Quasi-intrinsic thiobase derivatives as potential targeted photosensitizers in two-photon photodynamic therapy.

In this study, a set of potential quasi-intrinsic photosensitizers for two-photon photodynamic therapy (PDT) are proposed based on the unnatural 2-amino-8-(1'-β-ᴅ-2'-deoxyribofuranosyl)-imidazo[1,2-ɑ]-1,3,5-triazin-4(8H)-one (P), which is paired with the 6-amino-5-nitro-3-(1'-β-ᴅ-2'-deoxyribofuranosyl)-2(1H)-pyridone (Z) and can specifically recognize breast and liver cancer cells. Herein, the effects of sulfur substitution and electron-donating/electron-withdrawing groups on the photophysical properties in aqueous solution are systematically investigated. The one- and two-photon absorption sp

Pathogenic Variants Associated with Epigenetic Control and the NOTCH Pathway Are Frequent in Classic Hodgkin Lymphoma.

Classic Hodgkin lymphoma (cHL) constitutes a B-cell neoplasm derived from germinal center lymphocytes. Despite high cure rates (80-90%) obtained with the current multiagent protocols, a significant proportion of cHL patients experience recurrences, characterized by a lower sensitivity to second-line treatments. The genomic background of chemorefractory cHL is still poorly understood, limiting personalized treatment strategies based on molecular features. In this study, using a targeted next-generation sequencing (NGS) panel specifically designed for cHL research, we compared chemosensitive and

Kidney Amyloidosis: Updates on Pathogenesis and Therapeutic Frontiers.

BACKGROUND: Amyloidosis includes a diverse group of rare diseases characterized by the misfolding of native or mutant proteins, leading to extracellular accumulation in various organs. While 42 proteins have been identified to date, their distribution differs between systemic and localized forms. SUMMARY: Mass spectrometry analysis of tissue samples in the USA shows immunoglobulin light chain (AL) amyloidosis as the most prevalent systemic type, followed by transthyretin (ATTR). Heart and kidney involvements are common. Although there are 14 recognized types of kidney-related amyloidosis, cli

Resolvin D2-G-Protein Coupled Receptor 18 Enhances Bone Marrow Function and Limits Steatosis and Hepatic Collagen Accumulation in Aging.

Aging is associated with nonresolving inflammation and tissue dysfunction. Resolvin D2 (RvD2) is a proresolving ligand that acts through the G-protein-coupled receptor called GPR18. Unbiased RNA sequencing revealed increased Gpr18 expression in macrophages from old mice, and in livers from elderly humans, which was associated with increased steatosis and fibrosis in middle-aged (MA) and old mice. MA mice that lacked GPR18 on myeloid cells had exacerbated steatosis and hepatic fibrosis, which was associated with a decline in Mac2+ macrophages. Treatment of MA mice with RvD2 reduced steatosis an

Cytokines in Spondyloarthritis and Inflammatory Bowel Diseases: From Pathogenesis to Therapeutic Implications.

Spondyloarthritis and inflammatory bowel diseases are chronic immune disorders of the joints and the gut that often coexist in the same patient, increasing the burden of each disorder, worsening patients' quality of life, and influencing therapeutic strategies. Genetic predisposition, environmental triggers, microbiome features, immune cell trafficking, and soluble factors such as cytokines contribute to the pathogenesis of both articular and intestinal inflammation. Most of the molecular targeted biological therapies developed over the last two decades were based on evidence that specific cyt

The circulating proteomic signature of alcohol-associated liver disease.

Despite being a leading cause of advanced liver disease, alcohol-associated liver disease (ALD) has no effective medical therapies. The circulating proteome, which comprises proteins secreted by different cells and tissues in the context of normal physiological function or in the setting of disease and illness, represents an attractive target for uncovering novel biology related to the pathogenesis of ALD. In this work, we used the aptamer-based SomaScan proteomics platform to quantify the relative concentration of over 1300 proteins in a well-characterized cohort of patients with the spectrum

Case Report: Successful Therapy of Spontaneously Occurring Canine Degenerative Lumbosacral Stenosis Using Autologous Adipose Tissue-Derived Mesenchymal Stem Cells.

The management of degenerative lumbosacral stenosis (DLSS) in dogs usually requires aggressive, costly surgical treatments that may themselves present complications, while do not fully resolve the symptoms of the disease. In this study, the dog diagnosed with severe DLSS, with hind limb paresis, was treated using a new and least invasive treatment. Cultured autologous adipose tissue-derived mesenchymal stem cells (AT-MSCs) were injected bilaterally at the level of L7-S1, in the vicinity of the external aperture of the intervertebral foramen of DLSS patient. In the previously described treatmen

The diagnosis of lung cancer in the era of interventional pulmonology.

Advances in bronchoscopic and other interventional pulmonology technologies have expanded the sampling procedures pulmonologist can use to diagnose lung cancer and accurately stage the mediastinum. Among the modalities available to the interventional pulmonologist are endobronchial ultrasound-guided transbronchial needles aspiration (EBUS-TBNA) and transoesophageal bronchoscopic ultrasound-guided fine-needle aspiration (EUS-B-FNA) for sampling peribronchial/perioesophageal central lesions and for mediastinal lymph node staging, as well as navigational bronchoscopy and radial probe endobronchia

Severe adverse immunologic reaction in a patient with glioblastoma receiving autologous dendritic cell vaccines combined with GM-CSF and dose-intensified temozolomide.

Therapeutic vaccination of patients with cancer-targeting tumor-associated antigens is a promising strategy for the specific eradication of invasive malignancies with minimal toxicity to normal tissues. However, as increasingly potent modalities for stimulating immunologic responses are developed for clinical evaluation, the risk of inflammatory and autoimmune toxicities also may be exacerbated. In this report, we describe the induction of a severe (grade 3) immunologic reaction in a patient with newly diagnosed glioblastoma (GBM) receiving autologous RNA-pulsed dendritic cell (DC) vaccines ad

Autoimmune pancreatitis: an underdiagnosed autoimmune disease with clinical, imaging and serological features.

Since Sarles et al. [Sarles H, Sarles JC, Muratore R, Guien C. Chronic inflammatory sclerosis of the pancreas-an autonomous pancreatic disease? Am J Dig Dis 1961; 6: 688-698.] reported a case of pancreatitis associated with hypergammaglobulinemia, many cases have been described, which led to the current concept of "autoimmune pancreatitis (AIP)". Lymphoplasmacytic infiltration and fibrosis on histology together with elevated IgG levels or the presence of autoantibodies on laboratory examinations supported the concept of AIP. In recent years, based on histological and immunohistochemical examin

Naphthalocyanine-reconstituted LDL nanoparticles for in vivo cancer imaging and treatment.

Low density lipoproteins (LDLs) are naturally occurring nanoparticles that are biocompatible, biodegradable and non-immunogenic. Moreover, the size of LDL particle is precisely controlled (approximately 22 nm) by its apoB-100 component, setting them apart from liposomes and lipid micelles. LDL particles have long been proposed as a nanocarrier for targeted delivery of diagnostics and therapeutics to LDL receptor (LDLR)-positive cancers. Here, we report the design and synthesis of a novel naphthalocyanine (Nc)-based photodynamic therapy (PDT) agent, SiNcBOA, and describe its efficient reconstit

Bio-hybrid scaffolds combining polyvinyl alcohol and decellularized articular cartilage for the treatment of focal chondral lesions in hemophilic patients.

INTRODUCTION: Hemophilic arthropathy (HA) is a major complication of hemophilia, being caused by recurrent joint bleeding (hemarthrosis) which leads to iron accumulation in joints and subsequent damage to the articular cartilage (AC) and subchondral bone. Current treatments slow osteochondral degradation but do not promote cartilage regeneration. Tissue engineering offers a promising alternative for addressing AC damage in HA. METHODS: This study developed bio-hybrid scaffolds based on polyvinyl alcohol (PVA) combined with decellularized human AC to enhance bioactivity and mechanical support.

Therapeutic oxygen delivery by perfluorocarbon-based colloids.

After the protocol-related indecisive clinical trial of Oxygent, a perfluorooctylbromide/phospholipid nanoemulsion, in cardiac surgery, that often unduly assigned the observed untoward effects to the product, the development of perfluorocarbon (PFC)-based O2 nanoemulsions ("blood substitutes") has come to a low. Yet, significant further demonstrations of PFC O2-delivery efficacy have continuously been reported, such as relief of hypoxia after myocardial infarction or stroke; protection of vital organs during surgery; potentiation of O2-dependent cancer therapies, including radio-, photodynamic

In situ swelling of hydrogel microneedles facilitates suprachoroidal space expansion for targeted delivery of TOP-V122 to the posterior segment.

Posterior segment eye diseases are leading causes of vision impairment, yet current treatments such as intravitreal injections require frequent administration and carry risks of complications, including endophthalmitis and retinal detachment, highlighting the need for minimally invasive and controllable alternatives. Here, we present an improved hydrogel-forming microneedle (HFMN) platform composed of 20% poly(methyl vinyl ether-alt-maleic acid) and 7.5% poly(ethylene glycol) for minimally invasive, targeted drug delivery to the posterior segment via the suprachoroidal space. Engineered mesenchymal stem cell-derived small extracellular vesicles delivering miR-124 suppress colorectal cancer progression through EZH2 downregulation and attenuation of STAT3 signaling.

Colorectal cancer (CRC) progression is driven by dysregulated molecular pathways and epigenetic alterations, including enhancer of zeste homolog 2 (EZH2)-mediated histone methylation and STAT3 hyperactivation. While microRNA-124 (miR-124) acts as a tumor suppressor by targeting EZH2, its clinical translation is hindered by delivery challenges. Here, we engineered mesenchymal stem cell-derived small extracellular vesicles (MSC-sEVs) to deliver miR-124, exploiting their tumor-homing capacity and biocompatibility to suppress CRC progression. Bioinformatic analysis of clinical datasets revealed mi

The efficacy and safety of MSCs in GVHD prevention and the treatment of SR-aGVHD: a systematic review and meta-analysis of randomized controlled trials.

BACKGROUND: Hematopoietic stem cell transplantation (HSCT) is a cornerstone in the treatment of hematological disorders. However, its application is frequently complicated by acute and chronic graft-versus-host disease (aGVHD/cGVHD), pathological conditions in which donor-derived immune cells attack host tissues. With suboptimal survival rates and limited therapeutic options, GVHD remains a major clinical challenge. Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic modality due to their immunomodulatory capabilities, yet standardized protocols for their use in preventing or

YTHDF2-orchestrated m6A methylation of BECN1 induces Scoparone-mediated hepatic stellate cell ferroptosis to attenuate liver fibrosis.

BACKGROUND: Liver fibrosis represents a dynamically reversible pathological process arising as an adaptive repair response to chronic hepatic insults. Scoparone (SCO), an active constituent of artemisia, has demonstrated therapeutic potential across diverse liver diseases, but its antifibrotic mechanism remains unclear. PURPOSE: This study aims to elucidate the molecular mechanism by which SCO ameliorates liver fibrosis through m6A epitranscriptomic regulation of hepatic stellate cell (HSC) ferroptosis. METHODS: Murine liver fibrosis models and human HSC cells were employed to evaluate the t

The role of Ca2+/calmodulin-dependent protein kinase II in oxaliplatin resistance of colorectal cancer: In vitro and in vivo studies.

BACKGROUND: Oxaliplatin (Oxa) resistance is a major barrier in colorectal cancer (CRC) treatment. The Ca2+/CaM/CaMKII pathway is involved in tumor progression, but its role in chemoresistance remains unclear. METHODS: Oxa-resistant HCT116 and HT29 sublines (HCT116-Oxa and HT29-Oxa) were established through gradual dose escalation. The activation status of the Ca2+/CaM/CaMKII pathway was assessed by measuring intracellular Ca2+ levels and phosphorylation of CaMKII. Functional studies such as CCK-8, clonogenic assays, and flow cytometry-based apoptosis analysis were conducted to assess the impa
