Mouse embryo cells (C3H 10T1/2) were transfected with a plasmid (pAL8A) containing the HRas oncogene and neomycin resistance gene. Five transfected cell clones were isolated and established as cell lines, and these showed neomycin resistance. Two of these cell lines expressed a normal morphology while three showed a transformed morphology. Four of the cell lines produced tumours in nude mice. Flow cytometry measurements showed that exponentially growing cell cultures of the five transfected cell lines had the same cell cycle distribution as the normal parental cell line. The sensitivity to hyp

Breast cancer patients have high serum reactive oxygen species (ROS) levels, which exert toxicity on the ovary. However, it is still unclear whether tumor-derived ROS play a role in endometrial development and function in breast cancer. Breast cancer patients and healthy controls were recruited and endometrial thickness was measured by transvaginal ultrasound (TVUS). Xenograft tumors of the breast cancer cell line MDA-MB-231 in a female BALB/c nude mice model were established, and the therapeutic mechanism of vitamin C (VC) was investigated on uterine pathology in vivo and the contribution of

Cancer immunotherapies have induced long-lasting responses in cancer patients including those with melanoma and head and neck squamous cell carcinoma (HNSCC). However, the majority of treated patients does not achieve clinical benefit from immunotherapy because of systemic tumor-induced immunosuppression. Monocytic myeloid-derived suppressor cells (M-MDSCs) are implicated as key players in inhibiting anti-tumor immune responses and their frequencies are closely associated with tumor progression. Tumor-derived signals, including signaling via STAT3-COX-2, induce the transformation of monocytic

A minority of breast cancers is characterized by lymphoplasmacytic infiltrates that have been correlated with improved patient survivals. The association of improved prognosis with plasmacytic infiltrates has been classically linked with the rare medullary carcinoma subtype but is also evident in the smaller infiltrated fraction of the more abundant nonmedullary (not otherwise specified) tumors. It is our hypothesis that these plasma cell (PC) infiltrates represent a host humoral response driven by one or more tumor-derived neoantigens. As the index study group, two primary medullary carcinoma

Granulysin is a protein present in the granules of human CTL and NK cells, with cytolytic activity against microbes and tumors. Previous work demonstrated that granulysin caused cell death through mitochondrial damage with release of AIF and cytochrome c. However, the molecular mechanism and, especially, the type of cell death were still not well defined. In the present work we show that granulysin-induced cell death is apoptotic, with phosphatidylserine exposure preceding membrane breakdown and with caspase 3 activation. Granulysin-induced apoptosis is prevented in Jurkat cells over-expressin

Epithelioid hemangioma is a rare benign vascular tumor that consists of capillary-sized vessels lined by epithelioid endothelial cells. Diffuse cavernous hemangioma is a congenital benign vascular neoplasm consisting of increased dilated vessels. We report a case of epithelioid hemangioma and diffuse cavernous hemangioma that co-occurred in the rectum. To our knowledge, this is the first report in which two rare vascular lesions coexisted. Because both epithelioid hemangioma and diffuse cavernous hemangioma are often clinically confounded by malignant tumors, differentiating these benign lesio

Oncolytic viruses are of growing importance in cancer therapeutics since they combine direct oncolytic effect and the stimulation of antitumor immunity. Emerging evidences showed that the function of oncolytic viruses is dependent on immune response in tumor microenvironment, and the modulation of immunity could influence their efficacy. Here we combined the interleukin 10 (IL-10) and oncolytic adenovirus Ad-hTERT to treat lung cancer and explored the underlying mechanism under combination therapy. Lewis lung carcinoma (LLC) and B16F10 tumor-bearing immunocompetent C57BL/6 mice that received A

BACKGROUND: The immunological checkpoint known as Inducible T Cell Costimulatory Factor (ICOS, Cluster of Differentiation, CD278) is activated and expressed on T cells. Both somatic cells and antigen-presenting cells expressed its ligand, ICOSL (including tumor cells in the tumor microenvironment). It is important for immunosuppression. Uncertainty surrounds the function of ICOS in tumor immunity.
METHODS: Several bioinformatics techniques were employed by us to thoroughly examine the expression and prognostic value of ICOS in 33 cancers based on data collected from TCGA and GTEx. In addition,

Inhibitory receptors on natural killer cells and on a minority of T lymphocytes are major histocompatibility complex class Ia or Ib specific. We have previously reported several tumor-specific cytotoxic T cell clones infiltrating a CD4(+) V beta 13(+) cutaneous T cell lymphoma. These clones mediated a specific major histocompatibility complex class I-restricted cytotoxic activity toward the uncultured tumor cells and autologous long-term tumor T cell lines. In this study, we cultured with interleukin-2 the peripheral blood lymphocytes of the same patient a few weeks before invasion of the bloo

Although multi-hit TP53 mutations correlate with rapid disease progression, treatment resistance, recurrence, and poor survival, their occurrence within an individual TP53 gene molecule (cbTP53) remains unexplored. Consequently, the impact of cbTP53 on protein conformation, transcriptional architecture, and oncogenicity is unknown. We show that cbTP53 with combined mutations (Y220C+R248Q+R282Q) does not drastically alter local stability or structural conformation, but significantly affects TP53 binding to DNA promoters comparing it to a single Y220C mutation. Functionally, cbTP53 promotes dive

BACKGROUND: Thyroid hormone receptor-interacting protein 13 (TRIP13), an enzyme from the AAA-ATPase family, facilitates the assembly or disassembly of protein complexes and participates in various biological processes. However, its impact on cancer immune infiltration and pan-cancer prognosis remains largely unexplored.
METHODS: Pan-cancer multi-omics data from publicly available resources were systematically analyzed to evaluate TRIP13 expression across various cancer types and its association with patient prognosis. In addition, functional enrichment analyses were conducted to investigate T

Glioblastoma (GBM) is the most common and aggressive primary malignant brain tumor in adults, characterized by rapid progression and exceptionally poor prognosis. Identifying novel molecular drivers and therapeutic targets is urgently needed. This study reports a previously unrecognized MET-YANK2 signaling axis that drives glioma progression. Analysis of glioma patient samples reveals that high co‑expression of MET and YANK2 is positively correlated and significantly associated with poor survival outcomes. Mechanistically, MET directly phosphorylates YANK2 at tyrosine 282 (Y282), a conserved r

BACKGROUND: PTPN22, a protein tyrosine phosphatase (PTP) family member, has recently emerged as an intracellular regulator in adaptive immune cells. However, its function in natural killer (NK) cells remains unclear despite its high expression in innate immune cells.
METHODS: We analyzed single-cell RNA-sequencing (scRNA-seq) datasets and The Cancer Genome Atlas (TCGA) to evaluate PTPN22 expression in NK cells and its association with clinical outcomes. We established a CRISPR-mediated PTPN22 knockout platform for NK cell gene editing and evaluated PTPN22 function using both genetic deletion

Tumor-associated macrophages (TAMs) are key drivers of tumor progression, metastasis, and immune evasion in triple-negative breast cancer (TNBC). Yet, most treatment strategies focus solely on tumor cells, neglecting the immunosuppressive tumor microenvironment (TME). Given the strong correlation between TAMs infiltration and poor prognosis, innovative therapeutic strategies that modulate TAMs dynamics are urgently needed. Here, we introduce MPsomes, macrophage biomimetic nanoparticles engineered to disrupt TAMs recruitment and alter the TME. Fabricated via a microfluidic approach, MPsomes int

Cholesterol metabolism influences prostate cancer (PCa) progression, especially by affecting the tumor microenvironment. The present study demonstrated that cancer cell-intrinsic cholesterol promoted the S-palmitoylation of specificity protein 1 (SP1), enhancing SP1 nuclear translocation and driving the transcription and secretion of midkine (MDK), which in turn facilitated the differentiation of macrophages into a lipid-associated phenotype. Furthermore, targeting cholesterol metabolism with simvastatin significantly reduced MDK levels, inhibited immunosuppressive macrophage polarization, and

The dense microenvironment of triple-negative breast cancer (TNBC) poses a formidable barrier to nanomedicine penetration, rendering selective tumor retention critical for effective drug delivery. Cell-penetrating peptides (CPPs)-PEG polymers improve SN38 cellular uptake but lack tumor selectivity. We employed a quantitative structure-activity relationship (QSAR)-driven strategy to screen a combinatorial CPPs library for tumor microenvironment responsiveness. We elucidated that proline residues induce peptide bending and steric effects, resulting in PEG shield to limit cargo cellular entry. El

The immunosuppressive tumor microenvironment and vascular abnormalities are important factors affecting the anticancer effects of chemotherapeutic drugs. Previous studies have shown that moxibustion combined with cisplatin can improve tumor immunity and vascular normalization. The aim is to further explore the pathways and molecular mechanisms by which moxibustion combined with cisplatin exerts anti-tumor effects through regulating the tumor immune-vascular microenvironment. Through RNA sequencing and bioinformatics analysis, we identified related signaling pathways and key molecules, which we

BACKGROUND/AIM: The acquisition of stem-like properties and increased cellular plasticity is thought to drive tumor progression and therapeutic resistance, but grade-specific molecular trajectories across the epigenetic landscape in bladder cancer remain poorly defined. This study examined the dynamic rewiring of signaling networks and proteomic landscapes during reprogramming of low-grade (HTB-2) and high-grade (HTB-5) bladder cancer cells, and during their subsequent differentiation into embryoid bodies.
MATERIALS AND METHODS: Six experimental models were analyzed, including the parental HT

Adult T-cell leukemia/lymphoma (ATL) is an aggressive T-cell malignancy with a poor prognosis that is caused by human T-cell leukemia virus type 1 infection. We previously demonstrated that N-myc downstream-regulated gene 2 (NDRG2) is significantly downregulated in ATL, resulting in aberrant activation of the signal transduction pathways through the dissociation of serine/threonine protein phosphatase 2A. To identify potential targets of NDRG2, we performed comprehensive mass spectrometry of differentially phosphorylated peptides in ATL cells with overexpression of NDRG2 using a TiO2-based enr
