Pituitary adenylate cyclase activating polypeptide stimulates gallbladder motility in conscious dogs.

We investigated whether pituitary adenylate cyclase activating polypeptide (PA-CAP27 and PACAP38) had any effect on gallbladder motility in conscious dogs, in which force transducers were chronically implanted in the gastric antrum, duodenum and gallbladder. PACAP27 and PACAP38 were administered intravenously during the digestive and interdigestive states at doses of 30, 100 and 300 pmol/kg. By way of comparison, cholecystokinin octapeptide (CCK-OP) was administrated at doses of 3, 9 and 27 pmol/kg. As a result, each peptide evoked transient and tonic contractions both in the digestive and int

A PEG-mediated protoplast transformation system for EGFP labeling of the shot-hole pathogen Wilsonomyces carpophilus.

Wilsonomyces carpophilus is a fungal pathogen causing shot-hole disease in stone fruit trees, including wild apricot populations in the Tianshan wild fruit forests. The absence of an effective genetic transformation system has limited molecular studies of this pathogen. In this study, we established a PEG-mediated protoplast transformation system for W. carpophilus strain G052 5 m2. Key factors affecting protoplast preparation, including mycelial culture duration, enzyme combination, digestion time, and osmotic stabilizer, were evaluated. The optimal conditions consisted of 24-h-old mycelia, e…

Discovery of garlic-derived peptides as natural DPP4 inhibitors: An integrated computational, network pharmacology, and in situ evaluation approach.

Type 2 diabetes mellitus (T2DM), marked by impaired glucose homeostasis and hyperglycemia, remains a major global health burden. Although dipeptidyl peptidase-IV (DPP4) inhibitors improve glycemic control, safety concerns about synthetic agents drive the search for safer food-derived alternatives. Garlic (Allium sativum L.), a widely consumed functional food, is underexplored as a source of antidiabetic peptides. Here, simulated gastrointestinal digestion, virtual screening, molecular docking, and experimental validation were integrated to identify garlic-derived DPP4 inhibitory peptides. Five…

Transforming RNA Degradation into Accurate RNA Detection: RNase I-Assisted Rolling Circle Amplification for Clinical Samples.

Ribonucleases are powerful degradative enzymes that rapidly cleave RNA and have long been regarded as obstacles to RNA diagnostics. Detecting large, structured RNA molecules in biological samples is further complicated by extensive secondary structures and abundant background RNA. Here we transform this destructive activity into a diagnostic advantage through a RNase I-assisted rolling circle amplification (RI-RCA) strategy that integrates enzymatic RNA digestion with circular DNA templates (CDTs) to achieve direct, reverse-transcription-free RNA detection. In this system, Escherichia coli RNa…

The microbiome in reptile health, disease, and ecology.

SUMMARYReptiles are a diverse and speciose class of animals that are broadly threatened by habitat loss, climate change, and other factors. From a microbiological perspective, reptiles have historically been examined as a source of disease, particularly salmonellosis, with most studies being culture-based investigations into causative agents of disease and potential for zoonoses. More recent work has sought to characterize the oral, skin, and gut microbiomes of reptiles more broadly to understand their contribution to reptile health and digestion. Non-avian reptiles are particularly interestin…

A new strategy for steady-state butyric acid encapsulation: Mechanism of starch-lauric acid-protein-butyric acid complex formation, release characteristics and prebiotic properties.

To address the low endogenous production and short half-life of butyric acid (BA), this study successfully constructed an AM-LA-WP-BA quater … starch content increasing from 36.51% to 58.41%. This endowed the complex with strong resistance to digestion, thereby enabling colon-specific release. Furthermore, in vitro fermentation studies revealed that SLPB not only directly released butyric acid but also served as a fermentation substrate to promote the proliferation of butyrate-producing bacteria ( …

Goji Leaves and Buds as Food Resources: Composition, Processing, Safety, and Links to Side-Stream Valorization.

Goji leaves and buds are used as vegetables and teas, but their composition and behavior in formulated foods differ from those of the berries. This critical narrative review examines their phytochemistry, processing, food applications, digestion, and safety, with selected comparisons to berry-processing residues and orchard biomass. The literature includes leaf-tea processing, polysaccharide-modified dough, emulsion delivery systems, and bud-tea or leaf-extract additions to savory foods. Genotype, harvest stage, and extraction account for substantial compositional variation. In food matrices, …

Activation of chicken macrophages by isomalto/malto-polysaccharide (IMMP) is facilitated by toll-like receptor 4 (TLR4).

Isomalto/malto-polysaccharide (IMMP) is a digestion resistant starch that has shown to increase SCFA production in the gut microbiota of human and mice to promote intestinal health. Like other prebiotics, it escapes enzymatic digestion in the small intestine and reaches the colon intact where it can interact with host epithelial and immune cells. Recently it was shown that in vitro IMMP can bind TLR4 on human macrophages resulting in their activation. The purpose of the present study was to investigate putative immune activation effects of IMMP upon direct interaction with chicken macrophages.…

PI3-kinase has multiple functions in asexual blood stages of Plasmodium falciparum.

All symptoms of malaria are caused during the replication of the parasite Plasmodium falciparum in human red blood cells. The parasite digests the host cell cytoplasm in its food vacuole, a pathway of particular interest as drug target. The Vps34-type PI3-kinase in P. falciparum produces PI3-monophophate (PI3P) and has been linked to haemoglobin digestion, to resistance to the current first line antimalarial artemisinin and to biology of the apicoplast. Here we dissect the functions of PfPI3-kinase by inducible deletion of its gene using the loxP-DiCre system and find that PfPI3-kinase is esse…

Screening study of hydroxytyrosol metabolites from in vitro fecal fermentation and their interaction with intestinal barrier repair receptor AhR.

Olive oil polyphenol hydroxytyrosol (HT) significantly repairs intestinal barrier function, but its absorption in the stomach and small intestine is limited. The metabolites of unabsorbed HT that reach the colon are crucial, yet their effects on colonic microbiota and intestinal barrier repair remain unclear. This study utilized in vitro simulated digestion and colonic fecal fermentation to investigate HT's digestion and fermentation. Results indicated that 79.25% of HT potentially reached the colon intact. Further 16S rDNA, targeted, and untargeted metabolomics analyses showed that HT can be …

Oral Protein Supplements Might Improve Nutritional Status and Quality of Life in Elderly Patients after Standard Pancreatic Resection.

Elderly patients who undergo pancreaticoduodenectomy (PPPD) or distal pancreatectomy (DP) experience not only a reduction in protein intake but also a decrease in protease secretion, leading to impaired protein digestion and absorption. This increases the risk of malnutrition and creates a dual burden of sarcopenia. This randomized, double-blind, placebo-controlled trial examined the impact of protein supplements on the nutritional status and quality of life (QoL) of elderly patients after PPPD and DP surgeries. For six weeks, the case group (CG; n = 23) consumed protein supplements containing…
