The regulatory mechanisms of cerium oxide nanoparticles in oxidative stress and emerging applications in refractory wound care.

Cerium oxide nanoparticles (CeNPs) have emerged as a potent therapeutic agent in the realm of wound healing, attributing their efficacy predominantly to their exceptional antioxidant properties. Mimicking the activity of endogenous antioxidant enzymes, CeNPs alleviate oxidative stress and curtail the generation of inflammatory mediators, thus expediting the wound healing process. Their application spans various disease models, showcasing therapeutic potential in treating inflammatory responses and infections, particularly in oxidative stress-induced chronic wounds such as diabetic ulcers, radi

Recent advances in leishmaniasis treatment.

About 1.5 million new cases of cutaneous leishmaniasis and 500,000 new cases of visceral leishmaniasis occur each year around the world. For over half a century, the clinical forms of the disease have been treated almost exclusively with pentavalent antimonial compounds. In this review, we describe the arsenal available for treating Leishmania infections, as well as recent advances from research on plants and synthetic compounds as source drugs for treating the disease. We also review some new drug-delivery systems for the development of novel chemotherapeutics. We observe that the pharmaceuti

Detection and determination of surface levels of poloxamer and PVA surfactant on biodegradable nanospheres using SSIMS and XPS.

The surface chemical characterisation of sub-200 nm poly(DL-lactide co-glycolide) nanospheres has been carried out using the complementary analytical techniques of static secondary ion mass spectrometry (SSIMS) and X-ray photoelectron spectroscopy (XPS). The nanospheres, which are of interest for site-specific drug delivery, were prepared using an emulsification-solvent evaporation technique with poly(vinyl alcohol), Poloxamer 407 and Poloxamine 908 respectively as stabilisers. The presence of surfactant molecules on the surface of cleaned biodegradable colloids was confirmed and identified on

Nanomedicines for neurodegenerative ageing: nasal delivery innovations for Alzheimer's and Parkinson's disease.

Alzheimer's disease and Parkinson's disease are progressive, age-related neurodegenerative disorders with increasing global prevalence, yet their treatment remains challenging despite the availability of multiple therapeutic agents. Conventional formulations are often limited by poor solubility, restricted blood-brain barrier penetration, extensive first-pass metabolism, short elimination half-life, low brain bioavailability, and systemic adverse effects. The nose-to-brain route has emerged as a promising strategy for delivering therapeutics directly to the brain. This approach offers non-inva

Beyond Biomimetics: Pathology-Informed Engineering Rescues Reconstituted HDL From Inflammatory Dysfunction for Sepsis Immunotherapy.

Biomimetic therapies hold great promise for treating intractable diseases, yet their translation is often hindered by dysfunction-the loss of intended biological function upon exposure to pathological microenvironments. This challenge is well exemplified by reconstituted high-density lipoprotein (rHDL), a clinically advanced HDL mimetic and representative biomimetic drug delivery platform. Here, guided by a pathology-informed identification-design-evaluation-validation workflow, we identified representative sepsis-relevant dysfunction liabilities of HDL-based systems under pathology-relevant c

Nanomedicine Targeting Cancer-Associated Fibroblasts in Prostate Cancer: From Biological Mechanisms to Integrated Theranostic Strategies.

Prostate cancer (PCa) is the second most common malignancy in men worldwide. Advanced stages are characterized by tumor heterogeneity, metastasis, and resistance to androgen deprivation therapy and chemotherapy. Cancer-associated fibroblasts (CAFs), the predominant stromal cells in the PCa tumor microenvironment (TME), critically drive tumor progression, metastasis, and therapeutic resistance. Nanomedicine represents a transformative strategy for targeting CAFs. It leverages engineered nanomaterials to achieve precise drug delivery, improved bioavailability, and multimodal theranostic capabili

Structure, corrosion behavior and mechanical property of a novel poly(vinyl alcohol) composite in simulated body fluid.

The data for long-term drug-delivery systems are scarce compared to the short-term systems because the required research efforts are more time-consuming. In this study, we report a novel cross-linked composite based on poly(vinyl alcohol) (PVA) containing cupric ions for long-term delivery, which is helpful for contraception and trace element balance in the human body. The composition, corrosion products, crystal structure, chemical structure and mechanical stability of the composite, after being immersed in simulated body fluid (SBF) for one year, were studied by X-ray fluorescence spectrosco

Drug delivery to brain and the role of carrier-mediated transport.

In summary, the results suggest that carrier-mediated transport can be used to augment the brain delivery of a wide variety of hydrophilic therapeutic drugs. A large number of carriers are now known to be present at the brain capillary endothelium, and in many instances these carriers have been shown to mediate the brain uptake of exogenous drugs. The findings with D,L-NAM demonstrate that brain delivery can be improved through design of selective, high affinity agents. Although NAM was developed for the large neutral amino acid carrier, high affinity drugs could be produced for other systems,

Local Delivery of Azithromycin Nanoformulation Attenuated Acute Lung Injury in Mice.

Humanity has suffered from the coronavirus disease 2019 (COVID-19) pandemic over the past two years, which has left behind millions of deaths. Azithromycin (AZ), an antibiotic used for the treatment of several bacterial infections, has shown antiviral activity against severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) as well as against the dengue, Zika, Ebola, and influenza viruses. Additionally, AZ has shown beneficial effects in non-infective diseases such as cystic fibrosis and bronchiectasis. However, the systemic use of AZ in several diseases showed low efficacy and potential c

Hyperthermia-targeted rectal delivery of thermosensitive liposomal doxorubicin via intra-arterial and intravenous administration.

INTRODUCTION: Lyso-thermosensitive liposomal doxorubicin (LTLD) is a thermosensitive nanomedicine designed to release doxorubicin rapidly at mild hyperthermic temperatures. Unlike systemic doxorubicin, which is limited by cardiotoxicity and poor tumor penetration, LTLD enables targeted drug delivery enhanced by localized hyperthermia through heat-triggered release. While LTLD has demonstrated improved drug delivery with tumor-localized hyperthermia, comparative analyses of intravenous (IV) versus intra-arterial (IA) delivery routes for rectal targeting remain unexplored. This study evaluates d

Poly(lactic acid)-hyperbranched polyglycerol nanoparticles enhance bioadhesive treatment of esophageal disease and reduce systemic drug exposure.

The effective treatment of esophageal disease represents a significant unmet clinical need, as existing treatments often lead to unnecessary systemic drug exposure and suboptimal concentrations at the disease site. Here, surface-modified bioadhesive poly(lactic acid)-hyperbranched polyglycerol nanoparticles (BNPs), with an average 100-200 nm diameter, were developed for local and sustained esophageal drug delivery. BNPs showed significantly higher adhesion and permeation into ex vivo human and rat esophageal tissue than non-adhesive nanoparticles (NNPs) and had longer residence times within th

Multi-Channel Neural Interface for Neural Recording and Neuromodulation.

Neural interfaces have emerged as pivotal platforms for advancing digital neurotherapies by enabling the real-time acquisition and monitoring of neural signals. Traditional single-channel systems are inherently limited in their capacity to capture the complex and large-scale interactions among diverse neuronal populations. In contrast, multi-channel systems provide the high spatiotemporal resolution necessary to decode the dynamic activity of neural circuits across multiple brain and spinal cord regions. This review provides a comprehensive overview of recent advances in multi-channel neural i

Glucose sensitive konjac glucomannan/concanavalin A nanoparticles as oral insulin delivery system.

Compared with injection, oral drug delivery is a better mode of administration because of its security, low pain and simplicity. Insulin is the first choice for clinical treatment of type 1 diabetes, but, because insulin inability to resist gastrointestinal (GI) digestion results in poor oral bioavailability of insulin. Herein, we developed a targeted oral delivery system for diabetes. ConA-INS-KGM nanoparticles were prepared, loaded with insulin, fabricated from konjac glucomannan (KGM) and concanavalin A (ConA) through a crosslinking method, as an insulin oral delivery system in response to
