Association of serum pentraxin 3 with rheumatoid arthritis disease activity and synovial blood flow.

Rheumatoid arthritis (RA) is characterized by persistent synovitis. Conventional inflammatory markers such as C-reactive protein (CRP) and erythrocyte sedimentation rate (ESR) inadequately reflect synovial inflammation, particularly under interleukin-6 pathway inhibition. Pentraxin 3 (PTX3), an extrahepatic acute-phase protein produced in inflamed tissues, may better reflect local inflammation and angiogenesis in RA joints. We enrolled 80 RA patients and 80 age-sex-matched healthy controls. Clinical data, inflammatory markers, and disease activity scores (DAS28) were collected. Serum PTX3 leve

Therapeutic efficacy of upadacitinib in refractory juvenile idiopathic arthritis: a case report.

BACKGROUND: Juvenile idiopathic arthritis (JIA) remains a clinical challenge when conventional and biological therapies fail. CASE PRESENTATION: This report describes a case of highly refractory JIA and evaluates the efficacy of the Janus kinase (JAK) inhibitor upadacitinib, providing a potential therapeutic reference for difficult-to-treat pediatric autoimmune diseases. A 16-year-old female presented with a 3-year history of polyarticular swelling and pain that worsened over 1 month. Physical examination revealed extensive tenderness in the small joints of the hands, feet, and knees. Functio

Vitamin D Receptor BsmI Polymorphism and Inflammatory Features in Juvenile Idiopathic Arthritis.

BACKGROUND: Vitamin D and vitamin D receptor (VDR) gene polymorphisms influence immune regulation, but their role in juvenile idiopathic arthritis (JIA) remains unclear. We aimed to examine the association between BsmI VDR gene polymorphism and JIA susceptibility in children and evaluate its relationship with inflammatory biomarkers. METHODS: Sixty-nine children with JIA and 69 healthy controls were enrolled over two years. Serum vitamin D levels were analyzed using the calcidiol 25-hydroxyvitamin D (25(OH)D) enzyme-linked immunosorbent assay (ELISA). Participants' biochemical and inflammator

Comprehensive Risk Factor Control Attenuates Excess Cardiovascular Risk in Rheumatoid Arthritis: A Prospective Cohort Study.

OBJECTIVE: Rheumatoid arthritis (RA) confers substantially elevated cardiovascular disease (CVD) risk. We quantified the association between joint risk factor control and incident CVD among RA patients and examined modification by genetic susceptibility. METHODS: In this prospective UK Biobank cohort, 4,426 RA patients and 17,704 matched non-RA controls were followed for CVD, coronary heart disease (CHD), stroke, and heart failure. A nine-component risk factor control score (0-9) was constructed, incorporating healthy diet, smoking, physical activity, blood pressure, glycated hemoglobin, low-

Expression characteristics of C-reactive protein in autoimmune diseases and their complications.

BACKGROUND: Autoimmune diseases involve disruptions in immune tolerance, persistent systemic inflammation, and progressive multi-organ involvement, with increasing global prevalence. Accurate prediction of disease flares and complications, alongside tools for activity stratification, represents a significant clinical challenge. C-reactive protein (CRP), a canonical pattern recognition molecule of the pentraxin family, is a widely employed inflammatory biomarker; however, its expression patterns, cellular origins, and disease-specific roles across autoimmune conditions have not been comprehensi

Periodontal Disease and Cardiovascular Risk Factors.

Various studies have suggested a bidirectional association between periodontal disease and systemic health conditions. Periodontitis is caused by polymicrobial infection, and it affects 7 - 11% of adults worldwide in an age-dependent manner. Besides its local manifestations, including gingival inflammation, periodontal pocket formation, and alveolar bone loss, periodontitis is linked to several systemic conditions. Emerging evidence points to associations with obesity, diabetes, Cardiovascular (CV) Disease (CVD), chronic renal disease, inflammatory bowel disease, respiratory diseases, rheumato

Case Report: Triple autoimmune overlap: rheumatoid arthritis, systemic lupus erythematosus, and hypereosinophilic asthma with systemic manifestations.

BACKGROUND: Overlap between rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE) ("rhupus") is recognized, but coexistence with a severe eosinophilic asthma syndrome is exceptionally rare. We describe a triple autoimmune overlap of RA, SLE, and hypereosinophilic asthma with systemic manifestations (HASM), initially managed as ANCA-negative eosinophilic granulomatosis with polyangiitis (EGPA) and subsequently re-classified in light of evolving concepts. CASE PRESENTATION: A 44-year-old woman with a 10-year history of seropositive RA developed alopecia, Coombs-positive hemolytic ane

Cimicifuga and its component actein attenuate acute gouty arthritis by targeting neutrophil activation.

Neutrophil infiltration and abnormal activation are key pathological processes in the development of acute gouty arthritis (AGA). Cimicifuga and its components exhibit substantial anti-inflammatory properties and demonstrate therapeutic potential in a range of inflammatory diseases; however, their effects on AGA remain unclear. This study aimed to investigate the anti-arthritic effect of Cimicifuga and actein, and further explore the mechanisms. In the in vitro study, neutrophils were isolated and treated with Cimicifuga-containing serum or actein to investigate their regulatory effects on neu

Decoupling musculoskeletal damage from antiviral control: the NLRP3 inflammasome in arthritogenic alphavirus infection.

Mosquito-borne arthritogenic alphaviruses, including chikungunya virus (CHIKV), Ross River virus (RRV), and Mayaro virus (MAYV), cause acute febrile polyarthralgia and can leave a substantial proportion of patients with persistent or relapsing musculoskeletal disease. In many cohorts, chronic arthritis continues after systemic viremia has declined, suggesting that late tissue pathology is sustained largely by host inflammatory programs rather than by ongoing productive viral replication. This Mini Review examines the NLRP3 inflammasome as a pathogenic node linking viral danger sensing to chron

Synergistic Effects of LPS and MSU on NF-κB/NLRP3-mediated Inflammation in Fibroblast Cells.

Inflammation is a biological response implicated in various chronic diseases, including arthritis, fibrosis, and autoimmune disorders. Human dermal fibroblasts (HDF) play a significant role in inflammatory processes, making them a valuable model for studying inflammation-related mechanisms. This study investigates the inflammatory response of HDF cells upon stimulation with lipopolysaccharide (LPS) and monosodium urate (MSU) crystals, which mimic pathogen- and danger-associated molecular patterns, respectively. Cell viability assays demonstrated that MSU crystals did not significantly reduce f

Inflammation Beyond the Prostate: The Role of Systemic Inflammatory Disorders in Prostate Carcinogenesis.

Prostate cancer remains the most frequently diagnosed malignancy in men worldwide, yet the mechanisms driving its pathogenesis extend far beyond organ-specific factors confined to prostatic tissue. Emerging evidence increasingly demonstrates that systemic inflammatory disorders and body-wide inflammatory processes function as fundamental drivers of prostate cancer initiation and progression. This comprehensive review examines the multifaceted mechanisms by which systemic inflammation contributes to prostate carcinogenesis, synthesizing evidence from diverse inflammatory disease contexts, inclu

Promoter Methylation of MMP9 and RUNX3: Diagnostic Potential and Clinical Associations in Rheumatoid Arthritis.

BACKGROUND: Rheumatoid arthritis (RA) is an autoimmune disease characterized by chronic inflammation and joint damage. Conventional diagnostic markers for RA have limitations, prompting the exploration of DNA methylation analysis as a potential biomarker. This study focuses on evaluating the methylation levels of two genes, MMP9 and RUNX3, in peripheral blood mononuclear cells (PBMCs) as potential biomarkers for RA. MATERIALS AND METHODS: We assessed the methylation levels of the MMP9 and RUNX3 promoters in PBMCs from 72 RA patients and 72 healthy controls using the methylation-quantification
