Optogenetic Dissection of Sensory Neuron-Macrophage Interactions in Psoriasiform Skin Inflammation: Advances Based on In Vivo Calcium Imaging Techniques.

Psoriasis represents a chronic inflammatory skin disorder driven by bidirectional neuro-immune crosstalk between cutaneous sensory neurons and tissue macrophages. Tissue-resident macrophages exhibit a continuous functional spectrum rather than rigid binary M1/M2 subsets, and their dynamic reprogramming critically shapes inflammatory onset, perpetuation and resolution; however, conventional experimental approaches cannot readily provide causal, high spatiotemporal-resolution evidence for neuron-macrophage communication in intact skin. Optogenetics combined with intravital in vivo calcium imagin

Nociceptor neurons suppress antitumor immunity in breast cancer.

Peripheral nerves are emerging regulators of the tumor microenvironment, but how sensory innervation shapes breast cancer immunity remains poorly defined. Here we show that triple-negative breast cancers (TNBCs) co-opt nociceptor neurons to suppress antitumor immunity and promote disease progression. Across orthotopic TNBC models, we found that primary tumors and tumor-draining lymph nodes were densely innervated by CGRP+ sensory fibers. Tumor-derived cues directly activated dorsal root ganglion neurons, increased calcium responsiveness, induced Ngfr and Atf3, and triggered release of CGRP and

GLP-1R/NPY2R regulate gene expression, ovarian and adrenal morphology in HFD mice.

Glucagon-like peptide-1 receptor (GLP-1R) and neuropeptide Y receptors (NPYRs) are expressed in reproductive tissues contributing to the regulation of gonadal function. This exploratory study examines the potential impact of their modulation by assessing the effects of exendin-4 (Ex-4) and peptide YY (PYY) (3-36) on endocrine ovaries and adrenals in high-fat diet (HFD) mice. Ex-4 and PYY(3-36) reduced blood glucose and energy intake, with no effects on body weight. While HFD did not impact the estrous cycle, Ex-4 increased metestrus frequency and decreased diestrus frequency resulting in 0% mi

Pre-pregnancy treatment of semaglutide improves high-fat-diet-induced placental and fetal hypothalamic alterations in a sex-specific manner in rats†.

INTRODUCTION: The impact of obesity on pregnancy outcomes and offspring health has received increasing attention. Semaglutide have been shown to have significant effects on weight management. This study aims to evaluate whether Pre-pregnancy Semaglutide Intervention (PpSI) improves maternal metabolic disorders before and during pregnancy, affects placental function and fetal hypothalamic neuropeptide expression during late-gestation and its underlying mechanisms. METHODS: In this study, female SD rats were divided into three groups: control diet (CHOW), high-fat diet (HFD), and high-fat diet

Potential Modulatory Role of Phoenixin-14 in Epithelial-Mesenchymal Transition of Endometriotic 12Z Cells.

Background/Objectives: Endometriosis is a painful chronic condition in which the endometrium grows outside the uterus. The epithelial-mesenchymal transition (EMT) is critical to endometriosis progression, where cells lose epithelial traits and gain invasiveness. Methods: This study investigates the effects of phoenixin-14 (PNX-14), a neuropeptide found at reduced levels in endometriosis patients, on the expression of two molecular EMT markers, CDH1 (E-cadherin) and THBS2 (thrombospondin 2), as well as cell viability in the endometriosis-derived 12Z cell line. Cells were treated with physiologi

Orexin-A and Circadian Disruption in Alzheimer's Disease: Implications for Amyloid-Beta Pathology.

Alzheimer's disease (AD) is characterized by cognitive decline, circadian rhythm disruptions, and accumulation of Aβ plaques. Orexin-A, a neuropeptide involved in regulating sleep and circadian rhythms, has been implicated in these processes, although its specific role in modulating β-amyloid (Aβ) aggregation remains unclear. This study investigates how orexin-A influences Aβ aggregation and its impact on cognitive and circadian dysfunctions in AD mice subjected to acute sleep deprivation (ASD). Behavioural assessments showed significant cognitive deficits following ASD, including impaired rec

Comparative Transcriptomics Provides Insight into the Neuroendocrine Regulation of Spawning in the Black-Lip Rock Oyster (Saccostrea echinata).

The black-lip rock oyster, Saccostrea echinata, is an emerging aquaculture species; however, difficulties in regulating their gonad conditioning to full maturation and spawning have impacted industry progress. Addressing this challenge requires a deeper understanding of the molecular mechanisms underlying reproduction, particularly the signalling molecules (e.g., neuroendocrine hormones) that regulate gonad development and spawning, which remains poorly understood in this species. Therefore, we investigated the molecular neuroendocrine regulation of gonad maturation in S. echinata through the

Pleiotropic effects of oxytocin receptor polymorphisms: influencing striatocortical connectivity in bipolar disorder.

BACKGROUND: Oxytocin (OXT), a neuropeptide involved in social behaviors and emotions, exhibits bidirectional effects depending upon positive or negative environments. Our previous report highlighted dysregulation of OXT on striatocortical functional connectivity (FC) in bipolar disorder (BD) patients. We hypothesized that: (1) in healthy controls (HC), carriers of a "sensitive" OXTR allele would show altered FC, particularly in association with childhood trauma; and (2) this gene-brain relationship would be fundamentally altered or reversed in BD patients, reflecting a gene-disease interaction

High-fat diet-induced obesity enhances stress vulnerability and promotes a PTSD-like phenotype in rats.

The association between post-traumatic stress disorder (PTSD) and subsequent obesity is well-established in humans, however, whether obesity exacerbates vulnerability to PTSD remains underexplored. To investigate this, we employed a rat model fed either a high-fat diet (HFD; 60 % kcal from fat) or a control diet (CD). After confirming significant body mass index differences between HFD and CD groups, rats were exposed to predator scent stress (PSS) or a sham-PSS control. Behavioral phenotyping was conducted using the elevated plus maze (EPM) and acoustic startle response (ASR) to classify stre

Mapping cerebral blood perfusion and its links to multi-scale brain organization across the human lifespan.

Blood perfusion delivers oxygen and nutrients to all cells, making it a fundamental feature of brain organization. How cerebral blood perfusion maps onto micro-, meso- and macro-scale brain structure and function is therefore a key question in neuroscience. Here we analyze pseudo-continuous arterial spin labeling (ASL) data from 1305 healthy individuals in the HCP Lifespan studies (5-22 and 36-100 years) to reconstruct a high-resolution normative cerebral blood perfusion map. At the cellular and molecular level, cerebral blood perfusion co-localizes with granular layer IV, biological pathways

Sex-Specific Effects of Hypocretin Receptor Signaling in Corticotropin-Releasing Factor Neurons on Alcohol Drinking, Anxiety, and Extended Amygdala Neuronal Excitability.

BACKGROUND: Alcohol use disorder (AUD) is characterized by compulsive alcohol consumption and negative emotional states during withdrawal, often perpetuating a cycle of addiction through arousal dysfunction. The hypocretin/orexin (HCRT) neuropeptide system is a key regulator of arousal that is implicated in these processes, particularly in its interactions with corticotropin-releasing factor (CRF) neurons within the bed nucleus of the stria terminalis (BNST). METHODS: Using CRF-specific genetic deletion of Hcrt r 1 and/or Hcrt r 2 receptors in mice combined with behavioral and electrophysiolo

A peptide-neurotensin conjugate that crosses the blood-brain barrier induces pharmacological hypothermia associated with anticonvulsant, neuroprotective, and anti-inflammatory properties following status epilepticus in mice.

Preclinical and clinical studies show that mild to moderate hypothermia is neuroprotective in sudden cardiac arrest, ischemic stroke, perinatal hypoxia/ischemia, traumatic brain injury, and seizures. Induction of hypothermia largely involves physical cooling therapies, which induce several clinical complications, while some molecules have shown to be efficient in pharmacologically induced hypothermia (PIH). Neurotensin (NT), a 13 amino acid neuropeptide that regulates body temperature, interacts with various receptors to mediate its peripheral and central effects. NT induces PIH when administe

The intricate relationship between circadian rhythms and gastrointestinal peptides in obesity.

There are different molecular pathways that regulate appetite, particularly the role of the hypothalamus, circadian rhythms, and gastrointestinal peptides. The hypothalamus integrates signals from orexigenic peptides like neuropeptide Y (NPY) and agouti-related protein (AgRP), which stimulate appetite, and anorexigenic peptides such as pro-opiomelanocortin (POMC) and cocaine- and amphetamine-regulated transcript (CART), which promote satiety. These signals are influenced by peripheral hormones like leptin, ghrelin, insulin, and cortisol, as well as gut peptides including glucagon-like peptide-

The oxytocin system mediates behavioral and neurobiological alterations associated with early adversity.

Early life adversities (ELA) can significantly impact brain development and adult behavior, potentially increasing vulnerability to psychopathologies. Evidence shows that ELA exposure is significantly associated with dysfunctional Oxytocin (OXT), a neuropeptide strongly engaged in social behavior and linked to the processing of rewarding stimuli, such as drugs of abuse. Moreover, it has been recently demonstrated that peripheral OXT may be transported to the brain through several mechanisms, including Receptors for Advanced Glycation End-Products (RAGE), and the RAGE-mediated OXT transport has

Calcitonin gene-related peptide in the audiovestibular system.

OBJECTIVE: This study aimed to summarize and analyze the current literature related to calcitonin gene-related peptide (CGRP), the vestibular system, and vestibular migraine. BACKGROUND: CGRP is a neuropeptide that has been implicated in the pathophysiology of migraine. Vestibular migraine (VM) is a subtype of migraine that causes recurring episodes of vestibular symptoms like vertigo and is often associated with migrainous symptoms. Although the pathophysiology of VM is not completely understood, CGRP expression in the central and peripheral vestibular systems has suggested that it may also

Enterobacterial caseinolytic protease B (ClpB) and ClpB antibodies in adolescents with anorexia nervosa.

Caseinolytic protease B (ClpB) produced by the gut enterobacteria displays anorexigenic effects possibly due to its molecular mimicry with α-melanocyte-stimulating hormone (α-MSH), an anorexigenic neuropeptide. ClpB is known to induce α-MSH cross-reactive antibodies previously associated with eating disorders. In the present study we analyzed whether long-term changes of serum ClpB, anti-ClpB antibodies and fecal clpB gene content can be associated with disease progression in anorexia nervosa (AN). For this purpose, female adolescents diagnosed with the restrictive type of AN according to DSM-

Antagonizing NK-1R modulates pain perception following corneal injury.

Substance P (SP) expressed by corneal nerves, is an 11-amino acid long neuropeptide from the tachykinin family, encoded by the Tac1 gene, and binds to neurokinin receptors. SP overexpression is associated with various pathological responses in the cornea including vasodilation, pain, inflammation, and angiogenesis in the normally avascular tissue. This study investigates the role of neurokinin-1 receptor (NK-1R) mediated signaling in nociception, nerve regeneration, and neuronal activation following mechanical corneal injury in mice. Corneal injuries were induced in age- and sex-matched C57BL/
