cSCC), and investigate characterization of the cellular components and their changes which is crucial to understanding the pathologic process of HPV-induced cSCC, we diagnosed and followed up on a very rare HPV-induced cSCC patient who progressed at a very fast rate and transferred to death quickly. We performed single-cell RNA sequencing (scRNA-seq) of 11 379 cells from the skin tissues of this patient with four different skin statuses after HPV infection.

Different expression patterns of p63 and p73 in Felis catus papillomavirus type 2-associated feline Merkel cell carcinomas and other epidermal carcinomas.

Merkel cell carcinoma (MCC) is a cutaneous neuroendocrine tumor, and more than 90% of feline MCC cases test positive for Felis catus papillomavirus type 2 (FcaPV2). In the present study, basal cell markers p40, p63, and p73 and the stem cell marker SOX2 and cytokeratin 14 (CK14) were immunohistochemically examined in normal fetal, infant, and adult feline skin tissues. The expression of these proteins was examined in tumors positive for FcaPV2, including MCC, basal cell carcinoma (BCC), Bowenoid in situ carcinoma (BISC), and squamous cell carcinoma (SCC). Infant and adult feline skin tissues h
