Coadministration of bicuculline and NMDA induces paraplegia in the rat.

Motor neurons (MNs) of an adult rat are normally insensitive to the neurotoxic action of NMDA. Meanwhile, the experiments in non-motor neurons showed that sensitivity to NMDA can be increased by bicuculline, an antagonist at GABA(A) receptors. The aim of the present work was to examine whether bicuculline would produce such an effect in the adult MNs. In adult Wistar rats, intrathecal injection of bicuculline and NMDA individually failed to affect motor activity of the extremities. In contrast, bicuculline-NMDA combination dose-dependently impaired hindlimb functions. At the 9th day after inje

Assessment of the seizure susceptibility of Wistar Audiogenic rat to electroshock, pentyleneterazole and pilocarpine.

This work evaluates the seizure susceptibility of nai;ve female Wistar Audiogenic rats (WARs), a genetic model of reflex epilepsy in which seizures are induced by high-intensity sound stimulation (120 dB SPL), to other pro-convulsive stimuli: transauricular electroshock (ES), pentylenetetrazole (PTZ) and pilocarpine (PILO). Normal Wistar rats from the main breeding stock of the Institute of Biological Sciences, UFMG were taken as controls. Electroshock seizures were induced through a pair of ear-clip electrodes (10 mA, at a frequency of 60 Hz, applied for 1 s). In order to test WAR susceptibil

Developmental changes in seizure susceptibility during ethanol withdrawal.

It has recently been established that adolescence may represent a developmentally sensitive period with respect to the effects of ethanol, particularly within the NMDA neurotransmitter system. However, the same may also be true of the GABA system. There is evidence to suggest that the number of GABA receptors and their kinetics may change across development. The purpose of this study was to determine if GABA-mediated seizure susceptibility during ethanol withdrawal differed between adolescent and adult animals. Results indicate that adult animals pretreated with ethanol were more likely to ach

Establishment of a risk prediction model for hydrocephalus complicated by neonatal bacterial meningitis.

BACKGROUND: Hydrocephalus is a severe complication of neonatal bacterial meningitis (NBM), threatening the health and quality of life of neonates, affecting the outcome of nervous system development, and leading to neurological sequelae, such as movement disorders, hearing impairment, mental retardation, and epilepsy. Improvement in prognosis is closely related to early identification and active treatment. OBJECTIVE: To find the independent risk factors of NBM complicated with hydrocephalus, to construct the related risk prediction model and validate it, in order to provide help for clinician

A new derivative of valproic acid amide possesses a broad-spectrum antiseizure profile and unique activity against status epilepticus and organophosphate neuronal damage.

PURPOSE: sec-Butyl-propylacetamide (SPD) is a one-carbon homolog of valnoctamide (VCD), a central nervous system (CNS)-active amide derivative of valproic acid (VPA) currently in phase II clinical trials. The study reported herein evaluated the anticonvulsant activity of SPD in a battery of rodent seizure and epilepsy models and assessed its efficacy in rat and guinea pig models of status epilepticus (SE) and neuroprotection in an organotypic hippocampal slice model of excitotoxic cell death. METHODS: The anticonvulsant activity of SPD was evaluated in several rodent seizure and epilepsy mode

Sex specific correlation between GABAergic disruption in the dorsal hippocampus and flurothyl seizure susceptibility after neonatal hypoxic-ischemic brain injury.

Since neonatal hypoxia-ischemia (HI) disrupts the hippocampal (Hp) GABAergic network in the mouse and Hp injury in this model correlates with flurothyl seizure susceptibility only in male mice, we hypothesized that GABAergic disruption correlates with flurothyl seizure susceptibility in a sex-specific manner. C57BL6 mice were exposed to HI (Vannucci model) versus sham procedures at P10, randomized to normothermia (NT) or therapeutic hypothermia (TH), and subsequently underwent flurothyl seizure testing at P18. Only in male mice, Hp atrophy correlated with seizure susceptibility. The number of

Immunovirological correlates in human rabies treated with therapeutic coma.

A 37-year-old woman was admitted to hospital and over the next 5 days developed a progressive encephalitis. Nuchal skin biopsy, analyzed using a Rabies TaqMan(c) PCR, demonstrated rabies virus RNA. She had a history in keeping with exposure to rabies whilst in South Africa, but had not received pre- or post-exposure prophylaxis. She was treated with a therapeutic coma according to the "Milwaukee protocol," which failed to prevent the death of the patient. Rabies virus was isolated from CSF and saliva, and rabies antibody was demonstrated in serum (from day 11 onwards) and cerebrospinal fluid (

[Model of Huntington's disease induced with 3-nitropropionic acid].

INTRODUCTION: Huntington's disease is an autosomal dominant hereditary disorder. This neurodegenerative illness is characterized by mutation of the huntingtin protein gene, causing the formation of intracellular protein aggregates. DEVELOPMENT: Intensive research efforts have been made to investigate the molecular mechanism involved. For this reason, the development of animal and cellular models of Huntington's disease has offered alternative approaches to study of this disease. The alteration of succinate dehydrogenase activity has been linked to Huntington's disease. 3-nitropropionic acid i

Fibrous and protoplasmic astrocytes express GABAA receptors that differ in benzodiazepine pharmacology.

Astrocytes cultured from spinal cord contain two morphologically distinguishable types of astrocytes: fibrous and protoplasmic cells. Both astrocyte subtypes, in culture, are able to express GABAA receptors, and their activation results in inward currents at the resting potential. Using patch-clamp electrophysiology we characterized their basic receptor pharmacology and compared it to spinal cord neurons that were also present in small numbers in these cultures. As in neuronal GABAA receptors, the local anesthetic pentobarbital effectively potentiated GABA-induced currents in both astrocyte su

[Effect of psychopharmacotherapy on the course of forced insulin-coma treatment of patients with paranoid schizophrenia].

The course of forced insulin-coma therapy in cases of preliminary employment of psychotropic drugs (mainly derivatives of phenothiazine or leponex) may be attended by variable forms of deviations including serious complications whereas they are not encountered in cases of treatment with haloperidol and pharmacologically similar long-acting drugs. These findings justify the employment, when necessary, of the above drugs in combination with forced insulin-coma therapy.

A benzodiazepine agonist and contragonist have hypothermic effects in rodents.

The effects on body temperature of the benzodiazepine contragonists FG 7142 (N-methyl-beta-carboline-3-carboxamide) and DMCM (methyl-6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate), the benzodiazepine antagonist Ro 15-1788 (ethyl-8-fluoro-5,6-dihydro-5-methyl-7-oxo-4H-imidazo-(1,5-a)(1,4)benzod iazepin e -3-carboxylate) and the benzodiazepine agonist, flurazepam, were investigated in mice. Both of the contragonists and flurazepam reduced the body temperature. The antagonist Ro 15-1788 did not alter body temperature alone but significantly antagonised the effects of FG 7142 and of flurazepa
