BACKGROUND: Hypermelanosis includes a diverse group of genetic and acquired skin anomalies that appear as darker, hyperpigmented areas. Melasma, in particular, is a hypermelanotic condition that affects sun-exposed skin in females. Whether this condition is acquired or genetic is still controversial. However, it clearly correlates with exposure to UV light, a genetic predisposition, and hormonal variations (from pregnancy or oral contraceptives).
METHODS: Between October 2006 and March 2008, 38 patients with melasma were treated with intense pulsed light (IPL) at the LASER Center of the Depar

BACKGROUND: An objective demonstration of lesions disseminated in time and space remains the core of the last revision of diagnostic criteria for multiple sclerosis (MS), but this update is now empowered by a weighted use of magnetic resonance imaging (MRI), which results in an earlier and more unambiguous diagnosis ("MS," "not MS," or "possible MS"). Nevertheless, the exclusion of other entities still remains an integral element of the diagnostic process.
REVIEW SUMMARY: Exclusion of genetic disorders can be challenging in some cases with familial recurrence of MS, particularly when the tran

BACKGROUND: The recent identification of genes responsible for syndromes of periodic fever with amyloidosis has opened the way to a molecular diagnosis of hereditary AA amyloidosis.
METHODS: A Belgian woman presented for genetic counseling. Three first-degree relatives had a diagnosis of renal amyloidosis with a history of recurrent fever and inflammatory episodes. Medical records and pathological specimens were obtained from all physicians who had been in charge of her three relatives. Immunohistochemical staining was performed on paraffin-embedded material. A mutation search was performed i

Migraines are a common and often undiagnosed and undertreated problem in children of all ages. Migraine symptoms can vary dramatically in terms of character and severity, from brief self-limited headaches to prolonged events with complex neurologic and systemic symptoms. Identification of migraines requires an index of suspicion in any child with acute recurrent headaches or neurologic symptoms. Diagnosis remains predominately based on the patient's history of symptoms and supported by a positive family history. Neurodiagnostic tests are often unnecessary and of value predominately to exclude

OBJECTIVES: Diseases due to inherited hemoglobin disorders represent serious medical, social, and economic problems in the region of Marseille. The only effective treatment for such diseases is allogenic bone marrow transplantation. About 200 patients with either thalassemia, sickle cell or sickle cell-beta thalassemic diseases are regularly seen in local hospitals. All of these patients come from parts of the world where genetic hemoglobin disorders are endemic.
METHODS: At this time, the only approach for reducing the number of affected children born is preventive. This depends upon educati

Recent developments in recombinant DNA research promise to have a substantial impact on medicine. The technology now allows for DNA pieces to be isolated and ligated to other DNA, such as that from bacteria, and be replicated in foreign hosts. The replicated DNA can then be isolated and used to study its structure, to study gene expression and its regulation, to program microorganisms to synthesize medically, agriculturally or industrally important proteins, to transfer genes back into mammalian cells and to diagnose genetic diseases. The DNA used for cloning can be obtained by copying mRNA, b

BACKGROUND: Interstitial cystitis/bladder pain syndrome (IC/BPS) is a poorly understood and underdiagnosed syndrome of chronic bladder/pelvic pain with urinary frequency and urgency. Conflicting data exist regarding associated phenotypes, and little is known of its genetic aetiology.
METHODS: We conducted a retrospective case-control analysis of electronic medical record (EMR) and retrospective case-control analysis of exome sequencing (ES) to identify phenotypes related to and rare variant risk factors for IC/BPS. Retrospective EMR data were collected from a national network of research site

There is clear evidence of genetic modulation of bone phenotype parameters including bone density, quantitative ultrasound, bone size, and bone turnover. At any particular age and phase of life, genetic factors explain about 70% of the variance in bone phenotype after adjustment for major medical and disease factors. Hormonal factors, diet, and lifestyle interact with those genetic factors over time. Common allelic variation in the VDR was the first of several genes and now chromosomal loci to be implicated in the genetic determination of bone phenotype. The VDR polymorphisms have an effect we

The management of radiation exposure (RE) is critical for protecting human health, ensuring environmental safety, and optimizing industrial and medical applications. Inadequate RE management can lead to cancer risks, genetic effects, environmental contamination, and occupational hazards. RE arises from natural, medical, and occupational sources, and assessing these factors individually fails to provide a comprehensive understanding. This study systematizes RE management by estimating RE at national and regional levels, presenting it as Radiation Exposure Footprint (REF) to identify key influen

Opioid use disorder (OUD) is a chronic disease that remains difficult to treat, even with significant improvements in available medications. While current treatments work well for some, they often do not account for the unique needs of individual patients, leading to less-than-ideal results. Precision medicine offers a new path forward by tailoring treatments to fit each person's genetic, psychological, and social needs. This review takes a close look at medications for OUD, including methadone, buprenorphine, and naltrexone, as well as long-acting options that may improve adherence and conven

Prader-Willi syndrome (PWS) is a rare neuroendocrine genetic syndrome. Characteristics of PWS include hyperphagia, hypotonia, and intellectual disability. Pituitary hormone deficiencies, caused by hypothalamic dysfunction, are common and hypogonadism is the most prevalent. Untreated hypogonadism can cause osteoporosis, which is already an important issue in PWS. Therefore, timely detection and treatment of hypogonadism is crucial. To increase understanding and prevent undertreatment, we (1) performed a cohort study in the Dutch PWS population, (2) thoroughly reviewed the literature on female h

Specific ethnic genetic backgrounds are associated with the risk of Stevens-Johnson syndrome / toxic epidermal necrolysis (SJS/TEN) especially in Asians. However, there have been no large cohort, multiple-country epidemiological studies of medication risk related to SJS/TEN in Asian populations. Thus, we analyzed the registration databases from multiple Asian countries who were treated during 1998-2017. A total 1,028 SJS/TEN cases were identified with the algorithm of drug causality for epidermal necrolysis. Furthermore, those medications labeled by the US Food and Drug Administration (FDA) as

Although many human diseases have a genetic component involving many loci, the majority of studies are statistically underpowered to isolate the many contributing variants, raising the question of the existence of alternate processes to identify disease mutations. To address this question, we collect ancestral transcription factor binding sites disrupted by an individual's variants and then look for their most significant congregation next to a group of functionally related genes. Strikingly, when the method is applied to five different full human genomes, the top enriched function for each is

Amifostine is a broad-spectrum cytoprotective agent approved for protection against cisplatin toxicities and radiation-induced xerostomia; strong clinical evidence exists that amifostine protects normal mucosa and lung from radiation damage. Hypotension, nausea/vomiting, fatigue and fever/rash are the main side-effects associated with amifostine administration. The present study summarizes our experience on daily amifostine administration to cancer patients with various coexisting medical conditions and diseases. BACKGROUND: The evaluation of genetic mutations is crucial for personalized therapy in colorectal cancer (CRC), but the invasive tissue biopsy is subject to sampling bias and other complications. Radiomics has emerged as a non-invasive tool to predict these mutations from standard medical images. In this systematic review and meta-analysis, we aimed to evaluate the diagnostic accuracy and methodological quality of radiomics models for predicting key genetic mutations in CRC.
METHODS: A comprehensive search of PubMed, Scopus, Web of Science, and Embase was conducted in accordance with PRISMA g
