[Treatment of ANCA-associated vascularitides].

Treatment for ANCA-associated vasculitides is now well defined, but must be adjusted for each patient according to the type of vasculitis, its precise form (e.g., limited versus systemic Wegener's granulomatosis) and severity, and patients' characteristics, such as age and renal function. The therapeutic decision must also take into account the risk of adverse events inherent to each treatment. The efficacy of adequate induction treatment has been demonstrated: more than 80% of patients now achieve remission. Relapse rates nonetheless remain high, especially in Wegener's granulomatosis. Primary defect in UVB-induced systemic immunomodulation does not relate to immature or functionally impaired APCs in regional lymph nodes.

UVB irradiation of the shaved dorsal skin of mice can cause both local and systemic suppression of contact hypersensitivity responses; the former demonstrated by administration of the sensitizing Ag/hapten to the irradiated site and the latter by its administration at least 72 h later to distal unirradiated sites. The immunological basis of systemic immunomodulation is not clear. When haptens (trinitrochlorobenzene, FITC) were administered to the shaved ventral skin 4 days after irradiation (8 kJ/m(2)) to the shaved dorsum of BALB/c mice, CD11c(+)/FITC(+) cells in the skin-draining lymph nodes

MMP-3 expression and release by rheumatoid arthritis fibroblast-like synoviocytes induced with a bacterial ligand of integrin alpha5beta1.

Fibroblast-like synoviocytes (FLSs) play a major role in the pathogenesis of rheumatoid arthritis (RA) by secreting effector molecules that promote inflammation and joint destruction. How these cells become and remain activated is still elusive. Both genetic and environmental factors probably play a role in transforming FLSs into inflammatory matrix-degrading cells. As bacterial products have been detected in the joint and shown to trigger joint inflammation, this study was undertaken to investigate whether a bacterial ligand of integrin alpha5beta1, protein I/II, could contribute to the aggre

CD4+ T cells can induce airway hyperresponsiveness to allergen challenge in the brown norway rat.

Airway hyperresponsiveness to inhalational challenge with methacholine (MCh) develops by 32 h after allergen challenge of actively sensitized BN rats. To test the hypothesis that CD4+ T cells mediate allergen-induced hyperresponsiveness independent of IgE-mediated mechanisms, we administered CD4+ T cells, CD8+ T cells, and a mixture of CD4+ and CD8+ T cells (total T cells) isolated from the cervical lymph nodes of rats sensitized with ovalbumin (OA) to naive BN rats that underwent aerosol challenge with either OA or bovine serum albumin (BSA) 2 d later. Responsiveness to MCh was measured 2 d b

Lactic acid bacteria enhance autophagic ability of mononuclear phagocytes by increasing Th1 autophagy-promoting cytokine (IFN-gamma) and nitric oxide (NO) levels and reducing Th2 autophagy-restraining cytokines (IL-4 and IL-13) in response to Mycobacterium tuberculosis antigen.

BACKGROUND AND OBJECTIVES: Control of the intracellular Mycobacterium tuberculosis (Mtb), mainly requires an appropriate ratio of Th1/Th2 cytokines to induce autophagy, a physiologically, and immunologically regulated process that has recently been highlighted as an innate defense mechanism against intracellular pathogens. Current vaccines/adjuvants induce both protective Th1 autophagy-promoting cytokines, such as IFN-gamma, and immunosuppressive Th2 autophagy-restraining cytokines, such as IL-4 and IL-13. TB infection itself is also characterized by relatively high levels of Th2 cytokines, wh

Amelioration of diet-induced hyperglycemia by wheat peptides in a multi-target manner: integrated modulation of inflammation, lipid metabolism, gut microbiota, and DPP-IV inhibition.

Wheat peptides (WP) have been claimed to have the potential to regulate metabolism and effectively prevent/mitigate gut microbiota dysbiosis. However, many studies into the effects of WP on hyperglycemia have provided conflicting findings, and the underlying mechanism has been elusive. In this study, WP intervention (50-1000 mg kg-1) dose-dependently attenuated high fat diet (HFD)-induced weight gain, fasting hyperglycemia, glucose intolerance and insulin resistance. WP suppressed systemic inflammation by normalizing serum levels of lipopolysaccharide (LPS), tumor necrosis factor-α (TNF-α), an

Medin-Induced Pro-inflammatory and Prothrombotic Activation of Coronary Artery Endothelial Cells: A Potential Novel Mediator Linking Aging and Atherosclerosis.

BACKGROUND: Age is the most important risk factor for coronary artery disease (CAD) independent of traditional risk factors. Aging induces classic pro-inflammatory and prothrombotic vascular phenotypic changes whose molecular mediators remain poorly understood. Medin is a common cleavage product protein that accumulates in vasculature with aging and shown to cause endothelial dysfunction. Its role in CAD is unknown. The study aimed to evaluate the effects of medin on human coronary artery endothelial cell (HCAEC) pro-inflammatory and prothrombotic activation and establish the relationship betw

Protective Role of Apigenin-PEGylated Chitosan Nanoparticles Against Pesticide Deltamethrin-Induced Liver Injury via Antioxidant Activation, NF-κB Inhibition, and Bcl-2/Bax/Caspase-3 Signaling Modulation.

The present investigation evaluated the liver-protective effect of apigenin encapsulated in PEGylated chitosan nanoparticles (APG-PCHNPs) against hepatotoxicity induced by pesticide deltamethrin (DLM) in rats. Sixty rats were randomly allocated into six groups: a control group, groups treated with APG (20 mg/kg BW) or APG-PCHNPs (20 mg/kg BW), a group exposed to DLM (5 mg/kg BW), and two combination treatment groups receiving DLM (5 mg/kg BW) plus either APG or APG-PCHNPs at 20 mg/kg BW for 35 days. DLM administration significantly decreased serum total protein and its fractions. Simultaneousl

Direct inhibition of the TXNIP-NLRP3-GSDMD pathway reduces pyroptosis in colonocytes and alleviates ulcerative colitis in mice by the small compound PEITC.

Ulcerative colitis (UC) is a chronic inflammatory bowel disease. The etiology of UC is multifaceted, and the underlying pathogenesis remains incompletely understood. Pyroptosis, programmed cell death mediated by the gasdermins, is a pivotal driver of UC pathology due to its dual role in epithelial barrier disruption and inflammatory amplification. We previously showed that phenethyl isothiocyanate (PEITC), an isothiocyanate derived from cruciferous vegetables, alleviated acute liver injury in mice by suppressing hepatocyte pyroptosis. In this study we evaluated the therapeutic potential of PEI

Trimethylamine-N-oxide damages astrocytes and lymphatic endothelial cells in the cerebral lymphatic system.

BACKGROUND: Trimethylamine-N-oxide (TMAO), as a gut microbiota dependent metabolite, is involved in the occurrence and progression of many neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease, which are related to the disruption of the cerebral lymphatic system. However, the relationship between TMAO and cerebral lymphatic system remains to be elucidated. This study aimed to investigate the effects of TMAO on the astrocytes of the brain glymphatic system, and endothelial cells of the meningeal lymphatic vessels. METHODS: Astrocytes and lymphatic endothelial cells wer

Innate and T-cellular immune responses to sequential vaccination with chimeric hemagglutinin split influenza virus vaccines in mice.

Chimeric hemagglutinin (cHA)-based vaccines represent a promising strategy toward the development of a broadly protective universal influenza virus vaccine. Previous studies show that sequential administration of cHA split influenza vaccines stimulates broadly cross-reactive antibodies targeting the HA stalk and induces systemic and localized CD4+ T-cell responses to the HA, neuraminidase (NA), and nucleoprotein (NP). However, the exact role of the T-cellular immune response in the protection after vaccination with cHA constructs remains underinvestigated. Here we provide an in-depth character

USP5 Deletion Inhibits KD Serum Induced-Human Coronary Artery Endothelial Cell Dysfunction by Regulating the NFATC1/TLR4-Mediated NF-κB Signaling Pathway in Kawasaki Disease.

Kawasaki disease (KD) is an acute febrile illness characterized by systemic vasculitis, especially in coronary arteries. Previous studies have indicated that nuclear factor of activated T cells, cytoplasmic 1 (NFATC1, also known as NFAT2) plays a crucial role in the pathogenesis of KD. However, the molecular mechanism of NFATC1 involved in KD is poorly defined. Human coronary artery endothelial cells (HCAECs) were treated with 15% serum from KD patients to mimic the inflammatory injury model in vitro. NFATC1 mRNA level was determined using real-time quantitative polymerase chain reaction (RT-q
