Longitudinal analysis of the electroencephalogram and sleep phenotype in the R6/2 mouse model of Huntington's disease.

Deficits in sleep and circadian organization have been identified as common early features in patients with Huntington's disease that correlate with symptom severity and may be instrumental in disease progression. Studies in Huntington's disease gene carriers suggest that alterations in the electroencephalogram may reflect underlying neuronal dysfunction that is present in the premanifest stage. We conducted a longitudinal characterization of sleep/wake and electroencephalographic activity in the R6/2 mouse model of Huntington's disease to determine whether analogous electroencephalographic 's

Antibody response and antigen-specific gamma-interferon profiles of vaccinated and unvaccinated pregnant sheep experimentally infected with Brucella melitensis.

It is well known that the immune response in sheep against Brucella melitensis is subject to individual variation, depending on diverse factors. It bears asking whether these factors (e.g. clinical disease, active infection, state of previous immunity), when affecting a group, can cause variation in the performance of different diagnostic tests. To clarify some of the circumstances in which this immune response can vary, we examine the immune-response profile of sheep protected against the clinical disease by prior vaccination with strain Rev. 1 in comparison with the profile of unprotected fe

Rational Design of Live-Attenuated Vaccines against Herpes Simplex Viruses.

Diseases caused by human herpes simplex virus types 1 and 2 (HSV-1 and HSV-2) affect millions of people worldwide and range from fatal encephalitis in neonates and herpes keratitis to orofacial and genital herpes, among other manifestations. The viruses can be shed efficiently by asymptomatic carriers, causing increased rates of infection. Viral transmission occurs through direct contact of mucosal surfaces followed by initial replication of the incoming virus in skin tissues. Subsequently, the viruses infect sensory neurons in the trigeminal and lumbosacral dorsal root ganglia, where they are

Invisible poison attacks and holistic defence strategy for pyramid vision transformers in medical imaging.

Machine learning models, especially vision transformers in the domain of medical images, are highly prone to data poisoning attacks, in which a small proportion of adversarial samples is injected into the model's training dataset to manipulate its behavior. Existing data poisoning techniques have their limitations in terms of the presence of noticeable artifacts in the injected samples or their vulnerability to preprocessing transformations. Similarly, most defence techniques have their limitations in terms of robustness to different types of poisoning attacks. To overcome these challenges, th

MechanoAge, a machine learning platform to identify individuals susceptible to breast cancer based on mechanical properties of single cells.

BACKGROUND: Emerging evidence links cellular ageing and biophysical alterations with cancer susceptibility. Existing breast cancer risk models inadequately identify individuals at latent risk, particularly among women without known genetic mutations or family history. Risk is often underestimated or overestimated due to reliance on population-level data and absence of individualised tissue-based markers of breast cancer risk. METHODS: We profiled primary human mammary epithelial cells (HMECs) from women of varying ages and risk backgrounds using mechano-node-pore sensing (mechano-NPS), a high

Silent transmission of human African trypanosomiasis: isolation of a human-infective trypanosome from an asymptomatic pregnant woman in south-eastern Nigeria.

BACKGROUND: Human African trypanosomiasis (HAT) is an endemic neglected tropical disease in Sub-Saharan Africa targeted for elimination by 2030. However, asymptomatic human infections may sustain cryptic transmission. Nigeria has not reported confirmed HAT cases in over a decade, but emerging evidence suggests ongoing silent transmission. Infection during pregnancy raises additional concerns, including adverse outcomes and vertical transmission. This study therefore investigated the presence of trypanosome infection in pregnant women attending the antenatal clinic in Enugu, Nigeria. METHODS:

Community-Based Hereditary Breast and Ovarian Cancer Family History Assessment and Genetic Testing among Spanish-Speaking Hispanic/Latina Women in California.

UNLABELLED: Breast cancer is the most common cancer among women in the United States and the leading cause of cancer incidence and mortality among Hispanic/Latina (H/L) women. For women with a genetic predisposition to hereditary breast and ovarian cancer (HBOC), genetic counseling has been shown to aid in the decision-making process and engagement in risk-reducing strategies for cancer prevention and early detection. However, H/L women are less likely to receive genetic testing and undergo genetic counseling than non-Hispanic White women. We developed a promotores-led virtual HBOC outreach, e

Depression in Premanifest Huntington's Disease: Aberrant Effective Connectivity of Striatum and Default Mode Network.

BACKGROUND: Depression frequently precedes motor symptoms in Huntington's disease gene expansion carriers (HDGECs), yet the neural mechanisms remain poorly characterized. OBJECTIVE: We investigated effective connectivity between the default mode network (DMN) and striatal regions in HDGECs. METHODS: We analyzed 3-T resting-state functional magnetic resonance imaging data from 98 HDGECs (48.98% females; mean age, 42.82 years). Spectral dynamic causal modeling estimated subject-level connectivity, whereas parametric empirical Bayes determined group-level effective connectivity differences betw

Development of solid dispersions based on 3- (2,6-difluorobenzyl) -5- (5-bromo-1H-indol-3-ylmethylene) thiazolidine-2,4-dione for schistosomicidal treatment.

Schistosomiasis is an endemic disease in Brazil. It is important to broaden the treatment options to control and containment of the disease. Thiazolidine derivatives appear as important alternatives to treatment. In vitro studies have demonstrated excellent schistosomiasis activity for LPSF/GQ-238. The molecule, however, has poorly water-soluble. This study focused on increasing the aqueous solubility of LPSF/GQ-238 by obtaining solid dispersions. Were prepared by the solvent techniques, using Soluplus®, Polyethylene glycol (PEG), and Polyvinylpyrrolidone (PVP-K30) as carriers. Solubility test

The cost-effectiveness of HLA-B*5801 screening to guide initial urate-lowering therapy for gout in the United States.

OBJECTIVE: Positive HLA-B*5801 carriers are at greater risk of experiencing rare but severe allopurinol hypersensitivity syndrome (AHS) [i.e., Stevens-Johnson syndrome (SJS) and toxic epidermal necrolysis (TEN)]; however, HLA-B*5801 prevalence and AHS risk vary by race/ethnicity. We evaluated the cost-effectiveness of HLA-B*5801 testing according to race/ethnicity in the United States. METHODS: We determined the cost-effectiveness of universal testing for HLA-B*5801 compared to no testing prior to the initiation of allopurinol per US major race/ethnicity groups. Using US-specific data, SJS/TE

Follow-up compliance after endovascular abdominal aortic aneurysm repair in Medicare beneficiaries.

OBJECTIVE: Lifelong imaging follow-up is essential to the safe and appropriate management of patients who undergo endovascular abdominal aortic aneurysm repair (EVAR). We sought to evaluate the rate of compliance with imaging follow-up after EVAR and to identify factors associated with being lost to imaging follow-up. METHODS: We identified a 20% sample of continuously enrolled Medicare beneficiaries who underwent EVAR between 2001 and 2008. Using data through 2010 from Medicare Inpatient, Outpatient, and Carrier files, we identified all abdominal imaging studies that may have been performed

Prevalence of Stenotrophomonas maltophilia colonization among patients receiving mechanical ventilation in long-term care facilities in Maryland, United States.

There is increasing concern regarding the importance of Stenotrophomonas maltophilia as a nosocomial infection, given its inherent antibiotic resistance, particularly for patients who have risk factors such as being mechanically ventilated. The prevalence of colonization receiving mechanical ventilation in chronic care facilities has not been studied. The Multi-Drug-Resistant Organism (MDRO) Prevention Collaborative performed a point prevalence of patients receiving mechanical ventilation in all 18 chronic care facilities in Maryland between 7 March 2023 and 8 June 2023. Patients had at least

Antimicrobial-resistant organism carriage and metagenomic characterization of the microbiome and resistome in veterinary workers.

BACKGROUND: Veterinary personnel work in environments with high exposure to antimicrobials and antimicrobial-resistant pathogens. However, the extent to which these occupational exposures influence their carriage of antimicrobial-resistant (AMR) organisms and the composition of their microbiome and resistome remains poorly understood. The objective of this study was to characterize AMR pathogen carriage and microbiome/resistome composition in veterinary personnel and to assess whether workplace setting and patient-related exposures were associated with detectable differences in these outcomes.

Colonization of extended-spectrum beta-lactamase-producing bacteria in healthy pregnant women and its impact on perinatal care: A cross-sectional study.

BackgroundColonization of extended-spectrum beta-lactamase (ESBL)-producing organisms is increasing becoming more frequent not only in hospitalized patients but also in healthy individuals. Although these bacteria are thought to be transmitted to newborns on their way through the birth canal, molecular evidence for this is scarce. In this study, we aimed to survey the current prevalence of resistant bacterial colonization in this area by examining the colonization carriage of this organism before and after delivery.MethodsWe examined the colonization rate of ESBL-producing bacteria in healthy

Chronic High Epstein-Barr Viral Load Carriage Is Positively Correlated With Tacrolimus Intra-Patient Variability After Pediatric Liver Transplantation.

Chronic high Epstein-Barr virus (EBV) load (CHL) carriage has been closely associated with EBV infection after pediatric liver transplantation. Elevated tacrolimus (Tac) blood concentrations increased the risk of EBV-associated diseases. Tacrolimus intra-patient variability (Tac-IPV) help predict poor outcomes. This study examines whether Tac-IPV correlate with CHL carriage in living-donor-dominant pediatric liver transplantation. We analyzed the clinical data of 153 pediatric liver transplant recipients receiving Tac treatment with a 2-year follow-up period. Tac-IPV quantification as coeffici

Faecal microbiota transplant to ERadicate gastrointestinal carriage of Antibiotic-Resistant Organisms (FERARO): A feasibility randomised controlled trial.

OBJECTIVES: The gastrointestinal tract (GIT) is a reservoir of multidrug-resistant organisms (MDRO). Colonisation with MDRO precedes invasive infections, which can be challenging to treat with excess morbidity and mortality compared to antimicrobial-susceptible infections. Currently, there are no effective GIT decolonisation strategies. Whilst faecal microbiota transplant (FMT) has emerged as a potential therapeutic, there remains uncertainty about its feasibility, safety, and efficacy. METHODS: Population: Patients with invasive infection with extended-spectrum beta-lactamase (ESBL-) or carb

Pneumococcal genotype 23B1 as a driver of increased 23B serotype carriage, penicillin non-susceptibility, and invasive disease in Belgium: a retrospective analysis.

Streptococcus pneumoniae serotype 23B, a non-vaccine serotype, has shown an increasing prevalence and penicillin non-susceptibility among carriage and invasive pneumococcal disease (IPD) isolates. Recently, a novel penicillin non-susceptible genotype has emerged, named 23B1. In the framework of the Belgian pneumococcal carriage study, we studied the prevalence of 23B0/23B1 among 586 23B strains (2016-2022) in 172 day care centers from 6- to 30-month-old children and among 130 pediatric 23B IPD isolates (2007-2021).
