Human papillomavirus (HPV) genotype distribution in Malaysia: A systematic review.

BACKGROUND: Human papillomavirus (HPV) is a key etiological factor in cervical cancer in both Malaysia and globally. It continues to pose a significant public health challenge. This systematic review aims to delineate the distribution of HPV genotypes across different demographics in Malaysia to inform targeted prevention strategies. METHODS: We conducted a systematic review following PRISMA guidelines, analyzing observational studies published from 2000 onward that reported HPV genotypes in cervicovaginal samples from Malaysian women. The review utilized PubMed, SCOPUS, The Cochrane Library,

Prioritizing the risk of cervical cancer: findings from the SCCUT Multicentre Initiative.

Cervical cancer (CC) remains a significant global health burden, ranking eighth in incidence and ninth in cancer-related mortality among women worldwide. Persistent infection with high-risk human papillomavirus (HPVhr) is the primary etiological factor in CC development. The SCCUT program in Romania was designed to enhance access to primary screening and assess HPVhr prevalence among women aged 25-65 years in the South-Muntenia region. Between August 2022 and November 2023, a total of 36,813 women were enrolled in the SCCUT program after providing informed consent. Based on age, participants u

Short-term high-dose gemcitabine induces PERK-mediated immunogenic cell death and potentiates antitumor immunity in bladder cancer.

BACKGROUND: Gemcitabine is a widely used intravesical chemotherapeutic agent for non-muscle-invasive bladder cancer (NMIBC), yet its efficacy remains suboptimal, with high recurrence rates. While traditionally considered a cytotoxic agent, emerging evidence suggests that certain chemotherapeutics may exert immunomodulatory effects. This study investigated whether gemcitabine induces immunogenic cell death (ICD) and explored the underlying mechanisms involving endoplasmic reticulum (ER) stress. METHODS: A retrospective analysis was conducted to assess the association between gemcitabine-relate

Vulval squamous cell carcinoma: a review.

Vulval squamous cell carcinoma (VSCC) represents approximately 90% of all vulval cancers. Despite its rarity relative to other gynaecological or skin malignancies, the recent growing body of epidemiological and clinical evidence has resulted in important refinements to national guidelines. This review offers a comprehensive overview of the current evidence regarding the epidemiology, clinical features, diagnosis, and management of VSCC. There are two distinct aetiological pathways currently identified in the pathogenesis of VSCC. Human papillomavirus (HPV)-dependent VSCC is commonly found in y

[Psycho-oncological and psychosexual consequences of penile cancer: what can we offer our patients?].

Penile cancer is a rare but highly distressing form of cancer in men, associated with both physical and psychosocial challenges. With an incidence of 1-10 cases per 100,000 men, its prevalence varies worldwide, with higher rates observed in developing countries. The disease often leads to physical deficits, including loss of the penis and impairments in sexual life, which can result in significant psychological burdens such as anxiety, depression, and a diminished self-esteem. Stigmatization and social isolation play a central role, especially in cultures where masculinity and sexuality are cl

COVID-19 among lung cancer patients: Data from a real-life prospective French multicentric study.

BACKGROUND: COVID-19 started to spread early in 2020, the precise year that lung cancer (LC) patients were recruited into the prospective epidemiological cohort KBP-2020-CPHG in French hospitals. This provides a unique opportunity to study COVID-19 incidence, survival risk factors, and overall prognosis. METHODS: COVID data was collected before vaccination was made available. Clinical characteristics were compared (COVID vs non-COVID), incidence rate ratios were calculated based on clinical characteristics, survival (1 and 3 months) was estimated and the impact of COVID-19 on the overall prog

Evidence to Support Change of Clinical Pathway Following Colposcopy Treatment for Cervical Intraepithelial Neoplasia in Canada.

OBJECTIVES: Human papillomavirus (HPV) testing can be incorporated into the post-treatment pathway of cervical intraepithelial neoplasia (CIN) to confirm disease-free status. To inform a post-treatment strategy based on risk of recurrence, we modelled disease and economic outcomes. METHODS: The current Alberta, Canada, post-treatment care pathway-cytology testing with colposcopy assessment-was compared with 6 other scenarios incorporating cytology, HPV testing, or both tests at different time points in a modelling study based on a microsimulation program. Input parameter values for the screen

Anal cancer screening and prevention: a review for dermatologists.

The incidence of and mortality from anal cancer, predominantly squamous cell carcinoma (SCC), have been increasing since the 1980s, during an era when many common malignancies have seen decreases in mortality. Dermatologists may be more likely to see patients at an increased risk for anal SCC, such as those living with HIV, MSM and those presenting for management of anogenital warts, yet there is little guidance in the field on how to manage these patients. We underwent a project to review the evidence surrounding screening and prevention of anal SCC. HPV vaccination, the main preventative mea

Novel oligomannose liposome-DNA complex DNA vaccination efficiently evokes anti-HPV E6 and E7 CTL responses.

The aim of this study was to establish an efficient human papilloma virus (HPV) type 16-targeting cancer immunotherapy. Persistent high-risk HPV infection causes cervical intra-epithelial neoplasia (CIN) and subsequent cervical carcinoma. HPV type16 (HPV16) is one of the common carcinogenic types and is found in about 50% of invasive cervical carcinomas. HPV16-derived viral proteins E6 and E7 are expressed in cancerous cells through the progression of the disease and have a role in carcinogenesis but are not expressed in normal cells. Thus, these proteins are regarded as ideal antigens for cer

Low human papillomavirus prevalence in head and neck cancer: results from two large case-control studies in high-incidence regions.

BACKGROUND: Recent studies support an important role for human papillomavirus (HPV) in a subgroup of head and neck squamous cell carcinomas (HNSCC). We have evaluated the HPV deoxyribonucleic acid (DNA) prevalence as well as the association between serological response to HPV infection and HNSCC in two distinct populations from Central Europe (CE) and Latin America (LA). METHODS: Cases (n = 2214) and controls (n = 3319) were recruited from 1998 to 2003, using a similar protocol including questionnaire and blood sample collection. Tumour DNA from 196 fresh tissue biopsies was analysed for mult

Prevalence and type distribution of human papillomavirus in women with cervical lesions in Liaoning Province, China.

INSTRUCTION: Type-specific persistence of human papillomavirus (HPV) infection can cause invasive cervical cancer. The distribution and prevalence of HPV genotypes depend on the geographic region and on demographic factors. METHODS: This study aimed to investigate the prevalence and distribution of HPV genotypes in uterine cervical lesions in Liaoning Province, China. A total of 1444 cervical swabs from patients with cervical cancer (CC, n = 134), cervical intraepithelial neoplasia (CIN) II/III (n = 517), and CIN I (n = 180) were detected for HPV genotypes using the PGMY09/11 primer system an

Cervical cancer screening by cotesting method for Vietnamese women 25-55 years old: a cost-effectiveness analysis.

BACKGROUND: Cervical cancer (CC) is preventable through regular screening and vaccination against human papillomavirus (HPV). However, CC remains a significant public health issue in low-income and middle-income countries (LMICs) like Vietnam, where financial constraints hinder the widespread implementation of HPV vaccination and screening programmes. Currently, Vietnam lacks both a national CC screening intervention and an HPV vaccination programme for women and girls. To date, cost-effectiveness studies evaluating CC screening methods in Vietnam remain limited. OBJECTIVES: To evaluate the c

Lysosomal damage due to cholesterol accumulation triggers immunogenic cell death.

Cholesterol serves as a vital lipid that regulates numerous physiological processes. Nonetheless, its role in regulating cell death processes remains incompletely understood. In this study, we investigated the role of cholesterol trafficking in immunogenic cell death. Through cell-based drug screening, we identified two antidepressants, sertraline and indatraline, as potent inducers of the nuclear translocation of TFEB (transcription factor EB). Activation of TFEB was mediated through the autophagy-independent lipidation of MAP1LC3/LC3 (microtubule associated protein 1 light chain 3). Both com

Analogies between HPV Behavior in Oral and Vaginal Cavity: Narrative Review on the Current Evidence in the Literature.

Human genital papilloma virus infection is the most prevalent sexually transmitted infection in the world. It is estimated that more than 75% of sexually active women contract this infection in their lifetime. In 80% of young women, there is the clearance of the virus within 18-24 months. In developed countries, oral squamous cell carcinoma (OSCC) is now the most frequent human papilloma virus (HPV)-related cancer, having surpassed cervical cancer, and it is predicted that by 2030 most squamous cell carcinomas will be the HPV-related rather than non-HPV-related form. However, there are current

Human Sapovirus Replication in Human Intestinal Enteroids.

Human sapoviruses (HuSaVs), like human noroviruses (HuNoV), belong to the Caliciviridae family and cause acute gastroenteritis in humans. Since their discovery in 1976, numerous attempts to grow HuSaVs in vitro were unsuccessful until 2020, when these viruses were reported to replicate in a duodenal cancer cell-derived line. Physiological cellular models allowing viral replication are essential to investigate HuSaV biology and replication mechanisms such as genetic susceptibility, restriction factors, and immune responses to infection. In this study, we demonstrate replication of two HuSaV str

A Systematic Review of the Emerging Role of Immune Checkpoint Inhibitors in Metastatic Castration-resistant Prostate Cancer: Will Combination Strategies Improve Efficacy?

CONTEXT: The role of immune checkpoint inhibition (ICI) in the treatment of prostate cancer (PC) still remains elusive. It has been proposed that combination of ICI with other molecules increases the efficacy of immunotherapy in PC. OBJECTIVE: To systematically review the literature to assess the potential role of ICI in combination with additional therapies for the management of metastatic castration-resistant PC (mCRPC). EVIDENCE ACQUISITION: A systematic review using Medline and scientific meeting records was carried out in September 2020 according to the Preferred Reporting Items for Sys

Characterization of Ty21a immunostimulatory effects in the mouse bladder.

Intravesical treatment with Salmonella enterica Ty21a, an oral typhoid-fever vaccine, has shown therapeutic potential against bladder tumors mainly through local immune-cell recruitment, particularly CD8+ T-cells. However, the mechanisms underlying its efficacy and the impact of bacterial formulation remain unclear. Here, we show that increased immune-cell infiltration was neither associated with modification in blood vessel density nor the generation of high endothelial venules, but rather with a transient increase in local vessel permeability, requiring live bacteria. Giving prior evidence t

Redesign of a rapid, low-cost HPV typing assay to support risk-based cervical screening and management.

Accelerated cervical cancer control will require widespread human papillomavirus (HPV) vaccination and screening. For screening, sensitive HPV testing with an option of self-collection is increasingly desirable. HPV typing predicts risk of precancer/cancer, which could be useful in management, but most current typing assays are expensive and/or complicated. An existing 15-type isothermal amplification assay (AmpFire, Atila Biosystems, USA) was redesigned as a 13-type assay (ScreenFire) for public health use. The redesigned assay groups HPV types into four channels with differential cervical ca

Potential therapeutic approaches of microRNAs for COVID-19: Challenges and opportunities.

The coronavirus disease 2019 (COVID-19) emerges as current outbreak cause by Novel Severe Acute Respiratory Syndrome Corona Virus-2 (SARS-CoV-2). This infection affects respiratory system and provides uncontrolled systemic inflammatory response as cytokine storm. The main concern about SARS-CoV-2 pandemic is high viral pathogenicity with no specific drugs. MicroRNAs (miRs) as small non-coding RNAs (21-25 nt) regulate gene expression. The SARS-CoV-2 encoded-miRs affect human genes that involved in transcription, translation, apoptosis, immune response and inflammation. Also, they alter self-ge

Extracorporeal photochemotherapy induces bona fide immunogenic cell death.

Extracorporeal photochemotherapy (ECP) is employed for the management of cutaneous T cell lymphoma (CTCL). ECP involves the extracorporeal exposure of white blood cells (WBCs) to a photosensitizer, 8-methoxypsoralen (8-MOP), in the context of ultraviolet A (UVA) radiation, followed by WBC reinfusion. Historically, the therapeutic activity of ECP has been attributed to selective cytotoxicity on circulating CTCL cells. However, only a fraction of WBCs is exposed to ECP, and 8-MOP is inactive in the absence of UVA light, implying that other mechanisms underlie the anticancer effects of ECP. Recen
