Investigations on the venom of the South Indian scorpion Heterometrus scaber.

The enzymes from the venom of Heterometrus scaber, the indole compounds present and the toxic protein of the venom have been studied. The venom contains acid phosphatase, ribonuclease, 5'-nucleotidase, hyaluronidase, acetylcholine esterase and phospholipase. A. The indole compounds present in the venom have been identified as 5-hydroxytryptophan, tryptophan, serotonin and tryptamine, along with two unidentified indole compounds. The venom produces hyperglycaemia in sublethal doses and this has been found to be due to increased adrenaline secretion. The toxic protein of the venom has been obtai…

[Role of cholinoreactive substances of the brain in synthesis and secretion of hypothalamic hormones exerting an influence on cardiac activity].

Our previous studies had shown that cholinergic substances such as acetylcholine, arecoline, prozerine, CoA and gangleron affect the release of neurohormones from brain into blood. Arecoline, prozerine and CoA were also found to stimulate oxidative phosphorylation in heart mitochondria, decrease suprarenal ascorbic acid and raise the level of blood glucose. In the present study these processes were studied following section of the branch of the vagus going to the heart and of the spinal cord, leaving only a humoral connection between the brain and the heart and other organs. Intracisternal inj…

Baseline Regional Cholinergic Denervation Predicts Cognitive Trajectories in Moderate Parkinson Disease.

OBJECTIVE: Cognitive decline is a disabling and variable feature of Parkinson disease (PD). While cholinergic system degeneration is linked to cognitive impairments in PD, most prior research reported cross-sectional associations. We aimed to fill this gap by investigating whether baseline regional cerebral vesicular acetylcholine transporter ligand [18F]-fluoroethoxybenzovesamicol ([18F]-FEOBV) binding predicts longitudinal cognitive changes in mild-moderate, non-demented PD subjects. METHODS: Seventy-five non-demented, mild-moderate PD subjects underwent baseline [18F]-FEOBV PET and standar…

Increasing viability of pathological liver grafts from brain dead donors via intestinal parasympathetic nerve stimulation, or exogenous serotonin.

Livers from donors after brain death (DBDs) with alcohol-associated liver disease (ALD) or steatosis, which undergo prolonged cold ischemia are often not considered for transplantation. We evaluate whether norepinephrine and acetylcholine (ACh) deficiencies impair intestinal serotonin (5-HT) delivery to the liver in extended-criteria DBDs liver transplantation (LT). A DBD rat model with ALD or severe steatosis was used. Norepinephrine, ACh and 5-HT levels were evaluated in liver and intestine. Pharmacological (ACh or 5-HT administration) and electrical stimulation of intestinal parasympathetic…

Trifluoro-icaritin mitigates spared nerve injury-induced neuropathic pain by upregulating spinal α7nAChR through suppressing ferroptosis.

ETHNOPHARMACOLOGICAL RELEVANCE: Epimedium spp. Has served as a traditional analgesic herbal medicine in Chinese medicine for over two thousand years. Its active metabolite, icariin (ICT), along with its fluorinated derivative, trifluoro-icaritin (ICTF), has been shown in our previous research to alleviate neuropathic pain induced by spared nerve injury (SNI) through an α7 nicotinic acetylcholine receptor (α7nAChR)-dependent pathway. AIM OF THE STUDY: Neuropathic pain remains a significant and unresolved health issue. Ferroptosis has recently been identified as a key, yet insufficiently explor…

Falls induced by optogenetic inhibition of basal forebrain cholinergic neurons after dorsomedial striatal dopamine depletion in a dual disruption model of falling vulnerability in Parkinson's disease.

Falls are a common and debilitating feature of Parkinson's Disease (PD) patients. Prefrontal acetylcholine (ACh) deficits, as well as nigrostriatal dopamine deficits, are implicated in vulnerability to falls. PD patients with loss of cortical ACh and associated cognitive dysfunction experience a higher rate of falls than PD patients without cortical ACh loss. In addition, chemogenetic inhibition of basal forebrain (BF) neurons in rats increases the vulnerability to falls on a balance beam task. Here, the impact of transient optogenetic inhibition specifically of BF cholinergic neurons was asse…

Cholinergic-dependent dopamine signals in mouse dorsomedial striatum are regulated by frontal but not sensory cortices.

Everyday decisions depend on associations between sensory stimuli, actions, and outcomes. The striatum supports these sensorimotor associations through dopamine-dependent plasticity. Recent work has characterized a local striatal microcircuit in which cholinergic interneurons (CINs) modulate dopamine release via activation of nicotinic receptors on dopamine axons. Here, we show that visual stimuli evoke dopamine in the dorsomedial striatum partly through this cholinergic mechanism. Using anatomical and functional methods to identify the pathways involved, we found that visual and auditory cort

Dysfunction in the SOCE/EDH-mediated vasorelaxation of mesenteric arteries impairs intestinal barrier integrity in irritable bowel syndrome.

Irritable bowel syndrome (IBS) is a prevalent functional gastrointestinal disorder with an incompletely understood pathogenesis. Although dy … relaxation in mesenteric small resistance arteries was assessed using wire myography. We found that acetylcholine (ACh)-induced endothelium-dependent hyperpolarization (EDH)-mediated vasorelaxation was significantly impaired in WAS-treated mice. This impairment was transient and reversible on days 1 and 3 post-WAS but became sustained by day 10. In contrast, in …

Immunohistochemical characterisation of dopaminergic and cholinergic alterations in the prefrontal cortex and hippocampus of MPTP-treated marmosets.

Motor symptoms traditionally characterise Parkinson's disease (PD), but cognitive dysfunctions have recently emerged as significant non-motor features. While dopamine deficiency in the substantia nigra primarily causes PD, recent evidence indicates disruptions in neurochemical pathways beyond the nigrostriatal system also contribute to cognitive dysfunction. This preclinical study examines the roles of dopamine and acetylcholine (ACh) within the hippocampus and prefrontal cortex (PFC), assessing how their combined reduction manifests as neurochemical alterations in brain regions relevant to co…

Blockage of the peri-synaptic Schwann cell nicotinic α7 receptor inhibitory drive increases the neuromuscular performance in rats with experimental autoimmune Myasthenia gravis: repurposing dipyridamole usage in myasthenics.

Acetylcholine spillover from the neuromuscular synapse triggers an inhibitory drive via the activation of nicotinic α7 receptors (α7nAChRs) on perisynaptic Schwann cells (PSCs). This mechanism involves the outflow of adenosine from PSCs via ENT1 transporters and subsequent activation of presynaptic inhibitory A1 receptors. Reinforcement of this sensing inhibitory pathway by cholinesterase (AChE) inhibitors may partially attenuate their efficacy for Myasthenia gravis treatment. Here, we investigated whether this neuromuscular break is still operative in an experimental autoimmune Myasthenia gra…

Nicotine regulates abnormal macrophage polarization and trophoblast invasion associated with preterm labor via the α7nAChR/SIRT1 axis.

INTRODUCTION: Preterm birth (PTB) frequently results from the syndrome of preterm labor (PTL). PTL is linked to an atypical maternal inflammatory response, as well as intrauterine inflammation and/or infection. In this study, we explored the mechanisms involved in nicotine-mediated abnormal macrophage polarization and trophoblast invasion associated with PTL. METHODS: First, THP-1-M0 macrophages were generated by treating the human monocytic leukemia cell line (THP-1) with phorbol 12-myristate 13-acetate for a duration of 24 h. Afterward, nicotine treatment was administered, followed by cocul

Bladder instillations vs onabotulinumtoxinA injection for interstitial cystitis/bladder pain syndrome: a randomized clinical trial.

BACKGROUND: Interstitial cystitis (IC)/bladder pain syndrome (BPS) is an unpleasant sensation related to the bladder with lower urinary tract symptoms lasting more than 6 weeks, unrelated to an otherwise identifiable cause. The etiology is likely multifactorial including urothelial abnormalities, neurogenic pain upregulation, and potentially bladder and vaginal microbiome alterations. Despite treatment effectiveness of both bladder instillations and intradetrusor onabotulinumtoxinA injection for this condition, a head-to-head comparison has not been performed. OBJECTIVE: To compare the effica

Evaluating the Efficacy of Chronic Galantamine on Sustained Attention and Cholinergic Neurotransmission in A Pre-Clinical Model of Traumatic Brain Injury.

Cholinergic disruptions underlie attentional deficits following traumatic brain injury (TBI). Yet, drugs specifically targeting acetylcholinesterase (AChE) inhibition have yielded mixed outcomes. Therefore, we hypothesized that galantamine (GAL), a dual-action competitive AChE inhibitor and α7 nicotinic acetylcholine receptor (nAChR) positive allosteric modulator, provided chronically after injury, will attenuate TBI-induced deficits of sustained attention and enhance ACh efflux in the medial prefrontal cortex (mPFC), as assessed by in vivo microdialysis. In Experiment 1, adult male rats (n = …

Aβ1-42 Accumulation Accompanies Changed Expression of Ly6/uPAR Proteins, Dysregulation of the Cholinergic System, and Degeneration of Astrocytes in the Cerebellum of Mouse Model of Early Alzheimer Disease.

Alzheimer disease (AD) is a widespread neurodegenerative disease characterized by the accumulation of oligomeric toxic forms of β-amyloid (Aβ1-42) and dysfunction of the cholinergic system in the different brain regions. However, the exact mechanisms of AD pathogenesis and the role of the nicotinic acetylcholine receptors (nAChRs) in the disease progression remain unclear. Here, we revealed a decreased expression of a number of the Ly6/uPAR proteins targeting nAChRs in the cerebellum of 2xTg-AD mice (model of early AD) in comparison with non-transgenic mice both at mRNA and protein levels. We …
