Incomplete microwave ablation (iMWA) remains a major challenge in the clinical management of hepatocellular carcinoma (HCC), as residual tumors often foster an immunosuppressive microenvironment and upregulate PD-L1 expression, thereby promoting immune evasion and recurrence. This study systematically elucidates the mechanisms underlying HCC progression and metastasis following iMWA and describes the design of a dual-ion/immune checkpoint nanoplatform (aP@Mn/Ca) based on a metal-organic framework (MOF). Leveraging the excellent biocompatibility and tumor microenvironment-responsive degradabili

Uncontrolled childhood asthma contributes to chronic airway inflammation and remodeling, primarily characterized by bronchial smooth muscle hyperplasia. Although the NLR family pyrin domain-containing 3 (NLRP3) inflammasome regulates airway inflammation, its role in abnormal airway smooth muscle cell (ASMC) proliferation remains poorly understood. Here, we evaluated the selective NLRP3 inhibitor GDC-2394 in a house dust mite (HDM)-induced murine model of asthma with a vehicle control group, focusing on the NLRP3-IL-6/JAK2/STAT3 signaling axis. HDM-challenged mice displayed increased airway hyp

Traumatic brain injury (TBI) is a serious disease characterized by a high incidence in young people and can lead to significant neurological impairment. Deterioration in cognitive function following TBI is often underpinned by a sustained neuroinflammatory state, which impairs the integrity of the blood-brain barrier (BBB) and accelerates neuronal loss. Mesenchymal stem cells (MSCs) have emerged as a promising therapeutic approach for TBI because of their regenerative and immunomodulatory properties. Here, we identified heat shock (HS) pretreatment as an effective method to potentiate the ther

Diabetic kidney disease (DKD) is the main cause of end-stage renal disease (ESRD) worldwide. Podocytes are important components of the glomerular filtration barrier, which is located between glomerular capillary endothelial cells and the basement membrane, and is essential for maintaining normal renal function. Its injury is a key factor in the progression of DKD. Lipid homeostasis plays an important role in maintaining the normal physiological function of cells. In recent years, many studies have revealed that lipid metabolism disorders play a core role in driving podocyte injury, but its spe

This study aimed to investigate the role of STOX1 in trophoblast pyroptosis during preeclampsia (PE) and elucidate its underlying molecular mechanisms. A trophoblast hypoxia cell model was established by exposing HTR-8/SVneo cells to hypoxic conditions. The results showed that STOX1 messenger RNA expression was significantly elevated in placental tissues from PE patients as well as in hypoxia-treated HTR-8/SVneo cells. Hypoxia markedly induced pyroptosis in HTR-8/SVneo cells, whereas STOX1 silencing enhanced cell proliferation and migration while suppressing programmed cell death. Moreover, ST

Vitamin D3 (VitD3) deficiency affects over one billion individuals globally, representing a critical modifiable risk factor for immune-mediated diseases. Beyond its classical role in calcium metabolism, Vitamin D3 orchestrates immune homeostasis through vitamin D receptor (VDR) signaling, exerting profound regulatory effects on both innate and adaptive immunity. Mechanistically, Vitamin D3 maintains the balance between antimicrobial defense and inflammatory suppression by inhibiting key pro-inflammatory pathways including nuclear factor κB (NF-κB) and the NOD-like receptor protein 3 (NLRP3) in

Myocardial injury and adverse remodeling following acute myocardial infarction (MI) drive heart failure progression, in which cardiomyocyte pyroptosis plays a critical pathogenic role. Bone morphogenetic protein 7 (BMP7) exerts anti-fibrotic and anti-inflammatory effects; however, its role in regulating pyroptosis during post-infarction cardiac repair remains unclear. We found that plasma BMP7 levels were markedly reduced in patients with chronic MI, showing a positive association with left ventricular ejection fraction and a negative correlation with myocardial fibrosis quantified by late gad

Osteoarthritis (OA) is a degenerative joint disorder marked by cartilage degradation and synovial inflammation, for which effective treatments remain limited. Bone marrow mesenchymal stem cell (BMSC)-derived exosomes have emerged as promising cell-free therapies due to their immunomodulatory properties. In this study, we investigated whether exosomes from BMSCs overexpressing miR-27a-3p alleviate OA by targeting the TXNIP/NLRP3 axis in both chondrocytes and macrophages. BMSCs were transfected with miR-27a-3p mimics, and exosomes (Exos-miR-27a-3p) were isolated and characterized. Exos-miR-27a-3

Neutrophils, the most abundant of the circulating leucocytes, play crucial roles in antimicrobial defence, tissue remodelling, and immune regulation. Traditionally regarded as predominantly glycolytic, relying on aerobic glycolysis (the Warburg effect) for rapid ATP generation, neutrophils are now recognised to possess broader metabolic flexibility. Emerging evidence has revealed their capacity for oxidative phosphorylation (OXPHOS), fatty acid oxidation (FAO), and glutaminolysis. As a result, mitochondrial metabolism is dynamically reprogrammed during differentiation and activation. These met

As central mediators bridging innate and adaptive immunity, macrophages exhibit functional plasticity that is precisely governed by dynamic epigenetic networks, The histone H3K27 demethylase KDM6B acts as a key epigenetic hub that mediates the epigenetic remodeling of macrophages in response to environmental signals and exerts pleiotropic regulatory roles across diverse pathophysiological contexts. In the tumor microenvironment, KDM6B drives the polarization of tumor-associated macrophages toward an M2-like immunosuppressive phenotype, reshapes cellular metabolism, and consequently promotes tu

Sepsis-associated acute kidney injury (S-AKI) is a frequent and severe complication of sepsis in which metabolic dysfunction and inflammatory activation develop in parallel and reinforce each other. Although mitochondrial dysfunction is widely recognized in S-AKI, the structural mechanisms linking mitochondrial bioenergetic failure to innate immune signaling remain incompletely understood. Emerging evidence suggests that mitochondrial lipid remodeling-particularly alterations in cardiolipin (CL)-may represent a structural interface connecting these processes. As a signature phospholipid of the

Varicocele is a common cause of male infertility, but the mechanisms by which it disrupts testicular homeostasis and impairs spermatogenesis remain incompletely elucidated. This article reviews current evidence on the multifactorial disturbances in the testicular microenvironment induced by varicocele, with a focus on hemodynamic, biochemical, and structural abnormalities. Anatomical predisposition, venous valve incompetence, and impaired venous return collectively lead to chronic venous hypertension, causing progressive dilation of the pampiniform plexus and severe hemodynamic dysregulation.

Liver fibrosis is a critical pathological process in chronic liver disease and is strongly associated with poor prognosis. This study investigated the anti-fibrotic potential of G22037, a novel oxadiazole-pyrazole derivative whose pharmacological properties have not been previously characterized. In the human hepatic stellate cell line LX-2, G22037 significantly suppressed TGFβ1-induced overexpression of COL1A1 and α-SMA, indicating a direct inhibitory effect on hepatic stellate cells (HSCs) activation. The in vivo anti-fibrotic activity of G22037 was validated in two complementary mouse model

Metabolic dysfunction-associated steatotic liver disease (MASLD) is common and clinically heterogeneous, with fibrosis stage serving as a major determinant of long-term liver-related risk. This focused narrative review evaluates ketogenic dietary interventions and intermittent fasting as metabolic-switching strategies while explicitly distinguishing conventional or eucaloric ketogenic diets, very-low-energy ketogenic therapy, non-ketogenic low-carbohydrate diets, and distinct fasting protocols. Both approaches can reduce body weight, improve insulin sensitivity, and decrease hepatic steatosis

Cyclophosphamide (CP) has long been employed in cancer treatment as well as in therapeutic regimens for autoimmune diseases. Despite its clinical value, one of its major drawbacks is testicular damage which may be due to disruption of antioxidant defenses, amplification of inflammatory responses, and suppression of autophagy. Febuxostat is a xanthine oxidase inhibitor with promising antioxidant, anti-inflammatory, and autophagy-inducing effects. The present study investigated whether febuxostat could mitigate CP-induced testicular toxicity in a rat model. Fifty male Sprague-Dawley rats were al

Endometritis-inflammation of the endometrial lining-imposes a substantial reproductive and economic burden in both human gynecology and livestock production, where it is a leading cause of recurrent implantation failure in humans and the costliest reproductive disorder in cattle. Conventional management is overwhelmingly antibiotic-based, yet escalating antimicrobial resistance, tissue and milk residues, microbiota disruption and high relapse rates have eroded its efficacy and acceptability, creating an urgent need for mechanism-based, host-directed alternatives. Here we synthesize the expandi

Post-stroke insomnia (PSI) is a critical biological barrier to neurorehabilitation afflicting over half of all stroke survivors. Traditional sedatives often force clinicians into a therapeutic dilemma between sleep efficacy and cognitive suppression. The microbiota-gut-brain (MGB) axis has recently emerged as a transformative target to resolve this impasse. Acute stroke triggers profound autonomic dysfunction, causing immediate intestinal barrier collapse. This "leaky gut" facilitates the systemic translocation of lipopolysaccharides (LPS) and activates the NLRP3 inflammasome. The resulting in

Chronic alcohol and other psychoactive substance use is accompanied not only by disturbances in classical neurotransmitter systems but also by persistent activation of innate and adaptive immunity, leading to neuroinflammation. This review summarizes experimental and clinical data on how microglia and astrocytes act as central mediators at the intersection of immune, metabolic, and neuronal processes in alcohol-related disorders. We discuss Toll-like receptor 4 (TLR4)-dependent pathways, activation of the NLRP3 inflammasome, impaired glutamate clearance, metabolic "reprogramming" of glia, and
