Active Heymann nephritis in complement component C6 deficient rats.

The mechanisms of renal injury that result in proteinuria in active Heymann nephritis (AHN) remain unclear, though data suggest that in analogy of the passive form of the disease the membrane attack complex C5b-9 may be involved. AHN was induced in an inbred strain of PVG/c-rats that are totally deficient in the C6 component of complement and are unable to form the lytic C5b-9 complex, as well as in non-complement deficient PVG/c+ rats that are immunologic identical to the deficient strain. In both groups of animals comparably high titers of anti-Fx1A autoantibodies were found after three week

Regeneration of T-cell helper function in zinc-deficient adult mice.

Diets deficient in zinc cause rapid atrophy of the thymus and loss of T-cell helper function in the young adult A/J mouse. Because zinc deficiency, as well as other nutritional deficiences, causes extensive damage to the immune system, the question arose as to whether zinc-deficient mice could repair the thymus and fully regenerate T-cell helper function if returned to diets containing adequate amounts of zinc. Five-week-old A/J female mice were fed either a zinc-deficient (<1 mug of Zn per g) or a zinc-adequate (50 mug of Zn per g) diet for 31 days. Histological examination of thymuses from t

Fibroblastic reticular cells direct the initiation of T cell responses via CD44.

The movement of dendritic cells and T cells within secondary lymphoid organs is critical for the development of adaptive immune responses1,2. Central to this process is the fibroblastic reticular cell (FRC) network, which forms a highly organized conduit system that facilitates the movement of and interactions between dendritic cells and T cells3-6. Previous studies have partly characterized how FRCs support these interactions7,8. However, the molecular mechanisms that operate under physiological conditions remain unknown. Here we show that the viral protein m11, encoded by the herpesvirus mur

Genomic and Immune Landscape of Recurrent and/or Metastatic Squamous Cell Carcinoma of the Head and Neck Progressing on Anti-PD-1 Treatment.

Anti-PD-1 therapies improve survival in recurrent/metastatic (R/M) squamous cell carcinoma of the head and neck (SCCHN), but only a minority of patients achieve durable responses. The mechanisms driving resistance to anti-PD-1 in SCCHN remain poorly understood. Using the IMMUcan multiomics workflow, we characterized the molecular and immune profiles of R/M SCCHN progressing on anti-PD-1 treatment and compared them with an anti-PD-1-naïve cohort. Tumor biopsies from patients with anti-PD-1-resistant SCCHN exhibited significantly more EGFR and MYCL amplifications, along with increased MYC pathwa

A mouse model to study persistent fatigue.

Fatigue is characterized as a feeling of exhaustion or lack of energy, is a common symptom of multiple chronic illnesses and interferes with daily activities and quality of life. There is a limited availability of animal models to examine potential underlying mechanisms ultimately limiting development of potential therapeutic strategies. The primary purpose of this study was to develop a mouse model of persistent fatigue. A secondary purpose was to characterize potential measures of "fatigue-like" behaviors. An exploratory goal was to examine for immune and metabolic changes in the model. We e

Adaptive immune responses are not causal to SGN death after kanamycin-induced hair cell loss.

Aminoglycoside antibiotics such as kanamycin induce sensorineural hearing loss by killing hair cells, resulting in secondary degeneration of spiral ganglion neurons (SGNs). Previous studies show that anti-inflammatory agents reduce SGN death, implicating a causal role of the immune response. This is consistent with observations of increased numbers of macrophages and lymphocytes, including T and NK cells, in the spiral ganglion after exposure to aminoglycosides. Here, we directly test the role of T cells and other lymphocytes in SGN degeneration in kanamycin-deafened rats. Homozygous RNU nude

Pathogenic TET2 Variants and Autoinflammatory Manifestations in Myeloid Hematologic Malignancies: Case Report and Literature Review.

Emerging evidence suggests that somatic mutations in genes associated with innate immunity can trigger adult-onset autoinflammatory diseases. Notably, loss-of-function variants in the TET2 gene have been linked to both hematological malignancies and immune-mediated disorders. A case is presented of a 73-year-old woman with chronic myelomonocytic leukemia who developed severe pericardial effusion secondary to inflammatory serositis, associated with a pathogenic TET2 variant. Despite initial treatment with corticosteroids and diuretics, her condition worsened, which led to the need to implement

Effects of Metabolic Dysregulation on Macrophage Polarization in Rheumatoid Arthritis.

Rheumatoid arthritis (RA) is a systemic autoimmune disease driven by complex immune dysregulation in which macrophages play a central pathogenic role. Macrophages accumulate extensively in RA synovium, and their abundance and functional phenotypes are correlated with disease activity and severity. Disturbed macrophage polarization is a key feature of RA progression, characterized by an increased proportion of classically activated (M1) macrophages and a reduced proportion of alternatively activated (M2) macrophages. M1 macrophages produce proinflammatory mediators that sustain synovial inflamm

[PEMBROLIZUMAB-ASSOCIATED MYOCARDITIS FOLLOWED BY STEROID MYOPATHY IN A PATIENT WITH RENAL CELL CARCINOMA].

Immune checkpoint inhibitors (ICIs) are increasingly used as adjuvant therapy in high-risk renal cell carcinoma (RCC), but may cause serious immune-related adverse events, such as myocarditis. We report a rare case of myocarditis followed by steroid-induced myopathy during adjuvant pembrolizumab therapy. A 74-year-old man underwent partial and radical nephrectomy for bilateral clear cell RCC (pT1b and pT3a). Pembrolizumab was initiated postoperatively. On day 22, he developed exertional fatigue and cardiac enzyme levels were elevated. Myocardial biopsy confirmed myocarditis, and corticosteroid

Oncogenic EGFR rewires STING-TBK1 signalosomes to license DNA damage tolerance in NSCLC.

EGFR hotspot mutations (mEGFR), including primary L858R, exon 19 deletion, and secondary T790M, are pivotal oncogenic drivers in human non-small cell lung cancer (NSCLC). At the same time, NSCLC resistance to third-generation tyrosine kinase inhibitors (TKIs) is a major clinical challenge and remains mechanistically unresolved. Here, we uncover a previously unrecognized tumor cell-intrinsic mechanism in which mutant EGFR (mEGFR) exploits innate immune signaling via the cGAS-STING-TBK1 pathway to sustain oncogenic signaling and therapeutic resistance. Mechanistically, mutant EGFR kinase aberran

Loss of DHX36/G4R1, a G4 resolvase, drives genome instability and regulates innate immune gene expression in cancer cells.

G-quadruplexes (G4s) are four-stranded alternative secondary structures formed by guanine-rich nucleic acids and are prevalent across the human genome. G4s are enzymatically resolved by specialized helicases. Previous in vitro studies showed that DEAH-box helicase 36 (DHX36/G4R1/RHAU) has the highest specificity and affinity for G4 structures. Here, by mapping genome-wide DNA double-strand breaks (DSBs), we demonstrate that knockout of DHX36 helicase increases DSB enrichment at G4 sites and that the presence of the G4 motif is a significant mediator of genome instability at regulatory regions.

Tertiary Lymphoid Structures in Human Melanoma: Molecular Mechanisms and Therapeutic Opportunities.

Tertiary lymphoid structures (TLSs) are ectopic lymphoid aggregates often found in chronic inflammatory conditions, including cancer. These structures, which share many cellular and functional features with secondary lymphoid organs, can profoundly influence the tumor microenvironment by promoting local anti-tumor immune activation. TLSs have been observed in various cancers, including melanoma, and are associated with improved responses to immunotherapy and clinical outcomes. However, our understanding of the molecular mechanisms underlying TLS formation and function remains incomplete. This

The Effect of Plant Quercetin on Immune Function of Microtus Fortis.

Quercetin, a widely occurring phytogenic flavonoid, is the primary secondary metabolite in the diet of Microtus fortis. This study investigated the effects of dietary quercetin on immune organ development and cytokine levels in Microtus fortis. Microtus fortis were fed a standard diet containing 0, 1.5%, 3%, 5%, or 8% quercetin for 20 consecutive days. The results show that, compared to the control group, treatment with 5% quercetin increased the spleen and thymus indices by 14.47% and 11.67%, respectively. The proliferation of T lymphocytes, B lymphocytes and macrophages in Microtus fortis va

Pembrolizumab-Induced Hypophysitis: A Rare Immune-Related Adverse Event in a Patient With Metastatic Non-small Cell Lung Cancer.

Immune checkpoint inhibitors (ICIs), such as pembrolizumab, have revolutionized cancer treatment but are associated with immune-related adverse events (irAEs). Hypophysitis, though rare, is a serious endocrine complication that can lead to life-threatening adrenal insufficiency if not promptly recognized and treated. A 62-year-old man with metastatic non-small cell lung cancer presented with fatigue, headache, and hyponatremia after four cycles of pembrolizumab. Laboratory evaluation revealed secondary adrenal insufficiency (low cortisol and adrenocorticotropic hormone) and central hypothyroid

Cranial neuropathy during severe acute respiratory syndrome coronavirus 2 infection: a case report.

BACKGROUND: Cranial neuropathy has been reported in association with coronavirus disease 2019. Multiple reports have highlighted the possibility of Miller Fisher syndrome as a delayed immune-mediated response to severe acute respiratory syndrome coronavirus 2 infection. However, cranial neuropathy during the infectious phase of coronavirus disease 2019 with positive cerebrospinal fluid polymerase chain reaction has not been reported to date. CASE PRESENTATION: We report a 53-year-old Iranian woman who presented with right ptosis, diplopia, and headache without any respiratory symptoms of coro

The complement system in neurodegenerative diseases.

Complement is an important component of innate immune defence against pathogens and crucial for efficient immune complex disposal. These core protective activities are dependent in large part on properly regulated complement-mediated inflammation. Dysregulated complement activation, often driven by persistence of activating triggers, is a cause of pathological inflammation in numerous diseases, including neurological diseases. Increasingly, this has become apparent not only in well-recognized neuroinflammatory diseases like multiple sclerosis but also in neurodegenerative and neuropsychiatric

Defective kinase activity of IKKα leads to combined immunodeficiency and disruption of immune tolerance in humans.

IKKα is a multifunctional serine/threonine kinase that controls various biological processes, either dependent on or independent of its kinase activity. However, the importance of the kinase function of IKKα in human physiology remains unknown since no biallelic variants disrupting its kinase activity have been reported. In this study, we present a homozygous germline missense variant in the kinase domain of IKKα, which is present in three children from two Turkish families. This variant, referred to as IKKαG167R, is in the activation segment of the kinase domain and affects the conserved (DF/

SLAMF7 defines subsets of human effector CD8 T cells.

Long-term control of viral replication relies on the efficient differentiation of memory T cells into effector T cells during secondary immune responses. Recent findings have identified T cell precursors for both memory and exhausted T cells, suggesting the existence of progenitor-like effector T cells. These cells can persist without antigenic challenge but expand and acquire effector functions upon recall immune responses. In this study, we demonstrate that the combination of SLAMF7 with either CD27 or TCF-1 effectively identifies progenitor-like effector CD8 T cells, while SLAMF7 with GPR56

Comparison of the blood immune repertoire with clinical features in chronic lymphocytic leukemia patients treated with chemoimmunotherapy or ibrutinib.

Chronic lymphocytic leukemia (CLL) is characterized by the accumulation of CD19+ CD5+ clonal B lymphocytes in the blood, bone marrow, and peripheral lymphoid organs. Treatment options for patients range from historical chemoimmunotherapy (CIT) to small molecule inhibitors targeting pro-survival pathways in leukemic B cells, such as the Bruton's tyrosine kinase inhibitor ibrutinib (IBR). Using biobanked blood samples obtained pre-therapy and at standard response evaluation timepoints, we performed an in-depth evaluation of the blood innate and adaptive immune compartments between pentostatin-ba

Oncolytic T-VEC virotherapy plus neoadjuvant chemotherapy in nonmetastatic triple-negative breast cancer: a phase 2 trial.

Talimogene laherparepvec (T-VEC) is an oncolytic virus hypothesized to enhance triple-negative breast cancer (TNBC) responses to neoadjuvant chemotherapy (NAC). This article describes the phase 2 trial of T-VEC plus NAC (ClinicalTrials.gov ID: NCT02779855 ). Patients with stage 2-3 TNBC received five intratumoral T-VEC injections with paclitaxel followed by doxorubicin and cyclophosphamide and surgery to assess residual cancer burden index (RCB). The primary end point was RCB0 rate. Secondary end points were RCB0-1 rate, recurrence rate, toxicity and immune correlates. Thirty-seven patients we
