Transcriptome and metabolome analysis reveals that cuproptosis in bovine cumulus cells triggers the intercellular transmission of senescence and mitochondrial dysfunction to impair oocyte quality.

Cuproptosis, a newly discovered form of copper-driven regulated cell death, has been shown to be closely related to ovarian function. However, whether the oocyte dysfunction, decreased ovulation efficiency, and cumulus cell aging caused by copper overload or imbalance are associated with regulatory pathways related to cuproptosis remains unclear. In this study, the expression profiles of genes related to cuproptosis in cumulus cells were comprehensively analyzed through transcriptome sequencing and metabolome analysis, and key genes and pathways that affect oocyte maturation were identified in

Metabolomic Analysis of Intracellular Amino Acid Profile Differences Between Cisplatin-Resistant and Cisplatin- Sensitive CALU-1 Lung Cancer Cells.

Lung cancer remains one of the leading causes of cancer-related mortality worldwide, and resistance to cisplatin remains a major limitation in the treatment of non-small cell lung cancer (NSCLC). Accumulating evidence indicates that cancer cells reprogram amino acid metabolism to support proliferation, stress adaptation, redox balance, and therapeutic resistance. In this context, defining the amino acid-related metabolic alterations associated with cisplatin resistance may help identify potential metabolic vulnerabilities and biomarker candidates. In this study, intracellular free amino acid p

Design, Synthesis, and Antiproliferative Evaluation of C3/C12-Modified Panaxadiol Derivatives Against Gastric Cancer Cells: Integrated Transcriptomic and Metabolomic Analyses.

Panaxadiol (PD) is a bioactive dammarane triterpenoid with limited antiproliferative potency, and systematic optimization of its C3 and C12 positions remains underexplored. Here, a stepwise, site-differentiated strategy was established by combining selective C3 esterification with late-stage C12 diversification through a chloroacetyl-piperazine linker, affording 23 PD derivatives and enabling complementary C3/C12 structure-activity analysis. Antiproliferative screening in AGS gastric cancer cells identified compounds 6, 11, and 17 as the most active analogues, with IC50 values of 8.41, 5.08, a

Roe deer uterine fluid metabolome reveals elevated glycolysis, fatty acid breakdown, and spermidine synthesis upon reactivation from diapause†.

The blastocyst of the European roe deer (Capreolus capreolus) undergoes a period of decelerated growth and limited metabolism. During this period known as embryonic diapause, it floats freely in the uterus encircled by the histotroph. Prior to implantation, reactivation is marked by rapid embryonic growth and conceptus elongation. We hypothesized that the uterine fluid, which is known to undergo changes in its composition to support early embryonic development, contributes to controlling embryonic growth during diapause and elongation. We therefore characterized the pre-implantation uterine fl

Quercetin-based treatment improves eggshell quality in aged laying hens by modulating immune response, eggshell gland health and serum metabolome.

BACKGROUND: Eggshell quality is a critical determinant of egg product value and consumer preference. Deficient eggshells increase breakage susceptibility, leading to economic losses and reduced marketability. Dietary interventions hold promise for improving eggshell quality. Quercetin, a naturally occurring flavonoid, exhibits diverse biological activities relevant to eggshell formation. These include antioxidant and anti-inflammatory properties, and potential modulation of estrogen levels, which are known to influence eggshell quality. RESULTS: A total of 160 46-week-old laying hens were ran

Sibling species differently distributed around a CO2 vent show transplantation proteomic remodelling, while displaying metabolomic signatures associated with their origin.

The cellular homeostatic response (CHR) and cellular stress response (CSR) work together to maintain homeostasis. Studying phylogenetically closely-related species inhabiting different environments can help investigate the interplay between the CHR and CSR. We conducted reciprocal in situ transplant experiments in a natural CO2 vent (Ischia, Italy), using the sibling annelid species Platynereis cf.. dumerilii and Platynereis cf.. massiliensis which have been shown to have different preferential distributions around the CO2 vent. Following transplantations, we characterised the response of each

Metabolomic datasets of an apple progeny carrying resistance quantitative trait loci to apple scab before or after inoculation of the pathogen Venturia inaequalis.

Phytosanitary treatments are massively used in orchards to fight apple scab, a disease caused by the fungus Venturia inaequalis (Vi). To reduce these treatments, resistant varieties are largely deployed but their effectiveness can decrease over time. The combination of complementary molecular mechanisms within new varieties could enhance the durability of genetic resistance however, the underlying resistance mechanisms remain poorly understood. An apple pseudo-F1 progeny was previously widely investigated for its quantitative trait loci (QTL) controlling resistance to scab and at least three o
