Behaviourally modulated hippocampal theta oscillations in the ferret persist during both locomotion and immobility.

Theta oscillations are a hallmark of hippocampal activity across mammals and play a critical role in many hippocampal models of memory and spatial navigation. To reconcile the cross-species differences observed in the presence and properties of theta, we recorded hippocampal local field potentials in rats and ferrets during auditory and visual localisation tasks designed to vary locomotion and sensory attention. Here, we show that theta oscillations occur during locomotion in both ferrets and rats, however during periods of immobility, theta oscillations persist in the ferret, contrasting star

Deficiency of protocadherin 9 leads to reduction in positive emotional behaviour.

Protocadherin 9 (Pcdh9) is a member of the cadherin superfamily and is uniquely expressed in the vestibular and limbic systems; however, its physiological role remains unclear. Here, we studied the expression of Pcdh9 in the limbic system and phenotypes of Pcdh9-knock-out mice (Pcdh9 KO mice). Pcdh9 mRNA was expressed in the fear extinction neurons that express protein phosphatase 1 regulatory subunit 1 B (Ppp1r1b) in the posterior part of the basolateral amygdala (pBLA), as well as in the Cornu Ammonis (CA) and Dentate Gyrus (DG) neurons of the hippocampus. We show that the Pcdh9 protein was …

Activity disruption causes degeneration of entorhinal neurons in a mouse model of Alzheimer's circuit dysfunction.

Neurodegenerative diseases are characterized by selective vulnerability of distinct cell populations; however, the cause for this specificity remains elusive. Here, we show that entorhinal cortex layer 2 (EC2) neurons are unusually vulnerable to prolonged neuronal inactivity compared with neighboring regions of the temporal lobe, and that reelin + stellate cells connecting EC with the hippocampus are preferentially susceptible within the EC2 population. We demonstrate that neuronal death after silencing can be elicited through multiple independent means of activity inhibition, and that prevent…

White Fiber Correlates of Amygdalohippocampectomy Through the Middle Temporal Gyrus Approach.

OBJECTIVE: The white fiber and gross anatomy relevant for performing amygdalohippocampectomy through the middle temporal gyrus approach for mesial temporal sclerosis has been depicted by white fiber dissection. METHODS: Three previously frozen and formalin fixed cerebral hemispheres were studied. The Klingler method of fiber dissection was used to study the anatomy. The primary tools used were hand-made wooden spatulas, forceps, and microscissors. The anatomy of the amygdala and hippocampus and the landmarks for performing the disconnection during epilepsy surgery are presented. The white fib…

A Baldwin interpretation of adult hippocampal neurogenesis: from functional relevance to physiopathology.

Hippocampal adult neurogenesis has been associated to many cognitive, emotional, and behavioral functions and dysfunctions, and its status as a selected effect or an "appendix of the brain" has been debated. In this review, we propose to understand hippocampal neurogenesis as the process underlying the "Baldwin effect", a particular situation in evolution where fitness does not rely on the natural selection of genetic traits, but on "ontogenetic adaptation" to a changing environment. This supports the view that a strong distinction between developmental and adult hippocampal neurogenesis is ma

When Fear Shrinks the Brain: A Computational Model of the Effects of Posttraumatic Stress on Hippocampal Volume.

Post-traumatic stress disorder (PTSD) is a psychiatric disorder often characterized by the unwanted re-experiencing of a traumatic event through nightmares, flashbacks, and/or intrusive memories. This paper presents a neurocomputational model using the ACT-R cognitive architecture that simulates intrusive memory retrieval following a potentially traumatic event (PTE) and predicts hippocampal volume changes observed in PTSD. Memory intrusions were captured in the ACT-R rational analysis framework by weighting the posterior probability of re-encoding traumatic events into memory with an emotiona

Biomarkers of Pathological Dissociation: A Systematic Review.

Pathological dissociation is a severe, debilitating and transdiagnostic psychiatric symptom. This review identifies biomarkers of pathological dissociation in a transdiagnostic manner to recommend the most promising research and treatment pathways in support of the precision medicine framework. A total of 205 unique studies that met inclusion criteria were included. Studies were divided into four biomarker categories, namely neuroimaging, psychobiological, psychophysiological and genetic biomarkers. The dorsomedial and dorsolateral prefrontal cortex, bilateral superior frontal regions, (anteri

Expression of a heroin contextually conditioned immune effect in male rats requires CaMKIIα-expressing neurons in dorsal, but not ventral, subiculum and hippocampal CA1.

The physiological and motivational effects of heroin and other abused drugs become associated with environmental (contextual) stimuli during … plenic nitric oxide (NO) production in male rats, and this effect is mediated in part by the dorsal hippocampus (dHpc). However, specific dHpc output regions, whose efferents might mediate conditioned immune effects, have not been identified, nor has the contribution of ventral hippocampus (vHpc) been investigated. Here, we evaluated the role of CaMKIIα-expre …

Impact of Neuroinflammation on Hippocampal Neurogenesis: Relevance to Aging and Alzheimer's Disease.

The cognitive reserve is associated with the capacity of the brain to maintain cognitive performance in spite of being challenged by stressful degenerative insults related to aging. Hippocampal neurogenesis is a life-long process of continuous addition of functional new neurons in the memory processing circuits. Accordingly, adult hippocampal neurogenesis is increasingly seen as a key determinant of cognitive reserve robustness. On the other side, neuroinflammation, by releasing a plethora of proinflammatory cytokines and other inflammatory molecules, is increasingly shown to be one of the key

Targeting Neural Hyperactivity as a Treatment to Stem Progression of Late-Onset Alzheimer's Disease.

Sporadic late-onset Alzheimer's disease (LOAD), the most common form of dementia in the elderly, causes progressive and severe loss of cogni … ention in elderly individuals in an early prodromal stage of disease. Excess neural activity in the hippocampus, a recently described phenomenon associated with age-dependent memory loss, was first identified in animal models of aging and subsequently translated to clinical conditions of aging and early-stage LOAD. Review: Hippocampal sclerosis in epilepsy: a neuropathology review.

Hippocampal sclerosis (HS) is a common pathology encountered in mesial temporal lobe epilepsy (MTLE) as well as other epilepsy syndromes and in both surgical and post-mortem practice. The 2013 International League Against Epilepsy (ILAE) classification segregates HS into typical (type 1) and atypical (type 2 and 3) groups, based on the histological patterns of subfield neuronal loss and gliosis. In addition, granule cell reorganization and alterations of interneuronal populations, neuropeptide fibre networks and mossy fibre sprouting are distinctive features of HS associated with epilepsies; t

Factors affecting graded and ungraded memory loss following hippocampal lesions.

This review evaluates three current theories--Standard Consolidation (Squire & Wixted, 2011), Overshadowing (Sutherland, Sparks, & Lehmann, 2010), and Multiple Trace-Transformation (Winocur, Moscovitch, & Bontempi, 2010)--in terms of their ability to account for the role of the hippocampus in recent and remote memory in animals. Evidence, based on consistent findings from tests of spatial memory and memory for acquired food preferences, favours the transformation account, but this conclusion is undermined by inconsistent results from studies that measured contextual fear memory, probably the m…

Characterization of the in vitro antiepileptic activity of new and old anticonvulsant drugs.

1. The in vitro antiepileptiform effects of some old and new anticonvulsants in the experimental model of the "epileptiform" hippocampal slice have been reviewed. 2. On the basis of their influence on in vitro epileptogenesis and basal neuronal excitability, anticonvulsants can be classified into three main categories: (1) anticonvulsants (prototypical drug phenytoin) affecting basal neuronal excitability but not epileptogenesis; (2) anticonvulsants (prototypical drugs barbiturates) affecting basal neuronal excitability and epileptogenesis; (3) anticonvulsants (prototypical drug felbamate) aff

Distribution of acetylcholinesterase in the hippocampal region of the mouse. III. The area dentata.

The distribution of acetylcholinesterase (AChE) was examined in the area dentata of the adult mouse (Mus musculus domesticus). A distinctly stratified distribution of the enzyme was observed and was compared in detail with cytoarchitectural fields and layers. In the stratum moleculare, bands of relatively high AChE activity were seen immediately beneath the pia, at the borders between the outer, middle, and deep portions of the stratum moleculare, and superficial to the granule cell layer. AChE activity was low in the intervening parts of the stratum moleculare. In contrast to the rat, three s

Synaptic integration in hippocampal CA1 pyramids.

Excitatory synapses on hippocampal pyramids are exclusively located to dendritic spines, usually in a 1:1 proportion. The number of spines indicates a convergence of as many as 25,000-30,000 excitatory boutons per CA1 pyramidal cell in rats. Activation of a single afferent fibre produces a unitary excitatory postsynaptic potential (EPSP) of about 150 microV, probably produced by a single quantum of transmitter. The release probability is normally low, but may be increased by facilitatory processes. On the average, each afferent fibre has few boutons (mostly 1, but up to 5) in contact with a gi
