Age, Age-Related Comorbidities and Survival in Palbociclib, Ribociclib and Abemaciclib Users With Advanced Breast Cancer: A Nation-Wide Retrospective Cohort Study.

PURPOSE: Prevalence of breast cancer (BC) increases with age, but the external validity of data obtained from pivotal trials of medicinal products remains a concern due to the underrepresentation of frail and older adults. Cyclin-dependent kinase inhibitors (CDKi), palbociclib, abemaciclib, and ribociclib are considered an essential part of the standard-of-care in the management of advanced/metastatic breast cancer. We investigated age, comorbidities, and survival in a nationwide real-world cohort of CDKi users. METHODS: Data from the Finnish Cancer Registry, reimbursed dispensed prescription

Dual BRD4/AKT inhibition overcomes c-MYC-driven resistance in metastatic castration-resistant prostate cancer.

Metastatic castration-resistant prostate cancer (mCRPC) represents the terminal and most challenging stage of prostate cancer progression, characterized by limited therapeutic options and poor prognosis. Although AKT inhibitors have been explored as a potential treatment strategy for mCRPC, their efficacy as monotherapy is often constrained by the rapid development of resistance. Emerging evidence indicates that this acquired resistance is frequently associated with the elevated expression of the c-MYC oncogene. Bromodomain and extraterminal (BET) inhibitors, such as those targeting BRD4, can

NDGA alleviates oxidative stress and supports early embryonic development in porcine oocytes.

Nordihydroguaiaretic acid (NDGA) is a potent cell-permeable antioxidant that regulates cellular redox balance, but its role in porcine oocyte in vitro maturation (IVM) and embryonic development remains unclear. In this study, porcine oocytes were cultured in IVM medium supplemented with different concentrations of NDGA (0, 8, 10, 20, and 100 μM). The results showed that 8 μM NDGA significantly enhanced cumulus cell expansion and increased the first polar body extrusion rate, whereas a high concentration (100 μM) impaired these processes. Immunofluorescence analysis demonstrated that 8 μM NDGA

Tceal7 is a BRG1-regulated target of calcineurin signaling that promotes myoblast differentiation.

The transcriptional control of skeletal muscle differentiation requires the coordinated activity of lineage-defining transcription factors, signal-responsive regulators, chromatin modifiers, and ATP-dependent chromatin remodeling enzymes. Here, we identify TCEAL7, a member of the X-linked, poorly characterized TCEAL family of proteins, as a direct downstream target of BRG1-containing mammalian SWI/SNF (mSWI/SNF) complexes and calcineurin signaling during myoblast differentiation. Analyses of previously published datasets showed that pharmacological inhibition of mSWI/SNF bromodomains or knockd

DNA replication stress and translational repression converge to drive CDK1- and caspase-dependent apoptosis in Ewing sarcoma.

Despite aggressive multimodal therapy, including cytotoxic chemotherapy, surgery, and radiation, the prognosis for patients with Ewing sarcoma remains poor, particularly for those with metastatic or relapsed disease. Combining agents that increase DNA replication stress with ATR-CHK1-WEE1 pathway inhibitors, which disrupt the DNA damage response and cell cycle checkpoints, is a promising strategy under clinical investigation in Ewing sarcoma and other cancers. However, the mechanisms by which these drug combinations selectively kill cancer cells under replication stress remain incompletely und

Blockade of Presynaptic α2δ1-2 Subunits of Voltage-Gated Ca²⁺ Channels Attenuates Neurobiochemical and Sensorimotor Deficits After Traumatic Brain Injury in Mice.

Traumatic brain injury (TBI) to the motor cortex disrupts corticospinal tracts and induces persistent sensorimotor impairments, largely driven by secondary neurobiochemical cascades. Excessive synaptic glutamate release, mitochondrial Ca²⁺ overload, and progressive neurodegeneration critically shape these outcomes, with preclinical and clinical data revealing neuronal loss and proteinopathy resembling motor neuron disorders. Here, we investigated whether pregabalin blockade of the presynaptic α2δ1-2 subunit of voltage-gated Ca²⁺ channels could mitigate excitotoxicity and promote sensorimotor r

miR-122 Deficiency in Mice Enhances Regeneration in Healthy Liver but Drives Pathological Repair and Functional Decline in Fibrotic Liver.

MicroRNA-122 (miR-122) is the most abundant hepatic microRNA and a key regulator of hepatocyte proliferation, metabolism and differentiation. Although widely studied in hepatocellular carcinoma, its role in liver regeneration remains unexplored. This study investigated how miR-122 deficiency modulates liver regeneration under physiological conditions and during chronic liver injury. A miR-122-deficient mouse model (miR-122-/-) was generated using CRISPR/Cas9, and liver regeneration was assessed after two-thirds partial hepatectomy (PHx) in healthy and CCl4-induced fibrotic livers. In healthy l

Role of CNNM4 in the progression of cholangiocarcinoma: implications for ferroptosis and therapeutic potential.

BACKGROUND AND OBJECTIVE: Cholangiocarcinoma (CCA) is a heterogeneous neoplasm of the biliary epithelium that easily infiltrates, metastasises and recurs. Magnesium disbalance is a hallmark of CCA, with the magnesium transporter cyclin M4 (CNNM4) being a key driver of various hepatic diseases. This study aims to unravel the role of CNNM4 in the initiation and progression of CCA. DESIGN: CNNM4 protein and gene expression were assessed in vitro, in vivo and in patients with CCA. Silencing of CNNM4 was effectively achieved by using small interfering RNA (siRNA) or short hairpin RNA in CCA cell l

Using network pharmacology and structural biology to elucidate the molecular mechanism by which Macrolactin A protects against aphids.

Aphids are major agricultural pests that cause severe crop damage. While chemical pesticides remain the dominant control strategy, their associated environmental contamination and resistance issues necessitate the development of eco-friendly biopesticides. This study employed network pharmacology and structural biology to elucidate the molecular mechanism by which Macrolactin A protects against aphids, with validation through molecular docking, 100-ns molecular dynamics (MD) simulations, and enzymatic assays. Target prediction identified five aphid proteins as potential targets of Macrolactin

Targeted Therapeutics in Breast Cancer: Promise, Progress, and the Challenge of Triple-negative Subtype.

INTRODUCTION: Breast cancer is the most common cancer that affects women globally, with several molecular subtypes defined by hormone and growth factor receptor expression. While advances in understanding the biology of these subtypes have led to targeted therapies, aggressive forms, particularly Triple-Negative Breast Cancer (TNBC), remain challenging due to their heterogeneity and lack of approved targeted options. METHODS: This review explores current treatment approaches and emerging targeted therapies across the main breast cancer subtypes. We focus on the clinical development and transl

Miconazole as a Repurposed Anticancer Candidate for Prostate Cancer.

Prostate cancer is one of the major health concerns, with increasing incidence and persistent therapeutic resistance worldwide. Despite various available treatments (radiation therapy, chemotherapy, hormone therapy, cryotherapy, biological therapy, and high-intensity focused ultrasound), patients still show treatment-related side effects over many years. These challenges highlight the urgent need for novel and safe therapeutic strategies. In the present study, we repurposed five FDA-approved antifungal agents, namely natamycin (NM), terbinafine hydrochloride (TH), ketoconazole (KZ), miconazole

Estetrol potentiates progesterone-driven growth restraint while blunting pro-angiogenic activation in murine endometriosis lesions.

BACKGROUND: Endometriosis is sustained by ectopic endometrial implants that remain proliferative and vascularized, making endocrine modulation a cornerstone of therapy. Estetrol (E4) an endogenous estrogen produced during pregnancy, with selective tissue activity, may modulate progesterone (P4) outputs in ectopic tissue and potentially shift lesion biology toward growth restraint without reinforcing vascular support. OBJECTIVE: To test whether E4 modifies P4 effects on lesion proliferation, apoptosis-related signaling, and angiogenic/endothelial activation in vivo. METHODS: Hormonally intact

MCKG-SL: Knowledge graph-based multi-feature cross-aggregation synthetic lethality prediction for KRAS gene.

KRAS (Kirsten rat sarcoma viral oncogene homolog) is the most commonly mutated oncogene in human cancer. Targeting synthetic lethal (SL) partners in the setting of oncogenic KRAS is an alternative therapeutic strategy for KRAS-mutant malignancies. However, existing SL prediction algorithms are limited by incomplete understanding of complex biological system interaction networks or ignore some information in the local association of gene pairs. To overcome these challenges, we propose a novel Knowledge Graph-based Synthetic Lethality model named MCKG-SL, which learns the interaction information

Multifaceted Integrated Analysis of CDK1 and TOP2A Signaling Pathways for Multi-Target Therapeutic Intervention in Epithelial Ovarian Cancer.

Epithelial ovarian cancer (EOC) remains one of the most aggressive gynecological malignancies, largely due to late-stage diagnosis, therapeutic resistance, and molecular heterogeneity. This study aimed to identify biologically relevant hub genes and evaluate potential dual-target compounds against Cyclin-Dependent Kinase 1 (CDK1) and DNA Topoisomerase II Alpha (TOP2A) through an integrated computational framework. Transcriptomic datasets from GSE28799, GSE54388, and GSE14407 were analyzed to identify overlapping differentially expressed genes, followed by protein-protein interaction analysis,

Exercise and p21 signaling in central nervous system aging and senescence: A context- and dose-dependent relationship.

Physical exercise is a potent modifier of CNS aging, but its relationship with the cyclin-dependent kinase inhibitor p21 (CDKN1A) is neither uniformly suppressive nor equivalent to cellular senescence. Direct evidence remains sparse and preclinical. Sustained physiological exercise reduced hippocampal or cortical p21-associated signatures in models of metabolic distress, natural aging, and amyloid pathology. Conversely, acute treadmill exercise transiently increased hippocampal Cdkn1a, while spatial transcriptomics identified increased endothelial Cdkn1a during broader exercise-associated vasc

Isolation and functional characterization of novel neuropeptides regulating hippocampal neurogenesis.

Neuropeptides (NPs) are small peptides, produced and secreted by neurons, serving as neuromodulators and chemical messengers that influence neuronal growth, proliferation, and differentiation. Despite their diverse and important roles, the discovery and functional characterization of many NPs remain incomplete. This study aimed to extract and identify novel NPs from rat brain tissue. For this purpose, NPs were purified using gel filtration chromatography and sequenced by mass spectrometry (MS). After characterization, one candidate peptide (NP1) was selected for functional evaluation in primar

Dalpiciclib induced hepatitis B virus reactivation leading to liver failure: a rare case report and review of the literature.

Dalpiciclib is a cyclin-dependent kinase 4/6 (CDK4/6) inhibitor that plays a key role in hormone receptor (HR)-positive, human epidermal growth factor receptor 2 (HER-2)-negative breast cancer. However, there have been no reports of acute liver failure in Hepatitis B virus (HBV) carriers caused by HBV reactivation induced by dalpiciclib. We present a 50-year-old female with recurrent breast cancer and a history of asymptomatic HBV carriage. After dalpiciclib initiation, she developed progressive liver impairment, with hepatitis B virus DNA (HBV DNA) escalating from 1.02 × 103 to 2.13 × 106 IU/

High-resolution CRISPR/Cas9 screens identify PAK2 as a suppressor of macrophage proliferation.

Macrophage abundance is regulated by pathways controlling survival and proliferation, yet the genetic determinants of macrophage fitness remain poorly understood. Here, we performed replicate genome-wide CRISPR/Cas9 screens in primary murine bone marrow-derived macrophages (BMDMs) to identify positive and negative regulators of macrophage fitness. Validity of the screens is supported by finding previously characterized core essential genes such as those encoding translation machinery, while disruption of established tumor suppressors increased BMDM representation in the culture. As expected, C

GLUT1/SGLT1 balance associates with high-glucose buffering of metformin-induced lysosomal deacidification and mTORC1 inhibition in colorectal cancer.

Metformin has anticancer potential, but benefit in colorectal cancer (CRC) is heterogeneous and determinants of tumor cell response remain unclear. We tested whether extracellular glucose and the relative expression of glucose transporter 1 (GLUT1/SLC2A1) and sodium-glucose cotransporter 1 (SGLT1/SLC5A1), quantified as a glucose transporter ratio score (GTRS), are associated with buffering of metformin effects. We combined a 1,2-dimethylhydrazine (DMH)-induced CRC model, glucose-controlled cell-line experiments, and analyses of public bulk, single-cell, and spatial transcriptomic datasets. In
