Understanding the experience of women living with obesity and reproductive pathology.

INTRODUCTION: Obesity in women of reproductive age is a significant risk factor for infertility. Current treatment strategies primarily focu … rvey was administered and completed by 33 out of a total of 150 women with obesity and reproductive pathology who were invited to participate (response rate 22%). RESULTS: While some aspects of preconception care were positively evaluated, the overall patient journey is complex. We identified four areas for improvement: providing personalized information, …

Applications of artificial intelligence in pediatric surgical pathology: A systematic review.

BACKGROUND: Artificial Intelligence (AI) techniques can transform and enhance diagnosis and treatment response predictions in pediatric surgical pathology. AI offers the potential to reduce the workload of pathologists by automating routine and labor-intensive tasks. In this systematic review, we investigate current applications of computational pathology in pediatric surgical conditions. METHODS: Nine databases were searched from inception until January 2025 to retrieve articles looking at the use of machine learning, AI, or virtual reality in the pathological diagnosis of pediatric surgical…

DICER1 Syndrome and Tumor Pathology: An Updated Review for Diagnostic Practice.

DICER1 syndrome is an autosomal dominant tumor predisposition disorder caused by pathogenic variants in the microRNA-processing gene DICER1. Since its initial recognition in families with pleuropulmonary blastoma, the phenotypic spectrum has expanded to encompass a wide array of benign and malignant neoplasms-including thyroid follicular nodular disease, Sertoli-Leydig cell tumor, cystic nephroma, pituitary blastoma, and embryonal rhabdomyosarcoma of the cervix-often presenting in childhood or early adulthood. This updated review synthesizes current knowledge of DICER1 biology, the historical

ADAM19: biological characteristics and implications in disease pathology and clinical applications.

PURPOSE OF REVIEW: This article critically examines ADAM19-mediated inflammatory remodeling within five distinct subtypes of solid tumors prone to inflammation, employing non-neoplastic inflammatory states strictly as a comparative reference. As a transmembrane protease belonging to the ADAM family, ADAM19 undergoes maturation through plasmin-dependent cleavage, with its expression tightly regulated by both transcriptional and epigenetic controls. Although dysregulated ADAM19 levels are evident in various inflammatory lesions and malignancies, its context-specific dual biological roles necessi

CHCHD2: a bi-organellar fulcrum of mitochondrial homeostasis and reproductive pathology.

Mitochondrial dysfunction has been demonstrated with a role in pathologically driving various obstetric and gynecological (OB/GYN) diseases, but molecular mediators associating mitochondrial dysfunction with discrete pathologies have not been determined yet. CHCHD2, a bi-organellar protein called MNRR1 as well, integrates mitochondrial stress sensing with nuclear adaptive responses; nonetheless, its effect on reproductive disorder remains substantially paradoxical, which creates an important translational roadblock. Such ambiguity may not be arbitrary, instead, it may be associated with the un

Explainable AI for classifying vertebral fracture histology in digital spine pathology.

INTRODUCTION: Vertebral compression fractures (VCFs) commonly arise from osteoporosis, trauma, or malignancy. Accurate subtype differentiation is clinically essential but remains challenging using conventional imaging and histology. METHODS: We developed an explainable AI-driven digital pathology pipeline integrating whole-slide histopathology with clinical metadata and transcriptomic profiles to support fracture subtype classification, risk stratification, and therapy prediction. Model interpretability was assessed using Grad-CAM heatmaps and SHAP analysis, and the multi-omics risk score was…

From Pathology to Formulation: Designing Biodegradable Polymers for Personalized Drug Delivery.

Background/Objectives: Selection of polymer carriers for targeted drug delivery is typically guided by material availability or trigger responsiveness rather than disease-specific evidence. However, successful preclinical formulations may already encode implicit design rules linking polymer composition to particular pathological environments. This study aimed to identify reproducible material-disease associations across biodegradable polymer systems and to derive formulation-oriented guidance for disease-calibrated carrier selection. Methods: A structured synthesis of 65 preclinical in vivo st

The influence of spinal pathology on extremity pain: a clinical framework.

Musculoskeletal extremity pain is frequently diagnosed based on symptom location and region-specific musculoskeletal special tests despite evidence demonstrating limited diagnostic accuracy and increased risk of misdiagnosis. Literature suggests that a proportion of extremity pain is influenced by nociceptive spinally mediated somatic referred pain, which can occur in the absence of spinal symptoms, often misclassified as radicular pain or radiculopathy. Overreliance on pain location and special tests with poor clinometric properties increases the likelihood that spinal contributions are overl

Reduced ULK1 links impaired autophagy and mitophagy to Alzheimer's disease pathology.

ULK1 (Atg1) initiates macroautophagy and mitophagy, which support neuronal growth and survival, yet how this pathway is disrupted in aging and Alzheimer's disease (AD) remains unclear. Here we report reduced ULK1 in serum and cerebrospinal fluid during aging in cognitively unimpaired participants from the COGNORM study (n = 75) and in patients with AD from the NorCog Memory Clinic Cohort (n = 316). In AD mice, ULK1 overexpression stimulates autophagic flux, reduces AD pathology and delays cognitive decline alongside increased phagocytic degradation of amyloid-β, reduced tauopathy and improved …

Meniscus Pathology in the Setting of Revision ACL Surgery: Unique Considerations.

PURPOSE OF REVIEW: Similar to primary anterior cruciate ligament reconstruction (ACLR), meniscal pathology is frequently encountered during revision ACLR (RACLR). In the revision setting, however, meniscal disease often presents a greater challenge because tissue quality may be compromised, particularly when prior meniscal treatment was performed at the index ACLR. This review highlights key considerations and treatment strategies for managing extensive, complex meniscal lesions in the setting of RACLR. RECENT FINDINGS: Graft failure significantly increases the incidence of meniscal tears. Fu…

Deep learning via pathology-image integration for margin prediction after pancreatoduodenectomy.

INTRODUCTION: R1 margin status after pancreatoduodenectomy (PD) for pancreatic ductal adenocarcinoma (PDAC) remains common and is associated with inferior survival. Preoperative imaging inadequately predicts microscopic margin involvement, particularly at the superior mesenteric artery interface. This study evaluated whether deep learning applied to preoperative CT imaging could predict pathological margin status. METHODS: Patients who underwent PD for PDAC between 2007-2021 were identified from a prospectively maintained database. Preoperative triphasic CT scans were processed into standardi

Advancing Placental Pathology With Artificial Intelligence: Toward Reproducible and Predictive Diagnostics.

Placental pathology remains underdeveloped in its application of artificial intelligence (AI) and digital pathology, despite established diagnostic guidelines and increasing clinical demand. Determining which placentas require histologic examination continues to strain staffing and resources, while histologic interpretation is limited by high interobserver variability and a lack of scalable analytic tools. This review synthesizes current applications and future opportunities for AI in placental pathology across multiple domains, including gross examination, villous maturation, lesion detection…

Lip filler-related granulomatous inflammation mimicking oral pathology: A case report.

BACKGROUND: Injectable dermal fillers are widely used in esthetic practice for lip augmentation and perioral soft-tissue enhancement. Althou … ns associated with injectable dermal fillers may manifest as labial lesions that mimic primary oral pathology. Dentists should obtain a complete history of prior esthetic procedures when evaluating unexplained perioral or labial lesions. Appropriate surgical management and histopathologic confirmation are important when the diagnosis is uncertain.

N-terminal acetylation reduces synuclein pathology in models of Parkinson's disease.

α-synuclein protein is a major constituent of pathological intracellular inclusions such as Lewy bodies found in brains of patients with Parkinson's disease and other synucleinopathies. Whereas α-synuclein phosphorylation has been much studied, comparatively less work has been devoted to other post-translational modifications such as acetylation, especially given that N-terminally acetylated α-synuclein is the most abundant endogenous form of the protein in the brain. In this study, using multiple in vitro and in vivo models of Parkinson's disease, we sought to understand the pathological cons

Cinnamaldehyde improves the pathology of vascular cognitive impairment via astrocytic TRPA1.

Vascular cognitive impairment (VCI) comprises cognitive dysfunction caused by vascular pathology and is associated with various forms of dementia. VCI is typically caused by chronic cerebral hypoperfusion (CCH), which is accompanied by oxidative stress, central nervous system inflammation, microglial activation, and white matter injury. Therapeutic targets for VCI remain largely unknown. Transient receptor potential ankyrin 1 (TRPA1) is functionally expressed in the brain, where it senses changes in oxygen levels and is activated by hypoxia and hyperoxia. We investigated the pathophysiological…

Dual Pathology: A Case of Concurrent Lymphocytic Esophagitis and Crohn's Disease.

Crohn's disease (CD) is a chronic, systemic disease characterized by transmural inflammation of the gastrointestinal (GI) tract. Patients can present with a range of GI and extraintestinal features depending on the areas involved. Esophageal involvement can be a direct result of Crohn's or other co-existing diseases. Determining the cause of esophagitis in CD is important as it affects management and prognosis. Lymphocytic esophagitis (LE) is an example of a disease that can cause esophagitis on a background of CD. Patients presenting with the two conditions simultaneously are relatively rare.

Long-term locus coeruleus stimulation exacerbates tau pathology in PS19 mice.

BackgroundAlzheimer's disease (AD) is the most common form of dementia, characterized by the accumulation of amyloid-β (Aβ) plaques and hyperphosphorylated tau tangles. The locus coeruleus (LC) is among the first brain regions to show degeneration and tau pathology during the early stages of AD. Previous studies have demonstrated that short-term chemogenetic LC stimulation can improve memory performance in the TgF344-AD rat model, while long-term norepinephrine reuptake inhibition can worsen memory deficits in the ADLPTau mouse model. However, the effects of long-term LC stimulation in tau mou…
