Proteomic surveillance of autoimmunity in Behcet's disease with uveitis: selenium binding protein is a novel autoantigen in Behcet's disease.

Autoimmune response to retinal antigens is considered to be one of the pathogenesis of uveitis in Behcet's disease (BD). In the present study, we comprehensively compared retinal autoantigens recognized by sera from BD patients with uveitis or healthy donors using proteomic technique, 2-dimensional electrophoresis (2DE) followed by Western blotting (WB). Six protein spots showing high reactivity with the serum from the BD patients were detected as candidate retinal autoantigens, and three of them were identified by mass spectrometry. Two of them were previously reported BD autoantigens, S-anti

Utilizing Zebrafish Visual Behaviors in Drug Screening for Retinal Degeneration.

Zebrafish are a popular vertebrate model in drug discovery. They produce a large number of small and rapidly-developing embryos. These embryos display rich visual-behaviors that can be used to screen drugs for treating retinal degeneration (RD). RD comprises blinding diseases such as Retinitis Pigmentosa, which affects 1 in 4000 people. This disease has no definitive cure, emphasizing an urgency to identify new drugs. In this review, we will discuss advantages, challenges, and research developments in using zebrafish behaviors to screen drugs in vivo. We will specifically discuss a visual-moto

Simultaneous VEP and PERG investigations in early Parkinson's disease.

To evaluate whether visual evoked potential (VEP) and pattern electroretinogram (PERG) abnormalities may be detectable early in the course of Parkinson's disease (PD) and to assess a possible interdependence between retinal and cortical potentials, simultaneous VEP and PERG were carried out in nine patients with early PD. The mean (SD) duration of the disease was 10 (6) months. None of the patients had been previously treated with levodopa or other anti-parkinsonian drugs. The results show VEP and PERG abnormalities that are spatial stimulus dependent, with higher frequencies being more involv

Genotypic and Phenotypic Characterization of Hexokinase 1 p.Glu847Lys Variant Causing Autosomal Dominant Pericentral Retinitis Pigmentosa.

BACKGROUND AND OBJECTIVE: The aim of this study was to clinically and molecularly characterize the largest cohort of patients with HK1 associated retinal disease. Hexokinase 1 (HK1) variants have been reported to cause retinitis pigmentosa (RP), but a clearly defined genotype-phenotype correlation has not been well established. PATIENTS AND METHODS: A retrospective, consecutive, single-center case series was performed on patients with molecularly confirmed HK1-associated disease. Patients between 2016 and 2024 were included. RESULTS: Sixty eyes from 30 patients were assessed. Thirty patients

Comparison of CRISPR-Cas13b RNA base editing approaches for USH2A-associated inherited retinal degeneration.

CRISPR-Cas13 systems have therapeutic promise for the precise correction of point mutations in RNA. Using adenosine deaminase acting on RNA (ADAR) effectors, A-I base conversions can be targeted using guide RNAs (gRNAs). We compare the Cas13 effectors PspCas13b and Cas13bt3 for the repair of the gene USH2A, a common cause of inherited retinal disease and Usher syndrome. In cultured cells, we demonstrate up to 80% efficiency for the repair of the common c.11864 G > A and its murine equivalent c.11840 G > A, across different gRNAs and promoters. We develop and characterize a mouse model of Usher

Metabolic plasticity in a Pde6bSTOP/STOP retinitis pigmentosa mouse model following rescue.

OBJECTIVE: Retinitis pigmentosa (RP) is a hereditary retinal disease characterized by progressive photoreceptor degeneration, leading to vision loss. The best hope for a cure for RP lies in gene therapy. However, given that RP patients are most often diagnosed in the midst of ongoing photoreceptor degeneration, it is unknown how the retinal proteome changes as RP disease progresses, and which changes can be prevented, halted, or reversed by gene therapy. METHODS: Here, we used a Pde6b-deficient RP gene therapy mouse model and performed untargeted proteomic analysis to identify changes in prot

Mutations in the splicing regulator Prp31 lead to retinal degeneration in Drosophila.

Retinitis pigmentosa (RP) is a clinically heterogeneous disease affecting 1.6 million people worldwide. The second-largest group of genes causing autosomal dominant RP in human encodes regulators of the splicing machinery. Yet, how defects in splicing factor genes are linked to the aetiology of the disease remains largely elusive. To explore possible mechanisms underlying retinal degeneration caused by mutations in regulators of the splicing machinery, we induced mutations in Drosophila Prp31, the orthologue of human PRPF31, mutations in which are associated with RP11. Flies heterozygous mutan

Neuoroprotective efficacies by KUS121, a VCP modulator, on animal models of retinal degeneration.

Retinitis pigmentosa (RP) is one of the leading causes of adult blindness and has no established therapy. We have shown that valosin-containing protein (VCP) modulators, Kyoto University Substances (KUSs), ameliorated abnormally low ATP levels by inhibiting the ATPase of VCP, thereby protected several types of cells, including retinal neurons, from cell death-inducing insults. In this study, we found that KUS121, one of the VCP modulators, effectively protects photoreceptors both morphologically and functionally, in two animal models of retinal degeneration, rd12 mice and RP rabbits with a rho

Biallelic Loss-of-Function Variants in UBAP1L and Nonsyndromic Retinal Dystrophies.

IMPORTANCE: Inherited retinal dystrophies (IRDs) present a challenge in clinical diagnostics due to their pronounced genetic heterogeneity. Despite advances in next-generation sequencing (NGS) technologies, a substantial portion of the genetic basis underlying IRDs remains elusive. Addressing this gap seems important for gaining insights into the genetic landscape of IRDs, which may help improve diagnosis and prognosis and develop targeted therapies in the future. OBJECTIVE: To provide a clinical and molecular characterization of 6 patients with IRDs with biallelic disease-causing variants in

The role of prostanoids in the retina and optic nerve in health and disease.

Prostaglandin D2 (PGD2), PGE2, and other prostanoids play important roles in retinal and optic nerve (ON) health and disease, including neovascularization, neuroinflammation, and neurodegeneration. We reviewed the synthesis, expression, regulation, mechanisms, and functions of the principal prostanoids (PGF2α, PGI2, TXA2, PGE2, and PGD2) in the eye. Prostanoids can be difficult to study in-vivo or ex-vivo because they are small, unstable molecules. To address this, we incorporated lipidomics data clarifying the prostanoids present in the mouse retina and ON, and mined single-cell transcriptomi
