Mitochondrial diseases.

With the discovery of mitochondrial DNA (mtDNA) mutations in different neuromuscular disorders, investigations now seek to clarify how the mutant mtDNA induces biochemical and morphologic defects. In one of the most important approaches human mutant mtDNA is transferred into cells that lack mtDNA to examine the relationship between the amount of mutant mtDNA and defects in cell growth, respiration and enzyme activities. The resulting cells are 'cybrids'; these clonal cells contain the heteroplasmic mutant and normal mtDNA from patients with mitochondrial diseases. The mitochondria become funct…

Adrenal masses: evaluation with fast gradient-echo MR imaging and Gd-DTPA-enhanced dynamic studies.

Fast gradient-echo magnetic resonance (MR) imaging of 38 adrenal masses with proved diagnosis was performed during suspended respiration with various repetition times (TRs), echo times (TEs), and flip angles. Dynamic perfusion studies after gadolinium diethylenetriamine-pentaacetic acid (DTPA) administration were performed by repeated imaging at short time intervals. With more T2 weighting (TR = 60 msec, TE = 30 msec, and flip angle = 15 degrees), malignant tumors and pheochromocytomas had a significantly higher relative signal intensity than adenomas; overlap of signal intensity led to equivo…

M-mode echovenography: a new technique for the evaluation of venous wall and valve motion.

The M-mode technique which is widely used in echocardiography allows continuous recording of spontaneous venous wall movements. Compliance of the vein can be quantified. The diameter of the normal vein changes with respiration, while only minor changes are induced by cardiac function. Distensibility and compressibility of the common femoral vein (CFV) were documented. During Valsalva manoeuvre the mean diameter of the CFV increased from 1.05(SD 0.18) cm to 1.52(0.25) cm (p less than 0.01) in the recumbent position and from 1.50(0.20) cm to 1.63(0.17) cm (NS) in the upright position. A patent v…

Possible role of brain stem respiratory neurons in mediating vomiting during space motion sickness.

Vomiting is produced primarily by the coordinated action of the respiratory muscles. The object of this study was to determine if brain stem expiratory neurons control abdominal muscle activity during vomiting. We recorded the activity of 27 ventral respiratory group (VRG) expiratory neurons, which are known to be of primary importance for control of abdominal muscle activity during respiration. During fictive vomiting produced by emetic stimulants in decerebrate, paralyzed cats, only one-third of these neurons had the appropriate firing pattern to contribute to abdominal muscle activity durin…

Pattern of breathing in brachycephalic dogs.

The pattern of breathing was assessed in 19 brachycephalic dogs, using tidal breathing flow-volume loop (TBFVL) analysis. Fifteen dogs had T … act (n = 11) were similar to those of dogs with clinical signs of upper airway obstruction (n = 8). Respiration was monitored continuously for short periods (20 to 50 minutes) in 3 brachycephalic dogs resting in a cage in a quiet, darkened laboratory; 2 of these dogs had periodic breathing patterns characterized by multiple episodes of alternating hypopnea an …

[Studies on the periodicity of fetal and neonatal heart rate variability by fast Fourier transform analysis].

Heart rate changes were analyzed by fast Fourier transform technique in 68 normal fetuses of 23 to 41 weeks, 8 normal deliveries, 11 normal … ter periodicity. Neonatal 1.3 to 1.6 sec periodicity suggests a relationship between heart rate and respiration. No change in periodicity was found in fetal heart rate after the administration of 0.5 mg of atropine sulfate to the mother in 12 cases.

Transfusion reactions in cats due to AB blood group incompatibility.

Blood transfusion reactions were investigated in 70 unsensitised and sensitised cats. Twenty-five compatible transfusions with respect to the AB blood group system did not result in any immediate transfusion reactions. Transfusion of B erythrocytes in 12 group A cats did not produce any severe reactions because of low anti-B titres in the A recipients. However, 30 of 50 group B cats (60 per cent) suffered severe shock reactions, characterised by marked hypotension, cessation of respiration and sometimes atrio-ventricular blocks, within two minutes of the injection of incompatible A cells (phas…

Nonoperative measurement of pancreatic and common bile duct pressures with a microtransducer catheter and effects of duodenoscopic sphincterotomy.

Duodenoscopic manometry of the pancreatic duct (PD) and common bile duct (CBD) using a microtransducer catheter was distinct advantages over infusion manometry, giving absolute values of in situ intraluminal pressure. Microtransducer manometry was performed without medication in 49 patients with gallbladder stones (10), common bile duct stones (24), hepatic duct stones (6) and common bile duct dilatation (9), and was successful in 42 (86%) for PD and 36 (73%) for CBD. Ductal pressures showed respiration-synchronized biphasic variations superimposed by the arterial pulsation effect. Considerabl…

Anaesthesia for short-stay varicose vein surgery.

The study was undertaken in order to elucidate the suitability of four different types of general anaesthesia and of epidural block in short-stay varicose vein surgery. Special consideration was given to recovery after anaesthesia. The series consisted of 230 female patients subjected to varicose vein surgery of one lower extremity. The patients' recovery after surgery was observed through tests measuring visual-motor coordination and through questionnaires. The general anaesthesias used were halothane-nitrous oxide-oxygen anaesthesia with spontaneous respiration (Ha), relaxant-dominated combi…

Therapeutic problems in laryngo-tracheal stenoses.

History revealed iatrogenic injury (tracheostomy, intubation, artificial respiration) to be the reason for tracheal stenosis in each of our 32 cases (18 female, 14 male). So far 23 cases have been treated successfully and they live without a cannula; seven are still under treatment, while two patients died (severe heart-attack, spontaneous pneumothorax) and one refused further treatment. We were relatively successful in six cases of sleeve resection. All other 26 cases were treated by creating a tracheal groove, in which tracheal mucosa and skin of the neck were joined and the caudal end mostl…

Chemogenetic activation of mitochondria counteracts the Warburg effect in tumor cells.

Upregulation of glycolysis and downregulation of mitochondrial oxidative phosphorylation, termed as the Warburg effect, is characteristic of tumor cells. Restriction of pyruvate flux into the mitochondrial matrix is one of the major mechanisms underlying this phenomenon. Warburg metabolism is beneficial for rapidly proliferating cells; however, the metabolic consequences of activation of mitochondrial respiration in such cells remain unclear. Here we created a chemogenetic instrument, Grubraw, that generates pyruvate from ᴅ-alanine directly in the mitochondrial matrix, bypassing restricted pyr…

Application of Cu2+-chelated curcumin/chitosan coating in maintaining the storage quality of harvested passion fruit.

Passion fruit is susceptible to postharvest spoilage, requiring innovative preservation strategies. This study developed a novel Cu2+-chelated curcumin/chitosan composite coating (CuCur-CS) via self-assembly. Under ambient temperature, CuCur-CS treatment was the most effective, exhibiting the lowest weight loss rate (only 5.6% on day 18), along with superior fruit shape and surface color retention. The coating stabilized sugar-acid balance by suppressing respiration and alleviating oxidative stress. By day 18, H2O2 and MDA contents in the control increased by 53% and 28%, respectively, versus …

Astrocytic PI3Kα controls synaptic plasticity and cognitive function via serine metabolism.

Astrocytes modulate neuronal activity by gliotransmission and through metabolic regulation, yet the relationship between these functions rem … etion of p110α rewires astrocyte metabolism, reducing glycolytic flux while enhancing mitochondrial respiration. This, in turn, would limit L-serine biosynthesis through the phosphorylated pathway. Consistently, mice lacking astrocytic p110α display memory deficits that are rescued by in vivo L-serine administration. These findings identify astrocytic PI3K p1 …

Vertebrate endoderm development.

The endoderm forms the innermost layer of the embryonic gut tube. It gives rise to the epithelial and glandular components of many internal organs, including those of the digestive and respiratory systems. Endodermal organs have essential physiological processes such as nutrient absorption, respiration, detoxification and endocrine regulation. Over the past two decades, work in vertebrate embryos and human pluripotent stem cell models has defined the mechanisms that govern endoderm formation, regionalization and organogenesis. These processes are driven by conserved cell signaling pathways act…

Gas-sensing neurons prime mitochondrial fitness to offset metabolic stress.

The mitochondrial unfolded protein response (UPRmt) is triggered by cells to alleviate proteotoxicity in response to metabolic stress. The a … iption, confers mitochondrial stress resistance, and increases mitochondrial membrane potential and respiration. We show that starvation decreases GCY-9 expression and propose that the resultant cytoprotective program is launched to offset metabolic and proteotoxic risks. Thus, environmental sensing by peripheral neurons can preemptively enhance systemic mito …
