A novel molluscan CNS organ culture for the examination of adult nerve regeneration.

BACKGROUND: Central nervous system (CNS) regeneration is crucial for re-establishing neural connections following injury. Though some adult vertebrates have regeneration-competent CNS, invertebrate CNS also provide useful model systems for examining cellular and molecular mechanisms promoting regeneration. NEW METHOD: Using the mollusc Lymnaea stagnalis, we have developed a novel ex vivo CNS organ culture to examine regenerative neurite outgrowth from cut nerves floating at the air-liquid interface. RESULTS: We determined that single CNS organ cultures produced more outgrowth than co-culture

Rationale for the Use of 8-Aminoguanine for the Management of Cystitis.

Bladder pain and associated lower urinary tract symptoms (LUTSs) are hallmark features of interstitial cystitis/bladder pain syndrome (IC/BPS) and hemorrhagic cystitis (HC). IC/BPS is characterized by bladder-based pain and irritative bladder symptoms, with or without gross inflammatory changes (Hunner lesions) in the bladder wall, and the absence of well-described causes such as infection or neoplastic disease. The clinical features of HC are similar to IC/BPS, but in this instance, they are always associated with visible inflammatory disease with known etiologies such as infection or a compl

Identification and validation of growth hormone-reactive gene markers in brain tissue during the recovery period of stroke.

BACKGROUND: Functional recovery after ischemic stroke is often incomplete, highlighting the need for therapies that enhance neural repair. Growth hormone (GH) has shown potential neuroprotective effects, but its molecular mechanisms in the post-stroke brain remain unclear. This study aimed to identify and validate GH-responsive gene markers and explore their association with functional recovery. METHODS: A middle cerebral artery occlusion (MCAO) rat model was established. RNA sequencing of peri-infarct tissues was performed on days 7, 14, and 28 after recombinant human GH (rhGH) treatment to

Wnt5a-Induced Exosomes From Bone Marrow Mesenchymal Stem Cells Promote Spinal Cord Injury Repair by Modulating Immune Cell Phenotypes and Alleviating Neuroinflammation via the NF-κB Pathway.

OBJECTIVE: Spinal cord injury is a devastating neurological disorder. The transplantation of mesenchymal stem cell-derived exosomes has shown great promise. Wnt5a has been reported to promote neuronal differentiation and spinal cord regeneration. However, its mechanistic role when delivered via exosomes remains unclear. In the present study, the function and underlying mechanism of Wnt5a in promoting neuronal differentiation and spinal cord repair were investigated using bone marrow mesenchymal stem cell-derived exosomes. METHODS: This study initially constructed Wnt5a-overexpressing BMSCs an

Alzheimer disease: Amyloid peptide controversies and challenges of anti-Aβ immunotherapy.

Alzheimer disease (AD) is a progressive neurodegenerative disease marked by the accumulation of extracellular β-amyloid (Aβ) plaques and intracellular neurofibrillary tangles containing tau protein. Given the relentless deterioration in AD, strategies to slow its progression often focus on inhibiting Aβ production or aggregation. However, numerous clinical trials targeting Aβ in later disease stages have shown limited success in reversing cognitive decline, potentially due to intervening too late in the disease course. Emerging evidence suggests that addressing Aβ pathology during the earliest

The efficacy and safety of low-intensity focused ultrasound pulses for prolonged disorders of consciousness: a study protocol for a randomized controlled trial.

BACKGROUND: Recent randomized controlled trials (RCTs) in patients with prolonged disorders of consciousness (pDoC) have yielded limited success. Among them, only studies involving amantadine have provided Class II evidence. The effects of other non-invasive brain stimulation techniques remain inconclusive, largely due to patient heterogeneity and the clinical complexities of implementing such interventions. Low-intensity focused ultrasound pulses (LIFUP), as a novel, non-invasive, and safe neuromodulation technique, have the potential to both stimulate and inhibit deep subcortical structures.

Therapeutic effect of neural induced-stem cell-derived exosomes by regulating ERK/p38/NF-κB in traumatic brain injury.

Traumatic brain injury (TBI) can occur because of sudden external forces, such as falls or accidents, and it can cause immediate and long-term damage to the central nervous system. In this study, the therapeutic effects of neural-induced human adipose tissue-derived stem cell-derived exosomes (NI-Exo) on TBI were investigated. Exosomes were isolated and characterized through nanoparticle tracking analysis, cryo-transmission electron microscopy, and western blotting analysis. The therapeutic effects of NI-Exo were assessed in LPS-stimulated human microglial cells and TBI mice via behavioral tes

FGF21 maintains redox homeostasis and promotes neuronal survival after traumatic brain injury by targeting SLC25A39-mediated mitochondrial GSH transport.

BACKGROUND: Traumatic brain injury (TBI) represents a critical form of acute brain injury, characterized by considerable mortality and morbidity. Recently, fibroblast growth factor 21 (FGF21), a multifaceted hormone predominantly synthesized in liver, has emerged as a promising neuroprotective agent. In the study, we aim to investigate whether FGF21 exerts protective effects against TBI and to further elucidate its underlying molecular mechanisms. METHODS: To elucidate the role of FGF21 in regulating SLC25A39-dependent mitochondrial GSH transport and providing protection against TBI-induced n

Neuro-immune interactions: Exploring the anti-inflammatory role of the vagus nerve.

The vagus nerve, the longest cranial nerve, plays a critical role in maintaining homeostasis through its complex anatomical structure and diverse physiological functions. Recent studies highlight its unique anti-inflammatory mechanisms, showing that vagus nerve stimulation (VNS) can activate both the cholinergic anti-inflammatory pathway (CAP) and the hypothalamic-pituitary-adrenal (HPA) axis. This leads to a reduction in the release of pro-inflammatory cytokines and alleviation of inflammation. Such neuroimmune regulation provides valuable insights for managing chronic inflammatory diseases,

Edaravone dexborneol provides neuroprotective benefits by suppressing ferroptosis in experimental intracerebral hemorrhage.

Edaravone dexborneol (EDB) is widely recognized for its anti-inflammatory and antioxidant properties and is clinically applied in the treatment of acute cerebral infarction. Ferroptosis is a critical process in the pathophysiology of brain injury following intracerebral hemorrhage (ICH). However, it remains unclear whether EDB can ameliorate ICH through the modulation of ferroptosis. This study aimed to evaluate the function and mechanism of EDB in treatment of ICH. With a male rat ICH model, animal behavior tests, histopathological staining, magnetic resonance imaging and evans blue staining

Annexin 1 Reduces Dermatitis-Induced Itch and Cholestatic Itch through Inhibiting Neuroinflammation and Iron Overload in the Spinal Dorsal Horn of Mice.

The unclear pathogenesis of chronic itch originating from several systemic disorders poses challenges to clinical intervention. Recent studies recapitulate the spinal neurocircuits associated with neuroinflammation and synaptic plasticity responsible for pruriceptive sensations. The resolution of nociception and inflammation by Annexin 1 (ANXA1) has been identified. Given that pain and itch share many neural mechanisms, we employed two mice models of chronic itch to study the underlying targets and therapeutic potential of ANXA1, comprising allergic contact dermatitis-induced itch and cholesta

Docosahexaenoic acid ameliorates autistic-like behaviors by inhibiting oxidative stress and inflammatory response in neonatal maternal separation rats.

Autism spectrum disorder (ASD) is a pervasive neurodevelopmental disorder characterized by impaired social interactions and communication, repetitive, or stereotyped behavior. Docosahexaenoic acid (DHA), an essential polyunsaturated fatty acid, has been demonstrated to exert anti-oxidative stress and anti-inflammatory properties, while also promoting myelin development and neural differentiation and development. However, it remains uncertain whether DHA can ameliorate autistic-like behaviors, and if so, the underlying mechanisms are still unclear. Here, we established a neonatal maternal separ

Astaxanthin activates the Nrf2/Keap1/HO-1 pathway to inhibit oxidative stress and ferroptosis, reducing triphenyl phosphate (TPhP)-induced neurodevelopmental toxicity.

Triphenyl phosphate (TPhP) serves as a major organophosphorus flame retardant, and its induced neurodevelopmental toxicity has attracted widespread attention, but the mechanism remains unclear. In this study, we involved zebrafish to explore the new mechanism of TPhP inducing oxidative stress and ferroptosis to promote neurodevelopmental toxicity. The results suggested that TPhP affected the embryonic development, reduced the number of new neurons, and led to abnormal neural behavior in zebrafish larvae. TPhP also induced ROS accumulation, activated the antioxidant defense signal Nrf2 and Keap

A ganglionic intestinointestinal reflex activated by acute noxious challenge.

To investigate noxious stimulation-responsive neural circuits that could influence the gut, we recorded from intestinally directed (efferent) nerve filaments dissected from mesenteric nerves close to the small intestine in anesthetized rats. These exhibited baseline multiunit activity that was almost unaffected by vagotomy (VagX) and reduced only slightly by cutting the splanchnic nerves. The activity was halved by hexamethonium (Hex) treatment. When an adjacent gut segment received an intraluminal stimulus 2,4,6-trinitrobenzenesulfonate (TNBS) in 30% ethanol, mesenteric efferent nerve activit

Identification of a novel perifornical-hypothalamic-area-projecting serotonergic system that inhibits innate panic and conditioned fear responses.

The serotonin (5-HT) system is heavily implicated in the regulation of anxiety and trauma-related disorders such as panic disorder and post-traumatic stress disorder, respectively. However, the neural mechanisms of how serotonergic neurotransmission regulates innate panic and fear brain networks are poorly understood. Our earlier studies have identified that orexin (OX)/glutamate neurons within the perifornical hypothalamic area (PFA) play a critical role in adaptive and pathological panic and fear. While site-specific and electrophysiological studies have shown that intracranial injection and

Luteolin Attenuates Hypertension via Inhibiting NF-κB-Mediated Inflammation and PI3K/Akt Signaling Pathway in the Hypothalamic Paraventricular Nucleus.

BACKGROUND: Luteolin is widely distributed among a number of vegetal species worldwide. The pharmacological effects of luteolin are diverse and amongst antioxidant, free radical scavenging, and anti-inflammatory activities. Preliminary study showed that luteolin can ameliorate hypertension. However, the precise mechanism needs further investigation. There is no evidence that luteolin affects the paraventricular nucleus of the hypothalamus (PVN), a brain nucleus associated with a critical neural regulator of blood pressure. Our main aim was to explore the effect of luteolin on the PI3K/Akt/NF-κ

Perspectives on Agmatine Neurotransmission in Acute and Chronic Stressrelated Conditions.

Adaptive responses to stressful stimuli in the environment are believed to restore homeostasis after stressful events. Stress activates the hypothalamic-pituitary-adrenocortical (HPA) axis, which releases glucocorticoids (GCs) into the bloodstream. Recently, agmatine, an endogenous monoamine was discovered to have the potential as a pharmacotherapy for stress. Agmatine is released in response to certain stress conditions, especially those involving GCs, and participates in establishing homeostasis disturbed by stress following GC activation. The therapeutic potential of agmatine for the manage

Cephalopod learning and memory.

Cephalopod molluscs are renowned for their unique central nervous system - a donut-shaped brain organised around the oesophagus. This brain supports sophisticated learning and memory abilities. Between the 1950s and 1980s, these cognitive abilities were extensively studied in octopus (Figure 1A) - a now leading model for the study of memory and its neural substrates (approximately 200 papers during this period). The focus on octopus learning and memory was mainly due to their curious nature and the fact that they adapt to laboratory-controlled conditions, making them easy to test and maintain
