Aliphatic chains as physical realizations of one-dimensional spin chains supporting magnetically silent spin waves.

Spin waves are collective spin excitations that propagate through lattices with nearest-neighbor interactions. They are conventionally observed in magnetically ordered solids using inelastic scattering techniques or ferromagnetic resonance. Here, we show that analogous spin dynamics can occur in solution-state nuclear magnetic resonance (NMR) of molecules containing aliphatic chains. We show that singlet-triplet population imbalances of nuclear spins can propagate coherently along aliphatic chains. These dynamics correspond to spin waves that carry no net magnetization and are therefore referr

Shaping the Action Potential in Dorsal Root and Trigeminal Ganglia Neurons: Relevance to Pain Mechanisms.

Sensory neurons of the dorsal root ganglia (DRG) and trigeminal ganglia (TG) are critical for transmitting somatosensory information, including pain, to the central nervous system. In these neurons, as in other neurons, the shape of the action potential (AP) is a key determinant of excitability and signal transmission and is governed by the coordinated activity of voltage-gated sodium (NaV), potassium (KV), calcium (CaV), and calcium-activated potassium (BK) channels. This review synthesizes current knowledge about how these ion channels shape AP waveform and influence neuronal function across

Future horizons: Toward an integrated neuroscience of sleep.

Sleep is now recognized as a vital biological process with roles beyond rest, including neurodevelopment, memory, emotional regulation, metabolism, immune function, and neuroprotection. Advances in neuroscience and technology reveal that sleep is an active, multifaceted process involving multiple brain and body systems. This chapter explores how neural networks, waste clearance, circadian rhythms, immune responses, and cognition interact to influence brain health and disease. We examine sleep's connections to neurodegeneration, mental illness, aging, and cognitive resilience. We also highlight

A case of telangiectatic osteosarcoma arising from the frontal bone.

Osteosarcoma of the calvarium is extremely rare, specifically of the telangiectatic subtype, which has few reported cases. Herein, we report a case of telangiectatic osteosarcoma occurring in the frontal bone of a 33-year-old man without notable family or medical history. The patient presented with a painless mass in the frontal region, and computed tomography (CT) imaging and magnetic resonance imaging (MRI) revealed a tumor destructively involving the inner and outer tables of the skull. On T1- and T2-weighted magnetic resonance imaging (MRI) images, the tumor exhibited an isointense signal

JAK2-Positive Myeloproliferative Neoplasm: Polycythaemia Vera Presenting As Budd-Chiari Syndrome.

Budd-Chiari syndrome (BCS), caused by hepatic venous outflow obstruction, is a critical diagnosis often masking an underlying myeloproliferative neoplasm (MPN). We present the case of a 33-year-old female whose diagnosis was driven by hepatic decompensation rather than typical haematological symptoms of MPN. Initial imaging confirmed the anatomical pathology, revealing chronic BCS on computed tomography (non-visualised hepatic veins, hepatic vein stenosis, and classic caudate lobe hypertrophy). The subsequent work-up noted a paradoxical haematological picture (significant neutrophilic leucocyt

Integrating Single-Molecule Variant Phenotyping with Clinical Features Predicts Outcomes in KIF1A-Associated Neurological Disorder.

Rare monogenic diseases lack scalable prognostic frameworks, leaving patients and families without guidance after molecular diagnosis. Using KIF1A-associated neurological disorder (KAND) as a model system, we integrated single-molecule biophysical phenotyping of 91 pathogenic variants with longitudinal clinical characterization of 343 patients. Vineland Adaptive Behavior Scales (VABS) Adaptive Behavior Composite (ABC) and Growth Scales identified two divergent trajectories emerging after age 10: a stable group and a declining group characterized by seizures, abnormal EEGs, and optic nerve atro

Homology Modeling and Pharmacophore Based Inhibitor Design Targeting Isocitrate Lyase of Madurella mycetomatis-A Novel Approach Against Eumycetoma.

Eumycetoma, caused by pathogenic fungi, is one of the most neglected tropical disease worldwide. The causative agent of the disease is Madurella mycetomatis. Ethnomedicinal sources reveal that several medicinal plants are used for the phytotherapeutic management of eumycetoma symptoms. M. mycetomatis uses isocitrate lyase (ICL) to promote gluconeogenesis for growth and survival in adverse host conditions. This study aims to develop a natural inhibitor molecule of ICL to target against M. mycetomatis using computational techniques. Homology modeling was used to predict the 3D structure of ICL u

HPV18E6 and CDK5 virus-host interaction is a prospective therapeutic target for HPV-positive cervical cancer.

The E6 oncoprotein encoded by the cancer-causing human papillomavirus (HPV) is vital for maintaining the ability of the virus to promote cell proliferation and cancer progression. This study identified that cyclin-dependent kinase 5 (CDK5) is a previously unidentified host target of E6 encoded by cancer-causing HPV genotypes. Although E6 may not be a phosphorylation substrate of CDK5, interaction with CDK5 nonetheless increases the steady-state level of E6. To further elucidate the significance of CDK5-E6 interaction, we adopted pharmacological inhibition and silencing approaches. Leveraging t

Redefining the diagnostic pathway for pulmonary nocardiosis: The imperative for early metagenomic sequencing.

In this article, we comment on the pivotal article by Wang et al. We focus on the critical intersection of advanced imaging and molecular diagnostics highlighted by their findings. The study delineates specific high-risk computed tomography patterns, notably consolidation with nodules/cavities, particularly in immunocompromised hosts or patients with bronchiectasis, that should serve as immediate red flags for pulmonary nocardiosis. Traditionally, diagnosis has relied on slow-growing cultures, leading to dangerous therapeutic delays. This editorial argues that the presence of these defined rad

A sustainable functional food ingredient from Saussurea involucrata cell culture alleviates atopic dermatitis-like inflammation via arachidonic acid metabolic remodeling.

Nutritional interventions may complement established approaches for managing atopic dermatitis (AD). Here, we evaluated Saussurea involucrata cell culture (SIC), a sustainable functional-food material, in a DNCB-induced mouse model of AD-like inflammation. Chemical profiling established the phytochemical composition of SIC, and both topical and oral interventions improved macroscopic and histopathological outcomes. Under the tested conditions, the oral groups showed more consistent changes in systemic indices, providing the basis for subsequent mechanistic analyses. Integrated transcriptomic a

Quantitative modeling of rod outer segment phagocytosis and recycling.

There is continual turnover of macromolecules as part of maintenance. This allows removal of damaged macromolecules and their replacement. A prime example is the turnover of the rod photoreceptor outer segment. Every day, 10% of rod photoreceptor outer segments is phagocytosed by the neighboring retinal pigment epithelium cells where they undergo phagolysosomal degradation. For example, the proteins are broken down to their component amino acids. These components need to be efficiently recycled back to the rod photoreceptor, while the damaged components can be eliminated. These damaged molecul

A recursive enzymatic competition network capable of multitask molecular information processing.

Living cells understand their environment by combining, integrating and interpreting chemical and physical stimuli. Despite considerable advances in the design of enzymatic reaction networks that mimic hallmarks of living systems, these approaches lack the complexity to fully capture biological information processing. Here we introduce a scalable approach to design complex enzymatic reaction networks capable of reservoir computation based on recursive competition of substrates. This protease-based network can perform a broad range of classification tasks based on peptide and physicochemical in

Highly Specific Monoclonal Antibodies Combined with A Multiplex Quantum Dot Microsphere-based Fluorescence Lateral Flow Immunochromatographic Assay for Simultaneous Detection of Vanillin, Methyl Vanillin, and Ethyl Vanillin in Food Samples.

Vanillin (VAN), methyl vanillin (VER), and ethyl vanillin (EVA) are widely used as flavor enhancers in processed foods. Monitoring their levels is essential to prevent overuse or adulteration, especially in infant foods. Herein, a computational chemistry-aided molecular modeling technique was employed to design specific haptens for VAN, VER, and EVA. High-affinity monoclonal antibodies were generated via hybridoma technology with matrix effect-enhanced screening, yielding half-maximal inhibitory concentrations of 22.10, 49.94, and 40.84 ng/mL. Pressure-Induced Emission Enhancement of Multi-Resonance o-Carborane Derivatives via Exciton‒Vibration Coupling Suppression.

Polycyclic multiple resonance (MR) molecules reveal narrowband emission, making them very promising emitters for high color purity display. Nevertheless, they still have challenges such as aggregation-induced emission quenching and spectral broadening. Overcoming these obstacles requires an in-depth understanding of the correlations among the alterations in their geometries, packing structures, and molecular vibrations and their corresponding changes in their photoluminescence (PL) properties. Herein, it is demonstrated that high-pressure infrared, UV-visible absorption, and fluorescence spect

Impact of climate change on the toxicity of bisphenol A in Mytilus galloprovincialis and assessment of phycoremediation using Nannochloropsis salina via a multi-biomarker strategy and modeling.

In the current study, the mussels Mytilus galloprovincialis, exposed to four varying temperatures (17, 20, 23, and 26 °C), were contaminated with 50 μg/L of bisphenol A both with and without Nannochloropsis salina. The toxicity evaluation is determined by quantifying various biomarkers related to oxidative stress, neurotoxicity, and cellular damage. The key findings indicate that the toxicity of bisphenol A is heightened by rising temperature. The impact of bisphenol A is most evident at 26 °C, leading to excessive production of reactive oxygen species, depletion of non-enzymatic antioxidants,

Divergent Synthesis and Crystal Engineering of Room-Temperature Phosphorescent Carbonyl-Bridged Triphenylamines.

Organic materials with room temperature phosphorescence (RTP) are a growing research topic, with recent investigations revealing a strong effect of the crystal packing on the RTP properties. However, these relationships are difficult to decipher, as the crystal packing affects both the intermolecular interactions and the molecular conformation. Here, we report a divergent synthetic strategy to generate a family of rigid carbonyl-bridged triphenylamine (TPA)-based chromophores via metal-free transformations of the same starting material, and exploit this family to investigate the effects of cry

A Dual-Responsive Fluorescent Compound Exhibiting Turn-On Mechanoresponsive and Solvatochromism for Rewritable Optical Data Storage.

A novel mechanoresponsive and solvatochromic luminescent compound (MO-HBI) was designed by combining an ESIPT-active donor with a benzophenone acceptor, exhibiting reversible luminescence switching and aggregation-induced emission enhancement. The solvatochromic behavior of MO-HBI was systematically studied in solvents of varying polarity, revealing a distinct bathochromic shift in both absorption and fluorescence emission, with pronounced sensitivity attributed to the presence of a carbonyl group. Upon mechanical stimulation, MO-HBI undergoes a crystalline-to-amorphous phase transition, leadi

Multivalent Interactions Between the Picornavirus 3C(D) Main Protease and RNA Oligonucleotides Induce Liquid-Liquid Phase Separation.

The picornavirus 3CD protein is a precursor to the 3C main protease and the 3D RNA-dependent RNA polymerase. In addition to its functions in proteolytic processing of the virus polyprotein and cleavage of key host factors, the 3C domain interacts with cis-acting replication elements (CREs) within the viral genome to regulate replication and translation events. We investigated the molecular determinants of RNA binding to 3C using a wide range of biophysical and computational methods. These studies showed that 3C binds to a broad spectrum of RNA oligonucleotides, displaying minimal sequence and

Radiosynthesis and Biological Evaluation of a Small-Molecule Probe for Thyroid-Stimulating Hormone Receptor.

The thyroid-stimulating hormone receptor (TSHR) plays a pivotal role in regulating thyroid growth, function, synthesis, and secretion of thyroid hormones, with its overexpression linked to various diseases, especially in tumors. A novel TSHR-targeting small molecule agonist ML-109 with a nM potency has been developed. In this study, we radiolabeled ML-109 with radionuclide iodine-131 on the quinazolin-4-one ring to prepare 131I-ML-109, and its biological performance was further evaluated as a single-photon emission computed tomography radiotracer. The results displayed that 131I-ML-109 was pre

Novel Cellobiose 2-Epimerase from Thermal Aquatic Metagenome for the Production of Epilactose.

Epilactose is a prebiotic molecule that exerts a bifidogenic effect and increases calcium and iron absorption in the small intestine. This study identifies a novel cellobiose 2-epimerase gene (ceM) by investigating metagenomic data generated from a thermal aquatic habitat. The computation of secondary and tertiary structure analysis, molecular docking, and MD simulation analysis indicated the protein CEM to be a novel cellobiose 2-epimerase. The gene was expressed in Escherichia coli, followed by biochemical characterization of the purified protein. CEM is capable of transforming lactose into
