The association of parasitic infections in pregnancy and maternal and fetal anemia: a cohort study in coastal Kenya.

BACKGROUND: Relative contribution of these infections on anemia in pregnancy is not certain. While measures to protect pregnant women against malaria have been scaling up, interventions against helminthes have received much less attention. In this study, we determine the relative impact of helminthes and malaria on maternal anemia. METHODS: A prospective observational study was conducted in coastal Kenya among a cohort of pregnant women who were recruited at their first antenatal care (ANC) visit and tested for malaria, hookworm, and other parasitic infections and anemia at enrollment. All wo

Parasite-mediated selection drives an immunogenetic trade-off in plains zebras (Equus quagga).

Pathogen evasion of the host immune system is a key force driving extreme polymorphism in genes of the major histocompatibility complex (MHC … . Maladaptive GI parasite 'susceptibility alleles' were reduced in frequency, suggesting that these parasites may play a greater selective role at this locus. Heterozygote advantage, in terms of allele mutational divergence, also predicted decreased GI parasite burden in genotypes with a common allele. We conclude that an immunogenetic trade-off affects res …

The prevalence and diversity of intestinal parasitic infections in humans and domestic animals in a rural Cambodian village.

In Cambodia, intestinal parasitic infections are prevalent in humans and particularly in children. Yet, information on potentially zoonotic parasites in animal reservoir hosts is lacking. In May 2012, faecal samples from 218 humans, 94 dogs and 76 pigs were collected from 67 households in Dong village, Preah Vihear province, Cambodia. Faecal samples were examined microscopically using sodium nitrate and zinc sulphate flotation methods, the Baermann method, Koga Agar plate culture, formalin-ether concentration technique and Kato Katz technique. PCR was used to confirm hookworm, Ascaris spp., Gi…

Toxoplasma gondii relies on both host and parasite isoprenoids and can be rendered sensitive to atorvastatin.

Intracellular pathogens have complex metabolic interactions with their host cells to ensure a steady supply of energy and anabolic building … these mutants have only a mild growth phenotype and an isoprenoid composition similar to wild type parasites. However, when extracellular, the loss of the enzyme becomes phenotypically apparent. This strongly suggests that intracellular parasite salvage FPP and/or geranylgeranyl diphosphate (GGPP) from the host. We test this hypothesis using inhibitors of …

Characterisation of major vault protein during the life cycle of the human parasite Schistosoma mansoni.

Vaults are ribonucleoproteins (13 MDa) highly conserved among lower and higher eukaryotes. Their association produces a complex composed of three proteins named Major Vault Protein (MVP), vault (PolyADP-ribose) polymerase (VPARP) and Telomerase-associated protein (TEP1), plus a small untranslated RNA. The exact function of this complex is unknown, although the biological role of vaults has been associated with multidrug resistance phenotypes and signal transduction pathways. Genomic analysis showed that model organisms, such as Caenorhabditis elegans and Drosophila melanogaster, do not possess

Seasonal pattern of avian Plasmodium-infected mosquitoes and implications for parasite transmission in central Panama.

Aedeomyia squamipennis and Culex (Melanoconion) ocossa, two ubiquitous Neotropical mosquito species, are likely involved in the transmission of several bird pathogens in Gamboa, central Panama. However, knowledge on their eco-epidemiological profiles is still incomplete. Our goal in this study was to investigate temporal trends of vector density and their relationship with avian plasmodia prevalence. This information is central to identifying the risk posed by each vector species to the avian community locally. We found that A. squamipennis maintains stable population size across climatic seas

Ants affect the infestation levels but not the parasitism of honeydew and non-honeydew producing pests in citrus.

Ants act simultaneously as predators and as hemipteran mutualists, and thereby may affect the composition and population dynamics of a wide arthropod community. We conducted ant-exclusion experiments in order to determine the impact of ants on the infestation levels and parasitism of three of the most important citrus pests of western Mediterranean citrus: the honeydew producer Aleurothrixus floccosus Maskell (woolly whitefly) and the non-honeydew producers Aonidiella aurantii Maskell (California red scale; CRS) and Phyllocnistis citrella (Staiton) (citrus leafminer). The study was conducted i

Parasite prolyl oligopeptidases and the challenge of designing chemotherapeuticals for Chagas disease, leishmaniasis and African trypanosomiasis.

The trypanosomatids Trypanosoma cruzi, Leishmania spp. and Trypanosoma brucei spp. cause Chagas disease, leishmaniasis and human African trypanosomiasis, respectively. It is estimated that over 10 million people worldwide suffer from these neglected diseases, posing enormous social and economic problems in endemic areas. There are no vaccines to prevent these infections and chemotherapies are not adequate. This picture indicates that new chemotherapeutic agents must be developed to treat these illnesses. For this purpose, understanding the biology of the pathogenic trypanosomatid- host cell in

A scoring system approach for the parasite predictive assessment of fish lots: a proof of concept with anisakids.

A total of 982 individuals distributed in 11 lots belonging to 10 fish species from three Atlantic FAO fishing areas were sampled and examined to detect the presence of anisakid larvae in fish muscle. After hazard identification by genetic sequencing and exposure assessment by anatomic extent and demographic characterization of infection, all data were fitted for each fish species to a new proposed scoring schema of parasite prediction. In the absence of a criterion standard method for inspection and precise definition of the quantum satis for parasites in contaminated fish lots, the inspectio…

Expansion of host cellular niche can drive adaptation of a zoonotic malaria parasite to humans.

The macaque malaria parasite Plasmodium knowlesi has recently emerged as an important zoonosis in Southeast Asia. Infections are typically mild but can cause severe disease, achieving parasite densities similar to fatal Plasmodium falciparum infections. Here we show that a primate-adapted P. knowlesi parasite proliferates poorly in human blood due to a strong preference for young red blood cells (RBCs). We establish a continuous in vitro culture system by using human blood enriched for young cells. Mathematical modelling predicts that parasite adaptation for invasion of older RBCs is a likely

Colonial seabird's paralytic perfume slows lice down: an opportunity for parasite-mediated selection?

Selection for chemical signals in birds could be influenced by parasitism as has been previously suggested for visual and acoustic displays. Crested auklets (Aethia cristatella), colonial seabirds from Alaska and Siberia, offer an example of how this might occur. Crested auklets secrete lipids in plumage, possibly as an indicator of status and attractiveness. Prominent among these secretions are aldehydes, which are noticeable as a pungent citrus-like odour. Octanal and hexanal, the most abundant aldehydes in the plumage of crested auklets, are potent invertebrate repellents, reported from the

The antioxidant defence mechanisms of parasite and host after chronic Hymenolepis diminuta infestation of the rat.

The aim of the present study was to determine antioxidant defence mechanisms in the rat and Hymenolepis diminuta after long-term infestation. We determined levels of oxidative stress markers, and activity of antioxidant enzymes in the rat small intestine and in particular parts of H. diminuta. Observed changes in antioxidant enzymes activity in H. diminuta and the rat intestine indicate the defence against parasitic infestation and probably allowed parasite to adapt and live in oxidative stress.

Fine-scale analysis of parasite resistance genes in the red flour beetle, Tribolium castaneum.

Parasite infection impacts population dynamics through effects on fitness and fecundity of the individual host. In addition to the known roles of environmental factors, host susceptibility to parasites has a genetic basis that has not been well characterized. We previously mapped quantitative trait loci (QTL) for susceptibility to rat tapeworm (Hymenolepis diminuta) infection in Tribolium castaneum using dominant AFLP markers; however, the resistance genes were not identified. Here, we refined the QTL locations and increased the marker density in the QTL regions using new microsatellite marker…

Mosquito-parasite interactions can shape filariasis transmission dynamics and impact elimination programs.

The relationship between mosquito vectors and lymphatic filariasis (LF) parasites can result in a range of transmission outcomes. Anophelines are generally characterized as poor vectors due to an inability to support development at low densities. However, it is important to understand the potential for transmission in natural vectors to maximize the success of elimination efforts. Primary vectors in Papua New Guinea (n = 1209) were dissected following exposure to microfilaremic blood (range 8-233 mf/20 µl). We examined density dependent and species-specific parasite prevalence, intensity and y…

Evolution of the multi-domain structures of virulence genes in the human malaria parasite, Plasmodium falciparum.

The var gene family of Plasmodium falciparum encodes the immunodominant variant surface antigens PfEMP1. These highly polymorphic proteins are important virulence factors that mediate cytoadhesion to a variety of host tissues, causing sequestration of parasitized red blood cells in vital organs, including the brain or placenta. Acquisition of variant-specific antibodies correlates with protection against severe malarial infections; however, understanding the relationship between gene expression and infection outcome is complicated by the modular genetic architectures of var genes that encode v

Ecological and inhost factors promoting distinct parasite life-history strategies in Lyme borreliosis.

Understanding the ecology and evolution of tick-borne parasites is the foundation for preventing and managing tick-borne diseases. Tick-borne diseases such as Lyme borreliosis, are an emerging health threat in America, Europe, and Asia. Certain strains of Borrelia burgdorferi (the etiological agent of Lyme borreliosis) sampled in nature appear to be rapidly cleared by murine hosts. These strains, unlike their inhost-persistent counterparts, are unlikely to manifest severe disease. Their emergence and abundance in North America is unclear. Understanding why strains adopt a persistent or rapid-c…
