The subcellular distribution of methionine sulfoxide reductase A in mouse and human tissues.

Among all known repair enzymes, methionine sulfoxide reductase A (MsrA) plays a key role in protecting against oxidative stress and its dama … ochondria of mouse liver and kidney cells, in the cytoplasm of retina cells, and in the endoplasmic reticulum of glandular cells. Overall, the results indicate a correlation between the generation of reactive oxygen species (ROS) and the expression level of MsrA in subcellular compartments of the tissues examined.

The UFL1-AKT positive feedback loop promotes breast cancer progression by enhancing lipid synthesis.

UFMylation, a ubiquitin-like modification, is crucial for cellular processes and is linked to human diseases, including cancer. However, its role in cancer remains unclear. Here, we report that UFL1 promotes breast tumor growth by remodeling lipid metabolism. Mechanistically, UFL1 interacts with and UFMylates AKT, enhancing its localization at the endoplasmic reticulum and phosphorylation by PDK1 and mTORC2, thereby increasing AKT-mediated lipid synthesis. Moreover, AKT phosphorylates UFL1, boosting its activity. Thus, UFL1 and AKT form a positive feedback loop, accelerating lipid synthesis an…

Spermidine alleviates 2,2-dichloroacetamide/ 2,2-dibromoacetamide-exposed mouse oocyte toxicity by restoring autophagic homeostasis.

Environmental toxicants pose a major threat to human health, yet the reproductive toxicity of haloacetamide disinfection byproducts remains largely unclear. Here, we examined the effects of two haloacetamides, 2,2-dichloroacetamide (DCAcAm) and 2,2-dibromoacetamide (DBAcAm), on mouse oocyte maturation and identified the underlying mechanisms of toxicity. Both compounds markedly impaired meiotic spindle assembly and compromised the function of multiple organelles, including mitochondria, the endoplasmic reticulum, and the Golgi apparatus, primarily through autophagy dysregulation at specific ex…

ERO1a fosters glioblastoma aggressiveness and metabolic flexibility by regulating mitochondria-associated membrane dynamics.

Despite the wealth of data generated in the omics era to investigate molecular drivers, glioblastoma (GBM) remains one of the most incurable cancers with a poor median of survival. Here we unravelled the dynamic crosstalk between the endoplasmic reticulum and mitochondria, known as mitochondria-associated membranes (MAMs) and define how modulation of calcium fluxes and MAM structure influences GBM cell plasticity and metabolic flexibility. We identified ERO1α, whose expression is significantly associated with poor GBM patient survival, as a critical MAM protein that regulates MAM structure, dy…

Stearoyl-CoA desaturases regulate intestinal stem and progenitor cell metabolism and function in response to nutrient abundance.

Diet composition and nutrient availability play critical roles in intestinal stem and progenitor cell function and proliferation. Here, we show that stearoyl-CoA desaturases (SCDs) are regulated by nutrient abundance and link intracellular lipid metabolism to intestinal homeostasis. Pharmacological inhibition or genetic deletion of Scd1 and Scd2 in intestinal mouse organoids disrupted lipid desaturation, induced endoplasmic reticulum (ER) stress, and impaired stem and progenitor cell proliferation, effects largely mitigated by oleic acid supplementation. Intestine-specific deletion of Scd1 and…

S100A16 as a potential therapeutic target in sepsis-induced acute lung injury.

Sepsis-induced acute lung injury (ALI) is a major contributor to mortality in critically ill patients, yet its molecular basis remains incom … nd improved survival. In vitro experiments showed that S100A16 overexpression increased endoplasmic reticulum (ER) stress-related markers in lung epithelial cells. Furthermore, the protective effects of S100A16 knockdown were partially counteracted by S100A11 overexpression. These results indicate that the S100A16-S100A11 axis represents one of the contribu …

Functional inactivation of oligosaccharyltransferase a isoform suppresses tumor metastasis.

Oligosaccharyltransferase (OST), which is a multi-membrane protein complex that catalyzes asparagine-linked glycosylation (N-glycosylation) in the endoplasmic reticulum (ER), is a potential target to eradicate refractory cancer. Mammals express two distinct OST isoforms (OST-A and OST-B) that exhibit different acceptor site specificity to maximize N-glycosylation efficiency; however, the role of individual OST isoforms in tumor progression is not fully understood. Here, using mouse melanoma model, we showed that gene-edited knockout of either one of the OST isoforms did not compromise subcutan…

TUG protein acts through a disordered region to organize the early secretory pathway.

The Endoplasmic Reticulum (ER)-Golgi Intermediate Compartment (ERGIC) is a network of tubules and vesicles known for producing COPI vesicles and receiving COPII vesicles from the ER. Much about its identity, stability, and regulation remains unknown. Here, we show that TUG (UBXN9, Aspscr1) protein, a central regulator of GLUT4 trafficking, localizes to the ERGIC, and that its deletion enhances anterograde flux of a model soluble cargo protein. TUG deletion redistributes ERGIC markers to the cis-Golgi and alters Golgi morphology. TUG forms biomolecular condensates in vitro and contains a centra…

Dual roles of TRPV2 in the innate immune response to cytosolic DNA: Arresting dormant and boosting activated STING.

The cGAS/STING-dependent innate immune pathway is central in the cellular response to cytosolic DNA derived from viral infections, genotoxic … toDNA but dissociates from STING and promotes its activation by releasing Ca2+ from the endoplasmic reticulum in the presence of cytoDNA, which facilitates STING translocation to Golgi. Consequently, TRPV2 governs type I interferon production and natural killer cell-mediated cytotoxicity through the cGAS/STING pathway. The dual roles of TRPV2 provide an ele …

Nitric oxide attenuates PI4P accumulation at the ER membrane to inhibit encephalomyocarditis virus replication selectively in β-cells.

Viral infection, particularly by members of the picornavirus family, has been associated with autoimmune diabetes (type 1 diabetes mellitus) … plication is by attenuating the accumulation of phosphatidylinositol-4-phosphate at the endoplasmic reticulum membrane. As a result, viral replication complex formation is prohibited, and viral replication is effectively prevented. In agreement with previous studies, we show that these observations are selective for β-cells and because of a loss of cellular …

Suppression of SREBP by a transmembrane protein mutated in cardiomyopathy.

Arrhythmogenic right ventricular cardiomyopathy (ARVC) is an inherited cardiac disorder characterized by the replacement of myocardial tissue with adipose tissue. One of the frequently mutated proteins in ARVC is TMEM43, which encodes a transmembrane protein primarily localized to the endoplasmic reticulum (ER) and nuclear membranes and stabilized by interacting with squalene synthase (SQS). However, its precise role in ARVC pathogenesis remains unclear. The present study demonstrates that TMEM43 suppresses the expression of SQS and other lipogenic proteins by inhibiting sterol regulatory elem…

Discovery of isoindoline-2(1H)-carboxamide STING inhibitors as anti-inflammatory agents.

STING (stimulator of interferon genes) is an endoplasmic reticulum-resident membrane-spanning protein that is widely expressed in mammalian cells and functions as a central regulator for the innate immunity. Aberrant activation of the STING axis due to loss-of-function or gain-of-function mutation leads to various autoimmune and autoinflammatory disorders such as Aicardi-Goutières syndrome, systemic lupus erythematosus, and STING-associated vasculopathy with onset in infancy. Here we report the design, synthesis, and structure-activity relationship (SAR) of the isoindoline-2(1H)-carboxamide ST…

Haemaphysalis longicornis subolesin controls the infection and transmission of severe fever with thrombocytopenia syndrome virus.

Severe fever with thrombocytopenia syndrome (SFTS) caused by the SFTS virus (SFTSV) is an emerging tick-borne disease with a high mortality rate. Haemaphysalis longicornis is the primary reservoir and vector of SFTSV. Here, we found that targeting subolesin (SUB), an anti-tick vaccine candidate, affects the infection and transmission of SFTSV in H. longicornis. RNAi-mediated knockdown of SUB repressed SFTSV infection in the salivary glands but not in the gut of H. longicornis, which may be associated with the modulation of protein processing in endoplasmic reticulum revealed by transcriptomic …

Surface Gene Mutations of Hepatitis B Virus and Related Pathogenic Mechanisms: A Narrative Review.

Liver cancer has high incidence and mortality rates worldwide, with hepatocellular carcinoma (HCC) being the main histological subtype, accounting for 90% of primary liver cancers. The high mutation rate of viruses combined with endoplasmic reticulum stress may lead to the occurrence of cancer. Hepatitis B virus (HBV) infection is one of the most important pathogenic factors of HCC. The carcinogenic mechanisms of HBV have been widely studied. Among these mechanisms, immune escape and vaccine escape caused by mutations in the HBV S gene have been reported in numerous studies of patients with ch…

Recent insights about autophagy in pancreatitis.

Acute pancreatitis is a common inflammatory gastrointestinal disease without any successful treatment. Pancreatic exocrine acinar cells have high rates of protein synthesis to produce and secrete large amounts of digestive enzymes. When the regulation of organelle and protein homeostasis is disrupted, it can lead to endoplasmic reticulum (ER) stress, damage to the mitochondria and improper intracellular trypsinogen activation, ultimately resulting in acinar cell damage and the onset of pancreatitis. To balance the homeostasis of organelles and adapt to protect themselves from organelle stress,…

Navigating the Landscape of CMT1B: Understanding Genetic Pathways, Disease Models, and Potential Therapeutic Approaches.

Charcot-Marie-Tooth type 1B (CMT1B) is a peripheral neuropathy caused by mutations in the gene encoding myelin protein zero (MPZ), a key component of the myelin sheath in Schwann cells. Mutations in the MPZ gene can lead to protein misfolding, unfolded protein response (UPR), endoplasmic reticulum (ER) stress, or protein mistrafficking. Despite significant progress in understanding the disease mechanisms, there is currently no effective treatment for CMT1B, with therapeutic strategies primarily focused on supportive care. Gene therapy represents a promising therapeutic approach for treating CM…
