Chronic inflammation drives the pathogenesis of diverse disorders, yet current anti-inflammatory strategies often lack mechanistic selectivity. Glycyrrhizic acid (GL), a major triterpenoid saponin from Glycyrrhiza species, exhibits broad anti-inflammatory effects, but its mechanism has been viewed as non-specific. In this review, we propose an integrated framework positioning high-mobility group box 1 (HMGB1) as one major molecular hub for GL's action. Mechanistically, our molecular docking analysis suggests that GL may interact with the Box B domain of HMGB1 andmay potentially interfere with

Rather than viewing classical neurodevelopmental disorders (NDDs) such as autism spectrum disorder (ASD) and severe psychiatric illnesses like schizophrenia (SCZ) as isolated diagnostic silos, modern biological psychiatry increasingly conceptualizes them along a shared neurodevelopmental continuum. Although altered synaptic connectivity and sustained immune dysregulation are both well-documented pathological features, the mechanistic crosstalk between them remains incompletely understood. In this review, we propose that microglia occupy a central position at the interface of these processes be

PANoptosis is a complex inflammatory programmed cell death pathway that integrates the key mechanistic features of pyroptosis, apoptosis, and necroptosis. It represents a crucial innate immune mechanism for eliminating infected or damaged cells; however, its dysregulation is implicated in the pathogenesis of a wide spectrum of diseases, including sepsis, myocardial infarction, acute organ injuries, and cancer. Consequently, targeting PANoptosis has garnered significant interest as a novel therapeutic strategy. This review explored the potent modulatory effects of herbal medicines and their bio

Coronaviruses (CoVs) pose serious threats to both animal and human health, underscoring the urgent need for broad-spectrum antiviral strategies that can provide rapid and immune-independent protection beyond vaccination. In this study, we developed zeolitic imidazolate framework-8 (ZIF8) as an innovative zinc-based anti-coronavirus nanoagent and revealed that ZIF8 exhibits markedly enhanced antiviral activity relative to the commonly used zinc oxide (ZnO). Using porcine epidemic diarrhea virus (PEDV) as a model α-coronavirus, mechanistic studies demonstrated that ZIF8 alleviates excessive reac

Asthma is a chronic respiratory disease characterized by airway inflammation, hyperresponsiveness, and remodeling. Its pathogenesis is mainly related to immune imbalance, including Th1/Th2 and Th17/Treg imbalance, ILC2 activation and macrophage polarization, all of these contribute to airway inflammation and epithelial damage. Existing glucocorticosteroids and biologics have limited efficacy in non-Th2 or steroid-resistant patients, therefore new immunomodulatory strategies are urgently needed. Mesenchymal stem cell-derived exosomes (MSC-Exos), especially human umbilical cord MSC-Exos (HUCMSC-

BACKGROUND: Ulcerative colitis (UC) is a chronic inflammatory bowel disease characterized by dysregulated immune responses in the gut. Macrophages, as key innate immune cells in the intestinal mucosa, play a central role in both maintaining homeostasis and driving pathology in UC.
SUMMARY: Under physiological conditions, intestinal macrophages exhibit a unique "inflammatory anergy" phenotype, supporting epithelial integrity and immune tolerance. In UC, however, persistent inflammatory signals promote monocyte recruitment and their polarization into pro-inflammatory M1-like macrophages. These

Ulcerative colitis (UC) is a chronic and idiopathic inflammatory bowel disease with limited therapeutic options, high relapse rates, and potential adverse effects. Although natural polysaccharides have shown promise in alleviating colitis-associated inflammation, the impact of structural modification on their bioactivity and underlying mechanisms remains insufficiently explored. In this study, the pharmacological activities and mechanisms of phosphorylated Cyclocarya paliurus polysaccharides (P-CPPs) were systematically investigated in a dextran sulfate sodium (DSS)-induced murine colitis mode

BACKGROUND: Inflammatory diseases represent a major global health burden, and current therapies frequently fail to achieve sustained control of inflammation or prevent progressive tissue damage. Cortistatin (CST), an endogenous neuropeptide structurally related to somatostatin, has emerged as a multifunctional regulator of neuroimmune and immunometabolic pathways.
METHODS: This scoping review was conducted in accordance with PRISMA-ScR guidelines and maps and synthesizes preclinical evidence on the biological roles and therapeutic potential of CST across experimental disease models.

Extracellular ATP is a central danger signal linking cellular stress to neuroinflammation, yet the mechanisms governing the transition from physiological purinergic signaling to pathological neuroimmune activation remain incompletely understood. In this review, we integrate current evidence into a conceptual framework describing how ATP-dependent purinergic signaling may contribute to the shift from adaptive neuron-glia communication to maladaptive glial crosstalk and inflammatory amplification. Under physiological conditions, controlled ATP turnover primarily engages lower-threshold purinergi

The neonicotinoid insecticide, imidacloprid (IMI), is one of the widely used pesticides with well-documented serious health effects that are noticeable with long-term exposure. However, the long-term effects of IMI on cardiac tissues have not been fully elucidated. Herein, we investigated the mechanisms of IMI-induced cardiotoxicity. Additionally, we examined the potential protective effects of the natural alkaloid, berberine (BBR), against IMI-induced cardiotoxicity. Rats received IMI (45 mg/kg/day, orally) for 30 days, alone or in combination with BBR-loaded liposomes (BBR-Lip) at a dose of

Immune cells possess an intrinsic covalent signalling system in which endogenous electrophilic species function as molecular regulators of cellular state that integrate metabolic activity, oxidative stress, and inflammatory signals. Lipid peroxidation-derived electrophiles, enzymatically generated electrophilic lipid mediators, nitro-fatty acids, and metabolite-derived electrophiles selectively modify reactive amino acid residues within signalling proteins to regulate immune cell differentiation, metabolic adaptation, stress responses, and tissue responses. This perspective highlights that end

Fine particulate matter (PM, diameter < 2.5 μm) is a significant public health concern because it can disrupt the bone marrow (BM) microenvironment through oxidative stress and systemic inflammation. Patients with type 2 diabetes mellitus (T2DM) exhibit heightened vulnerability to environmental stressors such as PM2.5, which exacerbates metabolic dysfunction. However, the effects of PM2.5 on BM resident stem cells in the context of diabetes have not been explored. In this study, we investigated the impact of atmospherically relevant PM2.5 exposure on BM-resident stem cells in T2DM mice. Diabet

Acute myocardial infarction (AMI) remains a major cause of morbidity and mortality globally, with percutaneous coronary intervention (PCI) serving as the primary treatment for revascularization of culprit vessels. However, non-culprit vessels (NCVs), which harbor significant atherosclerosis but are not directly responsible for acute ischemic events, have gained increasing attention owing to their role in future cardiovascular risks. Recent studies have highlighted the rapid progression of post-PCI, driven by factors such as inflammation, endothelial dysfunction, and hemodynamic changes. This p

Familial Mediterranean Fever (FMF) is an inherited autoinflammatory disease triggered by Mediterranean Fever (MEFV) gene mutations that lead to spontaneous pyrin inflammasome activation and exaggerated pro-inflammatory cytokine secretion. The V726A knock-in (KI) mouse model of FMF mimics most of the clinical and immunologic manifestations of the disease, such as recurrent febrile attacks and systemic inflammation. Here, we explored the proof-of-concept evaluation of miR-197-3p-loaded microvesicles (MVs) as a therapeutic approach in the FMF KI mouse model. MVs were purified and structurally cha

Secondary damage in remote brain regions following ischemic stroke significantly worsens patient outcomes, yet its underlying mechanisms remain poorly understood. Microglial activation is a central pathological feature of secondary damage, with the P2X7 receptor (P2X7R) emerging as a key regulator of neuroinflammatory processes. In this study, we employed a distal middle cerebral artery occlusion (dMCAO) model in rats to investigate the role of P2X7R in secondary damage in the ventral posterolateral nucleus (VPN) of the ipsilateral thalamus. We observed a spatiotemporal pattern of microglial a

Leucine-rich repeat kinase 2 (LRRK2) is a multidomain serine/threonine kinase and a major genetic contributor to Parkinson's disease (PD). Although LRRK2 has been extensively studied in neurodegeneration, emerging evidence indicates that it also plays a critical role in systemic metabolism. LRRK2 regulates glucose homeostasis through modulation of insulin signaling, vesicle trafficking, mitochondrial function, and inflammatory responses. Studies using LRRK2 knockout and knock-in models, including the pathogenic G2019S mutation, have revealed abnormalities in insulin sensitivity, adipose tissue

NOD-like receptor family pyrin domain-containing 6 (Nlrp6) is an inflammasome related molecule expressed in intestinal epithelial and immune cells. While previous work showed that Nlrp6 in host nonhematopoietic cells exacerbates gastrointestinal acute graft-versus-host disease (aGVHD), its role in donor T cells remained unclear. Here we show that donor T cell-intrinsic Nlrp6 exerts a protective effect in murine aGVHD models. Mice receiving Nlrp6 deficient (Nlrp6-/-) donor T cells had reduced survival and more severe GVHD than those receiving wild-type T cells. Mechanistically, Nlrp6-/- T cells

Neurodegenerative diseases (NDs) are marked by selective neuronal vulnerability and progressive failure of neural circuits. Increasing evidence indicates that neuronal loss is not driven by a single terminal event, but emerges from interacting regulated cell death (RCD) programs. These programs are closely coupled to mitochondrial injury, proteostatic collapse, lysosomal stress, metabolic imbalance, glial state transitions, and chronic neuroinflammation. Yet, how distinct death pathways are organized across cell types, disease stages, and disease-specific microenvironments remains unresolved.

Alzheimer's disease (AD), a prevalent neurodegenerative disorder characterized by cognitive impairment and neuronal degeneration, is increasingly recognized as being driven not only by the traditional amyloid-beta and tau pathologies but also by persistent neuroinflammation and systemic immune dysregulation. Emerging evidence implicates microglia senescence and gut microbiota dysbiosis is critical contributors to the neuroinflammatory landscape. Senescent microglia marked by reduced phagocytic ability and a pro-inflammatory secretory profile, are unable to clear pathogenic stimuli, thereby int
