The anti-allodynic effects of amitriptyline, gabapentin, and lidocaine in a rat model of neuropathic pain.

UNLABELLED: The management of patients with neuropathic pain is challenging. There are only a few reports regarding the acute effects of the commonly used adjuvant drugs amitriptyline (AMI), gabapentin (GBP), and lidocaine (LDC) on neuropathic pain behaviors in animal models. Thus, the purpose of this study was to investigate the acute effects of AMI, GBP, and LDC on behavioral signs of mechanical allodynia and the site of action of these drugs using a rat model of neuropathic pain. Under general anesthesia with halothane, neuropathic injury was produced in rats by tightly ligating the left L5

Movement disorders associated with the use of gabapentin.

PURPOSE: We report two cases of unusual movement disorders associated with the use of gabapentin (GBP) in patients being treated for epilepsy who were otherwise neurologically intact. METHODS: We describe two cases of unusual movement disorders associated with the use of GBP. RESULTS: There were significant differences in the clinical findings between the two cases. In the first case, movements were very pronounced and the patient was in oculogyric crisis. Movements in the second case were quite subtle but nonetheless problematic for the patient. In each case, discontinuation of GBP led to r

Adulterants causing false negatives in illicit drug testing.

Illicit-drug users may attempt to falsify results by in vitro adulteration of specimens. We investigated eight additives (NaCl, Visine, handsoap, Drano, bleach, vinegar, golden-seal tea, and lemon juice) claimed by drug users to invalidate enzyme immunoassay (EIA) drug assays. We also analyzed adulterated urine specimens to determine if they could be identified, adding adulterants at several concentrations to 222 EIA-positive specimens confirmed by gas chromatography and mass spectrometry (GC/MS) to contain illicit drugs. To identify adulterated urines, we monitored pH, relative density, and u

Short-chain fatty acids differentially regulate feed intake and hypothalamic appetite-related genes in layer-type chicks.

INTRODUCTION: Activation of hypothalamic neurons by short-chain fatty acids (SCFAs) has been recognized as one of the mechanisms of appetite regulation in mammals. However, their central effects on hypothalamic neuropeptides have not yet been fully investigated in chicks. In the present study, feed intake and mRNA expression of neuropeptides in chicks were evaluated under the influence of SCFAs. METHODS: The experiments included intracerebroventricular (ICV) injection of SCFAs into five-day-old chicks. Feed intake was recorded up to 2 h post-injection, and PCR was used to determine the gene e

Exploring fecal microbial activity in zoo felids of varying body mass on a similar diet.

Under human care, felids are typically fed similar diets, unlike wild counterparts whose diets vary by body mass and ecology. This study evaluated fecal microbiota and fermentation products in 18 zoo felids from Pairi Daiza Zoo, Belgium, grouped by body mass: under 100 kg ("small") and over 100 kg ("large"), with 9 animals in each group. Fresh feces were collected from the rectum under anesthesia. Microbial composition was assessed via 16S rRNA gene sequencing, while the fecal volatile fatty acids were quantified using gas chromatography. At the phylum level, regardless of body mass, the gut m

Reducing residual solvent levels in poly (D, L-lactic-co-glycolic acid) microspheres: a roadmap for scalable industrial production.

OBJECTIVE: Poly (D, L-lactic-co-glycolic acid) (PLGA) microspheres have garnered significant attention as biocompatible and biodegradable carriers for sustained drug delivery. However, the production of PLGA microspheres typically involves organic solvents, such as ethyl acetate and benzyl alcohol. Residual solvents are undesirable given their potential toxicity and adverse effects on product stability. Effective solvent removal is critical for ensuring the safety and functionality of microspheres. METHOD: In this study, 12 formulations were designed by altering the conditions of solvent extr

The Role of Changes in the Proportion of Fecal Short-Chain Fatty Acids on the Severity of Hepatic Encephalopathy in Cirrhosis Patients.

BACKGROUND: Short-chain fatty acids (SCFA) are the main metabolites of the intestinal microbiota, which play a role as colonocyte trophic factors and maintain the integrity of the gastrointestinal tract and blood-brain barrier. Microbiota dysbiosis that occurs in cirrhosis reduces SCFA production and plays a role in the pathogenesis of hepatic encephalopathy (HE). This study aims to compare the amount and composition of fecal SCFA in patients with cirrhosis, with and without HE. METHODS: This research is a cross-sectional study at the Hepatobiliary Clinic and Integrated Procedure Room, Dr. Ci

Towards syngas biorefineries: The potential of microbial consortia for syngas valorisation.

Gasification has emerged as a promising platform to cope with recalcitrant organic waste within the framework of biomass-based biorefineries, producing syngas that can be fermented into valuable bioproducts. Despite its potential, syngas fermentation is based predominantly on pure cultures, which faces significant challenges, including the limited portfolio of generated compounds (primarily acetate and ethanol) and their low productivity. To address these bottlenecks, the potential of microbial consortia as effective platforms for syngas conversion has been evaluated. Syngas biomethanation usi

Development of a CRISPR/Cas12a genome editing toolbox in Kluyveromyces marxianus and its application in succinic acid biosynthesis.

Kluyveromyces marxianus is a promising thermotolerant yeast for industrial biotechnology, but lacks efficient genome engineering tools. A CRISPR/Cas12a genome editing toolbox for K. marxianus was developed for the first time in this study. A plasmid-free transient system achieved single-gene knockout efficiencies of about 50 %-100 % in Δku70 strain. Even with homology arms as short as 35 bp, the knockout efficiency remained 66.67 %. Chromosomal integration of Cas12a enabled single-to-triple fragment knock-ins efficiency of 82.93-85.70 % and 94.50 % for large fragment (>5 kb) integrations. Appl

Synergistic acetate-butyrate co-utilization enhances caproate production by Clostridium kluyveri CICC 24509 under high substrate loads.

Microbial chain elongation presents a compelling circular economy strategy for converting low-value waste streams into valuable chemicals like caproate. However, realizing its full potential is hampered by challenges, including limited microbial tolerance to industrially relevant high substrate concentrations and suboptimal electron donor-acceptor utilization. This study addresses these limitations by evaluating the performance of a previously uncharacterized chain-elongating bacterium, Clostridium kluyveri CICC 24509, under high-substrate conditions using ethanol as the electron donor. We sys

Precisely Programmable Degradation and Drug Release Profiles in Triblock Copolyether Hydrogels with Cleavable Acetal Pendants.

Hydrogels hold significant promise as drug delivery systems due to their distinct advantage of sustained localized drug release. However, the challenge of regulating the initial burst release while achieving precise control over degradation and drug-release kinetics persists. Herein, we present an ABA-type triblock copolymer-based hydrogel system with precisely programmable degradation and release kinetics. The resulting hydrogels were designed with a hydrophilic poly(ethylene oxide) midblock and a hydrophobic end-block composed of polyethers with varying ratios of ethoxyethyl glycidyl ether a
