Identification of Mitochondrial Dynamics-Related Genes as Diagnostic Biomarkers for Osteoarthritis Using Weighted Gene Co-expression Network Analysis and Machine Learning.

BACKGROUND: Osteoarthritis (OA) is a chronic, painful joint disease whose early diagnosis is critical for effective treatment. Since mitochondrial dynamics have been implicated in the onset and progression of OA, this study aimed to explore the diagnostic value of mitochondrial dynamics-related genes (MDRGs) in OA. METHODS: Weighted gene co-expression network analysis and 3 machine learning algorithms, including least absolute shrinkage and selection operator regression, support vector machine-recursive feature elimination, and Boruta, were used to screen signature genes. The intersection of

Glycemic Variability as a Predictor of Mortality in Sepsis Patients With Concurrent Persistent Inflammation, Immunosuppression, and Catabolism Syndrome.

BACKGROUND: Sepsis is a life-threatening condition caused by infection, which triggers dysregulated systemic inflammatory responses. Among sepsis patients, those who concurrently develop persistent inflammation, immunosuppression, and catabolism syndrome (PICS) have a significantly poorer prognosis. It has been demonstrated that associations exist between elevated glycemic variation coefficient (GVC) levels and the development of PICS in septic populations. However, the association between GVC and adverse clinical outcomes in the subgroup of septic patients with PICS requires further investiga

Development and validation of a clinical nomogram for predicting suboptimal concentration of valproate in pediatric with epilepsy: a retrospective study.

BACKGROUND: Valproate, a first-line anti-seizure medication, has a narrow therapeutic range of 50-100 μg/mL. Many children are prescribed insufficient doses of valproate, resulting in inadequate seizure control or potential toxicity. Currently, no predictive algorithms are available to customize treatment according to the specific needs of children. Our objective was to develop a nomogram that predicts the likelihood of suboptimal valproate concentrations in pediatric patients with epilepsy. METHODS: We conducted a single-center retrospective cohort study of pediatric patients with epilepsy a

Morphologic patterns associated with BRCA1 and BRCA2 genotype in ovarian carcinoma.

This study was undertaken with the hypothesis that certain common morphologic features of ovarian carcinomas are predictably associated with BRCA1 and BRCA2 deficiencies. We selected 43 high-grade serous carcinomas diagnosed at Memorial Sloan-Kettering Cancer Center that were studied as part of The Cancer Genome Atlas pilot project. In addition to 12 randomly selected nonfamilial BRCA-unassociated cases, all 31 Memorial Sloan-Kettering Cancer Center cases with BRCA1 or BRCA2 abnormality were included (n=43). Slides were examined to assess tumor architecture, mitotic index, tumor-infiltrating l

Modelling of human acute toxicity from physicochemical properties and non-vertebrate acute toxicity of the 38 organic chemicals of the MEIC priority list by PLS regression and neural network.

Linear and non-linear modelling of human acute toxicity (as human lethal concentrations; HLCs) of the 38 organic chemicals from the 50 priority compounds of the Multicentre Evaluation of In Vitro Cytotoxicity (MEIC) programme was investigated. The models obtained were derived either from a set of 23 physicochemical properties of the compounds or from their acute toxicities to five aquatic non-vertebrates together with the physicochemical properties. For the linear type, modelling was performed using a partial least square projection to latent structures (PLS) regression method; for the non-lin

Estimating the prevalence and incidence of symptomatic hand OA in the Netherlands using primary care electronic health records.

OBJECTIVE: This study estimated the incidence and prevalence of symptomatic hand osteoarthritis (OA) in the Dutch population using a validated algorithm that integrates free-text data from primary care electronic health records (EHRs). METHODS: This retrospective cohort study used the Integrated Primary Care Information (IPCI) database, including EHRs from approximately 1.8 million Dutch patients aged ≥30 years between 2013 and 2023. As the International Classification of Primary Care 1 (ICPC-1) lacks a specific code for hand OA, cases were identified using an algorithm combining ICPC-1 codes

An Active Control Method for a Lower Limb Rehabilitation Robot with Human Motion Intention Recognition.

This study presents a method for the active control of a follow-up lower extremity exoskeleton rehabilitation robot (LEERR) based on human motion intention recognition. Initially, to effectively support body weight and compensate for the vertical movement of the human center of mass, a vision-driven follow-and-track control strategy is proposed. Subsequently, an algorithm for recognizing human motion intentions based on machine learning is proposed for human-robot collaboration tasks. A muscle-machine interface is constructed using a bi-directional long short-term memory (BiLSTM) network, whic

A novel machine learning architecture to improve classification of intermediate cases in health: workflow and case study for public health.

BACKGROUND: The practice of medicine has evolved significantly during the past decade, with the emergence of Machine Learning (ML) that offers the opportunity of personalized patient-tailored care. However, ML models still face some challenges when classifying patients where clear-cut boundaries between classes are hard to identify. In this work, we propose an ML architecture to improve the sensitivity of detecting patients in intermediate "hard-to-classify" classes. METHODS: The proposed architecture replaces a single classifier with a group of cascaded increasingly specialized classifiers:

Optimization design of cross border intelligent marketing management model based on multi layer perceptron-grey wolf optimization convolutional neural network.

The cross-border intelligent marketing algorithm based on traditional linear models is relatively single in information feature extraction, making it difficult to effectively handle complex scenarios containing a large amount of implicit information in users and markets, resulting in poor personalized marketing effectiveness. To address this issue, this article proposes a cross-border intelligent marketing model that integrates rating information and user labels using a multi-layer perceptron grey wolf optimization and convolutional neural network (MLP-GWO-CNN). This model extracts implicit hi

Optimization of Forward Collision Warning Algorithm Considering Truck Driver Response Behavior Characteristics.

Forward collision warning (FCW) systems have been widely used in trucks to alert drivers of potential road situations so they can reduce the risk of crashes. Research on FCW use shows, however, that there are differences in drivers' responses to FCW alerts under different scenarios. Existing FCW algorithms do not take differences in driver response behavior into account, with the consequence that the algorithms' minimum safe distance assessments that trigger the warnings are not always appropriate for every driver or situation. To reduce false alarms, this study analyzed truck driver behavior

A flexible generative algorithm for growing in silico placentas.

The placenta is crucial for a successful pregnancy, facilitating oxygen exchange and nutrient transport between mother and fetus. Complications like fetal growth restriction and pre-eclampsia are linked to placental vascular structure abnormalities, highlighting the need for early detection of placental health issues. Computational modelling offers insights into how vascular architecture correlates with flow and oxygenation in both healthy and dysfunctional placentas. These models use synthetic networks to represent the multiscale feto-placental vasculature, but current methods lack direct con

Confirming diagnoses of acute pancreatitis with commonly available electronic data.

BACKGROUND: Recorded diagnoses of acute pancreatitis (AP) are often inaccurate resulting in limited utility for case identification in large data sources, especially where electronic medical records (EMR) are not available. Our objectives were to validate diagnoses of AP and to identify an algorithm using additional data to enhance the identification of AP cases in different data sources. METHODS: We randomly sampled 550 persons with an AP diagnosis from inpatient data or outpatient or emergency department diagnoses immediately preceding a hospitalization and 150 negative controls with a diff

Using Big Data to Develop a Clinical Decision Support System for Tinnitus Treatment.

Tinnitus is a common symptom of a phantom sound perception with a considerable socioeconomic impact. Tinnitus pathophysiology is enigmatic and its significant heterogeneity reflects a wide spectrum of clinical manifestations, severity and annoyance among tinnitus sufferers. Although several interventions have been suggested, currently there is no universally accepted treatment. Moreover, there is no well-established correlation between tinnitus features or patients' characteristics and projection of treatment response. At the clinical level, this practically means that selection of treatment i

CT image segmentation for inflamed and fibrotic lungs using a multi-resolution convolutional neural network.

The purpose of this study was to develop a fully-automated segmentation algorithm, robust to various density enhancing lung abnormalities, to facilitate rapid quantitative analysis of computed tomography images. A polymorphic training approach is proposed, in which both specifically labeled left and right lungs of humans with COPD, and nonspecifically labeled lungs of animals with acute lung injury, were incorporated into training a single neural network. The resulting network is intended for predicting left and right lung regions in humans with or without diffuse opacification and consolidati

Enhancement of capnogram waveform in the presence of chest compression artefact during cardiopulmonary resuscitation.

BACKGROUND: Current resuscitation guidelines emphasize the use of waveform capnography to help guide rescuers during cardiopulmonary resuscitation (CPR). However, chest compressions often cause oscillations in the capnogram, impeding its reliable interpretation, either visual or automated. The aim of the study was to design an algorithm to enhance waveform capnography by suppressing the chest compression artefact. METHODS: Monitor-defibrillator recordings from 202 patients in out-of-hospital cardiac arrest were analysed. Capnograms were classified according to the morphology of the artefact.
