Learning and memory deficits in ecstasy users and their neural correlates during a face-learning task.

It has been consistently shown that ecstasy users display impairments in learning and memory performance. In addition, working memory processing in ecstasy users has been shown to be associated with neural alterations in hippocampal and/or cortical regions as measured by functional magnetic resonance imaging (fMRI). Using functional imaging and a face-learning task, we investigated neural correlates of encoding and recalling face-name associations in 20 recreational drug users whose predominant drug use was ecstasy and 20 controls. To address the potential confounding effects of the cannabis u

Anisotropy in the representation of direction preferences in cat area 18.

Higher visual cortical areas are involved in the perception of complex stimuli, such as the optic flow created by self-motion. On the other hand, area 18 is thought to extract primitive visual features, feeding higher cortical areas for further processing. In this study, we applied optical imaging of intrinsic signals in the central, lower visual field of cat area 18, and reconstructed direction preference and direction selectivity maps in each hemisphere. We observed a significant overrepresentation of downward and temporal directions, in accordance with previous electrophysiological results.

Memory for spatial location and object-place associations are differently processed by the hippocampal formation, parahippocampal areas TH/TF and perirhinal cortex.

To clarify the specific contribution of the medial temporal lobe structures in spatial memory, we tested monkeys (Macaca mulatta) with sham operations and with lesions of either the hippocampal formation, areas TH/TF or perirhinal cortex on two versions of the visual-paired comparison task, measuring Spatial Location, and Object-in-Place associations. In the Spatial Location version, the comparison was between two identical objects presented simultaneously in a familiar and a novel location. In the Object-in-Place version, the comparison was between an image consisting of five objects and anot

Recognition of emotions from visual and prosodic cues in Parkinson's disease.

OBJECTIVE: To assess whether Parkinson Disease (PD) patients are impaired at perceiving emotions from facial and prosodic cues and whether any putative defective performance concerns recognition of a particular emotion. BACKGROUND: Braak et al. [1] demonstrated that in different stages PD pathology involves the nigrostriatal system, the amygdala, and the insular cortex. Discrete brain lesions to these structures can cause selective deficits in recognising facial and prosodic stimuli expressing particular emotions. However, the investigation of facial and prosodic emotional processing in PD pa

fMRI evidence for sensorimotor transformations in human cortex during smooth pursuit eye movements.

Smooth pursuit eye movements (SP) are driven by moving objects. The pursuit system processes the visual input signals and transforms this information into an oculomotor output signal. Despite the object's movement on the retina and the eyes' movement in the head, we are able to locate the object in space implying coordinate transformations from retinal to head and space coordinates. To test for the visual and oculomotor components of SP and the possible transformation sites, we investigated three experimental conditions: (I) fixation of a stationary target with a second target moving across th

Cell-nonautonomous function of ceramidase in photoreceptor homeostasis.

Neutral ceramidase, a key enzyme of sphingolipid metabolism, hydrolyzes ceramide to sphingosine. These sphingolipids are critical structural components of cell membranes and act as second messengers in diverse signal transduction cascades. Here, we have isolated and characterized functional null mutants of Drosophila ceramidase. We show that secreted ceramidase functions in a cell-nonautonomous manner to maintain photoreceptor homeostasis. In the absence of ceramidase, photoreceptors degenerate in a light-dependent manner, are defective in normal endocytic turnover of rhodopsin, and do not res

Discrete parieto-frontal functional connectivity related to grasping.

The human inferior parietal lobule (IPL) is known to have neuronal connections with the frontal lobe, and these connections have been shown to be associated with sensorimotor integration to perform various types of movement such as grasping. The function of these anatomical connections has not been fully investigated. We studied the judgment of graspability of objects in an event-related functional MRI study in healthy subjects, and found activation in two different regions within IPL: one in the left dorsal IPL extending to the intraparietal sulcus and the other in the left ventral IPL. The f

Day-length encoding through tonic photic effects in the retinorecipient SCN region.

The circadian clock in the suprachiasmatic nucleus (SCN) plays a critical role in seasonal processes by sensing ambient photoperiod. To explore how it measures day-length, we assessed the state of SCN oscillators using markers for neuronal activity (c-FOS) and the clock protein (PER1) in Syrian hamsters housed in long (LD, 16 : 8 h light : dark) vs. short days (SD, 8 : 16 h light : dark). During SD, there was no detectable phase dispersion across the rostrocaudal extent of the nucleus. In contrast, during LD, rhythms in the caudal SCN phase led those in the mid- and rostral SCN by 4-8 h and 8-

Neural basis of Delta-9-tetrahydrocannabinol and cannabidiol: effects during response inhibition.

BACKGROUND: This study examined the effect of Delta-9-tetrahydrocannabinol (THC) and cannabidiol (CBD) on brain activation during a motor inhibition task. METHODS: Functional magnetic resonance imaging and behavioural measures were recorded while 15 healthy volunteers performed a Go/No-Go task following administration of either THC or CBD or placebo in a double-blind, pseudo-randomized, placebo-controlled repeated measures within-subject design. RESULTS: Relative to placebo, THC attenuated activation in the right inferior frontal and the anterior cingulate gyrus. In contrast, CBD deactivated

Simultanagnosia: effects of semantic category and repetition blindness.

When confronted with two identical stimuli in a very brief period of time subjects often fail to report the second stimulus, a phenomenon termed "repetition blindness". The "type-token" account attributes the phenomenon to a failure to individuate the exemplars. We report a subject, KE, who developed simultanagnosia (the inability to see more than one item in an array) as a consequence of bilateral parietal lobe infarctions. With presentation of two words, pictures or letters for an unlimited time, KE typically reported both stimuli on less than half of trials. Performance was significantly in

Delayed response to animate implied motion in human motion processing areas.

Viewing static photographs of objects in motion evokes higher fMRI activation in the human medial temporal complex (MT+) than looking at similar photographs without this implied motion. As MT+ is traditionally thought to be involved in motion perception (and not in form perception), this finding suggests feedback from object-recognition areas onto MT+. To investigate this hypothesis, we recorded extracranial potentials evoked by the sight of photographs of biological agents with and without implied motion. The difference in potential between responses to pictures with and without implied motio

Empathy and judging other's pain: an fMRI study of alexithymia.

Because awareness of emotional states in the self is a prerequisite to recognizing such states in others, alexithymia (ALEX), difficulty in identifying and expressing one's own emotional states, should involve impairment in empathy. Using functional magnetic resonance imaging (fMRI), we compared an ALEX group (n = 16) and a non-alexithymia (non-ALEX) group (n = 14) for their regional hemodynamic responses to the visual perception of pictures depicting human hands and feet in painful situations. Subjective pain ratings of the pictures and empathy-related psychological scores were also compared

Interaction of sensory and cognitive processes during visual recognition: the role of the associative areas of the cerebral cortex.

The first part of the present study used a model of Alzheimer's disease in two groups of animals (three monkeys in each), given injections of neurotoxins (monkeys of group I) and physiological saline (monkeys of group II). Before injections, all monkeys were trained to discriminate stimuli containing different types of information (spatial frequency grids and geometrical figures of different colors and with different spatial relationships between objects) and to perform spatial selection. The dynamics of impairments in the characteristics of working memory were identified using delayed differe

Coupling of cerebral blood flow and oxygen consumption during physiological activation and deactivation measured with fMRI.

The physiological basis of the blood oxygenation level dependent (BOLD) signal and its dependence on baseline cerebral blood flow (CBF) were investigated by comparing responses to a visual stimulus after physiological changes of the baseline. Eight human subjects were imaged with 3 and 4 T MRI scanners, and both BOLD signal and CBF were simultaneously measured. Subjects viewed a flickering radial checkerboard in a block design experiment, alternating between eyes open or closed during the off periods. Compared to a baseline state with eyes open in a darkened room, substantial deactivation (ave

Attentional cueing improves vision restoration therapy in patients with visual field defects.

BACKGROUND: In patients with postgenicular lesions of the visual system, areas of residual vision (ARVs) are the main predictor of recovery induced by vision restoration therapy (VRT). In these partially defective regions, the elevated perceptual thresholds can be acutely reduced by attentional cueing. OBJECTIVE: To examine whether directing attention to ARVs using a visuospatial cue also increases long-term neural plasticity and thus enhances permanent training outcome. METHODS: In a prospective, randomized clinical trial, treatment outcome was compared in patients with postgenicular visual

The light-flash-evoked response as a possible indicator of increased intracranial pressure in hydrocephalus.

Surgical treatment of infantile hydrocephalus by shunt implantation may result in suboptimal intracranial pressure. Major neurological impairments and death are usually prevented by shunt treatment, but minor sequelae may persist or develop. The introduction of adjustable shunts has improved the possibilities of optimizing shunt function and minimizing the risk of such impairments. However, it is still impossible to determine the intracranial pressure without invasive measurements. Clinical findings and procedures such as computed tomography (CT) are not always enough to allow a conclusion as

Behavioral and electrophysiological effects of phenylethanolamine and 2-phenylethylamine.

The electrophysiological and behavioral effects of phenylethanolamine (OHPEA) and of its precursor 2-phenylethylamine (PEA) were studied in mice and rabbits. In animals pretreated with MAOI, PEA was found to exert strong amphetamine-like effects, EEG alerting, reduction of visual evoked responses, increased locomotor activity, and blockade of tonic seizures induced by electroshock. OHPEA exerted weaker amphetamine-like effects. Inhibition of dopamine-beta-hydroxylase increased most of the effects of PEA. In non-pretreated animals, OHPEA was found to shorten electroshock latency and to prolong

Social priors and mentalizing connectivity across autistic and schizotypal traits.

Autism spectrum disorder (ASD) and schizophrenia spectrum disorder (SSD) both involve social-cognitive difficulties but may rely on opposite predictive mechanisms. Predictive-coding and diametrical accounts propose weak priors in ASD versus strong priors in SSD. We tested whether autistic and positive schizotypal traits differentially shape the use of social priors when interpreting ambiguous social cues. Two studies with non-clinical adults employed an adapted Animated Triangles Task (ATT) with three cueing conditions: Random-Uncued, Random-Cued, and ToM-Cued. Study 1 (n = 71) assessed behavi

Automated recognition of spontaneous facial expression in individuals with autism spectrum disorder: parsing response variability.

BACKGROUND: Reduction or differences in facial expression are a core diagnostic feature of autism spectrum disorder (ASD), yet evidence regarding the extent of this discrepancy is limited and inconsistent. Use of automated facial expression detection technology enables accurate and efficient tracking of facial expressions that has potential to identify individual response differences. METHODS: Children and adults with ASD (N = 124) and typically developing (TD, N = 41) were shown short clips of "funny videos." Using automated facial analysis software, we investigated differences between ASD a
