BACKGROUND AND OBJECTIVE: Insulin bolus calculators are simple decision support software tools incorporated in most commercially available insulin pumps and some capillary blood glucose meters. Although their clinical benefit has been demonstrated, their utilisation has not been widespread and their performance remains suboptimal, mainly because of their lack of flexibility and adaptability. One of the difficulties that people with diabetes, clinicians and carers face when using bolus calculators is having to set parameters and adjust them on a regular basis according to changes in insulin req

In a recent work by our group involving a transcriptomics approach applied to the venom glands from Tityus stigmurus we identified a new family of peptides called Hypotensins (TSTI0006C) (Almeida et al., 2012). The cluster TSTI0006C was analyzed in the main 25 amino acid residues and named T. stigmurus Hypotensin (TistH), showing a molecular mass of 2.7 kDa, an absence of cysteines and the presence of two C-terminal proline residues, which are a bradykinin-potentiating peptide (BPP) signature. Here, we describe the homology modeling of the three-dimensional structure of TistH. In addition, we

Mutations in the LIS1 (Lissencephaly-1) gene underlie classical lissencephaly. This neurodevelopmental disorder is characterized by a loss of cortical gyri and improper laminar formation of the brain due to impaired neuronal migration. Patients with type 1 lissecephaly present with mental retardation and an increased risk of developing other disorders resulting from abnormal neurodevelopment, such as epilepsy. LIS1 is a dynamic protein implicated in numerous cellular mechanisms important for brain development. We have cloned and characterized the orthologs of LIS1 in the zebrafish. The zebrafi

Six genes in the 1.5 Mb region of chromosome 22 deleted in DiGeorge/22q11 deletion syndrome-Mrpl40, Prodh, Slc25a1, Txnrd2, T10, and Zdhhc8-encode mitochondrial proteins. All six genes are expressed in the brain, and maximal expression coincides with peak forebrain synaptogenesis shortly after birth. Furthermore, their protein products are associated with brain mitochondria, including those in synaptic terminals. Among the six, only Zddhc8 influences mitochondria-regulated apoptosis when overexpressed, and appears to interact biochemically with established mitochondrial proteins. Zdhhc8 has an

Recent progress in human neurogenetics has led to the discovery of new modes of inheritance and disease expression, including 1) stably inherited duplications in Charcot-Marie-Tooth disease type 1a, 2) dynamic mutations in fragile X syndrome and myotonic dystrophy, and 3) identical mutations with different phenotypes in fatal familial insomnia and Creutzfeldt-Jakob disease. The mechanisms by which known mutations of the amyloid precursor protein lead to early-onset Alzheimer's disease remain unexplained, despite hundreds of recent studies of beta-amyloid.

OBJECTIVE: To develop regulatory network to explore and model the regulatory relationships of protein biomarkers and classify different disease groups.
METHODS: Regulatory network is constructed to be a hopfield-like network with nodes representing biomarkers and directional connections to be regulations in between. The input to the network is the measured expression levels of biomarkers, and the output is the summation of regulatory strengths from other biomarkers. The network is optimized towards minimizing the energy function that is defined as the measure of the disagreement between the i

BACKGROUND: Blood feeding is essential for the maintenance of laboratory colonies and for experimental studies involving sand fly-parasite interactions. However, artificial feeding systems for Lutzomyia longipalpis, the principal vector of Leishmania infantum, the etiological agent of visceral leishmaniasis in the Americas, remain limited by low engorgement rates and methodological constraints in blood meal quantification.
OBJECTIVES: This study evaluated the feeding performance of Lu. longipalpis on different membranes in an artificial feeder and applied an image-based approach to quantify b

Plants coordinate their growth and developmental programs through dynamic communication in the core of the nucleus, enabling them to adapt to persistently high-temperature environments. Upon heat stress, plants primarily perceive and transduce thermal signals between the nucleus and other organelles, a process defined as nuclear-organellar communication. The published research on plants in response to heat environments mainly focuses on sensing heat physical signals from the plasma membrane (PM)/cytosol to the nucleus, eliciting nuclear transcriptional reprogramming and decoding organelle-deri

The European research landscape for developing new diagnostic, preventive, and therapeutic interventions is fraught with challenges. Scarcity of qualified talent, limited funding for laboratories and research infrastructure, geopolitical competition, and increasing complexity of data systems all require creativity, leadership, and vision to advance science and translate research results into effective solutions. This paper aimed to identify research priorities across the seven member organisations of the European Science for Health (EurSci4Health) community. The leadership of the membership or

BACKGROUND: Metabolic Dysfunction-Associated Steatotic Liver Disease (MASLD) represents the most prevalent chronic liver disease worldwide. The absence of approved pharmacotherapies is largely attributed to their profound molecular heterogeneity. The identification of novel hub genes in MASLD is therefore critical for unraveling the complex molecular mechanisms driving disease pathogenesis and progression.
METHODS: We employed an integrative systems-biology approach utilizing Weighted Gene Co-expression Network Analysis (WGCNA) followed by multi-algorithm machine learning (LASSO, Random Fores

Sarcopenia is characterized by a progressive loss of muscle strength and physical performance that frequently exceeds the decline predicted by muscle mass alone. This discrepancy indicates that functional deterioration is shaped not only by the force-generating capacity of skeletal muscle, but also by the tissues responsible for transmitting and integrating that force. In this Review, we examine sarcopenia through the biology of the muscle-tendon unit, encompassing skeletal muscle, tendon, and their specialized interface. We discuss how systemic aging processes, including metabolic dysfunction

BACKGROUND: Cancer has traditionally been framed as a cell-intrinsic genetic disease and, more recently, as a disorder of the tumor microenvironment (TME). An emerging third view reframes malignancy as a systems-level neuroimmune disorder, in which neurons, glia, immune cells, and tumor cells form a continuously communicating network, the tumor neuroimmune niche.
SUMMARY: This review synthesizes evidence that the nervous system actively regulates tumor initiation, proliferation, angiogenesis, invasion, and metastasis through autonomic, sensory, and neuroendocrine inputs, while tumors in turn
