Bacterial soft rot, caused by Pectobacterium carotovorum subsp. carotovorum, affects numerous vegetable crops. Beneficial microbes like Bacillus amyloliquefaciens PMB05 offer a sustainable alternative to traditional copper- or antibiotic-based controls, which risks the development of pathogen resistance. Bacillus amyloliquefaciens PMB05 enhances disease resistance to soft rot by intensifying PAMP-triggered immunity (PTI). Although our previous research showed that PMB05-intensified PTI correlates with a reduced photosynthetic electron transport rate under high light conditions, it remains uncl

Whole-genome duplication (WGD) is a recurrent feature of plant evolution and crop improvement, yet its direct contribution to abiotic stress tolerance remains difficult to disentangle from hybridization effects. Using full-sibling diploid (2x) and neotetraploid (4x) Medicago sativa genotypes generated through bilateral sexual polyploidization, we investigated how WGD alone reshapes physiological performance and gene-miRNA regulatory networks under salt stress. Neotetraploid plants displayed enhanced tolerance to salinity, maintaining higher chlorophyll content, stomatal conductance, and shoot

Tree seedling regeneration after wildfire will determine the long-term trajectory of forests under climate change, yet increasing drought and heat extremes can suppress regeneration and result in forest type conversion. We combined field observations and a controlled drought-heat growth chamber experiment to investigate how these stressors independently and jointly affect drought- and heat-tolerance physiology in seedlings of montane conifer species from drought- and wildfire-prone regions in the southern Sierra Nevada. In both the field and growth chamber experiment, we found that exposure to

The m⁶A demethylase ALKBH10B emerges as a crucial post-transcriptional regulator of hormonal crosstalk, photosynthetic maintenance, and mitochondrial energy metabolism to fine-tune plant stress adaptation to drought. Epitranscriptomic N⁶-methyladenosine (m⁶A) mRNA modification has emerged as an important regulatory layer of gene expression and protein synthesis in plant stress adaptation. In this study, expression profiling revealed that drought stress induces a global reprogramming of the m6A machinery in Arabidopsis thaliana, with the m6A demethylase ALKBH10B exhibiting a more pronounced tim

Shade-tolerant herbs occurring across contrasting radiation environments may differ not only in photosynthetic capacity but also in how assimilated carbon is stored and used. We compared two varieties of Paris polyphylla associated with lower- and higher-elevation habitats: P. polyphylla var. chinensis (PPC) and P. polyphylla var. yunnanensis (PPY), respectively. Plants were grown for 40 d under light intensity of 50, 200, and 400 μmol m-² s-¹, and we measured leaf structure, gas exchange, chlorophyll fluorescence, carboxylation traits, non-structural carbohydrates, biomass allocation, and Par

Abies beshanzuensis M. H. Wu is a critically endangered conifer endemic to cool, high-elevation forests; its narrow range renders it vulnerable to climate warming and associated heat and water stress. We simulated climate warming by growing seedlings along an elevational gradient (500-1200 m) to assess physiological and molecular stress responses. Seedlings at mid-high elevation (1000-1200 m) grew optimally, whereas low-elevation (500 m) seedlings showed stress symptoms. At 500 m, Yield and ETR declined sharply; stomatal conductance also dropped, and NPQ collapsed entirely, dropping below the

Leaf gas exchange is the key driver of forest carbon uptake and directly determines forest carbon sink activity. Additionally, plants release a variety of biogenic volatile organic compounds (VOCs) acting as stress signals of trees. However, continuous hourly resolved measurements of leaf gas exchange and VOC emissions in tall tree canopies are challenging and remain scarce. To this end, we developed a complex in-situ leaf gas exchange measurement system with 24 cuvettes deployed on mature Fagus sylvatica L. (n = 3) and Pseudotsuga menziesii (MIRBEL) FRANCO (n = 3) individuals in a mixed tempe

During the last two decades, a number of research outcomes have vetted possible plant responses to elevated CO2 (eCO2) and other climate stressors in temperate trees and agricultural crops. However, such climate change repercussions (elevated CO2, O3, temperature, and drought) on tropical trees, especially on dominant trees of the Indian subcontinent, remain largely unexplored. This study investigates the productivity of selected species (Pongamia pinnata, Ficus benghalensis, Azadirachta indica, and Syzygium cumini) under three experimental settings, 1) Combined Stress Condition (SC) - Carbon

Drought stress is a major constraint on global forestry productivity and poses a significant threat to economically important tree species such as Populus. Abscisic acid (ABA) signaling plays a central role in drought tolerance by regulating molecular, physiological, and biochemical processes. This review summarizes current knowledge of ABA signaling pathways and their roles in drought resilience in Populus. It covers physiological responses, molecular regulatory networks, hormonal crosstalk, biotechnological advances, and emerging field applications. Core ABA signaling components, including P

BACKGROUND: Saline-alkali stress (SA) can significantly limit the growth and yield of soybean. The grain filling stage (R6) is a crucial growth period that determines the yield of soybeans and is also the most complex stage of sucrose-starch metabolism. Tryptophan (Trp) is an essential amino acid for protein synthesis and also an important signaling molecule in plants, plays an important role in maintaining osmotic regulation in plants and resisting adverse external environments. However, the mechanism of Trp regulation on sucrose-starch metabolism in R6-stage soybean leaves and seeds under SA

BACKGROUND: Water deficits, exacerbated by climate change and unpredictable weather, have become a significant global challenge to agricultural productivity. In this context, exogenous melatonin treatment is well documented as a stress alleviator; however, its effects on various biological processes, particularly in less-explored genotypes, remain understudied. This study aimed to enhance water deficit resilience in sweet corn by applying foliar melatonin to four genotypes-Messenger, Dessert, Royalty, and Tyson under two levels of water deprivation induced by polyethylene glycol at 8% and 12%

Lower atmospheric pressure affects biologically relevant physical parameters such as gas partial pressure and concentration, leading to increased water vapor diffusivity and greater soil water content loss through evapotranspiration. This might impact plant photosynthetic activity, resource allocation, water relations, and growth. However, the direct impact of low air pressure on plant physiology is largely unknown. This study examined the effects of low air pressure, alone and combined with two water inputs, on different functional traits of three plant species transplanted from montane grass

BACKGROUND: Rabies is a fatal, but vaccine-preventable disease. The conventional 'gold standard' recommended for estimating anti-rabies vaccine immune response is Rapid Fluorescent Focus Inhibition Test (RFFIT). Here, we describe a pseudovirus-based neutralization assay (PVNT) as an alternative method to detect the neutralizing antibody response.
METHODOLOGY/PRINCIPAL FINDINGS: We used a recombinant Vesicular Stomatitis Virus (rVSV)-based pseudotyping system in HEK293 cells transfected with rabies G glycoprotein to produce high titre pseudoviruses. These were confirmed by neutralization with …

BACKROUND: The utilization of high-quality water in agriculture is increasingly constrained by climate change, affecting availability, quality, and distribution due to altered precipitation patterns, increased evaporation, extreme weather events, and rising salinity levels. Salinity significantly challenges salt-sensitive vegetables like lettuce, particularly in a greenhouse. Hydroponics water quality ensures nutrient solution stability, enhances nutrient uptake, prevents contamination, regulates pH and electrical conductivity, and maintains system components. This study aimed to mitigate salt

Widespread lead (Pb) contamination of agricultural soils is a global issue stemming from human activities. The remediation of Pb-contaminated soils used for agricultural purposes is critically important to safeguard food crop safety. Despite the modulating effects of sulfur (S) on plant responses to toxic heavy metals, the ecological, physiological, and molecular mechanisms driving such modulation in the Pb hyperaccumulator Arabis alpina L. remain unclear. Here, we investigated the effects of five S concentrations (0, 50, 100, 150, and 200 mg kg-1) on A. alpina grown in Pb-contaminated soil fr

Maize leaf blight (MLB), caused by the fungus Bipolaris maydis, is an important disease affecting maize production. In order to minimize the use of fungicides in agriculture, nutrient-based resistance inducers may become a promising alternative to manage MLB. The goal of this study was to investigate the potential of Semia® (zinc (20%) complexed with a plant-derived pool of polyphenols (10%)) to hamper the infection of maize leaves by B. maydis by analyzing their photosynthetic performance and carbohydrate and antioxidative metabolism, as well as the expression of defense-related genes. Soil cadmium (Cd) contamination threatens food safety and human health, particularly in developing countries. Previously, we have proposed that boron (B) could reduce Cd uptake and accumulation in hot peppers (Capsicum annuum) by regulating the expression of genes related to Cd transport in roots. However, only few studies have examined the role of B in plant leaves under Cd stress. It is unclear how B induces the expression of relevant genes and metabolites in hot pepper leaves and to what extent B is involved in leaf growth and Cd accumulation. The purpose of this study was to investigate th
