Defining High-risk Retinoblastoma: A Multicenter Global Survey.

IMPORTANCE: High-risk histopathologic features of retinoblastoma are useful to assess the risk of systemic metastasis. In this era of globe salvage treatments for retinoblastoma, the definition of high-risk retinoblastoma is evolving. OBJECTIVE: To evaluate variations in the definition of high-risk histopathologic features for metastasis of retinoblastoma in different ocular oncology practices around the world. DESIGN, SETTING, AND PARTICIPANTS: An electronic web-based, nonvalidated 10-question survey was sent in December 2020 to 52 oncologists and pathologists treating retinoblastoma at ref

SMΝΔ7 mice show breathing and airflow defects with significant pathology of respiratory and oral tract tissues.

INTRODUCTION: Spinal muscular atrophy (SMA) is a neurodegenerative disorder caused by SMN1 mutations, leading to SMN protein deficiency and motor neuron loss. While progressive weakness, respiratory defects, and oral dysfunction are well-documented in patients, the underlying pathophysiology of breathing and bulbar deficits remains understudied in SMA animal models. METHODS: We evaluated breathing and oral function in the SMN∆7 mouse model of severe SMA. Respiratory parameters and chemoreflexes were assessed via whole-body plethysmography. To identify underlying structural changes, we perform

Reversal of Diabesity Through Modulating Sympathetic Inputs to Adipose Tissue Following Carotid Body Resection.

BACKGROUND AND AIMS: The development of innovative strategies to treat diabesity and its comorbidities is of major societal importance. The carotid bodies (CB), classically defined as O2 sensors, are also metabolic sensors whose dysfunction contributes to the genesis and progression of metabolic disturbances. Here, we tested the hypothesis that the CBs are key players in the neural hypothalamic-sympathetic circuit controlling glucose and energy homeostasis. Moreover, we investigated if abolishment of CB activity has an anti-diabesity effect in Wistar rats and C75BL/6J mice, associated with inc

Recurrented bilateral low-dose regional anesthesia under ultrasound guidance rather than general anesthesia in a high-risk patient: Rare a case.

Regional anesthesia practices are important because they have the advantages, for example, the patient's awareness is open, spontaneous breathing continues, airway reflexes are preserved, analgesia continues in the post-operative period, and the patient has early mobilization. Local anesthetic at high doses and volumes are used in brachial plexus blocks with nerve stimulator. However, due to the development in ultrasound (US) technology and the increase in image quality, reduced dose of limited anesthesia, and because of its advantages such as vascular and reduced risk of pleural puncture, it

Methanol-induced optic neuropathy: a still-present problem.

Methanol-induced optic neuropathy (Me-ION) is a serious condition that may result in long-term or irreversible visual impairment or even blindness secondary to damage and loss of function of the optic nerve and retina. Me-ION shows a tendency to occur as mass poisonings around the world with a clear predilection for poor societies in developing countries. The main mechanism underlying the molecular basis of Me-ION is the inhibition of the mitochondrial oxidative phosphorylation process through the binding of the toxic metabolite of methanol-formic acid-with the key enzyme of this process-cytoc

Lateral Hind-Foot Endoscopic Anterolateral/Posterolateral (LEAP) Subtalar Arthrodesis: An Effective Minimally Invasive Technique to Achieve Subtalar Fusion and Deformity Correction.

Arthrodesis surgery aims to provide relief for chronic joint pain and correct limb alignment by achieving a stable union between articulating bones. The key factors to achieving sound arthrodesis is adequate debridement of arthritic cartilage and creating well-apposed bleeding subchondral bone surfaces without compromising the surrounding soft tissue envelope. Arthroscopic subtalar arthrodesis is technically demanding but provides better visualization of the articular surfaces and is safer for the surrounding soft tissues compared to the open approach. Early published reports of the arthroscop

Localization of soluble guanylate cyclase activity in the guinea pig cochlea suggests involvement in regulation of blood flow and supporting cell physiology.

Although the nitric oxide/cGMP pathway has many important roles in biology, studies of this system in the mammalian cochlea have focused on the first enzyme in the pathway, nitric oxide synthase (NOS). However, characterization of the NO receptor, soluble guanylate cyclase (sGC), is crucial to determine the cells targeted by NO and to develop rational hypotheses of the function of this pathway in auditory processing. In this study we characterized guinea pig cochlear sGC by determining its enzymatic activity and cellular localization. In cytosolic fractions of auditory nerve, lateral wall tiss

Paraganglioma in the orbit.

A 56-year-old Japanese woman was treated surgically for orbital paraganglioma. She had had proptosis of the left eye and lowered visual acuity. Papilledema and many retinochoroidal folds were seen. CT-scan demonstrated a tumor shadow on the optic nerve in the muscle cone. Tumor extirpation by Krönlein-Berke's procedure resulted in complete loss of vision due to surgical damage to the optic nerve. The previously reported 24 cases of orbital paraganglioma are reviewed. According to these clinical descriptions, orbital paragangliomas occur unilaterally in males and females, at any age, especially

Comparison of immunohistochemical and functional reinnervation of skin and muscle after peripheral nerve injury.

In order to investigate the usefulness of immunohistochemical detection of regenerating axons as a correlate of functional recovery, reinnervation of mouse foot pads, hairy skin, and muscle were studied at several intervals along 3 months after sciatic nerve crush using immunohistochemical markers PGP 9.5 and CGRP. These histological results were compared with functional recovery of sweat glands (SGs), plantar muscles, and pain sensibility. One week after nerve injury all neural functions were abolished in the operated hindpaw of all mice, no CGRP-immunoreactive (-ir) fibers were seen in the s

Polymer encapsulated cell lines genetically engineered to release ciliary neurotrophic factor can slow down progressive motor neuronopathy in the mouse.

Ciliary neurotrophic factor (CNTF) has recently generated great interest due to its potential as a therapeutic agent for the treatment of human neurodegenerative diseases such as amyotrophic lateral sclerosis (ALS). Because the systemic half-life of CNTF is only in the order of a few minutes, continuous delivery of this trophic factor could be attractive or even necessary in the therapy of these diseases. One promising technique involves the polymer encapsulation of cells which have been genetically modified to secrete neurotrophic factors. The polymer capsules can be implanted into animals an

A novel technique to isolate adult Schwann cells for an artificial nerve conduit.

The use of an artificial nerve conduit containing viable Schwann cells (SCs) is one of the most promising approaches to repair nerve injuries. Obtaining a large number of viable SCs in a short period is demanded for the clinical use of this technique. However, the previous methods using mitogens are not clinically acceptable, and other methods that do not require mitogens, failed to isolate adult SCs effectively or required a long period of time. In this study, we have developed a novel technique to isolate SCs from adult rat peripheral nerves for an artificial nerve conduit without mitogens,

Reggie-1 and reggie-2, two cell surface proteins expressed by retinal ganglion cells during axon regeneration.

Fish--in contrast to mammals--regenerate retinal ganglion cell axons when the optic nerve is severed. Optic nerve injury leads to reexpression of proteins, which typically are first expressed in newly differentiated retinal ganglion cells and axons. Here we identified two new proteins of fish retinal ganglion cells, reggie-1 and reggie-2, with monoclonal antibody M802 and molecular cloning techniques. In normal fish, M802 stained the few retinal axons derived from newborn ganglion cells which in fish are added lifelong to the retinal margin. After optic nerve injury, however, M802 labeled all

Ultrastructural evidence for the lack of co-transport of B-50/GAP-43 and calmodulin in myelinated axons of the regenerating rat sciatic nerve.

Following peripheral nerve injury, neurons respond with synthesis of proteins required for axonal regeneration. Newly synthesized membrane proteins, like B-50/GAP-43, are transported with the fast component of anterograde axonal transport. Structural proteins and calmodulin are transported by the slow component. Since B-50/GAP-43 can bind calmodulin, it has been hypothesised that B-50/GAP-43 may act as a carrier for fast anterograde transport of calmodulin, so that both proteins are delivered rapidly to the distally outgrowing axons ('the fast carrier hypothesis'). We have investigated whether

Toxicologic Pathology Forum: Opinion on Considerations for Determination of Adverse Versus Nonadverse Nerve Fiber Degeneration in Nonclinical Toxicity Studies of Oligonucleotides Delivered Directly Into the Intrathecal Space.

Determination of adversity and the no observed adverse effect level (NOAEL) in nonclinical toxicity studies are important components in the overall assessment of the potential for human risk during pharmaceutical development. Resources exist to guide adversity decisions in general, for findings in ganglia of the peripheral nervous system (PNS), and for gliosis in the central nervous system (CNS). The objective of this opinion article is to describe considerations that may apply when determining the adversity (or lack thereof) of nerve fiber degeneration (NFD) in either the CNS or PNS following

Primary Intraosseous Malignant Nerve Sheath Tumor Of Humerus Presenting With Pathological Fracture: A Rare Case Report With Review Of Literature.

INTRODUCTION: Malignant peripheral nerve sheath tumors (MPNST) are rare, aggressive soft-tissue sarcomas arising from peripheral nerves, usually associated with neurofibromatosis Type-I. Intraosseous MPNSTs are exceedingly uncommon and are typically located in the maxillofacial region. There are only 45 documented cases of intraosseous MPNSTs outside the head and neck region, of which only two are of humeral origin and only one reported as pathological fracture. CASE REPORT: We present a rare case of a de novo MPNST of the humerus that presented with pathological fracture and radial nerve pal

Neuroprotective liver portal area macrophages attenuate hepatic inflammation.

Tissue macrophages have an important role in the maintenance of liver homeostasis, and their functions are closely related to spatial localization. Here, through integration of whole liver lobe imaging and single-cell RNA sequencing analysis of CX3CR1+ cells in the mouse liver, we identified a dense network of CX3CR1+CD63+ liver portal area macrophages (LPAMs) that exhibited transcriptional and spatial differences compared with CX3CR1+CD207+ liver capsular macrophages. The survival of LPAMs was dependent on colony-stimulating factor 1 receptor (CSF1R). LPAMs colonized the hepatic portal area o

Quantitative Sensory Testing in a Girl With Tangier Disease: A Case Report.

Tangier disease is a rare genetic disorder caused by a deficiency in the ABCA1 gene, resulting in impaired metabolism of high-density lipoprotein (HDL) cholesterol. This disorder affects multiple organs, including the nervous system, and presents with a range of neurological symptoms. The primary neurological manifestation is peripheral nerve dysfunction, characterized by numbness, tingling, and weakness in the extremities. Autonomic dysfunction may also occur, causing bowel and bladder abnormalities. Cholesterol accumulation in nerve tissues is believed to be responsible for these symptoms. R…

Topical application of calcitonin gene-related peptide as a regenerative, antifibrotic, and immunomodulatory therapy for corneal injury.

Calcitonin gene-related peptide (CGRP) is a multifunctional neuropeptide abundantly expressed by corneal nerves. Using a murine model of corneal mechanical injury, we found CGRP levels in the cornea significantly reduced after injury. Topical application of CGRP as an eye drop accelerates corneal epithelial wound closure, reduces corneal opacification, and prevents corneal edema after injury in vivo. CGRP promotes corneal epithelial cell migration, proliferation, and the secretion of laminin. It reduces TGF-β1 signaling and prevents TGF-β1-mediated stromal fibroblast activation and tissue fibr
