Synthesis and Evaluation of Antibacterial Activity of Bottromycins.

Total synthesis of bottromycin A2 can be accomplished through a diastereoselective Mannich reaction of a chiral sulfinamide, mercury-mediated intermolecular amidination, and cyclization of a constrained tetracyclic peptide. Exploitation of this process allowed the synthesis of several novel bottromycin analogs. The antimicrobial activity of these analogs was evaluated in vitro against Gram-positive bacteria, such as methicillin resistant Staphylococcus aureus (MRSA) and vancomycin resistant enterococci (VRE). Structure-activity relationships were explored taking into consideration the unique t…

Strange case of dimorphic skin rash in a patient with cirrhosis: atypical herpes simplex and sweet's syndrome.

A middle-aged man with decompensated cirrhosis and a dimorphic multisite skin rash is diagnosed with rare atypical herpes simplex infection, manifesting Sweet's syndrome (SS) in the absence of other described associations. SS, an acute febrile neutrophilic dermatosis, has three common forms-classical or idiopathic, malignancy associated and drug induced. Systemic autoimmune, connective tissue diseases and infections are also strong associations. The latter is commonly described in Gram-positive bacteria, salmonellosis and Yersinia Herpes infections are a rare cause of SS, reported only thrice …

Persistence of a Cervical Neck Mass, Not Just the Lymphoma.

Actinomycosis is a rare, chronic granulomatous infection caused by gram-positive, anaerobic to microaerophilic branching filamentous bacteria. In the cervicofacial region, it usually presents as an enlarging neck mass. It remains a diagnostic challenge due to the fact that cultures show no growth in more than 50% of cases. We report a case of a 67-year-old patient known to have a neck mass secondary to lymphoma in which the neck mass persisted despite therapy. Upon evaluation, the diagnosis of culture-negative actinomycosis was based on histopathology findings, and the patient received antibio

First genetic detection of Coxiella burnetii in Zambian livestock.

Q fever is a widespread zoonosis caused by Coxiella burnetii, an obligate intracellular gram-negative bacterium. The investigation of C. burnetii infection in Zambian livestock was carried out using molecular detection techniques. A total of 489 cattle and 53 goat blood samples were collected from Chama, Chongwe, Monze, and Petauke districts in Zambia. Molecular screening by polymerase chain reaction was performed using C. burnetii-species-specific primers. In total, 38 cattle and 4 goat samples were positive. The prevalence of C. burnetii differed among the four sites, with Chama (Eastern pro

A case report of anaphylaxis to chlorhexidine during urinary catheterisation.

Chlorhexidine gluconate is a chemical antiseptic that is effective against Gram positive and negative bacteria and on certain viruses and fungi. A bacteriocidal and bacteriostatic agent, this cationic drug is absorbed on to negatively charged cell surfaces of organisms, disrupting the cell membrane, which results in increased permeability. Its use is mainly topical as a surgical hand antisepsis, site preparation/cleansing and for pre-genitourinary procedures such as urethral catheterisation. Like any drug, caution should be employed with its use as hypersensitivity reactions are being document

Acute progressive mitral regurgitation resulting from chordal rupture in infants.

This report describes six previously healthy infants with acute mitral valve chordal rupture. Although their initial symptoms were mild respiratory distress and fever, all the infants presented with acute progressive left heart failure and required urgent surgical repair. Mitral valve annuloplasty and mitral valve replacement were performed (for three patients each). Two patients died, both of whom had cardiac arrest and required resuscitation before surgery. Histologic examination of excised mitral valve leaflets showed infiltration of gram-positive bacteria in these two patients. All the pat…

Isolation of Shewanella algae from rectal swabs of patients with bloody diarrhoea.

Shewanella algae is an emerging bacteria rarely implicated as a human pathogen. It was infrequently recovered from clinical specimens probably because of inadequate processing of non-fermenting oxidase-positive gram-negative bacilli. We report here isolation of S. algae in pure culture and mixed with E. coli from two cases of acute gastroenteritis with bloody mucous containing diarrhea occurring at the same time. As this organism is not a normal flora of the gut, the possible source of infection may be fish contaminated with the organism. Whether this bacterium can be considered an enteric pat

Bacillus anthracis secretes proteins that mediate heme acquisition from hemoglobin.

Acquisition of iron is necessary for the replication of nearly all bacterial pathogens; however, iron of vertebrate hosts is mostly sequestered by heme and bound to hemoglobin within red blood cells. In Bacillus anthracis, the spore-forming agent of anthrax, the mechanisms of iron scavenging from hemoglobin are unknown. We report here that B. anthracis secretes IsdX1 and IsdX2, two NEAT domain proteins, to remove heme from hemoglobin, thereby retrieving iron for bacterial growth. Unlike other Gram-positive bacteria, which rely on cell wall anchored Isd proteins for heme scavenging, B. anthraci…

Nocardia tenerifensis genome identification in a cutaneous granuloma of a cat.

An adult Domestic Longhair cat developed a subcutaneous mass in its tail. Histologically, this mass consisted of ill-defined pyogranulomas centered around aggregates of gram-positive, acid-fast filamentous bacteria, consistent with Nocardia. Due to the lack of fresh samples, DNA was extracted from formalin-fixed paraffin-embedded tissue sections and subjected to polymerase chain reaction amplification and DNA sequencing of 16S ribosomal RNA gene encompassing Nocardia sp.-specific sequences. Sequences analyzed using the GenBank database revealed 99.5% homology with Nocardia spp. and had the hig

Listeriology (1926-2007): the rise of a model pathogen.

Listeria monocytogenes is an ubiquitous gram positive bacterium responsible for a severe food borne disease in human and animals which has become a multifaceted model. Indeed, it is a facultative intracellular bacterium that induces an amazingly rapid and sterilizing T-cell response and has been--and still is--widely used by immunologists. Listeria's strategies to invade non-phagocytic cells and spread from cell to cell have been and still are outstandingly instrumental to address key questions in cell biology. Orally acquired listeriosis can now be studied in relevant animal models. Finally,

Actinomycosis of the temporal bone: a report of a case.

Actinomycosis is a chronic suppurative infection of the cervicofacial region caused by Actinomyces species, which are anaerobic, gram-positive filamentous bacteria. Although actinomycosis has a propensity for involving the oral cavity, rare cases of actinomycosis involving the temporal bone have been published. We report the case of a 14-year-old girl who presented with clinical, audiometric, and radiologic findings consistent with right chronic suppurative otitis media that persisted despite tympanomastoidectomy. Altering tissue tropism of Listeria monocytogenes by ectopically expressing human E-cadherin in transgenic mice.

The Gram-positive bacterium Listeria monocytogenes invades a variety of cells in vitro and in vivo. Here, we show that L. monocytogenes is able to enter muscle cells if provided with the appropriate host receptor. Using a cardiac-specific promoter, human E-cadherin was misexpressed in the heart of transgenic mice. First, we demonstrate that L. monocytogenes is capable of infecting the E-cadherin-expressing, but not the N-cadherin-expressing cardiomyocytes in vitro. Second, we show that L. monocytogenes infects the heart of transgenic animals after administering the bacteria intravenously. This

Diagnosis by polymerase chain reaction of Erysipelas septicemia in a flock of ring-necked pheasants.

A flock of 810 pheasants experienced 6.2% mortality over 6 days. Affected birds were weak and lethargic for up to 24 hr before death. Examined birds were thin, and gross lesions consisted of thick opaque crops and cecal cores. Histologically, there was capillariasis of the crop and multifocal ulcerative typhlitis with Heterakis spp. infection, and numerous systemic intravascular monocytes were filled with clusters of blue rod-shaped organisms. The organisms were gram-positive bacilli by Brown and Brenn staining and ultrastructural analysis. Liver bacterial cultures were negative for pathogenic

A Staphylococcus aureus plasmid that specifies constitutive macrolide-lincosamide-streptogramin B resistance contains a novel deletion in the ermC attenuator.

By restriction enzyme anal … id pA22 is different from that found in similar 2.5kb constitutive MLS-resistance plasmids in other Gram-positive bacteria. It is suggested that plasmids conferring the constitutive phenotype have evolved from an inducible ancestor on several independent occasions.

Microbiology and pathogenesis of acute salpingitis as determined by laparoscopy: what is the appropriate site to sample?

Acute salpingitis is a polymicrobial disease. Neisseria gonorrhoeae and anaerobic gram-positive cocci were the predominant microorganisms isolated from the fallopian tubes of salpingitis patients. Gonococci were isolated from the fallopian tubes in eight of 35 (23%) patients; anaerobic bacteria were recovered from 10 of 35 (28.5%). Although Chlamydia trachomatis was not recovered from the fallopian tube exudate, there was abundant serologic evidence of chlamydial infection in the salpingitis patients. Twenty-three percent of patients with paired sera had a fourfold rise in IgM and IgG titer, w

The Microbiology of Phane, an Edible Caterpillar of the Emperor Moth, Imbrasia belina.

The larvae of Imbrasia belina (Westwood) are cooked and sun dried to make a product known as phane, which is consumed as a delicacy. A study was conducted to determine the sanitary quality of phane and the kinds of microorganisms associated with it. It also looked into the potential for the existence of health risk associated with its consumption. Laboratory- and field-processed phane and that from open markets were subjected to microbiological analyses. The total microbial population for the larvae was in the range of 3 × 105 to 2 × 107 CFU/g. Species belonging to seven genera of bacteria and

A comparison of plate assay methods for detecting extracellular cellulase and xylanase activity.

Identification of microorganisms for the production of carbohydrolytic enzymes is extremely important given the increased demand for these enzymes in many industries. To this end, dye-polysaccharide interactions which provide a visual indication of polymer hydrolysis (clear zones or halos) have been used for decades. For the detection of extracellular cellulase or xylanase activity many laboratories use Gram's iodine as the chromogenic dye, as it is a more rapid initial screening method compared to the use of other dyes. Here, we compared Gram's iodine and Congo red as indicators of polysaccha
