Effects of neonatal handling on basal and stress-induced monoamine levels in the male and female rat brain.

Neonatal handling has pervasive effects on the rat brain leading to increased ability to cope with and adapt to stressful stimuli. We determined the effects of neonatal handling on the dopaminergic and serotonergic system, in the male and female rat brain, under basal conditions before and after puberty and after short- and long-term forced swimming stress. Exposure of animals to neonatal handling resulted in sex-dependent changes in the concentration and turnover of monoamines in the different brain areas. In the prepubertal brain, the effect of neonatal handling was manifested as an increase…

Imaging Agonist-Induced D2/D3 Receptor Desensitization and Internalization In Vivo with PET/fMRI.

This study investigated the dynamics of dopamine receptor desensitization and internalization, thereby proposing a new technique for non-invasive, in vivo measurements of receptor adaptations. The D2/D3 agonist quinpirole, which induces receptor internalization in vitro, was administered at graded doses in non-human primates while imaging with simultaneous positron emission tomography (PET) and functional magnetic resonance imaging (fMRI). A pronounced temporal divergence between receptor occupancy and fMRI signal was observed: occupancy remained elevated while fMRI responded transiently. Anal…

Pharmacological perturbation reveals deficits in D2 receptor responses in Thap1 null mice.

The primary dystonia DYT6 is caused by mutations in the transcription factor Thanatos-associated protein 1 (THAP1). To understand THAP1's functions, we generated mice lacking THAP1 in the nervous system. THAP1 loss causes locomotor deficits associated with transcriptional changes. Since many of the genes misregulated involve dopaminergic signaling, we pharmacologically challenged the two striatal canonical dopamine pathways: the direct, regulated by the D1 receptor, and the indirect, regulated by the D2 receptor. We discovered that depleting THAP1 specifically interferes with the D2 receptor r…

Pharmacological analysis of zebrafish lphn3.1 morphant larvae suggests that saturated dopaminergic signaling could underlie the ADHD-like locomotor hyperactivity.

Polymorphisms in the gene coding for the adhesion G-protein coupled receptor LPHN3 are a risk factor for attention-deficit/hyperactivity disorder (ADHD). Transient down-regulation of latrophilin3.1 (lphn3.1), the zebrafish LPHN3 homologue, causes hyperactivity. Zebrafish injected with a lphn3.1-specific morpholino are hyperactive and display an impairment in dopaminergic neuron development. In the present study we used lphn3.1 morphants to further characterize the changes to dopaminergic signaling that trigger hyperactivity. We applied dopamine agonists (Apomorphine, Quinpirole, SKF-38393) and…

Effect of chronic treatment with (+)-PHNO, a D2 agonist in MPTP-treated monkeys.

A group of four drug naive Macaca fascicularis were rendered parkinsonian with the neurotoxin 1-methyl-4-phenyl-1,2,3,6- tetrahydropyridine and then treated chronically with (+)-PHNO, a potent D2 agonist. After several days, dyskinesia appeared in all animals. At this point, the daily dose of (+)-PHNO was replaced by a dose of the D1 agonist CY 208-243. The substitution by CY 208-243 reproduced the same dyskinesia observed with (+)-PHNO. The administration of the DA synthesis inhibitor AMPT (alpha-methyl-p-tyrosine methyl ester) blocked the dyskinetic and antiparkinsonian effect of (+)-PHNO, a

Long-term management of prolactinomas.

Prolactinomas are a frequent cause of gonadal dysfunction and infertility, especially in young women. The regulation of prolactin secretion and the efficacy of dopamine agonists in the therapy of prolactinomas are well established. The current challenges in management of prolactinomas are related to follow-up after successful therapy. Issues and questions to be addressed in this approach to long-term management of prolactinomas include the frequency of radiographic monitoring, effect of pregnancy and menopause, safety of estrogen in women taking oral contraceptives, and the potential for disco…

Effects of an AMPA receptor modulator on methamphetamine-induced hyperactivity in rats.

The present study tested if a positive modulator of AMPA-type glutamate receptors would counteract the behavioral effects of a drug that enhances the release of dopamine. BDP-29, a compound shown to increase AMPA receptor-mediated synaptic responses in hippocampal slices, markedly attenuated the amount of stereotypic rearings seen in rats after methamphetamine injections. These results suggest that AMPA receptor modulators ameliorate certain aberrant, dopamine-related behaviors and hence may be of interest with regard to schizophrenia.

Neuroleptic serum levels measured by radioreceptor assay and clinical response in schizophrenic patients.

Serum neuroleptic levels and clinical response have been compared serially in 10 schizophrenic patients treated with a variety of neuroleptics using a novel radioreceptor assay for neuroleptics. In this assay the drug in serum completes with [3H]spiroperidol for binding to dopamine receptors on membranes of the caudate nucleus. Serum neuroleptic levels, expressed in terms of dopamine receptor occupancy, were similar for most neuroleptics at therapeutic doses. Thioridazine levels were substantially higher than those of other neuroleptics.

Galactorrhea-amenorrhea: psychological interaction with neuroendocrine function.

Further substantiation of the interaction between psychological disease and the onset of galactorrhea-amenorrhea is presented in women who underwent intensive psychometric and neuroendocrine evaluation prior to the use of the dopamine against 2-Br-alpha-ergocryptine (CB-154). Normalization of a previously distorted psychometric test profile in each woman was accompanied by a profound decline in prolactin and an unexpected alteration in the peripheral concentrations of norepinephrine and dopamine-beta-hydroxylase. Cesation of galactorrhea and resumption of menses were observed in each instance.

Identification of DARPP-32 as a novel sleep regulator in physiological conditions and experimental Parkinsonism.

Sleep disorders are common in Parkinson's disease (PD) and respond poorly to current pharmacological treatments, partly due to limited understanding of their underlying mechanisms. Using polysomnographic recordings, we investigated the role of dopamine- and cAMP-regulated phosphoprotein 32 kDa (DARPP-32) in sleep-wake regulation and PD-related sleep dysfunction. In naive mice, the selective ablation of DARPP-32 in striatal projection neurons (SPN) co-expressing dopamine D2 and adenosine A2A receptors reduced NREM sleep during the active phase of the circadian cycle, whereas its deletion in dop…

A review of the neuropharmacological profile of schedule-induced polydipsia.

Schedule-induced polydipsia (SIP) arises when subjects consume excessive amounts of water during interval responding on operant tasks, a paradigm introduced and mostly developed in rats. Neuropharmacological studies conducted on SIP have mainly shown a prominent role of dopamine and 5HT in this adjunctive behavior. In particular, D1/D2 receptor antagonists decrease SIP, whereas low doses of dopamine reuptake blockers increase SIP. There is more specific evidence of a decline in SIP after lesions of the mesolimbic dopamine pathway. A decline in SIP is likewise observed after the injection of 5H…

Extinction-restraint intervention eradicates fear memory encoded in BLA engram cells via D2 receptor-mediated dopaminergic modulation from locus coeruleus.

Despite undergoing exposure therapy, individuals with post-traumatic stress disorder (PTSD) frequently experience relapses when confronted w … g the tyrosine hydroxylase (TH)-positive locus coeruleus neurons (LCTH) and BLAengram revealed that dopamine release, but not norepinephrine, is crucial for the deactivation of BLAengram following Ext-Res. Bridging migraine and psychosis: a neuropsychiatric review of shared dopaminergic mechanisms.

Migraine is increasingly recognized as a disorder with both neurological and psychiatric dimensions, with increasing evidence suggesting a potential link to psychotic symptoms through shared dopaminergic dysfunction. Both conditions exhibit dopamine receptor hypersensitivity, neurotransmitter imbalances, and altered cortical excitability, raising the question of whether migraineurs, particularly those with aura, may be more susceptible to transient psychotic-like symptoms. This review explores the neurobiological overlap between migraine and psychosis, focusing on shared pathophysiological mec…

rGO/ZIF-8 Aerogel for Effective Removal of Malachite Green from Wastewater.

In this study, dopamine-modified graphene aerogel (DGA) is synthesized through a one-step hydrothermal method using graphene oxide as the precursor and dopamine as the reducing agent. Subsequently, in situ immersion synthesis is conducted to obtain ZIF-8 loaded on a dopamine-modified graphene aerogel skeleton (ZDGA), featuring a regular honeycomb interconnected mesoporosity and a high specific surface area of 532.8 m2/g. The synthesized ZDGA exhibited exceptional adsorption performance for the cationic dye malachite green. At room temperature, ZDGA achieved an impressive equilibrium adsorption…

Antidepressant-Like Effect of β-Lactolin, a Glycine-Threonine-Tryptophan-Tyrosine Peptide.

The number of patients with mental illnesses, including depression, is rapidly increasing, and daily lifestyle is closely associated with the development of symptoms. Consequently, corrective measures, such as diet-based treatment for diseases, are receiving great attention. We previously showed that β-lactolin, a β-lactopeptide of glycine-threonine-tryptophan-tyrosine peptide, inhibits monoamine oxidase and improves memory impairment in mice, but the effects on depression have not been investigated. Here we showed that β-lactolin improved depression-like behavior via dopamine-D1-like receptor…

The new stimulant designer compound pentedrone exhibits rewarding properties and affects dopaminergic activity.

Cathinone derivatives are new recreational drugs known to produce psychostimulant effects. However, unlike other psychostimulants, the addictive potential of cathinone derivatives has not been widely studied. Here, we investigated the effects of pentedrone, a type of cathinone derivative, on the dopaminergic system using reverse transcription polymerase chain reaction and Western blot. We also evaluated the addictive potential of pentedrone using conditioned place preference and self-administration. We found that pentedrone increased the mRNA expression of dopamine 1 receptor, dopamine 2 recep…

Aripiprazole: a new risk factor for pathological gambling? A report of 8 case reports.

OBJECTIVE: It is commonly accepted that pathological gambling results from the interaction of multiple risk factors. Among these, dopamine replacement therapy (DRT) prescribed for Parkinson disease can be cited. Another dopamine agonist, aripiprazole, could be a new risk factor. We decided to explore this potential adverse drug reaction (ADR). METHOD: Based on a cohort of 166 pathological gamblers starting treatment in our department, data of each of the 8 patients treated by aripiprazole at inclusion were analyzed. RESULTS: The patients involved were schizophrenic or bipolar, mostly young m…

Association of DRD2 variants and Gilles de la Tourette syndrome in a family-based sample from a South American population isolate.

Gilles de la Tourette Syndrome (GTS) is a chronic neuropsychiatric disorder characterized by motor and vocal tics. Epidemiological evidence supports the importance of genetic factors in disease susceptibility, whereas pharmacological and neuroimaging studies have suggested a defect in the dopamine system. The dopamine receptor D2 gene (DRD2) has been reported to be associated with GTS and related phenotypes. Here, we evaluate genetic association between DRD2 and GTS in a sample from a South American population isolate (Antioquia, Colombia). We genotyped nine single nucleotide polymorphisms (SN…
