Integrated Transcriptome and Metabolome Analysis Reveals the Molecular Basis Underlying Anthocyanin Biosynthesis Associated with Different Flower Colors in Vicia faba L.

Faba bean (Vicia faba L.) exhibits significant variation in flower color. To elucidate the underlying regulatory mechanisms, transcriptomic and metabolomic analyses were conducted on pink, gray, and white flower germplasms. Anthocyanin content was higher in pigmented flowers (162.29 μg/g) than in white flowers (36.28 μg/g). Compared to white flowers, gray flowers showed upregulation of four key metabolites related to petunidin, cyanidin, and delphinidin. Pink flowers encompassed 15 derivatives derived from cyanidin, delphinidin, peonidin, petunidin, and malvidin. Across the three flower colors

Metabolomic aging and mortality clocks based on ensemble learning identify systemic aging risk across the human life span in 18,476 individuals.

Aging is characterized by progressive functional decline, yet metabolomic signatures across the human life span remain incompletely defined. Here, we profiled plasma metabolomes from 18,476 participants aged 1 to 96 years across six centers in China using nano-assisted laser desorption/ionization mass spectrometry (LDI-MS) and identified 56 representative age-associated metabolites. In a healthy cohort, we developed a stacking ensemble model to estimate metabolomic age and quantified metabolomic age acceleration (mAA) as each individual's deviation from chronological age. Higher mAA was associ

Concentration effects of proanthocyanidins on protein oxidation and quality deterioration of refrigerated golden pompano (Trachinotus blochii) fillets: Insights from metabolomic analysis.

This study investigated the effects of different proanthocyanidins (PA) concentrations (0-4.0 g/L) on protein oxidation, physicochemical quality, and metabolomic changes in refrigerated golden pompano (Trachinotus blochii) fillets. The results showed that PA exhibited concentration-related protective effects against quality deterioration, primarily by inhibiting lipid and protein oxidation. Notably, 2.0 g/L PA exhibited superior protective effects, suppressing the increase in free-radical intensity, carbonyl content, and disulfide bond content, while retaining more sulfhydryl content. A systematic profiling of chemicalome and metabolome of Citrus aurantium 'Changshan-huyou' flavonoids to provide insights into its potential in vivo pharmacodynamic material basis.

Citrus aurantium 'Changshan-huyou', as an authentic geo-characteristic Chinese medicinal material, has shown promising therapeutic potential for metabolic disorders. Although the abundant flavonoid constituents have been demonstrated as its main active substances, it is never an easy task for systematic profiling of its chemical constituents (chemicalome) and in vivo biotransformed metabolites (metabolome). Therefore, the unclear in vivo pharmacodynamic material basis of Citrus aurantium 'Changshan-huyou' flavonoids (CACF) is the key bottleneck that limits the full realization of its clinical

Hypothesis-free evaluation of circulating metabolome provides cell-specific insights regarding the role of energy substrate availability in amyotrophic lateral sclerosis.

BACKGROUND: Amyotrophic lateral sclerosis (ALS) is a neurodegenerative disease with limited therapeutic options. The circulating metabolome comprises small molecules present in plasma/serum which are the intermediates and end-products of cellular metabolism, and is linked to ALS pathogenesis. METHODS: We conducted hypothesis-free two-sample Mendelian randomisation (MR) analysis of the concentration of 575 plasma/serum metabolites, to determine which are causally linked to risk of ALS. Significant metabolites were validated in an independent GWAS of plasma/serum metabolite concentrations and e

Metabolomic signatures of probiotic secretomes reveal strain-specific biofunctional potential: insights from Lacticaseibacillus rhamnosus R0011 and Bifidobacterium longum R0175.

Probiotic secretomes are cell-free byproducts of bacterial growth, which contain bioactive metabolites. These metabolites have antioxidant and anti-inflammatory effects and mediate the host-microbe and microbe-microbe interactions. However, the metabolomic profiles of probiotic secretomes are strain-specific and have not been fully investigated. Untargeted metabolomic analysis was used to characterise the metabolomic profiles of Lacticaseibacillus rhamnosus R0011 (R11) secretome (LR) and Bifidobacterium longum R0175 (R175) secretome (BL). LR and BL had distinct metabolomic profiles. LR exhibit

Dulaglutide versus empagliflozin as add-on therapy to metformin and sulfonylurea in type 2 diabetes: a randomized pilot study with exploratory metabolomic and microbiome analyses.

In patients with type 2 diabetes mellitus (T2DM) inadequately controlled with metformin and sulfonylurea, evidence directly comparing glucagon-like peptide-1 receptor agonists and sodium-glucose cotransporter 2 inhibitors as add-on therapy is limited; therefore, we compared dulaglutide and empagliflozin in this setting. This 12-week, single-center, randomized, open-label, parallel-group pilot study included a 24-week observational extension. Patients with HbA1c≥7.0% receiving stable doses of metformin and glimepiride were randomized to dulaglutide 0.75 mg/week or empagliflozin 10 mg/day. Doses
