[Diagnosis of a "hereditary pancreatitis" by the detection of a mutation in the cationic trypsinogen gene].

HISTORY AND CLINICAL FINDINGS: A 71-year-old woman was admitted with the suspected diagnosis of pancreatic carcinoma. As a child she had had repeated attacks of abdominal pain of undetermined cause. When aged 48 years she had developed diabetes mellitus. Her now 42-year-old daughter had from the age of 9 years suffered from repeated attacks of acute pancreatitis that had finally led to chronic pancreatitis. The patient's 15-year-old grandchild was having recurrent bouts of abdominal pain. INVESTIGATIONS: Imaging procedures revealed calcifications in the pancreas and an infiltrating space-occu

Frequent deletion of chromosome 12p12.3 in children with acute lymphoblastic leukaemia.

Cytogenetic deletions of the short arm of chromosome 12 are common recurring alterations found in a wide range of haematological neoplasias, including childhood acute lymphoblastic leukaemia (ALL), the most frequent paediatric malignancy. Such a loss of genetic material suggests the presence of a tumour suppressor gene which plays an important role in growth regulation or in the differentiation of haemopoietic stem cells. To substantiate this hypothesis and to determine more precisely the chromosomal location of this putative gene, we applied a deletion mapping strategy based on the detection

Genetic factors predisposing to B-CLL and to autoimmune disease in spontaneous murine model.

The frequent occurrence of autoimmune diseases in patients with B cell chronic lymphocytic leukemia (B-CLL) or in their family members suggests the involvement of related predisposing genetic factors in the two distinct disorders. Because the majority of B-CLL is of CD5 B cell type, and because the majority of CD5 B cells produces polyreactive autoantibodies, certain regulatory abnormalities in the proliferation and differentiation of CD5 B cells appear to be involved in B-CLL and autoimmune disease, respectively. In studies using MHC(H-2)-congenic New Zealand mouse strains, NZB(H-2d), NZW(H-2

Accurate inference of relationships in sib-pair linkage studies.

Relative-pair designs are routinely employed in linkage studies of complex genetic diseases and quantitative traits. Valid application of these methods requires correct specification of the relationships of the pairs. For example, within a sibship, presumed full sibs actually might be MZ twins, half sibs, or unrelated. Misclassification of half-sib pairs or unrelated individuals as full sibs can result in reduced power to detect linkage. When other family members, such as parents or additional siblings, are available, incorrectly specified relationships usually will be detected through apparen

Coexistence of sudden cardiac death and end-stage heart failure in familial hypertrophic cardiomyopathy.

OBJECTIVES: The purpose of this study was to determine the occurrence of sudden cardiac death or end-stage heart failure, two phases of the natural history of hypertrophic cardiomyopathy, in closely related relatives. BACKGROUND: Hypertrophic cardiomyopathy is a genetically transmitted cardiac disease with a particularly diverse clinical and morphologic spectrum. Premature death usually occurs either suddenly or as a result of progressive congestive heart failure. METHODS: We describe seven families with genetically transmitted hypertrophic cardiomyopathy that were studied with echocardiogra

Myoclonic-astatic epilepsy.

Myoclonic-astatic epilepsy (MAE) belongs to the group of epilepsies with primarily generalized seizures as absence epilepsies, and juvenile myoclonic epilepsy, as well as infantile and juvenile idiopathic epilepsy with generalized tonic-clonic seizures. Like these types of epilepsy, MAE is polygenically determined with little non-genetic variability. The disease is characterized by the following criteria: genetic predisposition (high incidence of seizures and/or genetic EEG patterns in relatives); mostly normal development and no neurological deficits before onset; primarily generalized myoclo

Fetal pyelectasis and Down syndrome: is genetic amniocentesis warranted?

An association between pyelectasis and Down syndrome has recently been reported. The purpose of this investigation was twofold: 1) to test the hypothesis that pyelectasis is more common in fetuses with Down syndrome, and 2) to determine whether genetic amniocentesis should be offered when dilated renal pelves are identified during fetal ultrasound examination. The study population of 5944 fetuses was separated into two groups by infant outcome: 1) phenotypically and/or chromosomally normal, and 2) chromosomally abnormal. Each group was examined for the presence of pyelectasis, defined as an an

Loss of the 17p chromosomal region in a metastatic carcinoma of the prostate.

Genetic alterations of multiple loci that serve as markers for the induction and progression of disease have been identified in several adenocarcinomas, but not in adenocarcinoma of the prostate. To determine if similar genetic alterations occur in prostate carcinoma and could serve as markers for the extent of clinical disease, we have examined 23 predominantly moderately-differentiated, localized prostate carcinomas and one prostatic dysplasia for changes in the structure and copy number of ten selected genes. These genes include 1) those important to androgen metabolism in the prostate, the

Identification of a missense mutation in one allele of a patient with Pompe disease, and use of endonuclease digestion of PCR-amplified RNA to demonstrate lack of mRNA expression from the second allele.

Infantile-onset glycogen storage disease type II, or Pompe disease, results from a genetic deficiency of the lysosomal enzyme acid alpha glucosidase (GAA). Sequencing of the cDNA from a cell line (GM 244) derived from a patient with Pompe disease demonstrated a T953-to-C transition that predicted a methionine-to-threonine substitution at codon 318. The basepair substitution resulted in loss of restriction-endonuclease sites for NcoI and StyI. Analysis of genomic DNA revealed both a normal and an abnormal NcoI fragment, indicating that the patient was a genetic compound. NcoI and StyI digestion

Genetic mapping in phage group 2 Staphylococcus aureus.

Two regimens for transformation have been devised for the phage group 2 strains UT0002-19 and UT0017 of Staphylococcus aureus. Strain UT0002-19 produces exfoliative toxin, which is responsible for the clinical manifestations of the staphylococcal scalded skin syndrome, whereas strain UT0017 does not produce exfoliative toxin. A large pool of antibiotic-resistant and auxotrophic mutants from strains UT0002-19 and UT0017 were isolated by using (i) antibiotic gradient plates, (ii) trimethoprim selection, and (iii) nitrosoguanidine mutagenesis, which sometimes was coupled by enrichment with penici

High-throughput sequencing analyses of XX genital ridges lacking FOXL2 reveal DMRT1 up-regulation before SOX9 expression during the sex-reversal process in goats.

FOXL2 loss of function in goats leads to the early transdifferentiation of ovaries into testes, then to the full sex reversal of XX homozygous mutants. By contrast, Foxl2 loss of function in mice induces an arrest of follicle formation after birth, followed by complete female sterility. In order to understand the molecular role of FOXL2 during ovarian differentiation in the goat species, putative FOXL2 target genes were determined at the earliest stage of gonadal sex-specific differentiation by comparing the mRNA profiles of XX gonads expressing the FOXL2 protein or not. Of these 163 deregulat

Orthostatic hypotension in genetically related hypertensive and normotensive individuals.

OBJECTIVES: Prevalence and determinants of orthostatic hypotension remain largely unexplored in younger individuals without significant burden of chronic diseases. METHODS: We investigated frequency and main associations of impaired orthostatic response in a cohort of 469 middle-aged hypertensive patients and 453 of their normotensive first-degree relatives. RESULTS: 13.4% of hypertensive and 5.5% of normotensive study participants were found to have orthostatic hypotension. In a backward logistic regression the following determinants of orthostatic hypotension were identified: sex [female,

Genetic characterisation of adenovirus type 8 isolated in Hiroshima city over a 15 year period.

AIMS: To investigate the genetic differences among the strains of adenovirus type 8 (Ad8) circulating in Hiroshima city, Japan, and to study their circulation pattern. METHODS: One hundred and twenty nine strains of adenovirus type 8 (Ad8) were isolated in Hiroshima City over a 15 year period (1983-97) from patients with keratoconjunctivitis, and analysed with six restriction enzymes-BamHI, HindIII, PstI, SacI, SalI, and SmaI-to investigate possible relations among the isolates and their genetic variability. Seven hypervariable regions of the hexon gene that carry the type specific epitope we

Male reproductive genes influencing mating and fertility: enhancing the sterile insect technique and genetic control of Diptera pests.

Diptera include major agricultural pests and disease vectors, underscoring the need for species-specific population-control strategies. The sterile insect technique (SIT) relies on sterile males that remain sufficiently viable and competitive to mate successfully with wild females without producing viable offspring. This review synthesizes male reproductive genes and regulatory pathways in Diptera according to the reproductive processes they control and the failure modes caused by their disruption. Mechanistic understanding is based largely on studies of Drosophila melanogaster, complemented b

A chromosome 17 locus is involved in dorsal pigmentation in hooded Long-Evans Agouti rats.

The hooded coat pattern in laboratory rats is produced by recessive alleles of the h (Kit) locus on chromosome 14. A quantitative trait locus (QTL) on chromosome 17 has been previously identified as a potential modifier locus of dorsal pigmentation in hooded Long-Evans Agouti (LEA) rats. To validate the contribution of this chromosome 17 locus, we analyzed an (LEA × F344) F2 population and identified distinct genetic determinants of dorsal and ventral pigmentation. Genome-wide QTL mapping identified a major locus influencing dorsal pigmentation on chromosome 17 near D17Rat2 exceeding a highly

Constructed wetland treatment increases prevalence of multidrug-resistant Escherichia coli isolates but decreases overall concentrations.

AIMS: Antibiotic resistant bacteria (ARB) are high-risk pathogens of interest in the environment. Treatment wetlands are effective in reducing microbial contamination, but little is known of their ability to remove ARB such as methicillin resistant Staphylococcus aureus (MRSA), vancomycin resistant enterococci (VRE), and low-level ceftriaxone resistant Escherichia coli (LLCR). METHODS AND RESULTS: We measured putative MRSA, VRE, and LLCR E. coli from water column and sediments in five distinct sites within the Banklick Creek Wetland (BCW). Using a two-tier approach, we validated antibiotic re

The CsANT1-CsCUC2/CsCYCD3;1 regulatory module coordinates leaf margin development and cell proliferation in cucumber.

Leaf shape is a critical agronomic trait that determines plant architecture and cultivation value, but its genetic and molecular mechanisms remain elusive. Here, we found two natural cucumber mutants, jtll and 10yjy, characterized by little, deeply lobed leaves with reduced cell number and auxin levels. Despite originating from distinct genetic backgrounds, both mutants were fine-mapped to the same locus encoding the AP2-like transcription factor AINTEGUMENTA 1 (CsANT1), thereby revealing a novel role of ANT in leaf lobe formation. Furthermore, CsANT1 modulates auxin distribution along leaf ma

Heat stress in cereal crops: reproductive development and grain filling.

Increasingly frequent extreme heat events threaten cereal production and food security under a changing climate. The reproductive-to-grain formation continuum of cereals is particularly vulnerable to elevated temperatures, as heat stress disrupts developmental processes from inflorescence formation and fertilization to grain filling and quality establishment. These disruptions reduce reproductive success, impair yield formation, and compromise grain quality. A comprehensive understanding of the developmental, physiological, molecular, and genetic basis of cereal heat tolerance is therefore ess

Interactions between Mechanisms of Reproductive Isolation.

AbstractSpeciation is responsible for the diversity of species observed today and corresponds to the buildup of reproductive isolation between populations. Reproductive isolation can be generated by different mechanisms that have been extensively characterized, yet how their interactions affect speciation remains largely unknown. Here, we explicitly model the interaction of three key mechanisms (local adaptation, mate choice, and genetic hybrid incompatibilities), quantifying their relative contribution to the evolution of reproductive isolation. We modeled two populations exchanging migrants
