Tryptophan metabolism in colorectal cancer: From mechanistic insights to novel therapeutic strategies.

BACKGROUND: Tryptophan metabolism drives colorectal cancer (CRC) pathogenesis through three interconnected cascades: the kynurenine pathway, the serotonin pathway, and the gut-microbiota-dominated indole pathway. These pathways exert context-dependent dual activities determined by disease stage, metabolite concentration, cell type and tumor microenvironment. Substrate competition and metabolic flux redistribution occur across the three branches, while extra-intestinal tryptophan metabolites mediate multi-organ crosstalk. Despite substantial preclinical findings, clinical translation is hindere

Bladder Large Cell Neuroendocrine Carcinoma(LCNEC): A Case Report of Exfoliated Cytology and Pathological Characteristics.

Primary large cell neuroendocrine carcinoma (LCNEC) of the bladder is an extremely rare and highly aggressive malignancy with a poor prognosis, bringing major challenges to clinical diagnosis and treatment. We report the case of a 77-year-old man who presented with recurrent gross hematuria for more than 8 months, accompanied by urinary frequency, urinary urgency, and lumbosacral pain. Imaging revealed a large soft-tissue mass on the posterior bladder wall without pathognomonic imaging findings. Urine exfoliative cytology revealed atypical tumor cells with large cellular size, high nucleocytop

Urinary tract infections in postmenopausal women revisited (UTIr): a prospective observational cohort study to explore the urobiomes of postmenopausal women with and without recurrent urinary tract infections.

BACKGROUND: Recurrent urinary tract infections (RUTI) are prevalent, particularly among postmenopausal women, and place a significant burden on the affected individuals and the healthcare system. While Escherichia coli is the primary cause of most UTIs in premenopausal women, this may not hold true for postmenopausal women. To facilitate development of novel diagnostics, preventive interventions, and clinical management of RUTI in postmenopausal women, it is essential to strengthen the biological evidence base. METHODS: This observational prospective cohort study will enrol 20 postmenopausal

From bench to bedside: the role of gastrointestinal stem cells in health and disease.

The gastrointestinal (GI) tract constitutes a sophisticated system integral to digestion, nutrient absorption, and overall health, with its functionality predominantly hinging on the distinctive properties of diverse stem cell types. This review systematically investigates the pivotal roles of stem cells across the esophagus, stomach, small intestine, and colon, emphasizing their crucial contributions to tissue homeostasis, repair mechanisms, and regeneration. Each segment of the GI tract is characterized by specialized stem cell populations that exhibit distinct functional attributes, highlig

Facial Clefts and the Trigeminal Nerve: A Narrative Review of the Literature and Clinical Considerations in the Era of Personalized Medicine.

Background Facial clefts are rare congenital malformations, occurring in approximately 1 in 700 live births for cleft lip and palate and fewer than 1 in 100,000 for atypical Tessier clefts. They pose significant diagnostic and surgical challenges. While genetic, vascular, and environmental factors are well documented, growing embryological evidence suggests that the trigeminal nerve may also contribute to craniofacial development. This narrative review explores the association between trigeminal nerve development and facial clefts, aiming to provide a neurodevelopmental perspective with clinic

A survey of SARS-CoV-2 tropism.

The coronavirus disease 2019 (COVID-19) pandemic, caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), has significantly burdened global public health. However, the tropism of SARS-CoV-2 within the human body remains not fully understood. In this review, we overview the literature on SARS-CoV-2 infection across various human organs and tissues. We summarize the relevant specimen types, techniques for examining SARS-CoV-2 tropism, and findings at both organ/tissue and cellular levels. To systematically evaluate the evidence supporting SARS-CoV-2 tissue tropism, we establish a

Engineering a 3D platform for testis bioengineering: generation and proteomic profiling of decellularized fish testicular scaffolds.

Decellularization represents a robust strategy for generating biologically derived scaffolds that retain the native architecture and biochemical complexity of the extracellular matrix (ECM), thereby providing a conducive microenvironment for germ cell adhesion, proliferation, and differentiation-processes fundamental to the reconstitution of testicular function. While decellularized ECM (dECM) scaffolds have been extensively utilized in mammalian organoid systems for in vitro spermatogenesis and fertility-related research, the development of standardized protocols tailored to teleost models re

Hypoimmunogenic CD19 CAR-NK cells derived from embryonic stem cells suppress the progression of human B-cell malignancies in xenograft animals.

BACKGROUND: Chimeric antigen receptor (CAR) engineered natural killer (NK) cells exhibit advantages such as MHC-independent recognition and strong anti-tumor functions. However, allogeneic CAR-NK cells derived from human tissues are heterogeneous and susceptible to clearance by hosts. METHODS: We generated a B2M knockout, HLA-E and CD19 CAR ectopic expressing embryonic stem cell (ESC) line, which differentiated normally and gave rise to homogeneous CD19 CAR-NK (CD19 CAR-UiNK) cells using an organoid aggregate induction method. The CD19 CAR-UiNK were co-cultured with T cells or NK cells derive

Effects of oral prednisone after stenting in a rabbit model of established atherosclerosis.

OBJECTIVES: The aim of this study was to compare the effects of systemic prednisone in combination with a bare-metal stent (BMS) or a paclitaxel-eluting stent (Taxus, Boston Scientific Corp., Natick, Massachusetts) on neointimal inhibition and vessel healing in an atherosclerotic rabbit model. BACKGROUND: Inflammation plays a critical role in neointimal formation after coronary artery stenting. The efficacy of immunosuppressive doses of oral prednisone to inhibit in-stent neointimal proliferation was compared with BMS and with a commercially available paclitaxel-eluting stent (Taxus) in a rab

Shengxian decoction suppresses malignant progression of lung adenocarcinoma by enhancing CD8+ T cell function via the FYN-PI3K/AKT axis.

BACKGROUND: Shengxian Decoction (SXD), a traditional Chinese herbal formula, has demonstrated significant clinical efficacy against lung adenocarcinoma (LUAD). However, its exact mechanisms of action remain to be elucidated. This study aimed to establish an animal model capable of simulating the dynamic progression of LUAD to investigate the anti-tumor effects and immunomodulatory mechanisms of SXD. METHODS: A TK (Trp53-/-; KrasG12D) LUAD mouse model was established by subcutaneously transplanting CRISPR/Cas9-edited mouse lung organoids into immunocompetent C57BL/6 mice. The model was charact…

SLC22A3 regulates ferroptosis in the mesenchymal subtype of triple-negative breast cancer by modulating histone H3K4 serotonylation.

Triple-negative breast cancer (TNBC) remains a challenging and clinically aggressive subtype due to its heterogeneity and high mortality rat … optosis inducers or cisplatin, which suppress tumor growth in both MES-subtype TNBC patient-derived organoids and in vivo, offering a promising strategy for personalized therapy. These findings suggest that targeting SLC22A3, along with ferroptosis inducers, may offer a promising therapeutic strategy for patients with MES-subtype TNBC. SLC22A3 affects serot …
