Localization of lysine residues in the binding domain of the K99 fibrillar subunit of enterotoxigenic Escherichia coli.

Modification of lysine residues with 4-chloro-3,5-dinitrobenzoate results in the loss of the binding capacity of K99 fibrillae to horse eryt … t of K99, containing Lys-132 and Lys-133, and segments of three other sialic-acid-specific lectins; cholera toxin B subunit, heat-labile toxin B subunit of Escherichia coli and CFA1 fimbrial subunit, suggesting that these segments might also be part of the receptor-binding domain in these three proteins.

Observations questioning a protective role for breast-feeding in severe rotavirus diarrhea.

To investigate whether breast-feeding protects children against rotavirus diarrhea (RVD), we compared rates of breast-feeding by age and ent … g children with RVD than among children with non-RVD whereas it was less common among children with cholera and shigellosis. Twenty percent of breast milks consumed by infants less than 1 year of age had high levels of neutralizing activity (greater than or equal to 320) to the Wa strain of rotavirus but this activity did not appear to be protective since …

Factors controlling proliferation and differentiation of canine thyroid cells cultured in reduced serum conditions: effects of thyrotropin, cyclic AMP and growth factors.

Dog thyroid cells in primary culture in a low serum, hormone-supplemented medium represent a model system which allows the direct in vitro s … rotein. All the TSH effects were completely reproduced by specific activators of adenylate cyclase--cholera toxin and the diterpene forskolin--or by dibutyryl cyclic AMP, which indicates that they are mediated by cyclic AMP (cAMP). We showed that epidermal growth factor (EGF) and pituitary fibroblast growth factor (FGF) are potent mitogens for the dog thy …

Also, RO-20-1724 does not interfere with the ability of insulin to inhibit fully cholera toxin-mediated lipolysis. It is concluded that under these conditions the antilipolytic effect of insulin may be produced without demonstrable activation of cAMP phosphodiesterase. In vitro regulation of ornithine decarboxylase in dog thyroid slices.

Ornithine decarboxylase (ODC) activity in dog thyroid slices incubated in vitro disappears when protein or RNA synthesis is inhibited. Thus, as in other tissues, the level of the enzyme reflects a balance between rapid synthesis and catabolism. TSH, dibutyryl cAMP, prostaglandin E1 (PGE1), and cholera toxin all stimulated dog thyroid ODC activity. These actions were potentiated by the inhibitor of phosphodiesterases, 4-(3-butoxy-4-methoxy-benzyl)2-imidazolidinone (Ro 20-1724), and reproduced by high concentrations of this inhibitor, while 1-methyl-3-isobutylxanthine inhibited ODC induction. Molecular Diagnostics of Pathogenic Vibrio: Target Selection, Detection Platforms, and Challenges in Field Applications.

Pathogenic Vibrio species, including Vibrio parahaemolyticus, Vibrio cholerae, and Vibrio vulnificus, are major foodborne and environmental pathogens that threaten public health. Rapid and accurate detection is crucial for clinical diagnostics, food safety monitoring, and environmental surveillance. This review provides a comprehensive overview of molecular detection strategies for pathogenic Vibrio, integrating three key aspects: (i) selection of molecular targets, including species-specific, virulence-associated, and regulatory genes; (ii) analytical performance and limitations of major dete…

ABO gene polymorphisms: a molecular bridge linking disease susceptibility to therapeutic outcomes.

ABO blood group antigens represent more than surface markers on red blood cells. They are pivotal genetic determinants that affect susceptib … f contracting gastrointestinal infections-notably from Helicobacter pylori, noroviruses, and Vibrio cholerae-and tend to suffer from more severe symptoms. Furthermore, functional variations in ABO glycosyltransferases can directly modulate drug efficacy. Consequently, the development of predictive models for disease risk and treatment response based on AB …

Cryo-EM structure of Escherichia coli HPII catalase serendipitously co-purified with a recombinant adhesin construct.

Catalases are key antioxidant enzymes that protect aerobic organisms from reactive oxygen species generated through metabolic activities and host immune responses. In Escherichia coli, the monofunctional HPII catalase (encoded by katE) forms a stable tetrameric enzyme that contributes to oxidative stress resistance. During the purification of a recombinantly produced construct of the Vibrio cholerae FrhA adhesin, we unexpectedly observed a co-purifying protein species which peptide mass spectrometry identified as E. coli HPII catalase. Initial reconstruction was limited by severe preferred ori…

The role of cytochrome oxidases in bacterial virulence.

Bacterial physiology plays an important role in virulence. Being a pathogen is energetically expensive, requiring the production of speciali … e of bd-type cytochrome oxidases during infection has been investigated mechanistically, and Vibrio cholerae and Bordetella bronchiseptica, in which non-bd-type cytochrome oxidases contribute to infection. These examples indicate the need to assess what role cytochrome oxidases play in a wider array of bacterial pathogens.

NQR as a target for new antibiotics.

The rise in antimicrobial resistance has underscored the urgent need for identification of novel targets against antibiotic resistant bacteria, which pose enormous threats to public health. The respiratory enzyme NQR carries essential roles in pathogenic bacteria, producing an ion gradient across the plasma membrane that drives ATP generation by the oxidative phosphorylation system. The vital role of NQR in a multitude of pathogenic microbes for which drug development is a high priority, such as Vibrio cholerae, Chlamydia trachomatis, and Pseudomonas aeruginosa, makes it an ideal drug target m…

Recent developments in waterborne pathogen detection technologies.

Waterborne pathogens find their way into water bodies through contamination of fecal discharge, stormwater run-offs, agriculture and industrial activities, and poor water infrastructure. These organisms are responsible for causing diarrheal, gastroenteritis, cholera, and typhoid diseases which raise an alarming sense on public human health due to the high mortality rate, especially in children. Several studies have indicated that these waterborne diseases can be managed by monitoring pathogens in water using traditional culture-based and molecular techniques. However, these methods have shown …

Efficacy and safety review of tosufloxacin in the treatment of bacterial infection in children.

In recent years, the use of antibiotics in pediatric patients has received widespread. Tosufloxacin is a quinolone antimicrobial used to treat pneumonia, otitis media, cholera, and anthrax. Guidelines have recommended that quinolone antimicrobials can be used as alternatives for the treatment of bacterial infections in children. However, due to the potential adverse effects on bones and joints, the clinical application of fluoroquinolones requires further evaluation. This article reviewed the latest clinical evidence on the efficacy and safety of tosufloxacin for treating bacterial infections …

Protection of Rabbits Against Colonization and Morbidity Associated With Toxigenic Pasteurella multocida by Immunization With Inactivated Heat-labile Toxin.

BACKGROUND/AIM: Pasteurella multocida is a significant cause of morbidity and mortality in rabbits, as well as other species. Some isolates elaborate a heat-labile toxin (PMT) that has been shown to be an important virulence factor. Though previous studies have demonstrated protective immunity can be conferred via immunization of rabbits with heat-inactivated PMT (IPMT), we investigated the ability of immunization to impact colonization of P. multocida. MATERIALS AND METHODS: Rabbits were immunized at days 0, 7 and 14 with either phosphate buffered saline (PBS), the mucosal adjuvant cholera t…
