Sustained Delivery of a Shingles Subunit Vaccine Overcomes Age-Related Declines in Humoral and Cellular Immunity Relative to Shingrix.

The global aging population faces heightened vulnerability to infectious diseases due to immunosenescence, which diminishes the potency, durability, and breadth of vaccine-induced immunity. While the leading shingles vaccine, Shingrix®, provides protection against herpes zoster virus for adults aged 50 and older, it is often associated with severe local and systemic reactogenicity, which limits vaccine compliance. Here, we report an injectable polymer-nanoparticle (PNP) hydrogel platform for the sustained delivery of a shingles subunit vaccine to enhance immune responses while mitigating react

Sirolimus is effective and safe in childhood relapsed-refractory autoimmune cytopenias: A multicentre study.

Autoimmune cytopenias are a heterogeneous group of disorders characterized by immune-mediated destruction of haematopoietic cell lines. Effective and well-tolerated treatment options for relapsed-refractory immune cytopenias are limited. In this study, the aim was to evaluate the efficacy and safety of sirolimus in this disease group within the paediatric age group. The study enrolled patients in the paediatric age group who used sirolimus with a diagnosis of immune cytopenia between December 2010 and December 2020, followed at six centres in Turkey. Of the 17 patients, five (29.4%) were treat

Factoring the intestinal microbiome into the pathogenesis of autoimmune hepatitis.

The intestinal microbiome is a reservoir of microbial antigens and activated immune cells. The aims of this review were to describe the role of the intestinal microbiome in generating innate and adaptive immune responses, indicate how these responses contribute to the development of systemic immune-mediated diseases, and encourage investigations that improve the understanding and management of autoimmune hepatitis. Alterations in the composition of the intestinal microflora (dysbiosis) can disrupt intestinal and systemic immune tolerances for commensal bacteria. Toll-like receptors within the …

Protein kinase G-a key regulator of pathogenesis in Mycobacterium tuberculosis infection.

Mycobacterium tuberculosis (M. tuberculosis), the causative agent of tuberculosis (TB), remains a leading global health threat, exacerbated by rising drug resistance and the ability of this pathogen to persist within host macrophages. Central to the intracellular survival of M. tuberculosis is Protein Kinase G (PknG), a secreted, eukaryotic-like serine/threonine kinase that subverts host immune defenses and modulates bacterial physiology. This review provides a comprehensive overview of structural features and the multifaceted role of PknG in M. tuberculosis pathogenesis, including inhibition

Pathogenic autoantibody production requires loss of tolerance against desmoglein 3 in both T and B cells in experimental pemphigus vulgaris.

Mechanisms of tolerance break against desmoglein 3 (Dsg3) in patients with pemphigus vulgaris (PV) producing pathogenic anti-Dsg3 IgG autoantibodies are unclear. In this study, using a novel PV mouse model involving Dsg3 knockout mice, we investigated the mechanisms leading to production of autoantibodies against Dsg3. Adoptive transfer of Dsg3(-/-) splenocytes immunized with recombinant mouse Dsg3 to Rag2(-/-) recipient mice expressing Dsg3 resulted in the stable production of anti-Dsg3 IgG and development of PV phenotypes including oral erosions with suprabasilar acantholysis. When purified

Integrated transcriptomics and histological analysis reveal gill adaptation mechanisms to hypoxic stress in largemouth bass (Micropterus salmoides).

Largemouth bass (Micropterus salmoides) is a globally significant freshwater aquaculture species known for its rapid growth, high-quality flesh and excellent nutritional value. Given the increasing incidence of hypoxia in its aquaculture setting, understanding the molecular mechanisms underlying gill adaptation to hypoxic stress is critical for improving resilience in this species. To achieve this, we employed full-length transcript sequencing using PacBio Iso-Seq and high-throughput Illumina RNA-Seq to construct a comprehensive transcriptome profile. Histological analyses revealed apparent st

Restoring Microbial Signaling: A Metabolite-Immune-Redox Framework for Postbiotic Host-Directed Interventions.

Background/Objectives: Postbiotics are increasingly recognized as biologically active products of microorganisms with emerging potential as microbiome-inspired therapeutic interventions. While most microbiome-based strategies focus on modifying microbial composition, restoration of microbial signaling has received comparatively less attention. This review examines postbiotics through the lens of microbial signaling restoration and proposes a unified Metabolite-Immune-Redox (MIR) axis linking microbial-derived signals with immune regulation, redox homeostasis, endothelial integrity, and host re

Dietary vitamin E alleviates ammonia-induced effects on survival, antioxidant and immune response of post-larval Pacific white shrimp.

It remains unknown whether vitamin E (VE), a fat-soluble vitamin known for its potent antioxidant effects, alleviates the stress caused by ammonia-N exposure in post-larval Pacific white shrimp (Penaeus vannamei). Five isonitrogenous (crude protein: 32.5%) and isolipidic (crude lipid: 9.22%) diets were formulated containing DL-α-tocopheryl acetate at concentrations of 0, 40, 80, 120 and 160 mg/kg (E13.1, E50.6, E90.5, E130.6 and E163.9, respectively). Post-larval P. vannamei individuals (3.95 ± 0.21 mg) were randomly assigned to 20 tanks and fed the experimental diets for 34 days. After the fe

Determinants of response to neoadjuvant chemotherapy in breast cancer: Integrated roles of immune pathways, DNA damage response, and cell cycle regulators.

Breast cancer (BC) is the most commonly diagnosed malignant disease in women worldwide. Resistance to neoadjuvant chemotherapy (NAC) still limits treatment efficacy despite the enormous progress in systemic therapy. Because pathological complete response (pCR) is strongly associated with favorable clinical outcomes, especially in triple-negative and HER2-positive breast cancer, the discovery of reliable predictive biomarkers before treatment has become a key clinical goal. This narrative review summarizes the available evidence of genetic and transcriptomic biomarkers associated to responses t

Multi-tissue transcriptomics reveals age-dependent susceptibility to grass carp reovirus linked to metabolic alterations and extracellular matrix disruption in grass carp.

Grass carp (Ctenopharyngodon idella) is an important freshwater aquaculture species. Infection by grass carp reovirus (GCRV) can cause hemorrhagic disease and lead to major economic losses in aquaculture. Susceptibility to GCRV is strongly age dependent: juveniles (≤1 year old) are highly vulnerable, whereas fish aged three years or older show much greater tolerance. In this study, transcriptome sequencing was used to compare gene expression in the gill, intestine, head kidney, muscle, and spleen of three-year-old and six-month-old grass carp at pre-infection (Day 0) and at Days 4 and 7 post-i

Genistein in autoimmune diseases: from experimental evidence to emerging clinical data.

Autoimmune diseases are chronic inflammatory disorders characterized by loss of immune tolerance, activation of autoreactive immune cells, and progressive tissue injury. Despite advances in immunosuppressive and biologic therapies, treatment responses remain heterogeneous and long-term therapy is frequently associated with adverse effects and economic burden. These limitations have stimulated increasing interest in complementary immunomodulatory strategies, including naturally occurring bioactive compounds. Genistein, a soy-derived isoflavone belonging to the phytoestrogen family, has emerged …

Beyond rejection: engineering immune responses to redefine skin graft survival.

Skin grafting remains a fundamental reconstructive technique in reconstructive and dermatologic surgery for restoring skin integrity following burns, trauma, chronic ulcers, and excision of malignancies. However, a graft survival rate is determined by multiple factors and by a delicate balance between inflammatory and reparative immune system processes. Controlled inflammation promotes wound healing while excessive or prolonged immune activation has the potential to result in cytotoxicity and complement activation which will eventually lead to graft rejection. Early rejection is driven by anti
