LPS-Induced TNF-α and IL-1 Signaling in Periodontitis: Molecular Mechanisms and Therapeutic Opportunities.

Periodontitis is a chronic, dysbiosis-driven inflammatory disease, that, unlike gingivitis, results in irreversible, tooth-supporting soft tissue and bone loss. Clarifying the molecular pathways that sustain the host inflammatory response is therefore central to improving disease management. Lipopolysaccharide (LPS), a major pathogen-associated molecular pattern of Gram-negative bacteria, is one important-though not the sole-microbial stimulus that drives inflammation in the polymicrobial, dysbiotic periodontal environment. This narrative review summarizes how LPS-initiated signaling promotes

Search for Additional Pathogenic Variants to Explain Variation in PMP22-Related Neuropathies.

BACKGROUND AND OBJECTIVES: The aim of this study was to investigate whether the considerable phenotypic variation in Charcot-Marie-Tooth disease type 1A (CMT1A) and hereditary neuropathy with liability to pressure palsies (HNPP) is due to additional pathogenic coding variants in severely affected patients, by screening a panel of neuropathy-related genes. METHODS: In this cross-sectional study, the extremes of the spectrum of 742 patients with genetically confirmed CMT1A and HNPP were selected, based on disability as assessed using the Overall Neuropathy Limitation Scale (ONLS). The ONLS data

Addressing disparities in oral health access and outcomes for aging adults in the United States.

Oral health is essential for the overall well-being of aging adults, yet significant disparities persist in the United States, contributing to malnutrition, reduced quality of life, and social isolation. Despite advancements in preventive dentistry, older adults continue to face substantial oral health challenges. This study reviewed the current state of oral health among aging adults in the United States, analyzing national health surveys and existing preventive dentistry protocols to assess their effectiveness. National data indicated that a high percentage of older adults experience tooth l

Mechanosensitive Piezo channels in mineralized tissues: emerging roles in osteodental adaptation and disease.

Bone and dental tissues are highly mineralized and mechanically sensitive hard tissues. They detect and respond to mechanical forces via mechanosensitive Piezo channels, modulating physiological and pathological processes. While Piezo mechanobiology has been explored, systematic comparison of their roles across bone and dental tissues, particularly their potential crosstalk in adaptation and disease, remains underexamined in existing reviews. This review consolidates recent advances in Piezo channel biology, clarifying their structural properties, tissue-specific distribution, and functional r

ADGRU: Adaptive DenseNet with gated recurrent unit for automatic diagnosis of periodontal bone loss and stage periodontitis with tooth segmentation mechanism.

BACKGROUND: Periodontics and gingivitis are two of the most widely prevalent illnesses that affect people nowadays. The sixth most common disease in the world is periodontitis, and detecting periodontal bone loss is essential in the earlier condition and is crucial for the development of the proper diagnosis. Early bone loss detection can be assisted by using computer-assisted radiography examination. Understanding disease progression helps to select the most effective treatment action. OBJECTIVES: An effective deep model is suggested to detect periodontal bone loss at an earlier stage for pr

Erythropoietin receptor signal is crucial for periodontal ligament stem cell-based tissue reconstruction in periodontal disease.

Alveolar bone loss caused by periodontal disease eventually leads to tooth loss. Periodontal ligament stem cells (PDLSCs) are the tissue-specific cells for maintaining and repairing the periodontal ligament, cementum, and alveolar bone. Here, we investigated the role of erythropoietin receptor (EPOR), which regulates the microenvironment-modulating function of mesenchymal stem cells, in PDLSC-based periodontal therapy. We isolated PDLSCs from patients with chronic periodontal disease and healthy donors, referred to as PD-PDLSCs and Cont-PDLSCs, respectively. PD-PDLSCs exhibited reduced potency

A case report of adult Langerhans cell histiocytosis and review of the literature.

Langerhans cell histiocytosis (LCH) is an uncommon proliferative disease with an unknown cause. Its clinical manifestations vary and can involve a variety of organs. To diagnose LCH, radiographs, histopathological and immunohistochemical findings are essential. The gold standard for a definite diagnosis is positive CD1a/CD207 and S100 in the immunohistochemical results. Different treatment plans are available for patients struggling with LCH. To our knowledge, the LCH incidence rate is about 8.9 in one million children and 1-2 cases in one million adults. Our case shows the importance of early

Morc2a p.S87L mutant mice develop peripheral and central neuropathies associated with neuronal DNA damage and apoptosis.

The microrchidia (MORC)-family CW-type zinc finger 2 (MORC2) gene is related to DNA repair, adipogenesis and epigenetic silencing via the human silencing hub (HUSH) complex. MORC2 missense mutation is known to cause peripheral neuropathy of Charcot-Marie-Tooth disease type 2 Z (CMT2Z). However, there have been reports of peripheral and central neuropathy in patients, and the disease has been co-categorized with developmental delay, impaired growth, dysmorphic facies and axonal neuropathy (DIGFAN). The etiology of MORC2 mutation-mediated neuropathy remains uncertain. Here, we established and an

Characteristics of Clinical and Electrophysiological Pattern in a Large Cohort of Chinese Patients With Charcot-Marie-Tooth 4C.

The "Src homology 3 (SH3) domain and tetratricopeptide repeats 2" (SH3TC2) gene is mutated in individuals with Charcot-Marie-Tooth disease (CMT) and considered relevant to a demyelinating or intermediate subtype of CMT disease, CMT4C. In this study, we screened a cohort of 465 unrelated Chinese CMT patients alongside 650 controls. We used Sanger, next-generation, or whole-exome sequencing to analyze SH3TC2 and other CMT-related genes and identified 12 SH3TC2 variants (eight novel) in seven families. Of the eight novel variants, seven were likely pathogenic (c.280-2 A > G, c.732-1 G > A, c.1177

A Novel G542X CFTR Rat Model of Cystic Fibrosis Is Sensitive to Nonsense Mediated Decay.

Nonsense mutations that lead to the insertion of a premature termination codon (PTC) in the cystic fibrosis transmembrane conductance regulator (CFTR) transcript affect 11% of patients with cystic fibrosis (CF) worldwide and are associated with severe disease phenotype. While CF rat models have contributed significantly to our understanding of CF disease pathogenesis, there are currently no rat models available for studying CF nonsense mutations. Here we created and characterized the first homozygous CF rat model that bears the CFTR G542X nonsense mutation in the endogenous locus using CRISPR/

DYNC1H1-related disorders: A description of four new unrelated patients and a comprehensive review of previously reported variants.

Heterozygous variants in the DYNC1H1 gene have been associated chiefly with intellectual disability (ID), malformations in cortical development (MCD), spinal muscular atrophy (SMA), and Charcot-Marie-Tooth axonal type 20 (CMT), with fewer reports describing other intersecting phenotypes. To better characterize the variable syndromes associated with DYNC1H1, we undertook a detailed analysis of reported patients in the medical literature through June 30, 2019. In sum we identified 200 patients from 143 families harboring 103 different DYNC1H1 variants, and added reports for four unrelated patien
