Amelioration of Cavernosal Fibrosis and Erectile Function by Lysyl Oxidase Inhibition in a Rat Model of Cavernous Nerve Injury.

BACKGROUND: Cavernous nerve injury (CNI) causes fibrosis and loss of smooth muscle cells (SMCs) in the corpus cavernosum and leads to erectile dysfunction, and lysyl oxidase (LOX) activation has been found to play an important role in fibrotic diseases. AIM: To evaluate the role of LOX in penile fibrosis after bilateral CNI (BCNI). METHODS: Rats underwent BCNI or a sham operation and were treated with vehicle or β-aminopropionitrile, a specific LOX activity inhibitor. 30 days after BCNI, rats were tested for erectile function before penile tissue harvest. LOX and extracellular matrix compone

Knockout of Dual-Specificity Protein Phosphatase 5 Protects Against Hypertension-Induced Renal Injury.

Dual-specificity protein phosphatase 5 (DUSP5) is a member of the tyrosine-threonine phosphatase family with the ability to dephosphorylate and inactivate extracellular signal-related kinase (ERK). The present study investigates whether knockout (KO) of Dusp5 improves renal hemodynamics and protects against hypertension-induced renal injury. The renal expression of DUSP5 was reduced, and the levels of phosphorylated (p) ERK1/2 and p-protein kinase C (PKC) α were elevated in the KO rats. KO of Dusp5 enhanced the myogenic tone of the renal afferent arteriole and interlobular artery in vitro with

MSC-derived extracellular vesicles for spinal cord injury: inflammatory microenvironment modulation, stage-dependent repair mechanisms, and translational challenges.

Spinal cord injury is not only an acute mechanical insult but also the beginning of a prolonged secondary pathological process dominated by inflammatory microenvironmental imbalance. Because the primary traumatic event occurs within minutes and offers a narrow therapeutic window, persistent post-traumatic inflammation, blood-spinal cord barrier disruption, edema, oxidative stress, complement activation, inflammasome signaling, astrocyte reactivity, vascular dysfunction, and extracellular matrix remodeling represent more realistic therapeutic targets. In accordance with the Minimal Information

Effect of 38% Silver Diamine Fluoride on Salivary Matrix Metalloproteinase-9 Levels in Children with Early Childhood Caries: A Clinical Study.

BACKGROUND: Early childhood caries is a major oral health problem in most industrialized countries, affecting the overall quality of life of children. It is caused by the action of bacterial acids on the enamel surface thereby demineralizing it and progressively destroying the tooth. It presents initially as smooth-surface carious lesions affecting the primary maxillary incisors. With the advancement of the lesion, decay progresses further to involve other primary teeth as well and if not treated early, the caries may involve the pulp leading to irreversible pulpitis. Matrix metalloproteinases

ErpY-like Lipoprotein of Leptospira Outsmarts Host Complement Regulation by Acquiring Complement Regulators, Activating Alternative Pathways, and Intervening in the Membrane Attack Complex.

The survival of pathogenic Leptospira in the host depends on its proficiency to circumvent the immune response. These pathogens evade the complement system in serum by enticing and amassing the serum complement regulators onto their surface. ErpY-like lipoprotein, a surface-exposed protein of Leptospira spp., is conserved in the pathogenic Leptospira serovars. The recombinant form of this protein interacts with multiple extracellular matrix (ECM) components and serum proteins such as soluble complement regulators factor H (FH) and factor I (FI). Here, we document that the supplementation of rE

Negative regulation of Smad1 pathway and collagen IV expression by store-operated Ca2+ entry in glomerular mesangial cells.

Collagen IV (Col IV) is a major component of expanded glomerular extracellular matrix in diabetic nephropathy and Smad1 is a key molecule regulating Col IV expression in mesangial cells (MCs). The present study was conducted to determine if Smad1 pathway and Col IV protein abundance were regulated by store-operated Ca2+ entry (SOCE). In cultured human MCs, pharmacological inhibition of SOCE significantly increased the total amount of Smad1 protein. Activation of SOCE blunted high-glucose-increased Smad1 protein content. Treatment of human MCs with ANG II at 1 µM for 15 min, high glucose for 3

Protein-Anchoring Therapy of Biglycan for Mdx Mouse Model of Duchenne Muscular Dystrophy.

Duchenne muscular dystrophy (DMD) is a devastating muscle disease caused by loss-of-function mutations in DMD encoding dystrophin. No rational therapy is currently available. Utrophin is a paralog of dystrophin and is highly expressed at the neuromuscular junction. In mdx mice, utrophin is naturally upregulated throughout the muscle fibers, which mitigates muscular dystrophy. Protein-anchoring therapy was previously reported, in which a recombinant extracellular matrix (ECM) protein is delivered to and anchored to a specific target using its proprietary binding domains. Being prompted by a rep

Maternal protein restriction reduces perlecan at mid-metanephrogenesis in rats.

AIM: Maternal dietary protein restriction reduces nephron number in offspring and increases the risk of cardiovascular and chronic kidney diseases. Perlecan is the major basement membrane/extracellular matrix heparan sulfate proteoglycan (HSPG) that plays a crucial role in nephron formation. This study was to determine whether maternal dietary protein restriction during pregnancy leads to an abnormal perlecan expression pattern during kidney development and a correlation with aberrant cell proliferation and apoptosis. METHODS: Pregnant Sprague-Dawley rats were divided into two groups, maintai

Application of multiplexed ion mobility spectrometry towards the identification of host protein signatures of treatment effect in pulmonary tuberculosis.

RATIONALE: The monitoring of TB treatments in clinical practice and clinical trials relies on traditional sputum-based culture status indicators at specific time points. Accurate, predictive, blood-based protein markers would provide a simpler and more informative view of patient health and response to treatment. OBJECTIVE: We utilized sensitive, high throughput multiplexed ion mobility-mass spectrometry (IM-MS) to characterize the serum proteome of TB patients at the start of and at 8 weeks of rifamycin-based treatment. We sought to identify treatment specific signatures within patients as w

HIF2α-induced lysyl oxidase safeguards successful pregnancy by remodelling collagens at the feto-maternal interface.

During the initial stage of gestation, precise constructions of the microenvironment at the feto-maternal interface are critical for successful embryonic development to term. It consists of the fetal placenta and the maternal decidua, contributing to favorable pregnancy outcomes and long-term health in both mother and child. Remarkably, although our previous work demonstrated that physiological uterine hypoxia promotes blastocyst implantation into the decidua basalis and onset of trophoblastic invasion via the hypoxia-inducible factor (Hif) signaling pathway, it remains unclear which key regul

ECM1 protects against liver steatosis through PCBP1-mediated iron homeostasis.

BACKGROUND AND AIMS: Extracellular matrix protein 1 (ECM1) is known to inhibit transforming growth factor β signalling and HSC activation, thereby attenuating liver fibrosis. RNA-seq profiling of livers from wild-type and ECM1-deficient mice revealed different enrichments in metabolic changes in fatty acid synthesis and inflammatory pathways, suggesting a regulatory role for ECM1 in liver steatosis. Here, we studied the role of ECM1 in metabolic dysfunction-associated steatotic liver disease pathogenesis and underlying mechanisms. APPROACH AND RESULTS: Hepatic ECM1 expression was evaluated an

TSG-6-Mediated Extracellular Matrix Modifications Regulate Hypoxic-Ischemic Brain Injury.

Proteoglycans containing link domains modify the extracellular matrix (ECM) to regulate cellular homeostasis and can also sensitize tissues/organs to injury and stress. Hypoxic-ischemic (H-I) injury disrupts cellular homeostasis by activating inflammation and attenuating regeneration and repair pathways. In the brain, the main component of the ECM is the glycosaminoglycan hyaluronic acid (HA), but whether HA modifications of the ECM regulate cellular homeostasis and response to H-I injury is not known. In this report, employing both male and female mice, we demonstrate that link-domain-contain

Physiologic Reelin does not play a strong role in protection against acute stroke.

Stroke and Alzheimer's disease, two diseases that disproportionately affect the aging population, share a subset of pathological findings and risk factors. The primary genetic risk factor after age for late-onset Alzheimer's disease, ApoE4, has also been shown to increase stroke risk and the incidence of post-stroke dementia. One mechanism by which ApoE4 contributes to disease is by inducing in neurons a resistance to Reelin, a neuromodulator that enhances synaptic function. Previous studies in Reelin knockout mice suggest a role for Reelin in protection against stroke; however, these studies

Integrative Bioinformatic Identification and Molecular Docking of Quercetin and Sulforaphane-Associated Prognostic Targets in Pancreatic Adenocarcinoma.

Pancreatic adenocarcinoma (PAAD) remains a highly lethal malignancy with limited therapeutic options, motivating the search for robust prognostic markers and tractable therapeutic targets. In this study, we applied an integrative bioinformatic pipeline combining cross-cohort differential expression analysis, high-confidence protein-protein interaction network reconstruction, and topological hub-gene prioritization. Hub candidates were then intersected with curated target repertoires of multi-target chemicals (notably quercetin and sulforaphane [SFN]) to nominate pharmacologically accessible "e

Elucidating the mechanism of polycyclic aromatic hydrocarbons-induced osteoporosis through network toxicology and molecular docking approaches: BaP as a model compound.

This work showcases a network‑toxicology strategy to uncover the osteotoxic effects and molecular pathways of polycyclic aromatic hydrocarbons, using BaP‑induced osteoporosis as a model. Potential targets of BaP were extracted from the Comparative Toxicogenomics Database (CTD) and ChEMBL, while osteoporosis-related targets were obtained from GeneCards. A total of 93 overlapping genes between osteoporosis-related targets and BaP-perturbed genes were identified. Subsequent protein-protein interaction (PPI) network analysis was constructed using STRING and visualized with Cytoscape. Seven core ge

Pharmacological insights into fluvoxamine maleate: σ1 receptor agonism, multimodal mechanisms, and therapeutic repositioning.

Fluvoxamine maleate (FM) is a selective serotonin reuptake inhibitor (SSRI) that has attracted increased attention owing to its strong agonistic activity at the sigma 1 receptor (σ1R). This review consolidates the current understanding of its processes highlighting σ1R-mediated chaperone activity in the endoplasmic reticulum (ER), which alleviates protein misfolding, ER stress, and neuroinflammation in mental and neurological illnesses. Fluvoxamine demonstrates one of the highest affinities for σ1R among SSRIs, exceeding those of sertraline and fluoxetine, via dissociating σ1R from BiP chapero

Segment-Specific Functional Responses of Swine Intestine to Time-Restricted Feeding Regime.

Research indicates that TRF improves mammalian metabolism and health via the microbiota-gut-brain axis. Previous studies showed that TRF promotes pig growth, but the intestinal mechanisms remain unclear. This study explored the impact of TRF on pig intestinal functions. Twelve male pigs were split into ad libitum feeding (FA) and TRF groups. FA pigs had free access to feed, whereas TRF pigs were fed during 07:00-08:00, 12:00-13:00, and 17:00-18:00. TRF enhanced crude protein digestibility by 18.9% (p = 0.045) and increased pancreatic chymotrypsin and lipase activities, while reducing ileal amy

Periostin drives rheumatoid arthritis progression by regulating integrin αvβ3-mediated transforming growth factor-β1/SMAD signaling.

Periostin (POSTN), a non-structural extracellular matrix protein, plays a critical role in promoting tumor cell invasion, metastasis, and disease progression. We previously reported that POSTN mRNA and protein levels are markedly upregulated in the synovium of patients with rheumatoid arthritis (RA); however, its specific functions and underlying mechanisms remain unclear. In this study, we investigated the roles of POSTN in fibroblast-like synoviocytes (FLSs) and collagen-induced arthritis (CIA) model mice. POSTN mRNA and protein levels were markedly upregulated in primary RA-FLSs and closely
