BACKGROUND/OBJECTIVES: Pituitary neuroendocrine tumors (PitNETs) frequently exhibit invasive behaviors that complicate clinical treatment. While cyclin-dependent kinase 5 (CDK5) and lymphokine-activated killer T-cell-originated protein kinase (PBK, also known as PDZ-binding kinase) are implicated in tumor progression, their reciprocal regulatory mechanism remains unclear. This study aims to elucidate the CDK5-PBK interaction in PitNETs and identify potential therapeutic agents targeting this pathway.
METHODS: We utilized proximity labeling and phospho-specific assays to characterize the CDK5

Phenethyl isothiocyanate (PEITC) exerts anti-gastric cancer effect, however, the molecular mechanism has not yet been elucidated. KLF4 is an important regulatory molecule in gastric cancer. The programmed death ligand 1 (PD-L1) interacts with programmed cell death 1 (PD-1), inhibits function of cytotoxic T-lymphocyte and helps gastric cancer cells evade immune surveillance. The present study aimed to investigate the effect of PEITC against gastric cancer and the roles of KLF4 and PD-L1 in the anti-gastric cancer effect of PEITC. The effects of PEITC on gastric cancer cell proliferation and apo

BACKGRUOUND: The potassium voltage-gated channel subfamily Q member 1 (KCNQ1) gene has recently received much attention as a candidate susceptibility gene for type 2 diabetes mellitus, especially in Asian populations. We previously reported that Kcnq1 mutant mice exhibit reduced insulin secretion and hyperglycemia due to a decrease in pancreatic β-cell mass. Through in vivo and in vitro analyses, we ascertained that this mechanism is the result of the downregulation of the non-coding RNA 'Kcnq1ot1,' which is expressed in the paternal allele of the Kcnq1 gene region, causing an increase in the

Interleukin-21 (IL-21), a pivotal immune molecule, exerts an antitumor effect by regulating immune responses in vivo. Nevertheless, the short half-life of IL-21 protein and the difficulty of intratumoral injection of recombinant proteins for the majority of patients present obstacles. We have engineered an IL-21 gene expression vector and confirmed its ability to stably express IL-21 in tumor tissue, exhibiting a certain anti-melanoma effect. Meanwhile, the chemokine CCL2 (chemokine(C-C motif) ligand 2) promotes tumor progression in various cancers by binding to its receptor CCR2 (C-C Chemokin

The phenotypic switch of vascular smooth muscle cells (VSMCs), characterized by the tissue-specific expression of certain microRNAs (miRNAs), is a critical factor in the development of diabetic vascular diseases. Metformin, a widely prescribed anti-diabetic medication for type 2 diabetes treatment, activates the adenosine monophosphate-activated protein kinase (AMPK) pathway and exerts a protective effect on vascular endothelium. Although the regulatory effects of metformin on the switch of the vascular smooth muscle cell phenotype have been identified, the specific role of miRNAs in this proc

EZH2, the catalytic subunit of polycomb repressive complex 2 (PRC2), plays a critical role in neural development by regulating gene expression through the trimethylation of lysine 27 on histone H3 (H3K27me3), which promotes chromatin remodeling and transcriptional repression. Although PRC2 is known to regulate cell fate specification and gliogenesis, its in vivo functions during vertebrate neurodevelopment, particularly at the level of neuronal subtype differentiation, remain incompletely understood. Here, we investigated the consequences of ezh2 loss-of-function during zebrafish brain develop

The adaptor protein SRCIN1 is a novel Src-binding protein that regulates Src activation through C-terminal Src kinase. SRCIN1 has been shown to promote the development of colorectal cancer, while acting as a tumor suppressor in breast cancer. However, its role in the development of thyroid cancer has remained unknown. In this study, we analyzed the biological characteristics of SRCIN1 in thyroid cancer using public data (HPA, TIMER, GEPIA and UALCAN). Additionally, we investigated the biological impact of SRCIN1 knockdown and overexpression on thyroid cancer cells both in vitro and in vivo. Ou

KRAS -mutant non-small cell lung cancer (NSCLC) remains a major therapeutic challenge due to the paucity of effective targeted agents. Although covalent KRAS^G12C inhibitorsi such as sotorasib and adagrasib have demonstrated clinical activity, pooled analyses indicate only modest response rates and short progression-free survival, with no effective options for non-G12C subtypes. This highlights the need for novel therapeutic strategies with broader efficacy and improved durability. We evaluated the antiproliferative effects of SP09, a novel benzoin-Schiff base derivative, in KRAS -mutant (A549

Pancreatic cancer is a highly aggressive malignancy, with 10%-20% of cases linked to inherited genetic risk factors. Pathogenic variants (PVs) in the cyclin-dependent kinase inhibitor 2A (CDKN2A) gene have been associated with pancreatic cancer among Caucasians. However, data from South Asians are lacking. We investigated the prevalence of CDKN2A germline variants in 200 consecutively and prospectively enrolled, unselected pancreatic cancer patients from Pakistan. Comprehensive variant detection was performed using high-resolution melting analyses followed by DNA sequencing. Novel variants wer

Hepatocellular carcinoma (HCC) is the primary cause of cancer-related mortality in the world. Cytochrome P450 2E1 (CYP2E1) plays a key role in metabolizing xenobiotics and toxic substances. The attempts to target CYP2E1 with inhibitors have yielded limited success in cancer treatment. Loss and gain-of-function experiments were performed to investigate the effects of CYP2E1 on HepG2 and Hepa 1-6 cells. Migration, invasion, intracellular reactive oxygen species (ROS) accumulation, and mitochondrial membrane potential were assessed in HepG2 cells. In vivo histopathological alterations were studie

Hyphal formation is a key virulence trait in Candida albicans, playing a crucial role in the development and progression of candidiasis. As a result, it remains a significant area of interest for antifungal drug target studies. Conventional hyphal induction medium typically contain multiple components, making them complex to prepare. In this study, we have optimized a simple and cost-effective medium with minimal components that induces hyphal formation. Among different combination, MF8 with Peptone 0.16 %, Dextrose 0.4 % and BSA 0.25 % caused moderate hyphal formation. Further addition of N-a

Multidrug-resistant (MDR) variants of L1210 cells, selected for resistance to vincristine (R) or doxorubicin (D), exhibit elevated ABCB1 (P-glycoprotein) expression but differ in UVC sensitivity. D cells resemble parental L1210 (S) cells, whereas R cells are significantly more UVC vulnerable. To investigate this, we analyzed the expression of genes involved in DNA damage response, Trp53 family members, cyclin-dependent kinase inhibitors (p15, p21), Bcl-2 family genes, and DNA repair genes in S, R, and D cells before and after UVC irradiation. The most striking difference was the absence of Trp
