The present study aimed to test the effects of melatonin on oxidative stress in the yellowtail clownfish, Amphiprion clarkii, as produced by light emitting diodes (LEDs): red, green, and blue. We investigated the effects of the different LEDs on oxidative stress by measuring the mRNA expression of arylalkylamine N-acetyltransferase (AANAT2), the expression and activities of antioxidant enzymes (superoxide dismutase, SOD (EC 1.15.1.1); and catalase, CAT (EC 1.11.1.6)), and plasma H2O2 and plasma melatonin levels. In red light, the expression of AANAT2, SOD, and CAT mRNA was significantly higher

Although tubular necrosis in acute renal failure is associated with excessive production of reactive oxygen species (ROS), such as hydrogen peroxide (H2O2), the mechanism of ROS-induced cell necrosis remains poorly understood. In this study, we examined the role of the extracellular signaling-regulated kinase (ERK) pathway in H2O2-induced necrosis of renal proximal tubular cells (RPTC) in primary culture. Exposure of 60 to 70% confluent RPTC to 1 mM H2O2 for 3 h resulted in 44% necrotic cell death, as measured by trypan blue uptake, and inactivation of mitogen-activated protein kinase kinase (

In order to reduce the haemolytic susceptibility of glucose-6-phosphate dehydrogenase (G-6-PD) deficient erythrocytes, Ginkgo biloba extract (EGb 761) and vitamin E (vit E) were used as antioxidant agents and their effects compared. The erythrocyte suspensions from control and G-6-PD deficient patients were subjected to hydrogen peroxide (H2O2) incubation for 1 h. The produced thiobarbituric acid reactive substance (TBARS) levels measured as nmol/g Hb were compared with those of the erythrocytes administered 250 microg/mL EGb 761 or vit E previously or concomitantly with H2O2. The use of total parenteral nutrition (TPN) and intravenous fat emulsions in sick or preterm infants is often required to maintain adequate nutrition, yet recent research has shown that when exposed to light these nutrients are altered and deliver a high load of exogenous toxic hydroperoxides to already compromised infants. Hydroperoxides cause damage at the cellular level unless mediated by the body's antioxidant systems. NICU patients are, by definition, patients at risk. Preterm infants have low antioxidant reserves and, like sick term infants, typically suffer significant oxidative stress.

BACKGROUND: Metabolic-dysfunction-associated steatotic liver disease (MASLD) is a multifactorial liver disease in which mitochondrial dysfunction, oxidative stress, and inflammation play key roles in driving the progression toward metabolic dysfunction-associated steatohepatitis (MASH) and hepatocellular carcinoma (HCC). Dysfunctional mitochondria generate excess reactive oxygen species (ROS), impair antioxidant defenses, activate pro-inflammatory pathways and hepatic stellate cells, and perpetuate liver injury. Mitochondrial Complex I is a major ROS source, particularly under conditions of dy

Type 2 diabetes mellitus represents a major global health burden and is often preceded by a prediabetic state characterized by insulin resistance and metabolic dysfunction. Mitochondrial alterations, oxidative stress, and disturbances in lipid metabolism are central to the prediabetes pathophysiology. Melatonin, a pleiotropic indolamine, is known to regulate metabolic and mitochondrial processes; however, its therapeutic potential in prediabetes remains poorly understood. This study investigated the effects of melatonin on energy metabolism, oxidative stress, and mitochondrial function in a ra

In chronic kidney disease (CKD), oxidative stress (OS) plays a central role in the development of cardiovascular diseases. This pilot program aimed to determine whether an intradialytic aerobic cycling training protocol, by increasing physical fitness, could reduce OS and improve other CKD-related disorders such as altered body composition and lipid profile. Eighteen hemodialysis patients were randomly assigned to either an intradialytic training (cycling: 30 min, 55%-60% peak power, 3 days/week) group (EX; n = 8) or a control group (CON; n = 10) for 3 months. Body composition (from dual-energ

Intimal oxidation of LDL is considered an important early event in atherogenesis, and certain antioxidants are antiatherogenic. Dietary coenrichment with vitamin E (VitE) plus ubiquinone-10 (CoQ(10), which is reduced during intestinal uptake to the antioxidant ubiquinol-10, CoQ(10)H(2)) protects, whereas enrichment with VitE alone can increase oxidizability of LDL lipid against ex vivo oxidation. In the present study, we tested whether VitE plus CoQ(10) cosupplementation is more antiatherogenic than either antioxidant alone, by use of apolipoprotein E-deficient (apoE-/-) mice fed a high-fat di

Nonalcoholic fatty liver disease (NAFLD) caused by childhood obesity has become an important public health problem, leading to hepatic fat accumulation, inflammation, hepatocyte apoptosis, and potential liver dysfunction. Kynurenine monooxygenase (KMO) is an enzyme of the kynurenine (Kyn) pathway, which is associated with various chronic diseases. Here, we investigated the effect of KMO inhibitor (RO 61-8048) via the kynurenine pathway for the treatment of NAFLD. In this study, C57BL/6 mice were fed with high-fat diet (HFD) and fructose/sucrose (55%:45%) for to simulate fatty degeneration in v

BACKGROUND: The transient global cerebral hypoperfusion/reperfusion achieved by induction of Bilateral Common Carotid Artery Occlusion followed by Reperfusion (BCCAO/R) may trigger a physiological response in an attempt to preserve tissue and function integrity. There are several candidate molecules among which the endocannabinoid system (ECS) and/or peroxisome-proliferator activated receptor-alpha (PPAR-alpha) may play a role in modulating oxidative stress and inflammation. The aims of the present study are to evaluate whether the ECS, the enzyme cyclooxygenase-2 (COX-2) and PPAR-alpha are in

Cadmium (Cd) contamination threatens agro-ecological safety and food security; thus, elucidating mechanisms of Cd uptake and accumulation in plants is key to breeding low-Cd crops. Within the defensive network plants deploy against Cd stress, the plasma membrane and endomembrane system act as physical barriers to metal influx and as the core regulatory hub governing signal perception, transmembrane transport, and vacuolar compartmentalization for detoxification. This review examines the physiological and molecular mechanisms by which the cell membrane system influences Cd transport and accumul

Advanced glycation end-products (AGEs) accumulate in metabolic dysfunction-associated steatotic liver disease (MASLD); however, the contribution of individual AGE species remains poorly defined. We focused on Nε-(carboxymethyl)lysine (CML), a well-characterised AGE generated through glycoxidation and lipid peroxidation, and investigated its association with steatotic liver disease and its effects in multicellular liver models. AGE-RAGE pathway activity was analysed in public liver transcriptomic cohorts, and total AGEs and CML were quantified in human liver and serum. Free CML was then used as

Differentiated thyroid cancer (DTC), including papillary and follicular subtypes, is generally associated with favorable prognosis; however, a subset of patients develops recurrent, metastatic, or radioiodine-refractory diseases with limited therapeutic options. Ferroptosis, an iron-dependent form of regulated cell death driven by lipid peroxidation, has recently emerged as a biologically relevant process in thyroid cancer, yet its role in differentiated disease remains incompletely defined. Unlike many other malignancies, thyroid cancer arises within an organ intrinsically shaped by iodine-de

Cataract, the leading cause of blindness worldwide, results from progressive lens opacification driven by oxidative stress, protein aggregation, and metabolic dysregulation. Although aging remains the primary risk factor, growing evidence implicates systemic metabolic disorders, particularly metabolic syndrome (MetS), in accelerating cataractogenesis. MetS, characterized by central obesity, hypertension, hyperglycemia, and dyslipidemia, promotes chronic inflammation, oxidative stress, and metabolic imbalance. These systemic disturbances create a biology milieu that lowers the threshold for cry

Titanium dioxide nanoparticles (TiO2 NPs) are ubiquitous dietary additives, yet their impact on the intestinal barrier remains contentious, largely because conventional toxicological models overlook gastrointestinal digestive transformations. Characterization revealed that pristine TiO2 NPs exist as nanoscale aggregates, which, upon simulated digestion, lose electrostatic repulsion and expand into dense, biocorona-coated, micrometer-sized agglomerates enriched in nitrogen and phosphorus. While these transformed entities induce severe barrier dysfunction, the spatiotemporal mechanisms executing

The interplay between eicosanoids and reactive oxygen species (ROS) constitutes a bidirectional, self-amplifying "master switch" that drives pathophysiology ranging from inflammation to cell death. This review elucidates the molecular architecture of this axis, detailing how ROS activates the gatekeeper enzyme cytosolic phospholipase A₂α (cPLA₂α) to release arachidonic acid (AA), while downstream enzymes (COX, LOX, CYP450) inherently generate ROS as catalytic by-products. This review specifically examines the role of this axis as the central executioner of ferroptosis, where 15-LOX-mediated pe

Hepatocellular carcinoma (HCC), the most prevalent primary liver malignancy, is characterized by a rising global incidence, dismal prognosis, and high mortality rates attributed to its complex etiology and therapeutic resistance. Sorafenib is the first approved systemic therapy for advanced HCC. However, its clinical benefit is frequently limited by primary or acquired resistance, with most patients experiencing disease progression within approximately 6 months of treatment. This challenge highlights the urgent need to elucidate underlying mechanisms driving treatment failure. Ferroptosis, an

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a chronic liver condition with a rising global incidence, closely linked to metabolic risk factors such as dyslipidemia. Apolipoprotein E-deficient (ApoE-/-) mice fed a Paigen diet are an established model for MASLD and atherosclerosis research. This study investigated the effects of ellagic acid (EA) on MASLD progression in ApoE-/- mice fed a Paigen diet. Wild-type (WT) and ApoE-/- mice were fed a Paigen diet for 10 weeks to induce metabolic dysregulation. ApoE-/- mice were concurrently administered either EA (10 mg/kg/day or
