Molecular and Genetic Landscape of Intravenous Leiomyomatosis: A Narrative Review.

Intravenous leiomyomatosis (IVL) is a rare benign smooth muscle tumor originating from the uterus and characterized by intravascular growth along the venous system, with potential extension to the inferior vena cava, right heart, or pulmonary arteries, leading to life-threatening cardiorespiratory complications. IVL carries a 10%-31% recurrence rate and exhibits quasimalignant biological behavior despite its benign histology. Surgical resection is the main treatment. Current studies on IVL remain limited with regard to molecular and genetic mechanisms. This review systematically summarizes the

Early Onset Heart Failure due to RBM20 Variant: A Case Report Emphasizing Genetic Diagnosis and Arrhythmic Risk Stratification.

The RBM20 gene, located on chromosome 10q25.2, encodes a serine/arginine-rich protein essential for post-transcriptional splicing of several cardiac genes, including titin. Pathogenic variants in RBM20 are increasingly recognized as causes of familial dilated cardiomyopathy (DCM) with a high risk of heart failure and sudden cardiac death. We describe a 23-year-old man who presented with heart failure with mildly reduced ejection fraction secondary to DCM caused by a heterozygous missense variant in the RBM20 gene (c.1907G>A; p.Arg636His). Comprehensive clinical evaluation excluded non-genetic

Laboratory-Engineered Glioblastoma Organoid Culture and Drug Screening.

Glioblastoma (GBM) is described as a group of highly malignant primary brain tumors and stands as one of the most lethal malignancies. The genetic and cellular characteristics of GBM have been a focal point of ongoing research, revealing that it is a group of heterogeneous diseases with variations in RNA expression, DNA methylation, or cellular composition. Despite the wealth of molecular data available, the lack of transferable pre-clinic models has limited the application of this information to disease classification rather than treatment stratification. Transferring the patients' genetic in

Unraveling the role of mobile genetic elements in antibiotic resistance transmission and defense strategies in bacteria.

Irrational antibiotic use contributes to the development of antibiotic resistance in bacteria, which is a major cause of healthcare-associated infections globally. Molecular research has shown that multiple resistance frequently develops from the uptake of pre-existing resistance genes, which are subsequently intensified under selective pressures. Resistant genes spread and are acquired through mobile genetic elements which are essential for facilitating horizontal gene transfer. MGEs have been identified as carriers of genetic material and are a significant player in evolutionary processes. T

Simultaneous Presentation of Swimming-Induced Pulmonary Edema in a Set of Monozygotic Twin Elite Maritime Warfare Candidates: A Novel Case Report.

Swimming-induced pulmonary edema (SIPE) is an incompletely understood condition that is often seen in U.S. special operations candidates participating in maritime qualification training courses. We present a case of two monozygotic twins with the simultaneous onset of acute respiratory distress during a crucible event of a maritime assessment and selection course. Subsequent pulmonary ultrasonography in both candidates showed wedge-shaped hyperechoic lines (B-lines) extending from the pleural interface into the interstitium. Chest radiography of both candidates revealed bilateral asymmetric ha

Senior-Loken Syndrome: Ocular Perspectives on Genetics, Pathogenesis, and Management.

Senior-Loken syndrome (SLSN) is a group of rare autosomal recessive disorders caused by dysfunction of the primary cilium, primarily affecting the kidneys (typically leading to nephronophthisis) and eyes (typically leading to retinal degeneration). Moreover, patients with SLSN may experience additional multisystemic symptoms, such as developmental delay, intellectual disability, ataxia, and nystagmus. To date, eight genes have been demonstrated to cause SLSN, all encoding for proteins involved in the structure and functions of the primary cilium. This places SLSN within an expanding category o

Intrinsically Disordered Regions of Zinc Finger Protein 687 in Phase Separation: A Potentially Pathogenic Driver of Paget's Disease of Bone.

Paget's disease of bone (PDB) is a chronic skeletal disorder characterized by abnormal bone remodeling and structural deformities, yet its underlying molecular mechanisms remain poorly understood. Genetic mutations in regulatory genes have been implicated in the PDB, potentially disrupting osteoclast activity and leading to excessive bone resorption and abnormal bone formation. Current research has largely focused on clinical phenotypes and downstream pathways, relying on molecular biology approaches and omics analyses to elucidate mechanisms. However, the impact of genetic mutations on the bi

Case Report: A novel heterozygous variant of the TCOF1 gene in Treacher Collins syndrome.

Treacher Collins syndrome (TCS) is a craniofacial malformation caused by the abnormal development of the first and second pharyngeal arches during embryogenesis. While pathogenic variants in POLR1B, POLR1C, and POLR1D are implicated, the TCOF1 gene represents the primary causative locus. This case report describes a novel heterozygous frameshift variant, TCOF1:NM_001135243.2:c.3559delG (p.Ala1187GlufsTer21), identified in a child with TCS. This truncation disrupts the C-terminal nuclear localization signal (NLS), specifically ablating a phosphorylation site at residues 1,199-1,200. Consequentl

A Multi-Omics Perspective on Tritrichomonas foetus: From Genomics to Future Directions.

Tritrichomonas foetus is an anaerobic flagellated protozoan that infects multiple animal hosts, primarily cattle and cats, with occasional isolation from pigs. It causes bovine trichomonosis, a venereal disease associated with infertility, abortion, and economic losses in cattle herd. In cats, T. foetus infects the gastrointestinal tract, causing chronic diarrhea which can be difficult to treat. Despite its broad impact, the pathogen is difficult to control because it evades immune responses and persists in host tissues. Recent advances in omics technologies, including genomics, transcriptomic

The Human Monkeypox Virus and Host Immunity: Emerging Diagnostic and Therapeutic Challenges.

This article explores the Human Monkeypox Virus (MPV), a contagious virus that causes disease in both vertebrates and insects. It originated in Denmark in 1958 and expanded beyond Africa during the 1970s. The virus was initially detected in the United States in 2003 following the hospitalisation of a toddler who had been bitten by a prairie dog. The article examines the identification of the virus, its categorization into two genetic groups with different levels of harmfulness, and its genetic changes over time due to specific influences. Additionally, it investigates the immunological reactio
