Extra-articular arthrodesis of the subtalar joint: a clinical study and review.

A series of 73 Grice extra-articular arthrodeses in 54 children with hind foot valgus is presented with an overall satisfactory result in 79% of these cases. The diagnostic categories in which we have used this procedure include: cerebral palsy, poliomyelitis, meningomyelocele, pes plano valgus, convex pes valgus, osteogenesis imperfecta, congenital hydrocephalus, partial adactylia, and congenital calcaneovalgus. The results with 11 meningomyelocele deformities are encouraging as only 2 of these children required a triple arthrodesis; 5 out of 6 patients in whom the opposite foot could serve a…

Preserved scleral allografts in periodontal defects in man. II. Histological evaluation.

1. The scleral grafts appeared to be well accepted as there were no signs of antigenicity or untoward reactions. 2. The gingival connective tissue, periodontal ligament and the periosteum were observed intertwined with sclera at the interface. 3. Sclera was invaded by host fibroblasts, capillaries, and appeared in some areas to be raplaced by a dense connective tissue. 4. Areas of cementogenesis could be observed in all specimens. 5. There were no signs of osteogenesis within the scleral grafts. 6. The alveolar crest appeared relatively nonreactive to sclera. 7. There were no signs of external…

LIPUS inhibited the expression of inflammatory factors and promoted the osteogenic differentiation capacity of hPDLCs by inhibiting the NF-κB signaling pathway.

BACKGROUND AND OBJECTIVES: As a chronic infectious disease, periodontitis could lead to tooth and bone loss. Low-intensity pulsed ultrasound … min/d for 7 days could also inhibit IL-6 and IL-8 gene expression, upregulate the expression of the osteogenesis-related genes RUNX2, OPN, OSX, and OCN, and promote osteogenic differentiation capacity in osteogenic medium in inflamed hPDLCs. The NF-κB signaling pathway was inhibited with LIPUS (90 mW/cm2 ) via inhibition of the phosphorylation of IκBα and the …

Fourier transform infrared imaging spectroscopy (FT-IRIS) of mineralization in bisphosphonate-treated oim/oim mice.

Fourier transform infrared microscopy (FT-IRM) and imaging spectroscopy (FT-IRIS) are increasingly used to analyze the molecular components of mineralized tissues. A primary advantage of these techniques is the capability to simultaneously image the quantity and quality of multiple components in histological sections at 7 microm spatial resolution. In the current study, FT-IRM and FT-IRIS were used to characterize bone mineralization in a mouse model of osteogenesis imperfecta (OI) after treatment with the bisphosphonate alendronate (ALN). This application is currently relevant since recent st…

The Role of Vitamin D3 in Periodontal Health: Implications for Bone Metabolism, Immune Modulation and Inflammation Control.

Vitamin D3 is a fat-soluble steroid essential for bone metabolism, immune modulation, and inflammation control, all critical for periodontal health. Its active form, 1,25-dihydroxyvitamin D3, binds to the vitamin D receptor (VDR) in periodontal cells, including periodontal ligament stromal cells, fibroblasts, osteoblasts, and macrophages, enhancing osteogenesis, antimicrobial defenses, and anti-inflammatory responses. Clinical and experimental evidence demonstrates that adequate systemic vitamin D3 levels and local activation in gingival tissues improve outcomes of nonsurgical and surgical per…

Clostridium butyricum attenuates radiation-induced bone loss through gut microbiota and immune regulation in mice.

Radiation-induced bone loss remains clinically challenging due to limitations of existing treatments. Using a mouse model of fractionated total abdominal irradiation (TAI), we demonstrate that TAI triggers severe osteoporosis characterized by trabecular bone loss, suppressed osteogenesis, and elevated osteoclast activity. Through 16S rRNA sequencing and immune profiling, we found TAI induced gut dysbiosis, immune dysregulation such as Th17/Treg imbalance, and systemic inflammation. Oral supplementation with C. butyricum reversed bone loss, activated osteogenic signaling, suppressed osteoclasto…

The mode of interfragmentary movement affects bone formation and revascularization after callus distraction.

Callus distraction is sometimes associated with a delay in the maturation process and serious complications. It is believed that these complications are often caused by instability of the bone segment fixation. Typical fixation devices, such as ring-fixators, show significant deformations in all directions under external loading and muscle forces. This leads to axial compression and tension as well as shear movements in the healing area. Herein we investigated the hypothesis that the direction of interfragmentary movement after callus distraction affects the bone formation and revascularizatio

Co-option of a tendon-associated osteogenic program drives the evolutionary plasticity of intermuscular bones in teleosts.

Intermuscular bones (IBs) are critical yet poorly understood evolutionary specializations. These bones exhibit considerable morphological variability, proposed as adaptations to teleost diversification; however, the explicit adaptive and evolutionary mechanisms remain unresolved. By integrating comparative morphological analysis of 360 teleost species and eco-functional performance assays, we demonstrate that IB complexity is associated with ecological niche expansion and enhanced burst swimming capacity, underscoring IBs as versatile adaptations. Single-cell transcriptomic profiling across fo

Dynamic osteodistraction with springs - the Gothenburg experience of spring-assisted cranioplasty.

Surgical treatment of craniosynostosis includes a wide range of techniques. Although a variety of more extensive cranioplasties have been employed to enhance outcomes, efforts to identify less invasive surgical approaches with better results continue. Since the introduction of springs as a tool for craniosynostosis surgery in 1997 at Sahlgrenska University Hospital in Gothenburg, Sweden, several studies have established their utility as an efficient surgical method. Because springs are currently used in the majority of our surgeries addressing craniosynostosis in children, this review summariz

The central mechanotransducer in osteoporosis pathogenesis and therapy.

This review identifies the mechanosensitive ion channel Piezo1 as the central regulator of bone homeostasis. Piezo1 senses mechanical loads in osteocytes, osteoblasts, and bone marrow mesenchymal stem cells (BMSCs), converting them into Ca2+-dependent signals that activate key pathways, including CaMKII, YAP/TAZ, Wnt/β-catenin, and ERK. These cascades collectively promote osteoblast differentiation and suppress osteoclastogenesis via OPG/RANKL modulation. Age-related Piezo1 decline impairs bone mechanoresponsiveness, driving both senile and disuse osteoporosis. Piezo1 also integrates bone meta

Age estimation in the combined long bones and ribs by histomorphometry: Past, present, and future.

Numerous age estimation methods in unidentified bone have been a long time developing for application in forensic anthropology. The histomorphometric technique is one of the alternative methods that relied upon the evaluation of the cortical bone microstructure over the lifespan as a result of the remodeling process in bone. Remodeling is a sophisticated event occurring from the coupled function of bone formation and resorption cells for maintaining mineral homeostasis and repairment of microdamage in bone tissue. Products derived from remodeling are primary changes in the osteon or haversian

Mechanisms of preferential bone formation in myeloma bone lesions by proteasome inhibitors.

Proteasome inhibitors (PIs) can preferentially restore bone in bone-defective lesions of patients with multiple myeloma (MM) who respond favorably to these drugs. Most prior in vitro studies on PIs used continuous exposure to low PI concentrations, although pharmacokinetic analysis in patients has shown that serum concentrations of PIs change in a pulsatile manner. In the present study, we explored the effects of pulsatile treatment with PIs on bone metabolism to simulate in vivo PI pharmacokinetics. Pulsatile treatment with bortezomib, carfilzomib, or ixazomib induced MM cell death but only m
