Spiders can maintain a wide range of free-running periods while still being entrained to a 24-hour day. To investigate the underlying mechanism of this entrainment, we constructed the phase-response curve (PRC) for the orb weaver, Metazygia wittfeldae, by subjecting the spiders to one-hour light pulses at various times throughout the circadian day. The resulting type 0 PRC showed high amplitude (> 6 hour) phase advance and delays when the light pulse was applied during circadian time (CT) 16-18, including a break point of delays to advances. We then investigated the genetic mechanism of respon

Plant-derived lectins originating from plants perform essential functions in both plant growth and stress response processes. Rice jacalin-related mannose-binding lectin 1 (OsMBL1), a well-characterized canonical salt-responsive regulatory factor, acts as a core positive regulator that confers enhanced salt tolerance. In the present study, OsMBL1 was found to respond to multiple environmental stresses and signaling molecules, and it not only positively regulated salt tolerance at the seedling stage but also at the seed germination stage. To dissect the underlying regulatory network, we perform

Adeno-associated virus (AAV)-based gene therapies have garnered significant attention and investment due to their clinical success and potential to address underlying causes of many diseases. AAV vectors provide effective delivery of therapeutic genetic material to disease-relevant tissues. When evaluating safety and efficacy of recombinant AAV vectors, biodistribution profiles play a critical role in novel therapy development. Herein, a biodistribution metadata analysis was performed on ten studies involving 51 cynomolgus macaques (Macaca fascicularis). The macaques received a self-complement

A logical discovery method based on metabologenomic targeting was designed to search for new β-branched conjugated polyketides. PCR screening of a bacterial DNA library (1,638 strains) targeting the key enzyme hydroxymethylglutaryl-CoA synthase (HCS) homolog-encoding gene as a genomic hallmark identified 67 hit strains. Phylogenetic analysis of HCS homolog-encoding gene amplicons prioritized strains potentially producing new β-branched conjugated polyketides. Vibronic fine structures in ultraviolet spectra as a metabolomic hallmark clearly identified β-branched conjugated polyketides in bacter

Although copy number variants (CNVs) represent well-established genetic contributors to schizophrenia (SCZ), their role in bipolar disorder (BD), especially within non-European ancestries, has been inadequately explored. We evaluated the genome-wide load of rare CNVs, encompassing deletions and duplications, in a Han Chinese sample of 3915 BD cases and 7820 ethnically matched controls. We observed a marked overrepresentation of rare deletions in BD patients relative to controls, with affected genes showing enrichment in neural signaling and dosage-dependent networks, indicating that haploinsuf

Aspergillus fumigatus, a saprophytic fungus and the principal etiologic agent of aspergillosis, is increasingly implicated in drug-resistant infections, posing challenges to clinical management. RTA-like proteins, a distinctive class of transmembrane fungal proteins, are involved in several stress responses. Nonetheless, their role in fungal azole resistance is yet to be determined. Transcriptomic studies performed in this investigation revealed that azole stimulation significantly upregulates the expression of RTA-like proteins in A. fumigatus. Phenotypic analyses indicated that relative to t

Late leaf spot (LLS) is the most widespread and destructive fungal disease affecting peanut worldwide, yet relatively little is known about its genetic basis. In this study, QTL mapping using a 1327-bin map (derived from 9533 SNP/InDel markers) and multi-environment phenotypic data repeatedly detected two major LLS-resistance QTLs on chromosomes A02 and B06 in the Zhonghua 5 × ICGV 86699 recombinant inbred line (RIL) population. QTL-seq analysis confirmed qLLSA02, and integration of both mapping methods narrowed the A02 interval to a 1.0-3.4 Mb region. Transcriptome profiling identified thousa

Microtia is a common congenital craniofacial malformation characterized by the partial or complete absence of the external ear structure. Despite its relatively high incidence, the pathogenesis of microtia remain poorly understood. In this study, we analyzed both single-cell and bulk RNA sequencing data from microtia cases and identified a population of COL1 + HES1+mesenchymal stem cell in perichondrium with significantly higher expression of the CRABP2 gene, a gene that encodes a nuclear transporter of retinoic acid. Gene expression analysis further confirmed that the RA signaling intensity a

Skin pigmentation in teleosts is a complex process governed by molecular and cellular mechanisms during development. In this study, we investigated the pigmentation process in the tomato clownfish (Amphiprion frenatus) using an integrated approach combining histological analysis and transcriptomics (RNA-Seq and Ribo-Seq). Histological examination revealed the stepwise differentiation of chromatophores during post-embryonic development. Transcriptomic profiling at 14 days post-hatching (dph) identified extensive changes in transcript abundance, with 2063 of 2667 differentially expressed unigene

α-linolenic acid (ALA) has been reported to benefit testicular function. However, dietary ALA is extensively metabolized in the rumen, whereas isolated cell models do not retain seminiferous tubule structure or interactions among testicular cells. This pilot study examined the effects of ALA on seminiferous tubule morphology, UCHL1-positive cells, and gene expression in cultured sheep testicular tissue fragments. Tissue from one 3-month-old Hu ram (n = 1) was cultured for 50 d in control medium (CK) or medium containing 5 μM ALA. Three independently cultured fragments per condition were analyz

BACKGROUND: Long-chain non-coding RNAs (lncRNAs) play various roles in the regulation of gene expression at the levels of transcription and translation, and epigenetic modification. Dysregulation of lncRNAs is associated with various malignancies, including cancer. lncRNAs have been demonstrated to regulate critical biological processes in cancer cells, such as apoptosis, proliferation, migration, and invasion. They also play essential roles in the development of gastric cancer (GC). However, the clinical significance and biological function of many lncRNAs remain unexplored in GC progression.

The replication and mortality caused by the viral hemorrhagic septicemia virus (VHSV) in fish vary depending on temperature. VHSV causes mortality at the temperatures below 15 °C, while infection is not established in olive flounder at temperatures above 25 °C. However, how VHSV infection manifests at the cellular level under different temperature conditions is not understood. In this study, we aimed to elucidate the mechanism by which VHSV infection is controlled by comparing viral replication and immune responses in vitro and in vivo. In the in vitro experiment, viral mRNA levels on day 5 po

Patient and caregiver treatment preferences should be incorporated into the drug development process. We updated a 2018 survey to reflect current knowledge about gene therapy for Duchenne muscular dystrophy (DMD) and obtained new data in the U.S., U.K., and other countries. At data collection in 2023, there were no approved gene therapies for DMD. Using the threshold technique, we assessed the maximum acceptable risk of death (MAR) from non-curative gene therapy with time-limited benefit at different stages of DMD progression. Among 263 participants, mean MAR ranged from 2.4 % (treatment in ne

The treatment of infections by Acinetobacter baumannii, a clinically significant nosocomial gram-negative bacterial pathogen, is hampered by antibiotic resistance, which is exacerbated by its exceptional genetic plasticity. Knowledge of the dynamics and mechanisms underlying the acquisition of antibiotic resistance is essential for the proper stewardship of their utilization. Here, we used a continuous culture device (morbidostat) to characterize the evolutionary trajectories of two A. baumannii strains in response to the increasing pressure of last-resort drugs, tigecycline and colistin. Background/Objectives: Melanoma is an aggressive skin cancer influenced by genetic and immunological factors. The PDCD1 gene encodes PD-1, a receptor involved in immune evasion and therapeutic response. This study aimed to evaluate the association of PDCD1 variants (rs2227982, rs36084323, rs7421861) and its relative gene expression with melanoma in a Mexican population. Methods: An analytical cross-sectional study was conducted with 262 samples: 131 from melanoma patients (newly diagnosed and treatment-naïve) and 131 from cancer-free controls. Genotyping was performed using real-time PCR. PDCD

RNA hairpins may constitute a foundation of genetic evolution both in viruses and other organisms. Stem-loops theoretically comprise a stable part, the double-stranded stem, and a single-stranded loop allowing evolution. Here we tested for SARS-CoV-2 if "fertile" mutations were in loops while mutations in stems were poorly tolerated and rarely found in consensus genomes. We combined information on the frequencies of mutations, either "fertile" (present in ≥ 50 genomes) or "non-fertile" (neutral, weakly deleterious or lethal) in 61,397 SARS-CoV-2 genomes, and on whether these mutations occurred

Regions of sun-exposed skin are prone to carcinogenesis in geriatric individuals and exhibit a reduced ability to remove mutagenic UV photoproducts from DNA. To determine whether geriatric skin may be more reliant on translesion synthesis (TLS) DNA polymerases to replicate unrepaired UV photoproducts, we examined the expression of TLS polymerases in the epidermis of young and geriatric individuals. Although significant differences were not observed between these 2 age groups, the expression of the TLS polymerase genes POLH, POLI, POLK, REV1, and REV7 were found to be inversely correlated with
